1Clinical characteristics of 24 asymptomatic infections with COVID-19 screened among close contacts in Nanjing,China显示文摘Previous studies have showed clinical characteristics of patients with the 2019 novel coronavirus disease(COVID-19)and the evidence of person-to-person transmission.Limited data are available for asymptomatic infections.This study aims to present the clinical characteristics of 24 cases with asymptomatic infection screened from close contacts and to show the transmission potential of asymptomatic COVID-19 virus carriers.Epidemiological investigations were conducted among all close contacts of COVID-19 patients(or suspected patients)in Nanjing,Jiangsu Province,China,from Jan 28 to Feb 9,2020,both in clinic and in community.Asymptomatic carriers were laboratory-confirmed positive for the COVID-19 virus by testing the nucleic acid of the pharyngeal swab samples.Their clinical records,laboratory assessments,and chest CT scans were reviewed.As a result,none of the 24 asymptomatic cases presented any obvious symptoms while nucleic acid screening.Five cases(20.8%)developed symptoms(fever,cough,fatigue,etc.)during hospitalization.Twelve(50.0%)cases showed typical CT images of ground-glass chest and 5(20.8%)presented stripe shadowing in the lungs.The remaining 7(29.2%)cases showed normal CT image and had no symptoms during hospitalization.These 7 cases were younger(median age:14.0 years;P=0.012)than the rest.None of the 24 cases developed severe COVID-19 pneumonia or died.The median communicable period,defined as the interval from the first day of positive nucleic acid tests to the first day of continuous negative tests,was 9.5 days(up to 21 days among the 24 asymptomatic cases).Through epidemiological investigation,we observed a typical asymptomatic transmission to the cohabiting family members,which even caused severe COVID-19 pneumonia.Overall,the asymptomatic carriers identified from close contacts were prone to be mildly ill during hospitalization.However,the communicable period could be up to three weeks and the communicated patients could develop severe illness.These results highlighted the importance of close contact tracing and longitudinally surveillance via virus nucleic acid tests.Further isolation recommendation and continuous nucleic acid tests may also be recommended to the patients discharged.Zhiliang Hu Ci Song Chuanjun Xu Guangfu Jin Yaling Chen Xin Xu Hongxia Ma Wei Chen Yuan Lin Yishan Zheng Jianming Wang Zhibin Hu Yongxiang Yi Hongbing Shen 2020Science China(Life Sciences)2020,63,5:77
2Collagen-binding basic fibroblast growth factor improves functional remodeling of scarred endometrium in uterine infertile women: a pilot study显示文摘Intrauterine adhesion(IUA) is a common cause of uterine infertility and one of the most severe clinical features is endometrial fibrosis namely endometrial scarring for which there are few cures currently. Blocked angiogenesis is the main pathological change in the scarred endometrium. The fibroblast growth factor 2(b FGF), a member of FGF family, is usually applied to promote healing of refractory ulcer and contributes to angiogenesis of tissues. In this study, the sustained-release system of b FGF100 μg was administrated around scarred endometrium guiding by ultrasound every 4 weeks in 18 patients(2–4 times). Results showed that after treatment, the menstrual blood volume, endometrial thickness and the scarred endometrial area were improved.Histological study showed blood vessel density increased obviously. Three patients(3/18) achieved pregnancy over 20 gestational weeks. 5Cloud point extraction and flame atomic absorption spectrometry analysis of palladium, platinum, and gold ions from industrial polluted soil显示文摘Cloud point extraction (CPE) with Tergitol TMN-6 was applied for the extraction of trace amounts of palladium (Pd(Ⅱ)), platinum (Pt(Ⅳ)), and gold (Au(Ⅲ)) in the soil of industrial sewage. Ammonium pyrolysine dithiocarbamate (APDC) was adopted as the chelating agent prior to CPE and then was detected by atomic absorption spectrometry (AAS). Different parameters such as the concentration of surfactants, chelating agent and salt, sample pH, equilibration temperature and time, centrifugation time and rates, and the effect of foreign ions were studied. Under optimum conditions, the low limits of detections are 1.4, 2.8 and 1.2 ng·ml^-1 and the enrichment factors are 21, 12, and 24 for Pd(Ⅱ), Pt(Ⅳ), and Au(Ⅲ, respectively. The relative standard deviations vary from 0.6% to 1.0% (n=11). All correlation coefficients of the calibration curves are >0.9960. 