Design, methodology, and baseline of whole city‑million scale children and adolescents myopia survey(CAMS)in Wenzhou,China显示文摘Background:Myopia is the most common visual impairment in children and adolescents worldwide. This study described an economical and effective population-based screening pipeline and performed the project of a million scale children and adolescents myopia survey(CAMS),which will shed light on the further study of myopia from the level of epidemiology and precision medicine. Methods:We developed a novel population-based screening pattern,an intelligent screening process and internetbased information transmission and analysis system to carry out the survey consisting of school children in Wenzhou,China. The examination items include unaided distance visual acuity, presenting distance visual acuity, and noncycloplegic autorefraction.Myopia and high myopia were defined as spherical equivalent(SE)≤−1.00 diopters(D)and SE≤−6.00 D,respectively.Next, the reports of the vision checking were automatically sent to parents and the related departments. The CAMS project will be done two to four times annually with the support of the government. An online eyesight status information management system(OESIMS)was developed to construct comprehensive and efficient electronic vision health records(EVHRs)for myopia information inquiry, risk pre-warning,and further study. Results:The CAMS completed the first-round of screening within 30 days for 99.41%of Wenzhou students from districts and counties,in June 2019.A total of 1,060,925 participants were eligible for CAMS and 1,054,251(99.37%participation rate)were selected through data quality control, which comprised 1305 schools, and 580,609,251,050 and 170,967 elementary, middle, and high school students. The mean age of participants was 12.21±3.32 years(6-20 years),the female-to-male ratio was 0.82.The prevalence of myopia in elementary, middle, and high school students was 38.16%,77.52%,and 84.00%,respectively,and the high myopia incidence was 0.95%,6.90%,and 12.98%.Conclusions:The CAMS standardized myopia screening model involves automating large-scale information collection, data transmission, data analysis and early warning, thereby supporting myopia prevention and control. The entire survey reduced 90%of staff, cost, and time consumption compared with previous surveys. This will provide new insights for decision support for public health intervention. | Liangde Xu Youyuan Zhuang Guosi Zhang Yunlong Ma Jian Yuan Changseng Tu MiaoMiao Li Wencan Wang Yaru Zhang Xiaoyan Lu Jing Li Xinting Liu Zhengbo Xue Meng Zhou Jie Sun Jinhua Bao Ming Li Fan Lu Hong Wang Jianzhong Su Jia Qu | 2021 | Eye and Vision2021,8,1: | 7
Field trial of an HTS filter system on a CDMA base station显示文摘This paper presents the field trial results of a high temperature superconductor filter system (HTSFS) in a code division multiple access (CDMA) mobile communication base station (BTS) in Tangshan, China. The center frequency of the HTSFS is 830 MHz with 1.4% fractional bandwidth. The noise figure (NF) of the HTSFS, including connectors and cables, is 0.8 dB. The field trial results show that after the HTSFS was installed in the receiver front-end of a CDMA base station, the transmitting power of handset was decreased by 3.1 dB on average. Up to now, the HTSFS has been running steadily in the base station for more than 22 months, demonstrating the reliability and stability of the system. | YIN ZheSheng WEI Bin CAO BiSong WANG Xin GUO XuBo ZHANG XiaoPing GAO LongMa PIAO YunLong ZHU MaFeng LIANG Yong WANG Fan H. | 4 | Improving MapReduce Performance by Balancing Skewed Loads显示文摘MapReduce has emerged as a popular computing model used in datacenters to process large amount of datasets. In the map phase, hash partitioning is employed to distribute data that sharing the same key across data center-scale cluster nodes. However,we observe that this approach can lead to uneven data distribution, which can result in skewed loads among reduce tasks, thus hamper performance of MapReduce systems. Moreover, worker nodes in MapReduce systems may differ in computing capability due to(1) multiple generations of hardware in non-virtualized