2Diffusion tensor imaging assesses white matter injury in neonates with hypoxic-ischemic encephalopathy显示文摘With improvements in care of at-risk neonates, more and more children survive. This makes it increasingly important to assess, soon after birth, the prognosis of children with hypoxic-ischemic encephalopathy. Computed tomography, ultrasound, and conventional magnetic resonance imaging are helpful to diagnose brain injury, but cannot quantify white matter damage. In this study, ten full-term infants without brain injury and twenty-two full-term neonates with hypoxic-ischemic encephalopathy(14 moderate cases and 8 severe cases) underwent diffusion tensor imaging to assess its feasibility in evaluating white matter damage in this condition. Results demonstrated that fractional anisotropy, voxel volume, and number of fiber bundles were different in some brain areas between infants with brain injury and those without brain injury. The correlation between fractional anisotropy values and neonatal behavioral neurological assessment scores was closest in the posterior limbs of the internal capsule. 3Traditional Chinese medicine integrated with chemotherapy for stage Ⅳ non-surgical gastric cancer: a retrospective clinical analysis显示文摘OBJECTIVE: Traditional Chinese medicine(TCM) is regarded as an important treatment for gastric cancer patients, especially for those in advanced stage. To evaluate the effects of TCM treatment on gastric cancer patients, the authors performed a retrospective study to report the result of the integrated treatment of TCM with chemotherapy for stage IV non-surgical gastric cancer.METHODS: In this study, 182 patients with stage IV and non-surgical gastric cancer were retrospectively analyzed to evaluate the effects of TCM integrated with chemotherapy. Among the 182 cases, 88 cases received integrated therapy consisting of TCM and chemotherapy, while 94 cases received chemotherapy alone. The overall survival and Karnofsky performance status(KPS) score were measured as the main outcome.RESULTS: The median overall survival of the integrated therapy group and chemotherapy group were 16.9 and 10.5 months, respectively. The 1-, 3-and 5-year survival rates of integrated therapy group vs. chemotherapy group were 70% vs. 32%, 18% vs. 4%, and 11% vs. 0%, respectively. There was a significant difference between the two groups(χ2 = 42.244, P < 0.001). After six-month treatment, KPS scores of the integrated therapy group and the chemotherapy group were 75.00 ± 14.78 and 60.64 ± 21.39, respectively(P < 0.001). The Cox regression analysis showed that TCM treatment is a protective factor for patients' overall survival. 4Salt-responsive genes in rice revealed by cDNA microarray analysis显示文摘We used cDNA microarrays containing ~9,000 unigenes to identify 486 salt responsive expressed sequence tags (ESTs) (representing ~450 unigenes) in shoots of the highly salt-tolerant rice variety, Nona Bokra (Oryza sativa L. Ssp.Indica pv. Nona). Some of the genes identified in this study had previously been associated with salt stress. Howeverthe majority were novel, indicating that there is a great number of genes that are induced by salt exposure. Analysis of the salt stress expression profile data of Nona provided clues regarding some putative cellular and molecular processes that are undertaken by this tolerant rice variety in response to salt stress. Namely, we found that multiple transcription factors were induced during the initial salt response of shoots. Many genes whose encoded proteins are implicated in detoxification, protectant and transport were rapidly induced. Genes supporting photosynthesis were repressed and those supporting carbohydrate metabolism were altered. Commonality among the genes induced by salt exposure with those induced during senescence and biotic stress responses suggests that there are shared signaling pathways among these processes. We further compared the transcriptome changes of the salt-sensitive cultivar, IR28, with that of Nona rice. Many genes that are salt responsive in Nona were found to be differentially regulated in IR28. 5Blocking FSH inhibits hepatic cholesterol biosynthesis and reduces serum cholesterol显示文摘Menopause is associated with dyslipidemia and an increased risk of cardio-cerebrovascular disease.The classic view assumes that the underlying mechanism of dyslipidemia is attributed to an insufficiency of estrogen.In addition to a decrease in estrogen, circulating follicle-stimulating hormone (FSH)levels become elevated at menopause.In this study,we find that blocking FSH reduces serum cholesterol via inhibiting hepatic cholesterol biosynthesis.First,epidemiological results show that the serum FSH levels are positively correlated with the serum total cholesterol levels,even after adjustment by considering the effects of serum estrogen,in addition,the prevalence of hypercholesterolemia is significantly higher in peri-menopausal women than that in premenopausal women.Furthermore,we generated a mouse model of FSH elevation by intraperitoneally injecting exogenous FSH into ovariectomized (OVX)mice,in which a normal level of estrogen (E2)was maintained by exogenous supplementation. Consistently,the results indicate that FSH,independent of estrogen,increases the serum cholesterol level in this mouse model. 