6Exogenous hydrogen sulfide inhibits apoptosis by regulating endoplasmic reticulum stress-autophagy axis and improves myocardial reconstruction after acute myocardial infarction显示文摘During acute myocardial infarction,endoplasm ic reticulum(ER)stress-induced autophagy and apoptosis have been shown as im portant pathogeneses of myocardial reconstruction.Im portantly,hydrogen sulfide(H2S),as a third endogenous gas signaling molecule,exerts strong cytoprotective effect on anti-ER stress,autophagy regulation and antiapoptosis.Here,we showed that H2 S treatm ent inhibits apoptosis by regulating ER stress-autophagy axis and im proves myocardial reconstruction after acute myocardial infarction.We found that H2S intervention im proved left ventricle function,reduced glycogen deposition in myocardial tissue mesenchyme,and inhibited apoptosis.Moreover,the expressions of fibrosis indicators(Col3a1 and Col1a2),ER stress-related proteins(CHOP and BIP/ERP78),autophagy-related proteins(Beclin and ATG5),apoptosis protein(Bax),as well as fibrosis protein Col4a3bp were all decreased after treatm ent w ith H2 S.H2 S adm inistration also m aintained MMP/TIMP balance.Mechanistically,H2S activated the PI3K/AKT signaling pathway.In addition,H2S treatm ent also reduced the expressions of ER stress-related proteins,autophagy-related proteins,and apoptins in in wtro experiments.Interestingly,activation of ER stress-autophagy axis could reverse the inhibitory effect of H2S on myocardial apoptosis.Altogether,these results suggested that exogenous H2S suppresses myocardial apoptosis by blocking ER stress-autophagy axis,which in turn reverses cardiac rem odeling after myocardial infarction.Yaling Li Maojun Liu Jiali Yi Xiong Song Xia Zheng Da Liu Sen Wang Chun Chu Jun Yang 2020Acta Biochimica et Biophysica Sinica2020,52,12:5
7Chinese patent medicine for chronic obstructive pulmonary disease based on principles of tonifying Qi, promoting blood circulation by removing blood stasis, and resolving phlegm: a systematic review of randomized controlled trials显示文摘OBJECTIVE: To assess the efficacy and safety of Chinese patent medicine(CPM) with the principle of tonifying Qi, promoting blood circulation by removing blood stasis, and resolving phlegm(TQ-PBC-RP)in the management of stable chronic obstructive pulmonary disease(COPD).METHODS: A systematic review of randomized controlled trials(RCTs) identified from electronic databases and print was conducted. RCTs testing CPMs with TQ-PBC-RP against any type of controlled intervention in patients with stable COPD and assessing clinically relevant outcomes were included. Methodological quality was evaluated with the risk of bias tool according to systematic review handbook 5.0.2. Quality of evidence was estimatedby the rating approach developed by the Grading of Recommendations, Assessment, Development,and Evaluation Working Group.RESULTS: Thirteen eligible RCTs with 12 oral CPMs were tested. Significant differences between groups in favor of CPMs were not reported in all trials. Most trials included were deemed to be of low methodological quality with poor evidence quality.Because of large data heterogeneity, statistical pooling was not performed for all outcomes.CONCLUSION: The effectiveness of CPM in the treatment of stable COPD is not supported by evidence. Currently, evidence from RCTs is scarce and methodologically weak. 8A botanical medicine dragon’s blood exhibited clinical antithrombosis efficacy similar to low molecular weight heparin显示文摘Deep vein thrombosis(DVT)is a common complication following traumatic fracture with a 0.5%–1%annual incidence.Low molecular weight heparin(LMWH)is the most commonly used anticoagulation drug for DVT prevention,but treatment with LMWH is invasive.Our aim is to compare the antithrombotic effect of dragon’s blood,an oral botanical anticoagulant medicine approved by the Chinese FDA,with LMWH in patients undergoing hip fracture surgery and to explore the molecular mechanisms of anticoagulation treatment.Our study recruited patients and divided them into LMWH and dragon’s blood treatment group.Coagulation index tests,Doppler ultrasound and mRNA sequencing were performed before and after anticoagulation therapy.There was no significant difference in postoperative DVT incidence between the two