2SHP2 inhibition triggers anti-tumor immunity and synergizes with PD-1 blockade显示文摘Tyrosine phosphatase SHP2 is a promising drug target in cancer immunotherapy due to its bidirectional role in both tumor growth promotion and T-cell inactivation. Its allosteric inhibitor SHP099 is known to inhibit cancer cell growth both in vitro and in vivo. However, whether SHP099-mediated SHP2 inhibition retards tumor growth in vivo via anti-tumor immunity remains elusive. To address this, a CT-26 colon cancer xenograft model was established in mice since this cell line is insensitive to SHP099.Consequently, SHP099 minimally affected CT-26 tumor growth in immuno-deficient nude mice, but significantly decreased the tumor burden in CT-26 tumor-bearing mice with intact immune system.SHP099 augmented anti-tumor immunity, as shown by the elevated proportion of CD8tIFN-γtT cells and the upregulation of cytotoxic T-cell related genes including Granzyme B andPerforin, which decreased the tumor load. In addition, tumor growth in mice with SHP2-deficient T-cells was markedly slowed down because of enhanced anti-tumor responses. Finally, the combination of SHP099 and antiPD-1 antibody showed a higher therapeutic efficacy than either monotherapy in controlling tumor growthin two colon cancer xenograft models, indicating that these agents complement each other. 4Doping bioactive elements into a collagen scaffold based on synchronous self-assembly/mineralization for bone tissue engineering显示文摘Pure collagen is biocompatible but lacks inherent osteoinductive,osteoimmunomodulatory and antibacterial activities.To obtain collagen with these characteristics,we developed a novel methodology of doping bioactive elements into collagen through the synchronous self-assembly/mineralization(SSM)of collagen.In the SSM model,amorphous mineral nanoparticles(AMN)(amorphous SrCO3,amorphous Ag3PO4,etc.)stabilized by the polyampholyte,carboxymethyl chitosan(CMC),and collagen molecules were the primary components under acidic conditions.As the pH gradually increased,intrafibrillar mineralization occurred via the self-adaptive interaction between the AMNs and the collagen microfibrils,which were self-assembling;the AMNs wrapped around the microfibrils became situated in the gap zones of collagen and finally transformed into crystals.Srdoped collagen scaffolds(Sr-CS)promoted in vitro cell proliferation and osteogenic differentiation of rat bone marrow mesenchymal stromal cells(rBMSCs)and synergistically improved osteogenesis of rBMSCs by altering the macrophage response.Ag-doped collagen scaffolds(Ag-CS)exhibited in vitro antibacterial effects on S.aureus,as well as cell/tissue compatibility.Moreover,Sr-CS implanted into the calvarial defect of a rat resulted in improved bone regeneration.Therefore,the SSM model is a de novo synthetic strategy for doping bioactive elements into collagen,and can be used to fabricate multifunctional collagen scaffolds to meet the clinical challenges of encouraging osteogenesis,boosting the immune response and fighting severe infection in bone defects.Huanhuan Liu Mingli Lin Xue Liu Ye Zhang Yuyu Luo Yanyun Pang Haitao Chen Dongwang Zhu Xue Zhong Shiqing Ma Yanhong Zhao Qiang Yang Xu Zhang 2020Bioactive Materials2020,5,4:6

6Principle and application of three-band gradient difference vegetation index显示文摘Vegetation index is a simple, effective and experiential measurement of terrestrial vegetation activity, and plays a very important role in qualitative and quantitative remote sensing. Aiming at shortages of current vegetation indices, and starting from the analysis of vegetation spectral characteristics, we put forward a new vegetation index, the three-band gradient difference vegetation index (TGDVI), and established algorithms to inverse crown cover fraction and leaf area index (LAI) from it. Theoretical analysis and model simulation show that TGDVI has high saturation point and the ability to remove the influence of background to some degree, and the explicit functional relation with crown cover fraction and LAI can be established. Moreover, study shows that TGDVI also has the ability to partly remove the influence of thin cloud. Experiment in the Shunyi District, Beijing, China shows that reasonable result can be reached using the vegetation index to retrieve LAI. We also theoretically analyzed the reason why the normalized difference vegetation index (NDVI) owns the low saturation point, and show that it is determined by the definition of NDVI and the characteristic of vegetation spectra, and is unavoidable to some degree. 8Highly stable and antifouling graphene oxide membranes prepared by bio-inspired modification for water purification显示文摘Graphene oxide(GO)membranes show great potential in molecular separation for water treatment.However,the inferior stability of GO membranes is a major bottleneck for practical applications.In this study,bio-inspired polydopamine(PDA)deposition is reported for enhancing the stability of GO membranes.Through simple and mild immersion,PDA is self-polymerized on GO membranes.The blocking of PDA chains to membrane defects improves the rejections for various molecules.Because the inherently strong adhesion and crosslinking of PDA greatly strengthen the interactions of substrates to GO layers and the binding force of GO nanosheets,the prepared PDA-GO membranes exhibit impressive long-te rm stability in cross-flow filtration,and maintain good nanofiltration perfo rmance at various feed pressures,tangential velocities,and eve n after external scratching.Moreover,because the deposited PDA layers obstruct the direct contact between GO and contaminants,the antifouling property of the PDA-GO membranes increases substantially,with recovery ratio about 98%.Weiming Zhong Yuyu Zhang Ling Zhao Wanbin Li 2020Chinese Chemical Letters2020,31,10:3