data centers,or(2) co-location of virtual machines in virtualized data centers. The heterogeneity among cluster nodes exacerbates the negative effects of uneven data distribution. To improve MapReduce performance in heterogeneous clusters,we propose a novel load balancing approach in the reduce phase. This approach consists of two components:(1) performance prediction for reducers that run on heterogeneous nodes based on support vector machines models,and(2) heterogeneity-aware partitioning(HAP),which balances skewed data for reduce tasks. We implement this approach as a plug-in in current MapReduce system.Experimental results demonstrate that our proposed approach distributes work evenly among reduce tasks, and improves MapReduce performance with little overhead. | FAN Yuanquan WU Weiguo XU Yunlong CHEN Heng | 2014 | China Communications2014,11,8: | 4
A recombinant receptor-binding domain in trimeric form generates protective immunity against SARSCoV-2 infection in nonhuman primates显示文摘A safe and effective vaccine is critical to combat the COVID-19 pandemic. Here,we developed a trimeric SARS-CoV-2 receptor-binding domain(RBD)subunit vaccine candidate that simulates the natural structure of the spike(S)trimer glycoprotein.Immunization with the RBD trimer-induced robust humoral and cellular immune responses, and a high level of neutralizing antibodies was maintained for at least 4.5 months. Moreover, the antibodies that were produced in response to the vaccine effectively cross-neutralized the SARS-CoV-2501Y.V2 variant(B.1.351).Of note, when the vaccine-induced antibodies dropped to a sufficiently low level, only one boost quickly activated the anamnestic immune response, conferring full protection against a SARSCoV-2 challenge in rhesus macaques without typical histopathological changes in the lung tissues. These results demonstrated that the SARS-CoV-2 RBD trimer vaccine candidate is highly immunogenic and safe, providing long-lasting,broad,and significant immunity protection in nonhuman primates, thereby offering an optimal vaccination strategy against COVID-19. | Limin Yang Deyu Tian Jian-bao Han Wenhui Fan Yuan Zhang Yunlong Li Wenqiang Sun Yanqiu Wei Xiaodong Tian Dan-dan Yu Xiao-li Feng Gong Cheng Yuhai Bi Yong-tang Zheng Wenjun Liu | 2021 | The Innovation2021,2,3: | 3
Reprogramming of phytopathogen transcriptome by a non-bactericidal pesticide residue alleviates its virulence in rice显示文摘Bacteria equipped with virulence systems based on highly bioactive small molecules can circumvent their host's defense mechanisms.Pathogens employing this strategy are currently threatening global rice production. In the present study, variations in the virulence of the highly destructive Barkholderiaplantarii were observed in different rice-producing regions. The environment-linked variation was not attributable to any known host-related or external factors.Co-occurrence analyses indicated a connection between reduced virulence and 5-Amino-l,3,4-thiadiazole-2-thiol(ATT),a non-bactericidal organic compound.ATT,which accumulates in rice plants during metabolization of specific agrochemicals, was found to reduce virulence factor secretion by B.plantarii up to 88.8%and inhibit pathogen virulence by hijacking an upstream signaling cascade.Detailed assessment of the newly discovered virulence inhibitor resulted in mechanistic insights into positive effects of ATT accumulation in plant tissues.Mechanisms of virulence alleviation were deciphered by integrating high-throughput data, gene knockout mutants, and molecular interaction assays.TroK,a histidine protein kinase in a two-component system that regulates virulence factor secretion,is likely the molecular target antagonized by ATT.Our findings provide novel insights into virulence modulation in an important plant-pathogen system that relies on the host's metabolic activity and subsequent signaling interference. | Haruna Matsumoto Yuan Qian Xiaoyan Fan Sunlu Chen Yanxia Nie Kun Qiao Dandan Xiang Xinzhong Zhang Meng Li Bo Guo Peilin Shen Qiangwei Wang Yunlong Yu Tomislav Cemava Mengcen Wang | 2022 | Fundamental Research2022,2,2: | 2
