2Receptome profiling identifies KREMEN1 and ASGR1 as alternative functional receptors of SARS-CoV-2显示文摘Host cellular receptors play key roles in the determination of virus tropism and pathogenesis.However,little is known about SARS-CoV-2 host receptors with the exception of ACE2.Furthermore,ACE2 alone cannot explain the multi-organ tropism of SARS-CoV-2 nor the clinical differences between SARS-CoV-2 and SARS-CoV,suggesting the involvement of other receptor(s).Here,we performed genomic receptor profiling to screen 5054 human membrane proteins individually for interaction with the SARS-CoV-2 capsid spike(S)protein.Twelve proteins,including ACE2,ASGR1,and KREMEN1,were identified with diverse S-binding affinities and patterns.ASGR1 or KREMEN1 is sufficient for the entry of SARS-CoV-2 but not SARS-CoV in vitro and in vivo.SARS-CoV-2 utilizes distinct ACE2/ASGR1/KREMEN1(ASK)receptor combinations to enter different cell types,and the expression of ASK together displays a markedly stronger correlation with virus susceptibility than that of any individual receptor at both the cell and tissue levels.The cocktail of ASK-related neutralizing antibodies provides the most substantial blockage of SARS-CoV-2 infection in human lung organoids when compared to individual antibodies.Our study revealed an interacting host receptome of SARS-CoV-2,and identified ASGR1 and KREMEN1 as alternative functional receptors that play essential roles in ACE2-independent virus entry,providing insight into SARS-CoV-2 tropism and pathogenesis,as well as a community resource and potential therapeutic strategies for further COVID-19 investigations.Yunqing Gu Jun Cao Xinyu Zhang Hai Gao Yuyan Wang Jia Wang Juan He Xiaoyi Jiang Jinlan Zhang Guanghui Shen Jie Yang Xichen Zheng Gaowei Hu Yuanfei Zhu Shujuan Du Yunkai Zhu Rong Zhang Jianqing Xu Fei Lan Di Qu Guoliang Xu Yun Zhao Dong Gao Youhua Xie Min Luo Zhigang Lu 2022Cell Research2022,32,1:8
3Dual antiplatelet therapy may increase the risk of non-intracranial haemorrhage in patients with minor strokes: a subgroup analysis of the CHANCE trial显示文摘Aim:The aim of this study was to explore the difference between haemorrhagic events among those patients on either aspirin or aspirin plus clopidogrel who were enrolled in the Clopidogrel in High-Risk Patients with Acute Non-disabling Ischemic Cerebrovascular Events(CHANCE)trial.Methods:This was an ad hoc analysis of the CHANCE trial;data on all patients with any haemorrhagic event were reviewed and analysed.Cox proportional hazards regression was used to determine factors association with any bleeding.Results:In the CHANCE trial,there were a total of 101(2%)haemorrhagic events reported from 50 different hospitals.The clopidogrel–aspirin group had 60(2.3%)cases and the aspirin group had 41(1.6%,p=0.09).Moderate or severe haemorrhagic events occurred in 7 patients(0.3%)in the clopidogrel–aspirin group and in 8(0.3%)in the aspirin group(p=0.73).Of 36(0.7%)cases of intracranial haemorrhages,20(0.4%)were in the clopidogrel–aspirin group and 16(0.3%)in the aspirin group.Each group had 8(0.3%)cases of symptomatic haemorrhagic strokes.Other common haemorrhagic events included 24(0.5%)cases of skin bruises,13(0.3%)gastrointestinal haemorrhages,9(0.2%)gum haemorrhages and 8(0.2%)intraocular haemorrhages.Conclusions:There was no overall significant difference in haemorrhagic events(p=0.29),especially in the rate of intracranial haemorrhages between the 2 treatment groups.However,patients enrolled with minor strokes had an increased risk of haemorrhagic events regardless of treatment group,not seen in patients with high-risk transient ischaemic attacks.Being elderly,of male gender and with a history of aspirin or proton pump inhibitor usage were associated with increased risk of haemorrhage.Patients with higher body mass index had lower risk of haemorrhagic events.David Wang Li Gui Yi Dong Hao Li Shujuan Li Huaguang Zheng Anxin Wang Xia Meng Li-Ping Liu Yi-Long Wang Guangyao Wang Jing Jing Zixiao Li Xing-Quan Zhao Yong-Jun Wang on behalf of the investigators for the CHANCE trial 2016Stroke & Vascular Neurology2016,1,2:8
4The vernalization-induced long non-coding RNA VAS functions with the transcription factor TaRF2b to promote TaVRN1 expression for flowering in hexaploid wheat显示文摘Vernalization is a physiological process in which prolonged cold exposure establishes flowering competence in winter plants. In hexaploid wheat, TaVRN1 is a cold-induced key regulator that accelerates floral transition. However, the molecular mechanism underlying the gradual activation of TaVRN1 during the vernalization process remains unknown. In this study, we identified the novel transcript VAS (TaVRN1 alternative splicing) as a non-coding RNA derived from the sense strand of the TaVRN1 gene only in winter wheat, which regulates TaVRN1 transcription for flowering. VAS was induced during the early period of vernalization, and its overexpression promoted TaVRN1 expression to accelerate flowering in winter wheat. VAS physically associates with TaRF2b and facilitates docking of the TaRF2b-TaRF2a complex at the TaVRN1 promoter during the middle period of vernalization. TaRF2b recognizes the Sp1 motif within the TaVRN1 proximal promoter region, which is gradually exposed along with the disruption of a loop structure at the TaVRN1 locus during vernalization, to activate the transcription of TaVRN1. The tarf2b mutants exhibited delayed flowering, whereas transgenic wheat lines overexpressing TaRF2b showed earlier flowering. 