| 1 | Efficacy and safety of metformin and sitagliptin based triple antihyperglycemic therapy(STRATEGY):a multicenter,randomized,controlled,non-inferiority clinical trial显示文摘Despite the current guideline's recommendation of a timely stepwise intensification therapy,the 'clinical inertia',termed as the delayed treatment intensification,commonly exists in the real world,which may be partly due to the relatively little substantial evidence and no clear consensus regarding the efficacy and safety of triple oral agents in patients inadequately controlled with dual therapy.In this clinical trial performed in 237 centers in China,5,535 type 2 diabetic patients inadequately controlled by previous therapies were treated with a stable metformin/sitagliptin dual therapy for 20 weeks.The patients who did not reach the glycated hemoglobin A1c(HbA1c) goal were then further randomized into glimepiride,gliclazide,repaglinide,or acarbose group for an additional 24-week triple therapy.A mean HbAlc reduction of 0.85%was observed when sitagliptin was added to the patients inadequately controlled with metformin in 16 weeks.Further HbAlc reductions in the 24-week triple therapy stage were 0.65%in glimepiride group,0.70%in gliclazide group,0.61%in repaglinide group,and 0.45%in acarbose group.The non-inferiority criterion for primary hypotheses was met for gliclazide and repaglinide,but not for acarbose,compared with glimepiride,when added to metformin/sitagliptin dual therapy.The incidences of adverse events(AEs) were 29.2%in the dual therapy stage and30.3%in the triple therapy stage.Metformin/sitagliptin as baseline therapy,with the addition of a third oral antihyperglycemic agent,including glimepiride,gliclazide,repaglinide,or acarbose,was effective,safe and well-tolerated for achieving an HbAlc<7.0%goal in type 2 diabetic patients inadequately controlled with previous therapies.The timely augmentation of up to three oral antihyperglycemic agents is valid and of important clinical benefit to prevent patients from exposure to unnecessarily prolonged hyperglycemia. | Wen Xu Yiming Mu Jiajun Zhao Dalong Zhu Qiuhe Ji Zhiguang Zhou Bin Yao Anhua Mao Samuel S.Engel Bin Zhao Yan Bi Longyi Zeng Xingwu Ran Juming Lu Linong Ji Wenying Yang Weiping Jia Jianping Weng | 2017 | Science China(Life Sciences)2017,60,3: | 19 |
| 2 | Blocking FSH inhibits hepatic cholesterol biosynthesis and reduces serum cholesterol显示文摘Menopause is associated with dyslipidemia and an increased risk of cardio-cerebrovascular disease.The classic view assumes that the underlying mechanism of dyslipidemia is attributed to an insufficiency of estrogen.In addition to a decrease in estrogen, circulating follicle-stimulating hormone (FSH)levels become elevated at menopause.In this study,we find that blocking FSH reduces serum cholesterol via inhibiting hepatic cholesterol biosynthesis.First,epidemiological results show that the serum FSH levels are positively correlated with the serum total cholesterol levels,even after adjustment by considering the effects of serum estrogen,in addition,the prevalence of hypercholesterolemia is significantly higher in peri-menopausal women than that in premenopausal women.Furthermore,we generated a mouse model of FSH elevation by intraperitoneally injecting exogenous FSH into ovariectomized (OVX)mice,in which a normal level of estrogen (E2)was maintained by exogenous supplementation. Consistently,the results indicate that FSH,independent of estrogen,increases the serum cholesterol level in this mouse model. | 6 | Accelerating DNN-based 3D point cloud processing for mobile computing显示文摘3D point cloud data,which are produced by various 3D sensors such as LIDAR and stereo cameras,have been widely deployed by industry leaders such as Google,Uber,Tesla,and Mobileye,for mobile robotic applications such as autonomous driving and humanoid robots.Point cloud data,which are composed of reliable depth information,can provide accurate location and shape characteristics for scene understanding,such as object recognition and semantic segmentation.However,deep neural networks(DNNs),which directly consume point cloud data,are particularly computation-intensive because they have to not only perform multiplication-and-accumulation(MAC)operations but also search neighbors from the irregular 3D point cloud data.Such a task goes beyond the capabilities of general-purpose processors in realtime to figure out the solution as the scales of both point cloud data and DNNs increase from application to application.We present the first accelerator architecture that dynamically configures the hardware onthe-fly to match the computation of both neighbor point search and MAC computation for point-based DNNs.To facilitate the process of neighbor point search and reduce the computation costs,a grid-based algorithm is introduced to search neighbor points from a local region of grids.Evaluation results based on the scene recognition and segmentation tasks show that the proposed design harvests 16.4x higher performance and saves 99.95%of energy than an NVIDIA Tesla K40 GPU baseline in point cloud scene understanding applications. | Bosheng LIU Xiaoming CHEN Yinhe HAN Jiajun LI Haobo XU Xiaowei LI | 2019 | Science China(Information Sciences)2019,62,11: | 4 |