6Antioxidant and Anti-inflammatory Capacity of Ferulic Acid Released from Wheat Bran by Solid-state Fermentation of Aspergillus niger显示文摘Objective A strain of Aspergillus niger(A. niger), capable of releasing bound phenolic acids from wheat bran, was isolated. This strain was identified by gene sequence identification. The antioxidant and anti-inflammatory capacity of ferulic acid released from wheat bran by this A. niger strain(FA-WB) were evaluated. Methods Molecular identification techniques based on PCR analysis of specific genomic sequences were conducted; antioxidant ability was examined using oxygen radical absorbance capacity(ORAC), cellular antioxidant activity(CAA) assays, and erythrocyte hemolysis assays. RAW264.7 cells were used as a model to detect anti-inflammatory activity. Results The filamentous fungal isolate was identified to be A. niger. ORAC and CAA assay showed that FA-WB had better antioxidant activity than that of the ferulic acid standard. The erythrocyte hemolysis assay results suggested that FA-WB could attenuate AAPH-induced oxidative stress through inhibition of reactive oxy gen species(ROS) generation. FA-WB could significantly restore the AAPH-induced increase in intracellular antioxidant enzyme activities to normal levels as well as inhibit the intracellular malondialdehyde formation. TNF-?, IL-6, and NO levels indicated that FA-WB can inhibit the inflammation induced by lipopolysaccharide(LPS). 8Two-dimensional nanomaterials: fascinating materials in biomedical field显示文摘Due to their high anisotropy and chemical functions,two-dimensional(2D)nanomaterials have attracted increasing interest and attention from various scientific fields,including functional electronics,catalysis,supercapacitors,batteries and energy materials.In the biomedical field,2D nanomaterials have made significant contributions to the field of nanomedicine,especially in drug/gene delivery systems,multimodal imaging,biosensing,antimicrobial agents and tissue engineering.2D nanomaterials such as graphene/graphene oxide(GO)/reduced graphene oxide(r GO),silicate clays,layered double hydroxides(LDHs),transition metal dichalcogenides(TMDs),transition metal oxides(TMOs),black phosphorus(BP),graphitic carbon nitride(g-C3N4),hexagonal boron nitride(h-BN),antimonene(AM),boron nanosheets(B NSs)and tin telluride nanosheets(Sn Te NSs)possess excellent physical,chemical,optical and biological properties due to their uniform shapes,high surface-to-volume ratios and surface charge.In this review,we first introduce the properties,structures and synthetic strategies of different configurations of 2D nanomaterials.Recent advances and paradigms of 2D nanomaterials in a variety of biomedical applications,ranging from drug delivery,cancer treatment,bioimaging and tissue engineering to biosensing are discussed afterwards.In the final part,we foresee the development prospects and challenges of 2D nanomaterials after summarizing the research status of ultrathin 2D nanomaterials.Tingting Hu Xuan Mei Yingjie Wang Xisheng Weng Ruizheng Liang Min Wei 2019Science Bulletin2019,64,22:12
9Risk factors for hepatic decompensation in patients with primary biliary cirrhosis显示文摘AIM:To examine the clinical features and analyze prognostic factors in a prospective study of primary biliary cirrhosis(PBC) patients.METHODS:From 1995 to 2010,PBC patients without hepatic decompensation seen at the Peking Union Medical College Hospital were enrolled.Clinical signs and manifestations(pruritus,persistent fatigue,jaundice and pain in the right hypochondrium),laboratory parameters(auto-antibodies for autoimmune hepatic disease,biliary and hepatic enzymes,immunoglobulin,bilirubin,and albumin) and imaging findings were recorded at entry and at specific time points during follow-up.Cox regression and Kaplan-Meier analyses,respectively,assessed the risk factors for hepatic decompensation and survival.RESULTS:Two hundred and sixty-two PBC patients were enrolled with a median follow-up of 75.2 mo(range,21-201 mo).The 240 patients were aged 51.5 ± 10.2 years at diagnosis and 91.6% were female.Two hundred and forty-five(93.5%) were seropositive for anti-mitochondrial antibodies.At presentation,170 patients(64.9%) were symptomatic,while 96 patients(36.6%) had extra-hepatic autoimmune disease.During the follow-up period,62(23.7%) patients developed hepatic decompensation of whom four underwent liver transplantation and 17 died.The cumulative survival rate and median survival time were 83.9% and 181.7 mo,respectively.Cox regression analysis revealed that an incomplete ursodeoxycholic acid(UDCA) response or inconsistent treatment [P < 0.001;hazard risk(HR) 95%CI = 2.423-7.541],anti-centromere antibodies(ACA) positivity(P < 0.001;HR 95%CI = 2.516-7.137),alanine aminotransferase ratio(AAR) elevations(P < 0.001;HR 95%CI = 1.357-2.678),and histological advanced liver disease(P = 0.006;HR 95%CI = 1.481-10.847) were predictors of hepatic decompensation.The clinical features and survival of PBC in China are consistent with those described in Western countries.CONCLUSION:Incomplete UDCA response or inconsistent treatment,ACA positivity,AAR elevations,and advanced histological stage are predictors of decompensation.Tian-Yan Shi Li-Na Zhang Hua Chen Li Wang Min Shen Xuan Zhang Feng-Chun Zhang 2013World Journal of Gastroenterology2013,19,7:9