groups(23.1%versus 15.4%,P=0.694).D-dimer(D-D)and fibrinogen degradation product(FDP)showed significant reductions in both groups after anticoagulation treatments.We identified SLC4A1,PROS1,PRKAR2B and seven other genes as being differentially expressed during anticoagulation therapy in both groups.Genes correlated with coagulation indexes were also identified.Dragon’s blood and LMWH showed similar effects on DVT and produced similar gene expression changes in patients undergoing hip fracture surgery,indicating that dragon’s blood is a more convenient antithrombosis medicine(oral)than LMWH(hypodermic injection).Jiangbin Liang Song Mei Xiangyu Qiao Wei Pan Yan Zhao Shaohui Shi Yaling Zhai Haizhao Wen Guoping Wu Chengyu Jiang 2021Science China(Life Sciences)2021,64,10:4
9PER2-mediated ameloblast differentiation via PPARγ/AKT1/β-catenin axis显示文摘Circadian rhythm is involved in the development and diseases of many tissues.However,as an essential environmental regulating factor,its effect on amelogenesis has not been fully elucidated.The present study aims to investigate the correlation between circadian rhythm and ameloblast differentiation and to explore the mechanism by which circadian genes regulate ameloblast differentiation.Circadian disruption models were constructed in mice for in vivo experiments.An ameloblast-lineage cell(ALC)line was used for in vitro studies.As essential molecules of the circadian system,Bmal1 and Per2 exhibited circadian expression in ALCs.Circadian disruption mice showed reduced amelogenin(AMELX)expression and enamel matrix secretion and downregulated expression of BMAL1,PER2,PPARγ,phosphorylated AKT1 andβ-catenin,cytokeratin-14 and F-actin in ameloblasts.According to previous findings and our study,BMAL1 positively regulated PER2.Therefore,the present study focused on PER2-mediated ameloblast differentiation and enamel formation.Per2 knockdown decreased the expression of AMELX,PPARγ,phosphorylated AKT1 andβ-catenin,promoted nuclearβ-catenin accumulation,inhibited mineralization and altered the subcellular localization of E-cadherin in ALCs.Overexpression of PPARγpartially reversed the above results in Per2-knockdown ALCs.Furthermore,in in vivo experiments,the length of incisor eruption was significantly decreased in the circadian disturbance group compared to that in the control group,which was rescued by using a PPARγagonist in circadian disturbance mice.In conclusion,through regulation of the PPARγ/AKT1/β-catenin signalling axis,PER2 played roles in amelogenin expression,cell junctions and arrangement,enamel matrix secretion and mineralization during ameloblast differentiation,which exert effects on enamel formation.Wushuang Huang Xueqing Zheng Mei Yang Ruiqi Li Yaling Song 2021International Journal of Oral Science2021,13,2:3
10The Physiological Mechanism of Improved Formaldehyde Resistance in Petunia hybrida Harboring a Mammalian cyp2e1 Gene显示文摘Cytochrome P450 CYP2E1 is mainly present in hepatocytes in the livers of mammals,where it plays an important role in the metabolism of xenobiotic organic substances. Previous studies showed that transgenic petunia(Petunia hybrid) plants harboring a mammalian cyp2e1 gene(designated cyp2e1-transgenic petunia) exhibited increased resistance to formaldehyde stress. In this study,we used cyp2e1-transgenic petunia plants to analyze physiological indexes related to formaldehyde stress responses. The results indicated that under formaldehyde stress,the malondialdehyde content in cyp2e1-transgenic petunia plants was lower than in β-glucuronidase gene(gus)-transgenic and wild-type petunia plants. The activities of both superoxide dismutase and peroxidase in the cyp2e1-transgenic plants were higher than in gus-transgenic and wild-type plants. The alcohol dehydrogenase activity was slightly increased and more glutathione was consumed. Additionally,under formaldehyde stress,the levels of plant hormones including indole-3-acetic acid,zeatin and abscisic acid in cyp2e1-transgenic petunia plants displayed decreasing trends,whereas the level of gibberellic acid displayed an increasing trend. In contrast,the indole-3-acetic acid,zeatin and abscisic acid levels in gus-transgenic and wild-type petunia plants displayed increasing trends,whereas the gibberellic acid level displayed a decreasing trend. 