1Region-Specific Iron Measured by MRI as a Biomarker for Parkinson's Disease显示文摘The identification of sensitive and specific biomarkers for Parkinson's disease(PD) poses an important clinical challenge. A potential biomarker for early diagnosis and disease monitoring of PD is region-specific iron. Iron accumulation in the substantia nigra pars compacta is considered a main characteristic of PD.However, questions remain, such as the relationship between nigral iron and clinical indices of PD(motor impairment or disease duration). Further, previous studies have suggested the influence of iron on other nuclei. Iron quantification using magnetic resonance imaging(MRI)allows for studies of the relationship between regional iron and clinical symptoms in vivo. 2Geochemistry and Zircon U–Pb age of the Yao'an pseudoleucite porphyry,Yunnan Province,China显示文摘The Yao'an Pb–Ag deposit, located in the Chuxiong Basin, western Yangtze Block, is an important component of the Jinshajiang–Ailaoshan alkaline porphyry–related polymetallic intrusive belt. This complex suite of rock bodies includes a vein of pseudoleucite porphyry within deposits of syenite porphyry and trachyte.The pseudoleucite is characterized by a variable greyish,greyish-white, and greyish-green porphyritic texture. Phenocrysts are mainly pseudoleucite with small amounts of alkali feldspar and biotite. In an intense event, leucite phenocrysts altered to orthoclase, kaolinite, and quartz.Both the pseudoleucite porphyry and the syenite porphyry samples were typical alkali-rich, K-rich, al-rich rocks with high LaN/YbNratios; enriched in light rare earth elements and large-ion lithophile elements, and depleted in high field strength elements; and with strongly negative Ta, Nb, and Ti(TNT) anomalies and slightly negative Eu anomalies—all characteristics of subduction-zone mantle-derived rock.We obtained a LA-ICP-MS zircon U–Pb age of 34.1 ± 0.3 Ma(MSWD = 2.4), which is younger than the established age of the Indian and Eurasian Plate collision.The magma derived from a Type-II enriched mantle formed in a post-collisional plate tectonic setting. 3Mesoscale simulation of discontinuous dynamic recrystallization using the cellular automaton method显示文摘A dynamic recrystallization (DRX) cellular automaton (CA) model that can mark the microstructure with DRX circle was developed. The effects of initial grain size on the stress-strain curve, mean grain size and DRX fraction were mainly investigated, and the simulated results were compared with those obtained from previous researches. The results show that the shape of the stress-strain curve is sensitive, while the stress and mean grain size at the steady state are insensitive to the initial grain size. 4Network pharmacological prediction and molecular docking analysis of the combination of Atractylodes macrocephala Koidz.and Paeonia lactiflora Pall.in the treatment of functional constipation and its verification显示文摘Background:We aimed to reveal the mechanism of functional constipation in the treatment of Atractylodes macrocephala Koidz.(AMK)and Paeonia lactiflora Pall.(PLP).Methods:The main active ingredients of AMK and PLP were screened by the Traditional Chinese Medicine Systems Pharmacology(TCMSP)platform.A database of functional constipation targets was established by GeneCard and OMIM.An“ingredient-target”network map was constructed with Cytoscape software(version 3.7.1),and molecular docking analysis was performed on the components and genes with the highest scores.The rats in the normal group were given saline,and those in the other groups were given 10 mg/kg diphenoxylate once a day for 14 days.The serum and intestinal tissue levels of adenosine monophosphate(cAMP),protein kinase A(PKA),and adenylyl cyclase(AC)of the rats and aquaporin(AQP)1,AQP3,and AQP8 were measured.Results:AMK and PLP had a significant role in the regulation of targets in the treatment of functional constipation.After treatment with AMK,PLP,or mosapride,the serum and intestinal tissue levels of AC,cAMP,and PKA were significantly downregulated.Groups receiving AMK and PLP or mosapride exhibited a reduction in the level of AQP1,AQP3,and AQP8 to varying degrees.Conclusion:Molecular docking analysis revealed that AMK and PLP had a significant role in the regulation of targets in the treatment of functional constipation.Studies have confirmed that AMK and PLP can also affect AC,cAMP,and PKA.AC,cAMP,and PKA in model rats were significantly downregulated.AQP expression is closely related to AC,cAMP,and PKA.AMK and PLP can reduce the expression of AQP1,AQP3,and AQP9 in the colon of constipated rats.Yuxiao Meng Xiaojun Li Xiaoting Wang Lu Zhang Jiaqi Guan 2022Animal Models and Experimental Medicine2022,5,2:4
