1The acidic tumor microenvironment: a target for smart cancer nano-theranostics显示文摘The acidic tumor microenvironment(TME), which mainly results from the high glycolytic rate of tumor cells, has been characterized as a hallmark of solid tumors and found to be a pivotal factor participating in tumor progression. Recently, due to the increasing understanding of the acidic TME, it has been shown that the acidic TME could be utilized as a multifaceted target during the design of various pH-responsive nanoscale theranostic platforms for the precise diagnosis and effective treatment of cancers. In this article,we will give a focused overview on the latest progress in utilizing this characteristic acidic TME as the target of nano-theranostics to enable cancer-specific imaging and therapy. 2PIKA as an Adjuvant Enhances Specific Humoral and Cellular Immune Responses Following the Vaccination of Mice with HBsAg plus PIKA显示文摘An adjuvant is usually used to enhance the immune response induced by vaccines. The choice of adjuvant or immune enhancer determines the effectiveness of the immune response. Currently, aluminium (Alum, a generic term for salts of aluminium) is the only FDA-approved adjuvant. Alum predominantly induces the differentiation of Th2 cells and thus mediates an antibody immune response. Therefore, there is an urgent need for new adjuvants that enhance not only humoral but also cellular immune responses. In the present study, we demonstrates that PIKA (a stabilized dsRNA) as an adjuvant directly induces the activation and the proliferation of both B and NK cells in vitro. Injection of PIKA into mice results in the production of cytokines in vivo. In addition, the study demonstrates that PIKA promotes the maturation of bone marrow-derived dendritic cells (BMDCs) including up-regulation of the co-stimulatory molecules CD80, CD86 and CD40, and the induction of cytokines such as IL-12p70, IL-12p40 and IL-6. Importantly, after immunization of mice with HBsAg plus PIKA, the presence of PIKA enhances the titers of HBsAg-specific IgG and HBsAg-specific IFN-γ production. These results demonstrate that PIKA as an adjuvant can promote both humoral and cellular immune responses. 3Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 4The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 6Two-dimensional metal-organic-framework as a unique theranostic nano-platform for nuclear imaging and chemo-photodynamic cancer therapy显示文摘Nanoscale metal organic frameworks (NMOFs) with porous structure and inherent biodegradability are attractive nanomedicine platforms.In addition to conventional particulate NMOFs,two-dimensional (2D) NMOFs are emerging as a unique type of NMOFs which however have been relatively less explored for nanomedicine applications.Herein,2D NMOFs composed of Zn2+ and tetrakis(4-carboxyphenyl) porphyrin (TCPP) are fabricated and functionalized with polyethylene glycol (PEG).Compared to their particulate counterpart,such 2D NMOFs show greatly increased drug loading capacity and enhanced light-triggered singlet oxygen production,promising for chemotherapy and photodynamic therapy (PDT),respectively.Utilizing the porphyrin structure of TCPP,our 2D NMOFs could be labeled with a diagnostic radioisotope,99mTc,for single photon emission computer tomography (SPECT) imaging,which reveals efficient tumor homing of those 2D NMOFs upon intravenous injection.While offering a remarkable synergistic in vivo antitumor effect for the combined chemo-PDT,such 2D NMOFs show efficient biodegradation and rapid renal clearance.Our work presents the great promise of 2D NMOFs for nanomedicine applications.Wenjun Zhu Yu Yang Qiutong Jin Yu Chao Longlong Tian Jingjing Liu Ziliang Dong Zhuang Liu 2019Nano Research2019,12,6:9
7Nonlinear optical selection rule based on valley-exciton locking in monolayer ws2显示文摘Optical selection rules fundamentally determine the optical transitions between energy states in a variety of physical systems,from hydrogen atoms to bulk crystals such as gallium arsenide.These rules are important for optoelectronic applications such as lasers,energy-dispersive X-ray spectroscopy,and quantum computation.Recently,single-layer transition metal dichalcogenides have been found to exhibit valleys in momentum space with nontrivial Berry curvature and excitons with large binding energy.However,there has been little study of how the unique valley degree of freedom combined with the strong excitonic effect influences the nonlinear optical excitation.Here,we report the discovery of nonlinear optical selection rules in monolayer WS2,an important candidate for visible 2D optoelectronics because of its high quantum yield and large direct bandgap.We experimentally demonstrated this principle for second-harmonic generation and two-photon luminescence(TPL).Moreover,the circularly polarized TPL and the study of its dynamics evince a sub-ps interexciton relaxation(2p R 1s).The discovery of this new optical selection rule in a valleytronic 2D system not only considerably enhances knowledge in this area but also establishes a foundation for the control of optical transitions that will be crucial for valley optoelectronic device applications such as 2D valley-polarized THz sources with 2p–1s transitions,optical switches,and coherent control for quantum computing.Jun Xiao Ziliang Ye Ying Wang Hanyu Zhu Yuan Wang Xiang Zhang 2015Light(Science & Applications)2015,4,1:8