| 7 | Signal Detection of Large Volume Airgun Source Excitation in the Fixed Field of the Yangtze River显示文摘As this is the first time a large volume airgun has been excited in the 'Yangtse River Geoscience Project',it is necessary to study the time-frequency characteristic based on the linear stacked seismic data from records from portable stations near the fixed fields and seismic stations. Airgun signal propagation distances were detected using stacked seismic data to analyze the environmental impact on signal propagation distance. | 8 | Relationship among plasma endothelin, calcitonin gene-related peptide and blood flow rate of bilateral vertebral arteries in patients with cervical vertigo显示文摘Objective: To investigate the relationship among plasma endothelin(ET), calcitonin gene-related peptide(CGRP) and blood flow rate of bilateral vertebral arteries in patients with cervical vertigo(CV) and to assess the effect of ET and CGRP on the onset of CV. Methods:The concentration of ET and CGRP in 64 patients with CV and 30 controls was determined by radioimmunity method. The average blood flow velocity (Vm) of bilateral vertebral arteries was detected by Transcranial Doppler(TCD). Results:Plasma concen- tration of ET(91.48± 9.08 pg/ml) and ET/CGRP ratio value(2.88± 0.52) in vertebrobasilar arteriospasm group were both higher than those in vertebrobasilar non-arteriospasm group and in controls, while CGRP concentration(30.66± 6.05 pg/ml) in vertebrobasilar arteriospasm group was lower than that in vertebrobasilar non-arteriospasm group and controls respectively. The Vm of bilateral verte- bral arteries in vertebrobasilar arteriospasm group(67.97± 11.64 cm/s ) was higher than that in vertebrobasilar non-arteriospasm group and controls respectively, having a positive correlation with ET concentration and ET/CGRP ratio value(r1=0.52, P < 0.05; r2=0.59, P < 0.05), but a negative correlation with CGRP concentration(r3=-0.54, P < 0.05). There was no significant difference in ET and CGRP concentration, ET/CGRP ratio value and the Vm of bilateral vertebral arteries between vertebrobasilar non-arteriospasm group and the control group. | 13 | Design,Modeling and Experimentation of a Biomimetic Wall-climbing Robot for Multiple Surfaces显示文摘Wall-climbing robots can work on steep terrain and obtain environment information in three dimensions for human in real time,which can improve operation efficiency.However,traditional single-mode robots cannot ensure the stable attachment on complex wall surfaces.Inspired by the structure characteristics of flies and clingfishes,three bionic structures including the flexible spine wheel,the adhesive material and the adsorption system are proposed.Aiming at task requirements on multiple walls and based on the above three bionic structures,a wall-climbing robot with the composed mode of“grabbing+adhesion+adsorption”is presented v/a the law of mechanism configuration synthesis.Using static analysis,the safe attachment conditions for the robot on smooth and rough walls are that the adsorption force is 30 N or more.Based on Newton's Euler and Lagrange formulas,the dynamic equations of the robot on vertical walls are established to deduce that the maximum theoretical torque of the driving motor is 1.43 N·m at a uniform speed.Finally,the prototype of the wall-climbing robot is manufactured and tested on the vertical lime wall,coarse sandpaper wall and acrylic ceiling wall.Meanwhile,experiment results imply that the average maximum moving speed and the corresponding load are 7.19 cm·s-1 and 0.8 kg on the vertical lime wall,7.78 cm·s-1 and 0.6 kg on the coarse sandpaper wall,and 5.93 cm·s-1 and 0.2 kg on the acrylic ceiling wall respectively.These findings could provide practical reference for the robot’s application on walls. | Jinfu Liu Linsen Xu Jiajun Xu Tao Li Shouqi Chen Hong Xu Gaoxin Cheng Marco Ceccarelli | 2020 | Journal of Bionic Engineering2020,17,3: | 2 |

| 16 | Nanosecond pulsed laser-assisted modified copper surface structure:Enhanced surface microhardness and microbial corrosion resistance显示文摘Microbiologically influenced corrosion(MIC) is an unavoidable problem in several industries. Copper(Cu)and its alloys are widely used engineering materials. However, MIC of Cu remains a persistent challenge to their performance and functional lifetime under aggressive environments. This study investigated nanosecond pulsed laser processing(LP), which may enhance the corrosion resistance of Cu. The microstructural evolution and corrosion behavior of LP-Cu in the presence of sulfate-reducing bacteria(SRB) were evaluated. Typical deformation-induced microstructural features of high-density dislocations were analyzed on the top surface of LP-Cu coupon. Electrochemical measurements suggested that LP-Cu coupons exhibited better corrosion resistance in SRB-inoculated solution compared with their original counterpart. The enhanced corrosion resistance by LP primarily resulted from the combined influences of compressive residual stress and work hardening in the surface. However, overlap percentage played a key role in improving corrosion resistance. LP produced optimal corrosion resistance at 50% overlap. | 17 | Chromobox homolog 4 overexpression inhibits TNF-α-induced matrix catabolism