Energetic particles’fluxes and dose in the Radiation Gene Box measured by space radiation detector onboard SJ-10 satellite显示文摘Background To evaluate the hazard of space radiation posing to the tissues,it is important to obtain exact fluxes of different radiation particles. The Radiation Gene Box(RGB)onboard SJ-10 spacecraft was an instrument designed to investigate the effects of space environment on the mESCs and drosophila. To derive the dose received by the tissues inside the RGB,the Space Radiation Detector(SRD)was installed inside it.Purpose The SRD was designed to derive the fluxes of electron, proton, hellion and gamma rays around it.If the type of the particles, the energies, the fluxes and the conversion coefficients are known, the dose received by the tissues could be evaluated. Methods The SRDwas designed as a △E-E solid-state telescope.By measuring the energy deposited in the three subdetectors, the particles’type and their energies could be discriminated. The data of SRDwere divided into 15 bins by the types of particles and their energy ranges. Results The gamma ray flux was higher than any other particle flux inside the RGB,and the electron was the most intense charge particle, while the helium ion was the most harmful radiation to the cells inside the RGB.Conclusion The dose rate inside the Radiation Gene Box was much higher than in the ground, but the integral dose of 12 days inside the RGB was about 2.13 mSv.It seemed unlikely to have obvious biological effects on the tissues of mice and drosophila. | Xingzhu Cui Yaqing Liu Wenxi Peng Jinzhou Wang Min Gao Dongya Guo Xiaohua Liang Ruirui Fan Huanyu Wang Yunlong Zhang Zhongjian Ma Mingyang Yan Hong Xiao Yuanda Jiang Haiying Hang | 2018 | Radiation Detection Technology and Methods2018,2,2: | 2
Direct conversion of mouse astrocytes into neural progenitor cells and specific lineages of neurons显示文摘Background:Cell replacement therapy has been envisioned as a promising treatment for neurodegenerative diseases.Due to the ethical concerns of ESCs-derived neural progenitor cells(NPCs)and tumorigenic potential of iPSCs,reprogramming of somatic cells directly into multipotent NPCs has emerged as a preferred approach for cell transplantation. Methods:Mouse astrocytes were reprogrammed into NPCs by the overexpression of transcription factors(TFs)Foxg1,Sox2,and Brn2.The generation of subtypes of neurons was directed by the force expression of cell-type specific TFs Lhx8 or Foxa2/Lmx1a.Results:Astrocyte-derived induced NPCs(AiNPCs)share high similarities, including the expression of NPC-specific genes,DNA methylation patterns, the ability to proliferate and differentiate, with the wild type NPCs.The AiNPCs are committed to the forebrain identity and predominantly differentiated into glutamatergic and GABAergic neuronal subtypes.Interestingly, additional overexpression of TFs Lhx8 and Foxa2/Lmx1a in AiNPCs promoted cholinergic and dopaminergic neuronal differentiation, respectively. Conclusions:Our studies suggest that astrocytes can be converted into AiNPCs and lineage-committed AiNPCs can acquire differentiation potential of other lineages through forced expression of specific TFs.Understanding the impact of the TF sets on the reprogramming and differentiation into specific lineages of neurons will provide valuable strategies for astrocyte-based cell therapy in neurodegenerative diseases. | Kangmu Ma Xiaobei Deng Xiaohuan Xia Zhaohuan Fan Xinrui Qi Yongxiang Wang Yuju Li Yizhao Ma Qiang Chen Hui Peng Jianqing Ding Chunhong Li Yunlong Huang Changhai Tian Jialin C.Zheng | 2018 | Translational Neurodegeneration2018,7,1: | 2
Leakage current characteristics and ageing assessment technology of roof arrester under ultra harmonics overvoltage显示文摘The large leakage current of the roof arrester caused by the high‐frequency resonant overvoltage on the high‐speed train occurs intermittently, which will cause the arrester to overheat locally. In this work, the overheating issue of the arrester under Ultra Harmonics Overvoltage is studied. Then, the relationship between the active power and the voltage(including amplitude and frequency)of the arrester is investigated, and the impact mechanism of the harmonic voltage on the overheating fault of the arrester is analysed based on