1Receptome profiling identifies KREMEN1 and ASGR1 as alternative functional receptors of SARS-CoV-2显示文摘Host cellular receptors play key roles in the determination of virus tropism and pathogenesis.However,little is known about SARS-CoV-2 host receptors with the exception of ACE2.Furthermore,ACE2 alone cannot explain the multi-organ tropism of SARS-CoV-2 nor the clinical differences between SARS-CoV-2 and SARS-CoV,suggesting the involvement of other receptor(s).Here,we performed genomic receptor profiling to screen 5054 human membrane proteins individually for interaction with the SARS-CoV-2 capsid spike(S)protein.Twelve proteins,including ACE2,ASGR1,and KREMEN1,were identified with diverse S-binding affinities and patterns.ASGR1 or KREMEN1 is sufficient for the entry of SARS-CoV-2 but not SARS-CoV in vitro and in vivo.SARS-CoV-2 utilizes distinct ACE2/ASGR1/KREMEN1(ASK)receptor combinations to enter different cell types,and the expression of ASK together displays a markedly stronger correlation with virus susceptibility than that of any individual receptor at both the cell and tissue levels.The cocktail of ASK-related neutralizing antibodies provides the most substantial blockage of SARS-CoV-2 infection in human lung organoids when compared to individual antibodies.Our study revealed an interacting host receptome of SARS-CoV-2,and identified ASGR1 and KREMEN1 as alternative functional receptors that play essential roles in ACE2-independent virus entry,providing insight into SARS-CoV-2 tropism and pathogenesis,as well as a community resource and potential therapeutic strategies for further COVID-19 investigations.Yunqing Gu Jun Cao Xinyu Zhang Hai Gao Yuyan Wang Jia Wang Juan He Xiaoyi Jiang Jinlan Zhang Guanghui Shen Jie Yang Xichen Zheng Gaowei Hu Yuanfei Zhu Shujuan Du Yunkai Zhu Rong Zhang Jianqing Xu Fei Lan Di Qu Guoliang Xu Yun Zhao Dong Gao Youhua Xie Min Luo Zhigang Lu 2022Cell Research2022,32,1:8
2Inhibitory effects of the ferment liquids on sarcoma显示文摘Objective: To study the anti-tumor effects of the ferment liquid (FL) and to explore the anti-mutagenic effects induced by cyclophosphamide (CP). Methods: Anti-tumor effects and Anti-mutagenic effects induced by cyclophosphamide (CP) of the ferment liquid were observed in tumor-bearing mice in vivo. Spleen lymphocytes proliferation of mice was stimulated by ConA and NK cell activity was analyzed by the lactate dehydrogenase (LDH) release assay. Results: FL Ⅰ,FL Ⅱ and FL Ⅲ of 16.7, 33.3 and 66.6 ml/kg inhibited growth of Sarcoma 180 sighificanth by 11.6%, 24.2%, 28.0%, 4.8%, 15.2%, 2.2% and 2.1%, 8.5%, 5.2% respectively. Polychromatic erythrocytes (PCE) at all doses of FL Ⅰ and FL Ⅱ were greatly decreased compared with control group (P<0.05), but FL Ⅲ has no inhibition effents(P>0.05). Production of IL-2 showed no significant difference between all treatment groups. The NK cell killing activity in all treated mice increased by FL Ⅰ and FL Ⅱ was significantly potentiated compared to controls (P<0.05). Lymphocytes proliferation of ICR mice were stimulated by ConA 2.5 ml/L and there were no differences between groups. 3Si nanocrystals-based multilayers for luminescent and photovoltaic device applications显示文摘Low dimensional Si materials have attracted much attention because they can be developed in many kinds of new-generation nano-electronic and optoelectronic devices, among which Si nanocrystals-based multilayered material is one of the most promising candidates and has been extensively studied. By using multilayered structures, the size and distribution of nanocrystals as well as the barrier thickness between two adjacent Si nanocrystal layers can be well controlled, which is beneficial to the device applications. This paper presents an overview of the fabrication and device applications of Si nanocrystals, especially in luminescent and photovoltaic devices. We first introduce the fabrication methods of Si nanocrystals-based multilayers. Then, we systematically review the utilization of Si nanocrystals in luminescent and photovoltaic devices. 7High Sensitivity Plasmonic Metal-Dielectric-Metal Device With Two Side-Coupled Fano Cavities显示文摘In this paper, we propose a compact plasmonic sensor structure comprised of a metal-dielectric(MDM) waveguide, and a baffle plate in waveguide core and two side-coupled rectangular cavities. In this structure, two Fano resonances are achieved and can be tuned independently by changing the structural parameters of the cavities. Especially, when the resonant wavelengths of the two Fano resonances are the same, the sensing sensitivity can be enhanced by coupling between two Fano resonances. By investigating the transmission spectrum, the effect of structural parameters on Fano resonances and the refractive index sensitivity of the sensor structure are analyzed in detail. 