and senescence by suppressing activation of NF-κB signaling pathway in nucleus pulposus cells显示文摘Intervertebral disc degeneration(IDD)is featured as enhanced catabolism of extracellular matrix(ECM)in the nucleus pulposus(NP),in which tumor necrosis factor-alpha(TNF-α)-related cell senescence is involved.Chromobox homolog protein 4(CBX4)exhibits anti-inflammatory effects and shows promising therapeutic potential.Thus,in the present study,we explore the role of CBX4 in IDD.Immunohistochemistry staining reveals that CBX4 expression is decreased in severe degenerative NP tissues compared to mild degenerative tissues,and real-time PCR and western blot analysis results show that CBX4 expression is downregulated under TNF-αstimulation in NP cells.siRNA and adenoviruses are used to knockdown or overexpress CBX4,respectively.The results demonstrate that CBX4 knockdown augments the catabolism of ECM in human NP cells,while CBX4 overexpression in rat NP cells restores the ECM degradation induced by TNF-α,as illustrated by immunofluorescence and western blot analysis.In addition,transcriptome sequencing results reveal the regulatory effect of CBX4 on the cell cycle,and further western blot analysis and senescence-associatedβ-galactosidase staining assay indicate that CBX4 overexpression alleviates cell senescence in the presence of TNF-α.Moreover,the phosphorylation of p65,which indicates the activation of NF-κB signaling,is measured by western blot analysis and immunofluorescence assay,and the results reveal that CBX4 overexpression reduces the TNF-α-induced increase in the p-p65/p65 ratio.In addition,the effect of CBX4 overexpression in NP cells is suppressed by NF-κB agonist.In summary,our results indicate that CBX4 overexpression can suppress TNF-α-induced matrix catabolism and cell senescence in the NP by inhibiting NF-κB activation.This study may provide new approaches for preventing and treating IDD. | Yangyang Zhang Shuangxing Li Junmin Hong Jiansen Yan Zhengqi Huang Jiajun Wu Zhihuai Deng Tianyu Qin Kang Xu Wei Ye | 2022 | Acta Biochimica et Biophysica Sinica2022,54,7: | 2 |
| 18 | Source attribution for mercury deposition with an updated atmospheric mercury emission inventory in the Pearl River Delta Region, China显示文摘We used CMAQ-Hg to simulate mercury pollution and identify main sources in the Pearl River Delta (PR.D) with updated local emission inventory and latest regional and global emissions. The total anthropogenic mercury emissions in the PRD for 2014 were 11,939.6 kg. Power plants and industrial boilers were dominant sectors, responsible for 29.4 and 22.7%. We first compared model predictions and observations and the results showed a good performance. Then five scenarios with power plants (PP), municipal solid waste incineration (MSWI), industrial point sources (IP), natural sources (NAT), and boundary conditions (BCs) zeroed out separately were simulated and compared with the base case. BCs was responsible for over 30% of annual average mercury concentration and total deposition while NAT contributed around 15%. Among the anthropogenic sources, IP (22.9%) was dominant with a contribution over 20.0% and PP (18.9%) and MSWI (11.2%) ranked second and third. Results also showed that power plants were the most important emission sources in the central PRD, where the ultra-low emission for thermal power units need to be strengthened. In the northern and western PRD, cement and metal productions were priorities for mercury control. The fast growth of municipal solid waste incineration were also a key factor in the core areas. In addition, a coordinated regional mercury emission control was important for effectively controlling pollution. | 19 | Ursolic acid induces neural regeneration after sciatic nerve injury显示文摘In this study,we aimed to explore the role of ursolic acid in the neural regeneration of the injured sciatic nerve.BALB/c mice were used to establish models of sciatic nerve injury through unilateral sciatic nerve complete transection and microscopic anastomosis at 0.5 cm below the ischial tuberosity.The successfully generated model mice were treated with 10,5,or 2.5 mg/kg ursolic acid via intraperitoneal injection.Enzyme-linked immunosorbent assay results showed that serum S100 protein expression level gradually increased at 1-4 weeks after sciatic nerve injury,and significantly decreased at 8 weeks.As such,ursolic acid has the capacity to significantly increase S100 protein expression levels.Real-time quantitative PCR showed that S100 mRNA expression in the L4-6 segments on the injury side was increased after ursolic acid treatment.In addition,the muscular mass index in the soleus muscle was also increased in mice treated with ursolic acid.Toluidine blue staining revealed that the quantity and average diameter of myelinated nerve fibers in the injured sciatic nerve were significantly increased after treatment with ursolic acid.10 and 5 mg/kg of ursolic acid produced stronger effects than 2.5 mg/kg of ursolic acid.Our findings indicate that ursolic acid can dose-dependently increase S100 expression and promote neural regeneration in BALB/c mice following sciatic nerve injury. | Biao Liu Yan Liu Guang Yang Zemin Xu Jiajun Chen | 2013 | Neural Regeneration Research2013,8,27: | 2 |