experiments in this work.It is found that the total leakage current of the arrester is proportional to the amplitude and frequency of the applied voltage. The internal resistance of the arrester is independent of the amplitude of the voltage, but it will decrease with the increase of the frequency. The main reason that leads to thermal breakdown of the arrester valve under high‐frequency harmonic voltage is the heating power elevation of the arrester valve. The research study can provide theoretical basis and data support for the design, type selection and monitoring of the high‐speed train roof arrester. | Jixing Sun Fan Ding Yunlong Lv Jiayi Ren Sibo Song Tiance Li Qingyun Zhi Changwang Guo | 2022 | High Voltage2022,7,2: | 1

SAMD4 family members suppress human hepatitis B virus by directly binding to the Smaug recognition region of viral RNA显示文摘HBV infection initiates hepatitis B and promotes liver cirrhosis and hepatocellular carcinoma.IFN-α is commonly used in hepatitis B therapy, but how it inhibits HBV is not fully understood. We screened 285 human interferon-stimulated genes(ISGs)for anti-HBV activity using a cell-based assay, which revealed several anti-HBV ISGs.Among these ISGs,SAMD4A was the strongest suppressor of HBV replication. We found the binding site of SAMD4A in HBV RNA,which was a previously unidentified Smaug recognition region(SRE)sequence conserved in HBV variants.SAMD4A binds to the SRE site in viral RNA to trigger its degradation. The SAM domain in SAMD4A is critical for RNA binding and the C-terminal domain of SAMD4A is required for SAMD4A anti-HBV function.Human SAMD4B is a homolog of human SAMD4A but is not an ISG,and the murine genome encodes SAMD4.All these SAMD4 proteins suppressed HBV replication when overexpressed in vitro and in vivo.We also showed that knocking out the Samd4 gene in hepatocytes led to a higher level of HBV replication in mice and AAV-delivered SAMD4A expression reduced the virus titer in HBVproducing transgenic mice.In addition,a database analysis revealed a negative correlation between the levels of SAMD4A/B and HBV in patients. Our data suggest that SAMD4A is an important anti-HBV ISG for use in IFN therapy of hepatitis B and that the levels of SAMD4A/B expression are related to HBV sensitivity in humans. | Yuze Wang Xinrui Fan Yunlong Song Yifei Liu Ruixin Liu Jianfeng Wu Xiaoling Li Quan Yuan Guo Fu Ningshao Xia Jiahuai Han | 2021 | Cellular & Molecular Immunology2021,18,4: | 1
Formation of the Huajiang Grand Canyon (southwestern China) driven by the evolution of a Late Pleistocene tiankeng显示文摘Collapse is a common geomorphic process in karst areas, especially on the Yunnan-Guizhou Plateau, which has a tectonic background of integral uplift. The frequent occurrence of collapse processes in karst underground caves and canyons indicates that collapses play an important role in the formation of canyons.Through an analysis of the morphology of a semicircular cliff in the Huajiang Grand Canyon and an investigation of sediments at the bottom of the cliff,a large-scale collapse event was found to have occurred.U-series dating of secondary calcium carbonate cement in the collapse breccias indicates that collapse processes occurred approximately 200 ka.According to the geomorphological evolution of the Huajiang Grand Canyon, the following geomorphic evolutionary process is proposed:underground river-cave hall-collapse of a tiankeng-tiankeng degradation-canyon formation. These findings also show that the dating of collapsed breccia cement can be effectively used to determine the development times of karst canyons and the formation ages of tiankengs. | Yunlong Fan Andrea Columbu Kangning Xiong Guangjie Luo Song Li Xuefeng Wang Yangyang Wu | 2022 | Acta Geochimica2022,41,1: | 1