5FAT1, a direct transcriptional target of E2F1, suppresses cell proliferation, migration and invasion in esophageal squamous cell carcinoma显示文摘Objective: Growing evidence indicates that FAT atypical cadherin 1(FAT1) has aberrant genetic alterations and exhibits potential tumor suppressive function in esophageal squamous cell carcinoma(ESCC). However, the role of FAT1 in ESCC tumorigenesis remains not well elucidated. The aim of this study was to further investigate genetic alterations and biological functions of FAT1, as well as to explore its transcriptional regulation and downstream targets in ESCC.Methods: The mutations of FAT1 in ESCC were achieved by analyzing a combined study from seven published genomic data, while the copy number variants of FAT1 were obtained from an analysis of our previous data as well as of The Cancer Genome Atlas(TCGA) and Cancer Cell Line Encyclopedia(CCLE) databases using the cBioPortal. The transcriptional regulation of FAT1 expression was investigated by chromatin immunoprecipitation(ChIP) and the luciferase reporter assays. In-cell western, Western blot and reverse transcription-quantitative polymerase chain reaction(RT-qPCR) were used to assess the indicated gene expression. In addition, colony formation and Transwell migration/invasion assays were employed to test cell proliferation, migration and invasion.Finally, RNA sequencing was used to study the transcriptomes.Results: FAT1 was frequently mutated in ESCC and was deleted in multiple cancers. Furthermore, the transcription factor E2 F1 occupied the promoter region of FAT1, and depletion of E2 F1 led to a decrease in transcription activity and mRNA levels of FAT1. Moreover, we found that knockdown of FAT1 promoted KYSE30 and KYSE150 cell proliferation, migration and invasion;while overexpression of FAT1 inhibited KYSE30 and KYSE410 cell proliferation, migration and invasion. 6Phylogeography of regional fauna on the Tibetan Plateau: A review显示文摘The studies of uplift and glaciations of the Tibetan Plateau are summarized, and a series of recent case studies of the endemic species based on DNA sequences are detailed. In general, these molecular data show that all the organisms originated from Early Pliocene to Late Miocene, and then multi-stages of divergence/speciation occurred within each taxa following their original occupation on the plateau, mainly as a result of periodic glacial cycles and geographic isolation. The regional fauna may have undergone several range contractions and expansions during the Pleistocene glaciations. However, the population expansion and refugia may vary in space, time, and extent. The regional fauna of the Tibetan Plateau may be combinations of ancient movement from adjacent zoogeographical regions, speciation in situ, and postglacial colonization from adjacent areas. 7Alkali-activation reactivity of chemosynthetic Al_2O_3-2SiO_2 powders and their ^(27)Al and ^(29)Si magic-angle spinning nuclear magnetic resonance spectra显示文摘Pure Al_2O_3-2SiO_2 powders were prepared by sol-gel and coprecipitation methods,and their alkaliactivation reactivities were compared.The alkali-activation reactivity of the powder prepared by the sol-gel method was higher than that of the powder prepared by the coprecipitation method.The powders were investigated by ^(27)AI and ^(29)Si magic-angle spinning nuclear magnetic resonance spectroscopy(MAS NMR) to understand the relationship between their structure and alkali-activation reactivity.The ^(27)Al MAS NMR data showed that the five-coordinate AI content of the powder prepared by the sol-gel method was higher than that of the powder prepared by coprecipitation.The higher content of five-coordinate Al corresponded to higher alkali-activation reactivity.The ^(29)Si MAS NMR data showed that for the powder prepared by the sol-gel method,silicon was replaced by aluminum at secondary coordination sites of the central Si atoms during calcination.However,for the powder prepared by single-batch coprecipitation,the main change was from a low degree of polycondensation to a high degree of polycondensation.Guangjian Zheng Xuemin Cui Dong Huang Jinying Pang Guowei Mo Shujuan Yu Zhangfa Tong 2015Particuology2015,13,5:3