5Longitudinal Alterations of Local Spontaneous Brain Activity in Parkinson's Disease显示文摘We used resting-state fMRI to evaluate longitudinal alterations in local spontaneous brain activity in Parkinson's disease(PD) over a 2-year period. Data were acquired from 23 PD patients at baseline and follow-up,and 27 age-and sex-matched normal controls. Regional homogeneity(ReHo) and voxel-based-morphometry(VBM) were used to identify differences in local spontaneous brain activity and grey matter volume. With disease progression, we observed a progressive decrease in ReHo in the sensorimotor cortex, default-mode network, and left cerebellum, but increased ReHo in the supplementary motor area, bilateral temporal gyrus, and hippocampus.Moreover, there was a significant positive correlation between the rates of ReHo change in the left cerebellum and the rates of change in the Unified Parkinson's Disease Rating Scale-III scores. VBM revealed no significant differences in the grey matter volume among the three sets of acquisitions. 6Ethyl acetate fraction in ethanol extract from root of 'Dai-Bai-Jie' (Marsdenia tenacissima):anti-tumor activity in A549cancer cells显示文摘OBJECTIVE: To evaluate the anti-tumor activity of ethyl acetate fraction(EFA), extracted with ethanol from the root of 'Dai-Bai-Jie' in A549 cancer cells and its underlying mechanism.METHODS: 'Dai-Bai-Jie' was extracted with 95%ethanol-aqueous(DBJ-1), 50% ethanol-aqueous(DBJ-2), and water(DBJ-3) by reflux method. 95%ethanol-aqueous extract was separated byethyl acetate(EFA) and n-butyl alcohol(DBJ-5), consecutively. The SRB method was used to evaluate the cytotoxic activity. Annexin V-FITC staining was applied to observe the apoptosis and analyze the cell cycle activated by EFA in A549 tumor cell. Western blot was used to detect the apoptosis/related pro-teins expressions. A549 tumor cellsbearing nude mice model was employed to measure the tumor volume, mice weight, and tumor inhibition ratio in order to verify the antitumor activity in vivo.RESULTS: DBJ-1 and EFA showed better cytotoxic activity on A549 tumor cells with IC5025 and 3.5 μg/mL, respectively. EFA can exhibit the proliferation,arrest cell cycle at G0/G1 phase, and induce apoptosis in A549 tumor cells in vitro. The mechanisms of apoptosis induced by EFA may be associated with decreasing Bcl-2 protein expression and increasing p53, Bax, Caspase-3, and Caspase-8 proteins expression. EFA also possessed significant anti-tumor efficacy in nude mice, and little toxicity was observed in the host.CONCLUSION: EAF could induce A549 tumor cells apoptosis and G0/G1 cell cycle arrest. 13Effectiveness of E-SBIRT intervention in community healthcare institution in China:study proposal for a randomised controlled trial显示文摘Background Many studies have demonstrated the effectiveness of Screening,Brief Intervention and Referral to Treatment(SBIRT)in addressing substance use problem.However,owing to the shortage of counsellors,it has not been widely used in China.With the development of smart medicine,we developed a web-based electronic SBIRT(E-SBIRT)program and explored the effectiveness of E-SBIRT in reducing substance use in China.Methods A randomised controlled trial will be conducted in primary healthcare institutions.Four primary healthcare institutions will be selected and randomly divided into an intervention group and a control group(each institution will recruit 60 participants,and in total,240 participants will be recruited).The control group will get a pamphlet of drug abuse prevention,and