| 1 | One-year clinical study of NeuroR egen scaffold implantation following scar resection in complete chronic spinal cord injury patients显示文摘The objective of this clinical study was to assess the safety and feasibility of the collagen scaffold, Neuro Regen scaffold, one year after scar tissue resection and implantation. Scar tissue is a physical and chemical barrier that prevents neural regeneration. However, identification of scar tissue is still a major challenge. In this study, the nerve electrophysiology method was used to distinguish scar tissue from normal neural tissue, and then different lengths of scars ranging from 0.5–4.5 cm were surgically resected in five complete chronic spinal cord injury(SCI) patients. The NeuroR egen scaffold along with autologous bone marrow mononuclear cells(BMMCs), which have been proven to promote neural regeneration and SCI recovery in animal models, were transplanted into the gap in the spinal cord following scar tissue resection. No obvious adverse effects related to scar resection or Neuro Regen scaffold transplantation were observed immediately after surgery or at the 12-month follow-up. In addition, patients showed partially autonomic nervous function improvement, and the recovery of somatosensory evoked potentials(SSEP) from the lower limbs was also detected. | 5 | Effects of lanthanum on the growth and essential oil components of lavender under osmotic stress显示文摘The effects of lanthanum on growth, soluble sugar content, essential oil contents and quality were studied in lavender plant(Lavandula Angustifolia Mill, Variety 701) under osmotic stress by means of PEG-6000 solution. The results show that osmotic stress could reduce the growth rate of lavender, and increase the content of SSC(soluble sugar concentration) to some extent. Compared with control group,the contents of essential oil in flowers and leaves respectively increase by 45.6% and 48.3% in the osmotic stress group induced by 15% PEG-6000. However, the presence of lanthanum can make the contents of essential oil in the flowers and leaves of stressed lavender plants enhance by 19.4% and 18.6%, respectively. Under osmotic stress, the relative contents of four kinds of lavender essential oil compositions of camphor, linalool, linalyl acetate and lavandulol acetate in the lavender flowers were successively 22.63%,0.61%,25.46%,7.03%,and 20.17%,0.62%,20.72%,10.80%,respectively in the presence and absence of lanthanum. The contents of all four main components of lavender essential oils meet the requirements of the national standard under osmotic stress in the presence of lanthanum. However, in the absence of lanthanum, the contents of linalyl acetate and lavandulol acetate does not meet the requirements of the national standard under osmotic stress. Moreover, the contents of five components of borneol, camphor,eudesmol, caryophyllene oxide and bicyclic sesquiphellandrene from the essential oil of lavender leaves are 12.89%, 3.84%, 8.76%, 11.30% and 8.10%, respectively. The total content of above five components accounts for 44.89% of the essential oil of the lavender leaves. Particularly, the borneol content in leaf essential oil is up to 12.89%. It is 125.2 times the amount of borneol in the flower essential oil. | 12 | Establishment and Application of a SYBR Green I Real-time Quantitative PCR Assay for the Detection of LAT Gene of Pseudorabies Virus in Bama Miniature Pigs显示文摘[Objective] This study aimed to establish a rapid,sensitive and specific SYBR Green I real-time quantitative PCR assay for the detection of latent pseudorabies virus( PRV) infection. [Method]SYBR Green I real-time quantitative PCR conditions and system for early detection of latent pseudorabies virus infection were optimized and compared with conventional PCR to investigate the sensitivity and specificity. Subsequently,the established assay was applied to detect different clinical samples. [Result]The sensitivity of SYBR Green I real-time quantitative PCR assay( 52 copies / μl) was 1 000 times higher than that of conventional PCR( 5. 