8Association among lifestyle, clinical examination, polymorphisms in CDH1 gene and Traditional Chinese Medicine syndrome differentiation of gastric cancer显示文摘OBJECTIVE:To explore the association among lifestyle,clinical examination,polymorphisms in CDH1 gene and Traditional Chinese Medicine(TCM)syndrome differentiation of gastric cancer(GC). METHODS:A hospital-based population of 387 GC patients was investigated in Jiangsu province.Relevant information regarding lifestyle and clinical examination were collected by a standard questionnaire.Four known single nucleotide polymorphisms(SNPs)in CDH1 were investigated by polymerase chain reaction-ligation detection reaction methods.Statistical analysis was conducted by SPSS 16.0 software.RESULTS:The results showed that meal duration and the status of glutamic pyruvic transaminase were significantly associated with TCM syndrome differentiation of GC(both P<0.05).None of the four SNPs in the E-cadherin(CDH1)gene achieved significant differences in their distributions among the nine syndrome types of GC(both P>0.05).However,significant differences were observed in rs13689 genotype distributions between several pairs of syndrome types of GC,suggesting that rs13689 is correlated with the syndrome differentiation of GC.CONCLUSION:Integrated analysis of lifestyle,clinical examination and CDH1 gene polymorphisms can contribute to a better understanding of the GC syndrome types and may improve the efficacy of interventions by stratifying disease according to TCM criteria.Junfeng Zhang Zhen Zhan Juan Wu Chunbing Zhang Yaping Yang Shujuan Tong Ruiping Wang Xuewen Yang Wei Dong 2013Journal of Traditional Chinese Medicine2013,33,5:3
9Influence of synthesis temperatures on the crystalline grain growth and morphology of lanthanum magnesium hexaaluminate显示文摘Lanthanum magnesium hexaaluminate(LaMgAl_(11)O_(19), LMA) was prepared at different temperatures by solid-state reaction. Phase compositions and crystal morphologies of specimens synthesized at different temperatures were investigated using X-ray diffraction(XRD), scanning electron microscopy(SEM). It was observed that the crystalline grain size of LMA was not only dependent on the preparation temperature but also on its powder morphology. In the temperature range of 1300 e1550℃, LMA showed platelet grain and the average crystalline grain size increases with the increase in temperature. At1600℃, if the powder was sintered for two times, the equiaxed grain could be found with the decrease in grain space, resulting in the reduction of the crystalline grain size. Styles of specimens(powder or disk) might have no obvious influence on morphologies and sizes of LMA crystalline grains which were synthesized with the well-dispersed raw material mixtures. 11Cyclical helping hands: seasonal tailwinds differentially affect migrating Oriental Storks (Ciconia boyciana) travel speed显示文摘Background:The Oriental Stork(Ciconia boyciana)breeds in southeastern Siberia and parts of northeast China,and winters mainly in southeast China.Although the autumn migration pattern of Oriental Storks has been previ-ously described,differences between spring and autumn migration travel speed in relation to wind assistance were unknown.Methods:Using GPS/GSM transmitters,we tracked the full migrations of 18 Oriental Storks during 2015-2018 to compare differences in autumn and spring migration patterns,and combined the satellite telemetry data with the National Center for Environmental Prediction Reanalysis data to explain the relationship between 850 mbar wind vec-tors and seasonal differences in travel speed.Results:Differences in tailwinds contributed to significant differences in daily average Oriental Storks travel speed in spring(258.11±64.8 km/day)compared to autumn(172.23±49.7 km/day,p<0.001).Storks stopped significantly more often in autumn than spring(1.78±1.1 versus 1.06±0.9,p<0.05),but stopover duration(15.52±12.4 versus 16.30±15.1 days,respectively,p=0.3)did not differ significantly.Tailwinds at 850 mbar pressure level(extracted from the National Center of Environmental Prediction Reanalysis data archive)significantly affected daily flying speed dur-ing spring and autumn migration.Tailwind conditions in spring(mean 4.40±5.6 m/s)were always more favourable than in autumn when they received no net benefit(0.48±5.6 m/s,p<0.001).Despite mean spring migration dura-tion being less than autumn(27.52±15.9 versus 32.77±13.4 days,p=0.17),large individual variation meant that this duration did not differ significantly from each other.Conclusions:For long distance migratory soaring birds(such as storks),relative duration of spring and autumn migration likely relates to the interaction between imperative for earliest arrival to breeding grounds and seasonal meteorological conditions experienced en route.Shujuan Fan Qingshan Zhao Hongbin Li Baoguang Zhu Shubin Dong Yanbo Xie Lei Cao Anthony David Fox 2020Avian Research2020,11,2:2