11Floralozone protects endothelial function in atherosclerosis by ameliorating NHE1显示文摘Endothelial dysfunction is the pathological basis of atherosclerosis.Incomplete understanding of endothelial dysfunction etiology has impeded drug development for this devastating disease despite the currently available therapies.Floralozone,an aroma flavor,specifically exists in rabbit ear grass.Recently,floralozone has been demonstrated to inhibit atherosclerosis,but the underlying mechanisms are undefined.The present study was undertaken to explore whether floralozone pharmacologically targets endothelial dysfunction and therefore exerts therapeutic effects on atherosclerosis.The Na+/H+exchanger 1(NHE1),a channel protein,plays a vital role in atherosclerosis.Whether NHE1 is involved in the therapeutic effects of floralozone on endothelial dysfunction has yet to be further answered.By performing oil red staining and hematoxylin–eosin staining,vascular functional study,and oxidative stress monitoring,we found that floralozone not only reduced the size of carotid atherosclerotic plaque but also prevented endothelial dysfunction in atherosclerotic rats.NHE1 expression was upregulated in the inner membrane of carotid arteries and H2O2-induced primary rat aortic endothelial cells.Inspiringly,floralozone prevented the upregulation of NHE1 in vivo and in vitro.Notably,the administration of NHE1 activator LiCl significantly weakened the protective effect of floralozone on endothelial dysfunction in vivo and in vitro.Our study demonstrated that floralozone exerted its protective effect on endothelial dysfunction in atherosclerosis by ameliorating NHE1.NHE1 maybe a drug target for the treatment of atherosclerosis,and floralozone may be an effective drug to meet the urgent needs of atherosclerosis patients by dampening NHE1.Ning Huang Yue Qiu Yanhua Liu Tianheng Liu Xianjun Xue Ping Song Jian Xu Yutian Fu Ruili Sun Yaling Yin Peng Li 2021Acta Biochimica et Biophysica Sinica2021,53,10:2
12A urinary proteomic landscape of COVID-19 progression identifies signaling pathways and therapeutic options显示文摘Signaling pathway alterations in COVID-19 of living humans as well as therapeutic targets of the host proteins are not clear.We analyzed 317 urine proteomes,including 86 COVID-19,55 pneumonia and 176 healthy controls,and identified specific RNA virus detector protein DDX58/RIG-I only in COVID-19 samples.Comparison of the COVID-19 urinary proteomes with controls revealed major pathway alterations in immunity,metabolism and protein localization.Biomarkers that may stratify severe symptoms from moderate ones suggested that macrophage induced inflammation and thrombolysis may play a critical role in worsening the disease.Hyper activation of the TCA cycle is evident and a macrophage enriched enzyme CLYBL is up regulated in COVID-19 patients.As CLYBL converts the immune modulatory TCA cycle metabolite itaconate through the citramalyl-CoA intermediate to acetyl-CoA,an increase in CLYBL may lead to the depletion of itaconate,limiting its anti-inflammatory function.These observations suggest that supplementation of itaconate and inhibition of CLYBL are possible therapeutic options for treating COVID-19,opening an avenue of modulating host defense as a means of combating SARS-CoV-2 viruses.Yuntao Liu Lan Song Nairen Zheng Jinwen Shi Hongxing Wu Xing Yang Nianci Xue Xing Chen Yimin Li Changqing Sun Cha Chen Lijuan Tang Xiaotian Ni Yi Wang Yaling Shi Jianwen Guo Guangshun Wang Zhongde Zhang Jun Qin 2022Science China(Life Sciences)2022,65,9:1
13Clinical observation of capecitabine monotherapy in elderly patients with advanced breast cancer显示文摘Objective The aim of the study was to evaluate the safety and efficacy of capecitabine mono-chemotherapy in elderly patients with advanced breast cancer. Methods The data from 36 cases of capecitabine monotherapy in elderly patients with advanced breast cancer were retrospectively analyzed. Oral administration of capecitabine 2000 mg/m2 twice daily(D1–14) for 21 days constituted a cycle. The effect of the disease and main adverse reactions were evaluated every 2 cycles. Results The data from 36 elderly patients were studied. The median number of chemotherapy cycles was 4. The total effective rate was 30.6%(11/36) and the disease control rate was 72.2%(26/36). The number of patients with clinical complete remission was 2, clinical partial response was 9, stable disease was 15, and progressive disease was 10. Where treatment was effective, the median time to progression was 6 months and the median overall survival was 9.5 months. The main adverse events were gastrointestinal reactions, bone marrow suppression, and oral mucositis; most of the reactions were grade 1 to 2. 