the intervention group will get the E-SBIRT intervention and the pamphlet.Both groups will receive baseline and follow-up assessment at 1 and 3 months after the intervention.The primary outcome is the change in scores on the Alcohol,Smoking and Substance Use Involvement Screening Test,and the secondary outcomes include changes in motivation,depression,anxiety,positive/negative emotion,self-esteem,addiction knowledge and addiction severity index.Conclusions If the‘E-SBIRT’program is found to be effective,it will be an accessible,affordable and widely implementable intervention to help participants at moderate risk of substance use to reduce their consumption.The potential benefit is to provide early intervention to high-risk patients in time and reduce the harmful consequences to individuals and society.Rao Fu Chenxin Yuan Wei Sun Wenzheng Wang Lei Zhang Jing Zhai Qun Guan Xiaojun Wu Jiang Long Min Zhao Jiang Du 2021General Psychiatry2021,34,5:1

15A shape-persistent plasticine-like conductive hydrogel with self-healing properties for peripheral nerve regeneration显示文摘In recent years,electrically conductive hydrogel-based nerve guidance conduits(NGCs)have yielded promising results for treating peripheral nerve injuries(PNIs).However,developed ones are generally pre-manufactured and exhibit a limited ability to achieve good contact with nerve tissue with irregu-lar surfaces.Herein,we developed a plasticine-like electrically conductive hydrogel consisting of gelatin,conducting polypyrrole,and tannic acid(named GPT)and assessed its ability to promote peripheral nerve regeneration.The shape-persistent GPT hydrogel exhibited good self-healing properties and could easily be molded to form a conduit that could match any injured nerve tissue.Their electrical properties could be tuned by changing the PPy concentration.In vitro,the improved conductivity of the hydrogel pro-moted dorsal root ganglion(DRG)axonal extension.More importantly,we found that the GPT hydrogel enhanced axonal regeneration and remyelination in vivo,preventing denervation atrophy and enhancing functional recovery in a mice model of sciatic nerve injury.These results suggest that our plasticine-like NGC has huge prospects for clinical application in the repair of PNI.Xinchang Kang Xiaojun Li Can Liu Min Cai Pengfei Guan Yian Luo Youjun Guan Yu Tian Kunyu Ren Chengyun Ning Lei Fan Guoxin Tan Lei Zhou 2023Journal of Materials Science & Technology2023,,11:0

17Emulsifying property,antioxidant activity,and bitterness of soybean protein isolate hydrolysate obtained by Corolase PP under high hydrostatic pressure显示文摘Enzymatic hydrolysis of proteins can enhance their emulsifying properties and antioxidant activities.However,the problem related to the hydrolysis of proteins was the generation of the bitter taste.Recently,high hydrostatic pressure(HHP)treatment has attracted much interest and has been used in several studies on protein modification.Hence,the study aimed to investigate the effects of enzymatic hydrolysis by Corolase PP under different pressure treatments(0.1,100,200,and 300 MPa for 1-5 h at 50℃)on the emulsifying property,antioxidant activity,and bitterness of soybean protein isolate hydrolysate(SPIH).As observed,the hydrolysate obtained at 200 MPa for 4 h had the highest emulsifying activity index(47.49 m^(2)/g)and emulsifying stability index(92.98%),and it had higher antioxidant activities(44.77%DPPH free radical scavenging activity,31.12%superoxide anion radical scavenging activity,and 61.50%copper ion chelating activity).At the same time,the enhancement of emulsion stability was related to the increase of zeta potential and the decrease of mean particle size.In addition,the hydrolysate obtained at 200 MPa for 4 h had a lower bitterness value and showed better palatability.This study has a broad application prospect in developing food ingredients and healthy foods.Haining Guan Chunmei Feng Min Ren Xiaojun Xu Dengyong Liu Xiaoqin Diao 2024Food Science and Human Wellness2024,13,3:0

