| 4 | Impact of STAT4 gene silencing on the expression profile of proteins in EL-4 cells显示文摘The signal transducers and activators of transcription (STATs) have diverse biological functions and are involved in cell differentiation, proliferation, development, apoptosis and inflammation. Several constitutively activated STATs have been observed in a wide variety of human cancer cell lines and primary tumor cells, including blood malignancies and solid neoplasias. Although regulatory T (Treg) cells induce immune tolerance by suppressing host immune responses against selfor nonself-antigens, thus playing critical roles in preventing autoimmune diseases, they might inhibit antitumor im-munity and promote tumor growth. Our previous findings suggest that the supernatant from STAT4silenced tumor cell culture can significantly increase the ratio of CD4+ CD25+ Foxp3+ regulatory T cells among splenic cells in vitro, when compared to that from normal tumor cell culture. In the present study, we identified that the mouse lymphoma cell line EL-4 expressed a high level of STAT4, and silencing of STAT4 by siRNA did not change the expression levels of TGF-β and IL-10 in EL-4 cells. Two-dimensional electrophoresis was conducted to examine the difference of expression profiles of proteins between normal and STAT4-silenced EL4 cells. | 5 | Effects of different aperture-sized type I collagen/silk fibroin scaffolds on the proliferation and differentiation of human dental pulp cells显示文摘This study aimed at evaluate the effects of different aperture-sized type I collagen/silk fibroin(CSF)scaffolds on the proliferation and differentiation of human dental pulp cells(HDPCs).The CSF scaffolds were designed with 3D mapping software Solidworks.Three different aperture-sized scaffolds(CSF1-CSF3)were prepared by low-temperature deposition 3D printing technology.The morphology was observed by scanning electron microscope(SEM)and optical coherence tomography.The porosity,hydrophilicity and mechanical capacity of the scaffold were detected,respectively.HDPCs(third passage,1105 cells)were seeded into each scaffold and investigated by SEM,CCK-8,alkaline phosphatase(ALP)activity and HE staining.The CSF scaffolds had porous structures with macropores and micropores.The macropore size of CSF1 to CSF3 was 421627 lm,579636 lm and 707643 lm,respectively.The porosity was 69.862.2%,80.162.8%and 86.563.3%,respectively.All these scaffolds enhanced the adhesion and proliferation of HDPCs.The ALP activity in the CSF1 group was higher than that in the CSF3 groups(P<0.01).HE staining showed HDPCs grew in multilayer within the scaffolds.CSF scaffolds significantly improved the adhesion and ALP activity of HDPCs.CSF scaffolds were promising candidates in dentine-pulp complex regeneration. | Shihui Jiang Zhaoxia Yu Lanrui Zhang Guanhua Wang Xiaohua Dai Xiaoli Lian Yan Yan Linpu Zhang Yue Wang Ruixin Li Huiru Zou | 2021 | Regenerative Biomaterials2021,8,4: | 2 |
| 6 | Engineering and modification of microbial chassis for systems and synthetic biology显示文摘Engineering and modifying synthetic microbial chassis is one of the best ways not only to unravel the fundamental principles of life but also to enhance applications in the health,medicine,agricultural,veterinary,and food industries.The two primary strategies for constructing a microbial chassis are the top-down approach(genome reduction)and the bottom-up approach(genome synthesis).Research programs on this topic have been funded in several countries.The‘Minimum genome factory’(MGF)project was launched in 2001 in Japan with the goal of constructing microorganisms with smaller genomes for industrial use.One of the best examples of the results of this project is E.coli MGF-01,which has a reduced-genome size and exhibits better growth and higher threonine production characteristics than the parental strain[1].The‘cell factory’project was carried out from 1998 to 2002 in the Fifth Framework Program of the EU(European Union),which tried to comprehensively understand microorganisms used in the application field.One of the outstanding results of this project was the elucidation of proteins secreted by Bacillus subtilis,which was summarized as the‘secretome’[2].The GTL(Genomes to Life)program began in 2002 in the United States.In this program,researchers aimed to create artificial cells both in silico and in vitro,such as the successful design and synthesis of a minimal bacterial genome by John Craig Venter's group[3].This review provides an update on recent advances in engineering,modification and application of synthetic microbial chassis,with particular emphasis on the value of learning about chassis as a way to better understand life and improve applications. | Haotian Chi Xiaoli Wang Yue Shao Ying Qin Zixin Deng Lianrong Wang Shi Chen | 2019 | Synthetic and Systems Biotechnology2019,4,1: | 2 |

The 5-year event-free survival rate for high-risk (HR) NB is still poor, especially for patients with advanced NB withMYCN gene amplification. Chimeric antigen receptor T (CAR-T) cell therapy is a new treatment for HR-NB.Case presentation: A 55-month-old boy with stage IV HR-NB received 4th-generation CAR-T cells that target disialoganglioside GD2, as consolidation maintenance treatment after intensive chemotherapy, surgery, and autologous stem-cell transplantation. As of February 2019, his CAR-T follow-up time was 37.5 months, indicating prolonged survival. | 20 | Linear dichroism and polarization controllable persistent spin helix in two-dimensional ferroelectric ZrOI_(2) monolayer显示文摘Two-dimensional ferroelectric(2D-FE)materials that characterize the spontaneous ferroelectricity down to monolayer limit and rich ferroic properties arising from FE orderings,have been extensively explored as low-dimensional sensor,electric and memory devices.In current work,group-IV transition metal oxide dihalide MOX_(2)(M=Zr and Hf,X=Cl,Br and I)monolayers have been identified as a new group of 2D-FE materials.Using the comprehensive first-principles calculations combined with finite temperature Monte Carlo(MC)and ab initio molecular dynamics(MD)simulations,we investigate the temperature stability of FE polarization and further uncover the unique properties associated with spontaneous ferroelectricity of MOX_(2) monolayers.In particular,ZrOI_(2) monolayer,a promising 2D-FE material with room temperature stable ferroelectricity,semiconducting electronic structure and optoelectronic response under visible light,offers an ideal material platform to investigate the coupling of intrinsic anisotropy,optical absorption selectivity and spin degree of freedom with 2D ferroelectricity.Typically,significant optical absorption anisotropy and giant linear dichroism effect are predicted for a 2D optical polarizer device based on ZrOI_(2) monolayer,where the adsorption of incident monochromatic linearly polarized light(hv=3.23 eV)along two planar directions with a nearly 100%optical selectivity can be achieved.Moreover,the spin–orbit coupling(SOC)induced spin splitting of valence band edges and out-of-plane textured spin configuration occur in ZrOI_(2) monolayer.In the meanwhile,the unidirectional spin–orbit field protected by C2v wave-vector point group can further create the persistent spin helix(PSH)state,leading to the extraordinarily long spin carrier lifetime.More importantly,the nonvolatile control of PSH state via the electric field induced polarization reversal has also been demonstrated for FE-ZrOI_(2) monolayer,which manifests as a great advantage for applications of ZrOI_(2) as the lowdimensional spin-field effect transistor and all-electric spintronics devices. | Niuzhuang Yang Gaoyang Gou Xiaoli Lu Yue Hao | 2022 | Nano Research2022,15,7: | 0 |