18Feasibility of terahertz spectroscopy for hybrid purity verification of rice seeds显示文摘The purity of hybrid rice seeds reflects the typical consistency of seed varieties in characteristics.The accuracy and reliability of seed purity detecting are of great significance to ensure the quality of seeds.In this study,the feasibility of identifying the purity of hybrid rice seeds,Xinong 1A/89,by terahertz(THz)time-domain spectroscopy system combined with chemometrics was explored.Three quantitative identification models for testing the purity of Xinong 1A/89 hybrid rice seed were developed and compared by THz absorption spectroscopy with extreme learning algorithm(ELM),Principal cComponent Regression(PCR)and Partial Least Squares Regression(PLSR).Experimental results showed that comparing with classical PLSR and PCR models,ELM presents a better feasibility and stability.For the testing set,the quantitative prediction result of ELM(ELoo=2.005×10^(-5),R^(2)=96.75%)is significantly better than those of PCR(ELoo=7.346×10^(-5),R^(2)=88.10%)and PLSR(ELoo=8.007×10^(-5),R^(2)=87.03%).The results highlight the feasibility of THz spectroscopy combined with ELM as an efficient and reliable method for verification of hybrid rice seeds.Yaling Yang Shengling Zhou Jie Song Jie Huang Guanglin Li Shiping Zhu 2018International Journal of Agricultural and Biological Engineering2018,11,5:1
19Molecular dynamics simulation of alkaline electrolyte diffusion in anion exchange membrane显示文摘During the operation of alkaline direct liquid fuel cells,the alkaline electrolyte is usually needed in the anode electrode to accelerate the electrochemical reaction kinetics of the liquid fuel.However,the crossover of the alkaline solution in the anode through the anion exchange membrane to the cathode can increase the transfer resistance of the oxygen in the cathode.In order to reduce the crossover of the alkaline solution,the diffusion process of the alkaline solution in the anion exchange membrane needs to be fully understood.In this work,interface models of anion exchange membrane-alkaline electrolytes are established based on the cell structure of the quaternary ammonium polysulfone(QAPS)membrane to simulate the dynamic process of the alkaline solution in the membrane.The effect of the type and the concentration of the alkaline solution on the transportation of the metal ions and OH^-in the membrane are studied.The results show that the agglomeration of Na+is formed more easily than K^+in the interface model.Because of the strong interaction of Na^+on OH^-,OH^-ions appear to be concentrated,resulting in that the diffusion coefficients of the metal ion and OH^-in the in Na^+solution are lower than those in the K^+solution.In addition,with the raised concentration of electrolyte solution,the aggregation degrees of the metal ions and OH^-can be increased,which means an enlarged mass transfer resistance of the components.Furthermore,by adding a polytetrafluoroethylene(PTFE)layer on the QAPS membrane,the distribution of metal ions tends to be concentrated,and the number of hydrophilic channels in the QAPS membrane is reduced,which significantly increases the alkali resistance of the anion exchange membrane.SONG BingYe LI Dong HE YaLing HUANG Dong TONG ZiXiang 2020Science China(Technological Sciences)2020,63,11:0
20Functional characterization of novel NPRL3 mutations identified in three families with focal epilepsy显示文摘Focal epilepsy accounts for 60% of all forms of epilepsy, but the pathogenic mechanism is not well understood. In this study,three novel mutations in NPRL3(nitrogen permease regulator-like 3), c.937_945del, c.1514dup C and 6,706-bp genomic DNA(g DNA) deletion, were identified in three families with focal epilepsy by linkage analysis, whole exome sequencing(WES) and Sanger sequencing. NPRL3 protein is a component of the GATOR1 complex, a major inhibitor of m TOR signaling. These mutations led to truncation of the NPRL3 protein and hampered the binding between NPRL3 and DEPDC5, which is another component of the GATOR1 complex. Consequently, the mutant proteins enhanced m TOR signaling in cultured cells, possibly due to impaired inhibition of m TORC1 by GATOR1. Knockdown of nprl3 in Drosophila resulted in epilepsy-like behavior and abnormal synaptic development.