12N6-methyladenosine facilitates mitochondrial fusion of colorectal cancer cells via induction of GSH synthesis and stabilization of OPA1 mRNA显示文摘Mitochondria undergo fission and fusion that are critical for cell survival and cancer development,while the regulatory factors for mitochondrial dynamics remain elusive.Herein we found that RNA m^(6)A accelerated mitochondria fusion of colorectal cancer(CRC)cells.Metabolomics analysis and function studies indicated that m^(6)A triggered the generation of glutathione(GSH)via the upregulation of RRM2B-a p53-inducible ribonucleotide reductase subunit with anti-reactive oxygen species potential.This in turn resulted in the mitochondria fusion of CRC cells.Mechanistically,m^(6)A methylation of A1240 at 3'UTR of RRM2B increased its mRNA stability via binding with IGF2BP2.Similarly,m^(6)A methylation of A2212 at the coding sequence(CDS)of OPA1-an essential GTPase protein for mitochondrial inner membrane fusion-also increased mRNA stability and triggered mitochondria fusion.Targeting m^(6)A through the methyltransferase inhibitor STM2457 or the dm^(6)ACRISPR system significantly suppressed mitochondria fusion.In vivo and clinical data confirmed the positive roles of the m^(6)A/mitochondrial dynamics in tumor growth and CRC progression.Collectively,m^(6)A promoted mitochondria fusion via induction of GSH synthesis and OPA1 expression,which facilitated cancer cell growth and CRC development.Jiawang Zhou Haisheng Zhang Ke Zhong Lijun Tao Yu Lin Guoyou Xie Yonghuang Tan You Wu Yunqing Lu Zhuojia Chen Jiexin Li Xin Deng Qin Peng Zigang Li Hongsheng Wang 2024National Science Review2024,11,3:0
13Scenario-based assessment and multi-objective optimization of urban development plan with carrying capacity of water system显示文摘Environmental impact assessments and subsequent regulation measures of urban development plans are critical to human progress toward sustainability,since these plans set the scale and structure targets of future socioeconomic development.A three-step methodology for assessing and optimizing an urban development plan focusing on its impacts on the water system was developed.The methodology first predicted the pressure on the water system caused by implementation of the plan under distinct scenarios then compared the pressure with the carrying capacity threshold to verity the system status;finally,a multi-objected the pressure with the carrying capacity threshold to verity the system status;finally,a multi-objective optimization method was used to propose regulation solutions.The methodology enabled evaluation of the water system carrying state,taking socioeconomic development uncertainties into account,and multiple sets of improvement measures under different decisionmaker preferences were generated.The methodology was applied in the case of Zhoushan city in South-east China.The assessment results showed that overloading problems occurred in 11 out of the 13 zones in Zhoushan,with the potential pressure varying from 1.1 to 18.3 times the carrying capacity.As a basic regulation measure,and environmental efficiency upgrade could relieve the overloading in 4 zones and reduce 9%-63%of the pressure.The optimization of industrial development showed that the pressure could be controlled under the carrying capacity threshold if the planned scale was reduced by 24%and the industrial structure was transformed.Various regulation schemes including a more suitable scale and structure with necessary efficiency standards are provided for decisionmakers that can help the case city approach a more sustainable development pattern.Yilei Lu Yunqing Huang Siyu Zeng Can Wang 2020Frontiers of Environmental Science & Engineering2020,14,2:0

16Four-junction AlGaAs/GaAs laser power converter显示文摘Four-junction AlGaAs/GaAs laser power converters (LPCs) with n^+-Ga As/p^+-Al_(0.37)Ga_(0.63)As hetero-structure tunnel junctions (TJs) have been designed and grown by metal-organic chemical vapor deposition(MOCVD) for converting the power of 808 nm lasers. 17Anatomical morphology of the aortic valve in Chinese aortic stenosis patients and clinical results after downsize strategy of transcatheter aortic valve replacement显示文摘Background:The study aimed to describe the aortic valve morphology in Chinese patients underwent transcatheter aortic valve replacement(TAVR)for symptomatic severe aortic stenosis(AS),and the impact of sizing strategies and related procedural outcomes.Methods:Patients with severe AS who underwent TAVR were consecutively enrolled from 2012 to 2019.The anatomy and morphology of the aortic root were assessed.'Downsize'strategy was preformed when patients had complex morphology.The clinical outcomes of patients who performed downsize strategy were compared with those received annular sizing strategy.The primary outcome was device success rate,and secondary outcomes included Valve Academic Research Consortium-3 clinical outcomes variables based on 1-year follow-up.Results:A total of 293 patients were enrolled.Among them,95 patients(32.4%)had bicuspid aortic valve.The calcium volume(Hounsfield Unit-850)of aortic root was 449.90(243.15-782.15)mm3.Calcium is distributed mostly on the leaflet level.Downsize strategy was performed in 204 patients(69.6%).Compared with the patients who performed annular sizing strategy,those received downsize strategy achieved a similar device success rate(82.0%[73]vs.83.3%[170],P=0.79).Aortic valve gradients(downsize strategy group vs.annular sizing group,11.28 mmHg vs.11.88 mmHg,P=0.64)and percentages of patients with moderate or severe paravalvular regurgitation 2.0%(4/204)vs.4.5%(4/89),P=0.21were similar in the two groups at 30 days after TAVR.These echocardiographic results were sustainable for one year.Conclusions:Chinese TAVR patients have more prevalent bicuspid morphology and large calcium volume of aortic root.Calcium is distributed mostly on the leaflet level.Compare with annular sizing strategy,downsize strategy provided a non-inferior device success rate and transcatheter heart valve hemodynamic performance in self-expanding TAVR procedure.Guannan Niu Walid Ben Ali Moyang Wang Hasan Jilaihawi Haitong Zhang Qian Zhang Yunqing Ye Xinmin Liu Jing Yao Qinghao Zhao Yubin Wang Zheng Zhou Lizhi Zhang Xinshuang Ren Yunqiang An Bin Lu Thomas Modine Yongjian Wu Guangyuan Song 2022Chinese Medical Journal2022,135,24:0