10Numerical simulation of topography effects on the “00.7” severe rainfall in Beijing显示文摘In an effort to study the severe rainfall event of 4—5th July 2000 in Beijing (with 24h accumulated precipitation of 240 mm), we perform numerical simulations to investigate this event using the MM5v3.6 model. The model is initialized with the MM5/WRF 3DVAR analysis, which incorporates the ground-based GPS precipitable water vapor, automatic and conventional meteorological observations in its assimilation step. For 24 h accumulated precipitation forecast, the threat scores are 0.72, 0.76, 0.67 and 0.63, for thresholds of 1, 5, 10, and 20 mm, respectively. Holding other factors unchanged, sensitivity experiments were conducted with different topographic resolutions of 110, 50 and 3.7 km to investigate the topographic effects on precipitation over the Beijing area. In these sensitivity experiments, we attempt to preserve the realistic orographic distribution in the model topography under the condition of keeping the dynamic-thermodynamic consistency of the initial model atmosphere to the extent possible. 11Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. 12Application of deep learning in ecological resource research:Theories, methods, and challenges显示文摘Ecological resources are an important material foundation for the survival,development,and self-realization of human beings.In-depth and comprehensive research and understanding of ecological resources are beneficial for the sustainable development of human society.Advances in observation technology have improved the ability to acquire long-term,cross-scale,massive,heterogeneous,and multi-source data.Ecological resource research is entering a new era driven by big data.Traditional statistical learning and machine learning algorithms have problems with saturation in dealing with big data.Deep learning is a method for automatically extracting complex high-dimensional nonlinear features,which is increasingly used for scientific and industrial data processing because of its ability to avoid saturation with big data.To promote the application of deep learning in the field of ecological resource research,here,we first introduce the relationship between deep learning theory and research on ecological resources,common tools,and datasets.Second,applications of deep learning in classification and recognition,detection and localization,semantic segmentation,instance segmentation,and graph neural network in typical spatial discrete data are presented through three cases:species classification,crop breeding,and vegetation mapping.Finally,challenges and opportunities for the application of deep learning in ecological resource research in the era of big data are summarized by considering the characteristics of ecological resource data and the development status of deep learning.It is anticipated that the cooperation and training of cross-disciplinary talents may promote the standardization and sharing of ecological resource data,improve the universality and interpretability of algorithms,and enrich applications with the development of hardware.Qinghua GUO Shichao JIN Min LI Qiuli YANG Kexin XU Yuanzhen JU Jing ZHANG Jing XUAN Jin LIU Yanjun SU Qiang XU Yu LIU 2020Science China Earth Sciences2020,63,10:7
13Agglutinin isolated from Arisema heterophyllum Blume induces apoptosis and autophagy in A549 cells through inhibiting PI3K/Akt pathway and inducing ER stress显示文摘Arisaema heterophyllum Blume is one of the three medicinal plants known as traditional Chinese medicine Rhizoma Arisaematis(RA). RA has been popularly used to treat patients with convulsions, inflammation, and cancer for a long time. However, the underlying mechanisms for RA effects are still unclear. The present study was designed to determine the cytotoxicity of agglutinin isolated from Arisema heterophyllum Blume(AHA) and explore the possible mechanisms in human non-small-cell lung cancer A549 cells. AHA with purity up to 95% was isolated and purified from Arisaema heterophyllum Blume using hydrophobic interaction chromatography. AHA dose-dependently inhibited the proliferation of A549 cells and induced G_1 phase cell cycle arrest. AHA induced apoptosis by up-regulating pro-apoptotic Bax, decreasing anti-apoptotic Bcl-2, and activating caspase-9 and caspase-3. In A549 cells treated with AHA, the PI3K/Akt pathway was inhibited. Furthermore, AHA induced increase in the levels of ER stress markers such as phosphorylated eukaryotic initiation factor 2α(p-eIF2α), C/EBP-homologous protein(CHOP), inositol-requiring enzyme 1α(IRE1α), and phosphorylated c-Jun NH_2-terminal kinase(p-JNK). AHA also induced autophagy in A549 cells. Staining of acidic vesicular organelles(AVOs) and increase in the levels of LC3II and ATG7 were observed in AHA-treated cells. These findings suggested that AHA might be one of the active components with anti-cancer effects in Arisaema heterophyllum Blume. 