2 × 104 copies / μl) and the detection time was shortened by 1 /2. The established assay could be used to detect PRV but could not be used to detect PCV2,PPV,CSFV or PRRSV. Various tissues were collected from Bama miniature pigs with latent PRV infection under sterile conditions for real-time PCR detection. Results showed that viral copy number in the brain,nasal swab,inguinal lymph node,liver,lung and spleen was above 20,while PRV was not detected in the kidney and heart tissues. | 14 | Isolation and characterization of 49 polymorphic microsatellite loci for Decapterus maruadsi using SLAF-seq, and cross-amplifi cation to related species显示文摘Decapterus maruadsi is a commercially important species in China, but has been heavily exploited in some areas. There is a growing need to develop microsatellites promoting its genetic research for the adequate management of this ?shery resources. The recently developed speci?c-locus ampli?ed fragment sequencing(SLAF-seq) is an effcient and high-resolution method for genome-wide microsatellite markers discovery. In this study, 28 905 microsatellites(mono-to hexa-nucleotide repeats) were identi?ed using SLAF-seq technology, of which di-nucleotide was the most frequent(13 590, 47.02%), followed by mono-nucleotide(8 138, 28.15%), tri-nucleotide(5 727, 19.81%), tetra-nucleotide(1 104, 3.82%), pentanucleotide(234, 0.81%), and hexa-nucleotide(112, 0.39%). One hundred and thirty-two microsatellite loci(di-and tri-nucleotide) were randomly selected for ampli?cation and polymorphism, of which 49 were highly polymorphic and well-resolved. The average number of alleles per locus was 13.63, ranging from 4 to 25, and allele sizes varied between 110 bp and 309 bp. The observed heterozygosity( Ho) and expected heterozygosity( He) ranged from 0.233 to 1.000 and from 0.374 to 0.959, with mean values of 0.738 and0.836, respectively. The polymorphism information content(PIC) ranged from 0.341 to 0.941(mean=0.806).However, 12 loci deviated from Hardy-Weinberg equilibrium. Furthermore, transferability tests were also successful in validating the utility of the developed markers in ?ve phylogenetically related species of family Carangidae. In order to produce gE antigen in large quantities and at low cost,a gene fragment encoding PRV gE core epitopes was expressed in E. coli BL21 expression system. SDS-PAGE and Western Blotting revealed that the expression product in culture supernatant of E. coli BL21 was a recombinant protein,approximately 51. 8 Kd. At 5 h post-induction,protein concentration assay showed that the expression product amounted to 1. 65 mg /ml,accounting for 24. 17% of total proteins in the culture supernatant. An indirect PRV gE-ELISA was established by using the recombinant expression product as a coating antigen. Cross-reactivity assay showed that this antigen was PRV specific. In addition,the assay was consistently reproducible. | 18 | Melittin inhibited glycolysis and induced cell apoptosis in cisplatinresistant lung adenocarcinoma cells via TRIM8显示文摘Chemotherapy is widely used for non-small cell lung cancer(NSCLC)patients at a late stage;however,NSCLC patients often acquire resistance to chemotherapeutic drugs,thus limiting the therapy efficacy.Melittin,a major component of bee venom,possesses anti-tumor activity in various cancer cells.Here,we examined the effects of melittin on A549/DDP cisplatin-resistant lung adenocarcinoma cells and xenografts formed from this cell line and investigated the possible target of melittin.Treatment with melittin resulted in the induction of cell apoptosis,glycolysis inhibition,and reduction of phosphorylated AKT(p-AKT)in A549/DDP cells.We also identified that tripartite motif-containing 8(TRIM8)was a potential target of melittin.Moreover,we found that TRIM8 mRNA expression was elevated in NSCLC specimens as compared to adjacent normal tissues(N=25)and that patients with high expression of TRIM8 had a poor prognosis for lung adenocarcinoma.The knockdown of TRIM8 had a similar effect of melittin,while overexpression of TRIM8 reversed the effects of melittin in A549/DDP cells.More importantly,we revealed that melittin enhanced cisplatin sensitivity in A549/DDP cells and tumor growth in vivo using a xenograft model of A549/DDP cells.In conclusion,melittin appears to be a potential chemotherapy sensitization agent in NSCLC. | SUFANG ZHANG XIANG LV LI LI YINGBIN LUO HUINAN XIANG LIXIN WANG YAN LI | 2021 | BIOCELL2021,45,1: | 1 |