Excessive branched-chain amino acid accumulation restricts mesenchymal stem cell-based therapy efficacy in myocardial infarction显示文摘Mesenchymal stem cells(MSCs)delivered into the post-ischemic heart milieu have a low survival and retention rate, thus restricting the cardioreparative efficacy of MSC-based therapy.Chronic ischemia results in metabolic reprogramming in the heart, but little is known about how these metabolic changes influence implanted MSCs.Here,we found that excessive branched-chain amino acid(BCAA)accumulation,a metabolic signature seen in the post-ischemic heart, was disadvantageous to the retention and cardioprotection of intramyocardially injected MSCs.Discovery-driven experiments revealed that BCAA at pathological levels sensitized MSCs to stress-induced cell death and premature senescence via accelerating the loss of histone 3 lysine 9 trimethylation(H3K9me3).A novel mTORC1/DUX4/KDM4E axis was identified as the cause of BCAA-induced H3K9me3 loss and adverse phenotype acquisition.Enhancing BCAA catabolic capability in MSCs via genetic/pharmacological approaches greatly improved their adaptation to the high BCAA milieu and strengthened their cardioprotective efficacy. We conclude that aberrant BCAA accumulation is detrimental to implanted MSCs via a previously unknown metabolite-signaling-epigenetic mechanism, emphasizing that the metabolic changes of the post-ischemic heart crucially influence the fate of implanted MSCs and their therapeutic benefits. | Fuyang Zhang Guangyu Hu Xiyao Chen Ling Zhang Lanyan Guo Congye Li Hang Zhao Zhe Cui Xiong Guo Fangfang Sun Dandan Song Wenjun Yan Yunlong Xia Shan Wang Miaomiao Fan Ling Tao | 2022 | Signal Transduction and Targeted Therapy2022,7,7: | 1
Perivascular localized cells commit erythropoiesis in PDGF-B-expressing solid tumors显示文摘Background:Tumors possess incessant growth features, and expansion of their masses demands sufficient oxygen supply by red blood cells(RBCs).In adult mammals, the bone marrow(BM)is the main organ regulating hematopoiesis with dedicated manners.Other than BM,extramedullary hematopoiesis is discovered in various pathophysiological settings. However, whether tumors can contribute to hematopoiesis is completely unknown.Accumulating evidence shows that,in the tumor microenvironment(TME),perivascular localized cells retain progenitor cell properties and can differentiate into other cells. Here,we sought to better understand whether and how perivascular localized pericytes in tumors manipulate hematopoiesis. Methods:To test if vascular cells can differentiate into RBCs,genome-wide expression profiling was performed using mouse-derived pericytes.Genetic tracing of perivascular localized cells employing NG2-CreERT2:R26R-tdTomato mouse strain was used to validate the findings in vivo.Fluorescence-activated cell sorting(FACS),single-cell sequencing, and colony formation assays were applied for biological studies. The production of erythroid differentiationspecific cytokine, erythropoietin(EPO),in TME was checked using quantitative polymerase chain reaction(qPCR),enzyme-linked immunosorbent assay(ELISA,magnetic-activated cell sorting and immunohistochemistry. To investigate BM function in tumor erythropoiesis,BM transplantation mouse models were employed.Results:Genome-wide expression profiling showed that in response to plateletderived growth factor subunit B(PDGF-B),neural/glial antigen 2(NG2)+perivascular localized cells exhibited hematopoietic stem and progenitor-like features and underwent differentiation towards the erythroid lineage.PDGF-B simultaneously targeted cancer-associated fibroblasts to produce high levels of EPO,a crucial hormone that necessitates erythropoiesis.FACS analysis using genetic tracing of NG2+cells in tumors defined the perivascular localized cell-derived subpopulation of hematopoietic cells.Single-cell sequencing and colony formation assays validated the fact that, upon PDGF-B stimulation,NG2+cells isolated from tumors acted as erythroblast progenitor cells, which were distinctive from the canonical BM hematopoietic stem cells. Conclusions:Our data provide a new concept of hematopoiesis within tumor tissues and novel mechanistic insights into perivascular localized cell-derived erythroid cells within TME.Targeting tumor hematopoiesis is a novel therapeutic concept for treating various cancers that may have profound impacts on cancer therapy. | Kayoko Hosaka Chenchen Wang Shiyue Zhang Xue Lv Takahiro Seki Yin Zhang Xu Jing Jieyu Wu Qiqiao Du Xingkang He Yulong Fan Xuan Li Makoto Kondo Masahito Yoshihara Hong Qian Lihong Shi Ping Zhu Yuanfu Xu Yunlong Yang Tao Cheng Yihai Cao | 2023 | Cancer Communications2023,43,6: | 1