14A high-fiber diet synergizes with Prevotella copri and exacerbates rheumatoid arthritis显示文摘Both preclinical and established rheumatoid arthritis(RA)patients display alterations in the gut microbiome.Prevotella spp.are preferentially enriched in a subset of RA patients.Here,we isolated a Prevotella strain,P.copri RA,from the feces of RA patients and showed that colonization of P.copri RA exacerbated arthritis in a collagen-induced arthritis(CIA)model.With the presence of P.copri RA colonization,a high-fiber diet exacerbated arthritis via microbial alterations and intestinal inflammation.Colonization of P.copri together with a high-fiber diet enabled the digestion of complex fiber,which led to the overproduction of organic acids,including fumarate,succinate and short-chain fatty acids.Succinate promoted proinflammatory responses in macrophages,and supplementation with succinate exacerbated arthritis in the CIA model.Our findings highlight the importance of dysbiosis when evaluating the effects of dietary interventions on RA pathogenesis and provide new insight into dietary interventions or microbiome modifications to improve RA management.Lingjuan Jiang Mengmeng Shang Shengnan Yu Yudong Liu Hui Zhang Yangzhong Zhou Min Wang Tingting Wang Hui Li Zhihua Liu Xuan Zhang 2022Cellular & Molecular Immunology2022,19,12:6
15Proteomic and bioinformatic analyses of possible targetrelated proteins of gambogic acid in human breast carcinoma MDA-MB-231 cells显示文摘Gambogic acid(GA) is an anticancer agent in phase Ⅱb clinical trial in China but its mechanism of action has not been fully clarified. The present study was designed to search the possible target-related proteins of GA in cancer cells using proteomic method and establish possible network using bioinformatic analysis. Cytotoxicity and anti-migration effects of GA in MDA-MB-231 cells were checked using MTT assay, flow cytometry, wound migration assay, and chamber migration assay. Possible target-related proteins of GA at early(3 h) and late stage(24 h) of treatment were searched using a proteomic technology, two-dimensional electrophoresis(2-DE). The possible network of GA was established using bioinformatic analysis. The intracellular expression levels of vimentin, keratin 18, and calumenin were determined using Western blotting. GA inhibited cell proliferation and induced cell cycle arrest at G2/M phase and apoptosis in MDA-MB-231 cells. Additionally, GA exhibited anti-migration effects at non-toxic doses. In 2-DE analysis, totally 23 possible GA targeted proteins were found, including those with functions in cytoskeleton and transport, regulation of redox state, metabolism, ubiquitin-proteasome system, transcription and translation, protein transport and modification, and cytokine. Network analysis of these proteins suggested that cytoskeleton-related proteins might play important roles in the effects of GA. Results of Western blotting confirmed the cleavage of vimentin, increase in keratin 18, and decrease in calumenin levels in GA-treated cells. 16Glutamate Impairs Mitochondria Aerobic Respiration Capacity and Enhances Glycolysis in Cultured Rat Astrocytes显示文摘Objective To study the effect of glutamate on metabolism, shifts in glycolysis and lactate release in rat astrocytes. Methods After 10 days, secondary cultured astrocytes were treated with 1 mmol/L glutamate for 1 h, and the oxygen consumption rates(OCR) and extra cellular acidification rate(ECAR) was analyzed using a Seahorse XF 24 Extracellular Flux Analyzer. Cell viability was then evaluated by MTT assay. Moreover, changes in extracellular lactate concentration induced by glutamate were tested with a lactate detection kit. Results Compared with the control group, treatment with 1 mmol/L glutamate decreased the astrocytes' maximal respiration and spare respiratory capacity but increased their glycolytic capacity and glycolytic reserve. Further analysis found that 1-h treatment with different concentrations of glutamate(0.1-1 mmol/L) increased lactate release from astrocytes, however the cell viability was not affected by the glutamate treatment. 18Longitudinal Alterations of Local Spontaneous Brain Activity in Parkinson's Disease显示文摘We used resting-state fMRI to evaluate longitudinal alterations in local spontaneous brain activity in Parkinson's disease(PD) over a 2-year period. Data were acquired from 23 PD patients at baseline and follow-up,and 27 age-and sex-matched normal controls. Regional homogeneity(ReHo) and voxel-based-morphometry(VBM) were used to identify differences in local spontaneous brain activity and grey matter volume. With disease progression, we observed a progressive decrease in ReHo in the sensorimotor cortex, default-mode network, and left cerebellum, but increased ReHo in the supplementary motor area, bilateral temporal gyrus, and hippocampus.Moreover, there was a significant positive correlation between the rates of ReHo change in the left cerebellum and the rates of change in the Unified Parkinson's Disease Rating Scale-III scores. VBM revealed no significant differences in the grey matter volume among the three sets of acquisitions.