14Quantifying the response of surface urban heat island to urbanization using the annual temperature cycle model显示文摘Urban heat island(UHI),driving by urbanization,plays an important role in urban sustainability under climate change.However,the quantification of UHI’s response to urbanization is still challenging due to the lack of robust and continuous temperature and urbanization datasets and reliable quantification methods.This study proposed a framework to quantify the response of surface UHI(SUHI)to urban expansion using the annual temperate cycle model.We built a continuous annual SUHI series at the buffer level from 2003 to 2018 in the Jing-Jin-Ji region of China using MODIS land surface temperature and imperviousness derived from Landsat.We then investigated the spatiotemporal dynamic of SUHI under urban expansion and examined the underlying mechanism.Spatially,the largest SUHI interannual variations occurred in suburban areas compared to the urban center and rural areas.Temporally,the increase in SUHI under urban expansion was more significant in daytime compare to nighttime.We found that the seasonal variation of SUHI was largely affected by the seasonal variations of vegetation in rural areas and the interannual variation was mainly attributed to urban expansion in urban areas.Additionally,urban greening led to the decrease in summer daytime SHUI in central urban areas.These findings deepen the understanding of the long-term spatiotemporal dynamic of UHI and the quantitative relationship between UHI and urban expansion,providing a scientific basis for prediction and mitigation of UHI.Huidong Li Yuyu Zhou Gensuo Jia Kaiguang Zhao Jinwei Dong 2022Geoscience Frontiers2022,13,1:1

16The point-of-care-testing of nucleic acids by chip, cartridge and paper sensors显示文摘Point-of-care nucleic acid testing(POCNAT) has played an important role in the outbreak of infectious diseases(e.g., COVID-19) over recent years. POCNAT aims to realize the rapid, simple and automatic detection of nucleic acid. Thanks to the development of manufacturing technology, electronic information technology, artificial intelligence technology, and biological information technology in recent years, the development of the POCNAT device has led to significant advancement. Instead of the normal nucleic acid detection methods used in the laboratory, some novel experimental carriers have been applied, such as chips, cartridges and papers. The application of these experimental carriers has realized the automation and integration of nucleic acid detection. The entire process of nucleic acid detection is normally divided into three steps(nucleic acid extraction, target amplification and signal detection). All of the reagents required by the process can be pre-stored on these experimental carriers, without unnecessary manual operation. Furthermore, all of the processes are carried out in this experimental carrier, with the assistance of a specific control device. Although they are complicated to manufacture and precise in design,their application provides a significant step forwards in nucleic acid detection and realizes the integration of nucleic acid detection. This technology has great potential in the field of point-of-care molecular diagnostics in the future. 19Corrosion behavior,antibacterial ability,and osteogenic activity of Zn-incorporated Ni-Ti-O nanopore layers on NiTi alloy显示文摘Development of bone fixation devices with excellent corrosion resistance,antibacterial ability,and osteogenic activity is critical for promoting fracture healing.In this study,Zn-incorporated nanopore(NP)layers were prepared on the NiTi alloy through anodization and hydrothermal treatment.Results show that Zn can be evenly incorporated into the NP layers in the form of ZnTiO_(2).The Zn-incorporated samples exhibit good corrosion resistance and significantly reduce Ni^(2+)release.Meanwhile,the samples can continuously release Zn^(2+),which is responsible for excellent long-term antibacterial ability.Furthermore,the synergetic effect of Zn^(2+) release and nanoporous structure of the NP layers endues the NiTi alloy excellent osteogenic activity,as verified by upregulated alkaline phosphatase activity,secretion of type I collagen,and extracellular matrix mineralization.Therefore,Zn-incorporated Ni-Ti-O NP layers have great potential as biomedical coatings of NiTi-based implant materials.Yonghua Sun Yuyu Zhao He Zhang Youjie Rong Runhua Yao Yi Zhang Xiaohong Yao Ruiqiang Hang 2022Journal of Materials Science & Technology2022,,2:1