Laser-Induced Recoverable Fluorescence Quenching of Perovskite Films at a Microscopic Grain Scale显示文摘Understanding the fundamental properties of metal-halide perovskite materials is driving the development of novel optoelectronic applications. Here,we report the observation of a recoverable laser-induced fluorescence quenching phenomenon in perovskite films with a microscopic grain-scale restriction, accompanied by spectral variations. This fluorescence quenching depends on the laser intensity and the dwell time under Auger recombination dominated conditions. These features indicate that the perovskite lattice deformation may take the main responsibility for the transient and show a new aspect to understand halide perovskite photostability. We further modulate this phenomenon by adjusting the charge carrier recombination and extraction, revealing that efficient carrier transfer can improve the bleaching resistance of perovskite grains. Our results provide future opportunities to attain high-performance devices by tuning the perovskite lattice disorder and harvesting the energetic carriers. | Yuren Xiang Yameng Cao Wenqiang Yang Rui Hu Sebastian Wood Bowei Li Qin Hu Fan Zhang Jujie He Mozhgan Yavari Jinlai Zhao Yunlong Zhao Jun Song Junle Qu Rui Zhu Thomas P.Russell S.Ravi P.Silva Wei Zhang | 2022 | Energy & Environmental Materials2022,5,4: | 0
Mixing Dynamics and Synthesis Performance of Staggered Herringbone Micromixer for Limit Size Lipid Nanoparticles显示文摘Staggered herringbone micromixer has shown good efficiency of mixing and performance of synthesizing nanoparticles. To bring a detailed understanding of the mixing dynamics and syn-thesis performance of this kind of micromixer, this paper carries out a high-fidelity numerical simu-lation and a parametrial experimental study on a well-established design.A passive tracer is induced in the numerical simulation to analyze mixing dynamics induced by the staggered herring-bone structures.Three effects are identified to reveal the underlying mechanisms, including folding, stretching, and splitting. To the authors’knowledge, the splitting effect is identified for the first time by the isosurface of the passive tracer,to show the high efficiency of the staggered herring-bone design. The micromixer is then used to synthesize lipid nanoparticles by mixing a mixture of lipid and poly(lactic-co-glycolic acid)(PLGA)solutions with deionized water.Under a wide mass ratio of lipid and PLGA solutions, nanoparticles with good monodispersity are synthesized to reflect the good compatibility of the micromixer and the mixture. In addition,an optimized mass ratio is identified from the parametrical experiment. | Shuai Zhang Yunlong Fan Chenmu Xue Yuanzhan Yang Xiaoqiong Li Xiaodong Chen Yulin Deng | 2022 | Journal of Beijing Institute of Technology2022,31,3: | 0
Multimodal integrated strategy for the discovery and identification of quality markers in traditional Chinese medicine显示文摘With the modernization and internationalization of traditional Chinese medicine(TCM),the requirement for quality control has increased. The quality marker(Q-marker)is an important standard in this field and has been implemented with remarkable success in recent years. However, the establishment of Qmarkers remains fragmented and the process lacks systematicity, resulting in inconsistent quality control and insufficient correlation with clinical efficacy and safety of TCM.This review introduces four multimodal integrated approaches that contribute to the discovery of more comprehensive and accurate Qmarkers, thus aiding in the establishment of new quality control patterns based on the characteristics and principles of TCM.These include the whole-process quality control strategy,chemical-activity-based screening method, efficacy, safety, and consistent combination strategy, and TCM theory-guided approach.Furthermore, methodologies and representative examples of these strategies are described, and important future directions and questions in this field are also proposed. | Xiaoyan Lu Yanyan Jin Yuzhen Wang Yunlong Chen Xiaohui Fan | 2022 | Journal of Pharmaceutical Analysis2022,12,5: | 0