19Baizhu-Baishao herb pair ameliorates functional constipation and intestinal microflora disorder in rats显示文摘Background:In China,Rhizoma atractylodis macrocephalae-Paeonia lactiflora Pall(Biazhu-Baishao,BZBS)is a classic herb pair used to treat intestinal stress syndrome,ulcerative colitis and other diseases.However,the mechanism of BZBS in the treatment of functional constipation(FC)has been little studied and remains unclear.In this study,a behavioral investigation,colon tissue morphology,enzyme-linked immunosorbent assay(Elisa)and intestinal microflora analysis have been used to illuminate the potential mechanism of the effects of BZBS on FC in a rat model.Methods:A FC rat model was constructed and BZBS was given as treatment.Observations and recordings were made of the fecal moisture content,the defecation time of the first black stool,and the rate of intestinal propulsion.Elisa was used to detect the expression levels of substance P(SP),vasoactive intestinal peptide(VIP),5-hydroxytryptamine(5-HT)in the colon.To ascertain the composition of the microbial community,a high throughput 16S ribosomal RNA(16S r RNA)gene sequencing technique was employed.Results:Oral administration of BZBS significantly ameliorated several key excretion parameters,including the time to first black stool defecation,stool water content,and the propulsion rate in the small intestine in FC rats.It increased the expression of SP,VIP and 5-HT in the colon.16S r RNA gene sequencing results showed that BZBS changed the microbial community structure,decreased the Bacteroidetes/Firmicutes ratio,increased the relative abundance of Blautia and Fusicatenibacter,and decreased the relative abundance of Ruminococcus and Roseburia.Conclusions:BZBS effectively alleviates FC and improves dysbacteriosis.Xiaojun Li Xiaoting Wang Ziyan Wang Jiaqi Guan 2023Animal Models and Experimental Medicine2023,6,6:0
20Oscillation-specific nodal alterations in early to middle stages Parkinson’s disease显示文摘Background:Different oscillations of brain networks could carry different dimensions of brain integration.We aimed to investigate oscillation-specific nodal alterations in patients with Parkinson’s disease(PD)across early stage to middle stage by using graph theory-based analysis.Methods:Eighty-eight PD patients including 39 PD patients in the early stage(EPD)and 49 patients in the middle stage(MPD)and 36 controls were recruited in the present study.Graph theory-based network analyses from three oscillation frequencies(slow-5:0.01–0.027 Hz;slow-4:0.027–0.073 Hz;slow-3:0.073–0.198 Hz)were analyzed.Nodal metrics(e.g.nodal degree centrality,betweenness centrality and nodal efficiency)were calculated.Results:Our results showed that(1)a divergent effect of oscillation frequencies on nodal metrics,especially on nodal degree centrality and nodal efficiency,that the anteroventral neocortex and subcortex had high nodal metrics within low oscillation frequencies while the posterolateral neocortex had high values within the relative high oscillation frequency was observed,which visually showed that network was perturbed in PD;(2)PD patients in early stage relatively preserved nodal properties while MPD patients showed widespread abnormalities,which was consistently detected within all three oscillation frequencies;(3)the involvement of basal ganglia could be specifically observed within slow-5 oscillation frequency in MPD patients;(4)logistic regression and receiver operating characteristic curve analyses demonstrated that some of those oscillation-specific nodal alterations had the ability to well discriminate PD patients from controls or MPD from EPD patients at the individual level;(5)occipital disruption within high frequency(slow-3)made a significant influence on motor impairment which was dominated by akinesia and rigidity.Conclusions:Coupling various oscillations could provide potentially useful information for large-scale network and progressive oscillation-specific nodal alterations were observed in PD patients across early to middle stages.Xiaojun Guan Tao Guo Qiaoling Zeng Jiaqiu Wang Cheng Zhou Chunlei Liu Hongjiang Wei Yuyao Zhang Min Xuan Quanquan Gu Xiaojun Xu Peiyu Huang Jiali Pu Baorong Zhang Min-Ming Zhang 2019Translational Neurodegeneration2019,8,1:0