8The Low Energy X-ray telescope (LE) onboard the Insight-HXMT astronomy satellite显示文摘The Low Energy X-ray telescope(LE) is one of the three main instruments of the Insight-Hard X-ray Modulation Telescope(Insight-HXMT). It is equipped with Swept Charge Device(SCD) sensor arrays with a total geometrical area of 384 cm^2 and an energy band from 0.7 to 13 ke V. In order to evaluate the particle induced X-ray background and the cosmic X-ray background simultaneously, LE adopts collimators to define four types of Field Of Views(FOVs), i.e., 1.6°×6°, 4°×6°, 50°-60°×2°-6 oand the blocked ones which block the X-ray by an aluminum cover. LE is constituted of three detector boxes(LEDs) and an electric control box(LEB) and achieves a good energy resolution of 140 e V@5.9 ke V, an excellent time resolution of 0.98 ms, as well as an extremely low pileup(<1%@18000 cts/s). 9Genetic Types and Metallogenic Model for the Polymetallic Copper–Gold Deposits in the Tongling Ore District, Anhui Province, Eastern China显示文摘The Tongling ore district is one of the most economically important ore areas in the Middle–Lower Yangtze River Metallogenic Belt, eastern China. It contains hundreds of polymetallic copper–gold deposits and occurrences. Those deposits are mainly clustered(from west to east) within the Tongguanshan, Shizishan, Xinqiao, Fenghuangshan, and Shatanjiao orefields. Until recently, the majority of these deposits were thought to be skarn-or porphyry–skarn-type deposits; however there have been recent discoveries of numerous vein-type Au, Ag, and Pb-Zn deposits that do not fall into either of these categories. This indicates that there is some uncertainty over this classification. Here, we present the results of several systematic geological studies of representative deposits in the Tongling ore district. From investigation of the ore-controlling structures, lithology of the host rock, mineral assemblages, and the characteristics of the mineralization and alteration within these deposits, three genetic types of deposits(skarn-, porphyry-, and vein-type deposits) have been identified. The spatial and temporal relationships between the orebodies and Yanshanian intrusions combined with the sources of the ore-forming fluids and metals, as well as the geodynamic setting of this ore district, indicate that all three deposit types are genetically related each other and constitute a magmatic–hydrothermal system. This study outlines a model that relates the polymetallic copper–gold porphyry-, skarn-, and vein-type deposits within the Tongling ore district. 10Effect of a starch-based filter aid on the dewatering of fine clean coal显示文摘The dewatering of fine, flotation cleaned coals from Huaibei and Xuzhou (bituminous) and Yongcheng (anthracite) were studied. The supernatant and filter cake were examined to determine the rate and extent of flocculation and dewatering. A starch-based filter aid was used to increase flocculation and dewatering rates. The filtration constant, K, and compression index, s, of the Yongcheng slurry were measured under various conditions. A designed experiment was performed to determine optimum conditions for dewatering. The results showed that the filter aid enhanced flocculation and coagulation of the fine cleaned coal slurry, enhanced the structure of the filter cake and promoted dewatering of the cake. 12Frost heave and freezing processes of saturated rock with an open crack under different freezing conditions显示文摘Frost heave experiments on saturated sandstone and tuff with an open crack are conducted under uniform and unidirectional freezing conditions.Frost heave of crack in sandstone with high permeability is more significant under uniform freezing condition than that under unidirectional freezing condition.However,frost heave of crack in tuff with low permeability is more significant under unidirectional freezing condition.To illustrate the reasons for this phenomenon,a numerical model on the freezing processes of saturated rock with an open crack considering the latent heat of pore water and water in crack is proposed and confirmed to be reliable.Numerical results show that a frozen shell that blocks the migration of water in crack to rock develops first in the outer part of the rock before the freezing of water in crack under unifonn freezing condition.However,the migration path of water in crack to the unfrozen rock under freezing front exists under unidirectional freezing condition.The freezing process and permeability of rock together determine the migration of water in crack and lead to the different frost heave modes of crack for various permeable rocks under different freezing conditions.The frost heave modes of crack in rock with low or high permeability are similar under uniform freezing condition because water migration is blocked by a frozen shell and is irrelevant to rock permeability.For high permeability rock,the frost heave of crack will be weakened due to water migration under unidirectional freezing condition;however,the frost heave of crack would be more significant for low permeability rock because water migration is blocked under unidirectional freezing condition.Therefore,the freezing condition and rock permeability determine the frost heave of rock with crack together,and this should be concerned in cold regions engineering applications.Zhitao Lv Caichu XIA Yuesong WANG Ziliang LIN 2020Frontiers of Structural and Civil Engineering2020,14,4:5

14Preparation of monoclonal antibody based indirect competitive ELISA for detecting 19-nortestosterone residue显示文摘19-Nortestosterone (NT) has been illegally used in horse racing to boost physical performance, and in animal husbandry to accelerate weight gain. To monitor the abuse of NT, our goal was to develop a commercial enzyme linked immunosorbent assay (ELISA) kit. For this purpose, hybridomas were prepared by fusing NS0 mouse myeloma cells with splenocytes isolated from immunized BALB/c mouse. Noncompetitive and competitive indirect ELISA were used to screen positive cell clones. To optimize the indirect competitive ELISA (icELISA) method, various methanol concentrations in assay buffer were evaluated. Matrix effects in urine and spiking test were also investigated. Finally, five hybridoma cell lines named NT-1, NT-2, NT-3, NT-4 and NT-5 were screened out. The corresponding monoclonal antibodies (mAbs) were of the IgG 1 isotype with a k light chain, and the antibody affinity of all mAbs were between 2.6×10 9 and 4.7×10 9 L/mol. The titer and IC 50 values of purified ascites were in the range of 0.64×10 5 2.56×10 5 and 0.55-1.0 ng/mL, respectively. Based on the NT-1 hybridoma, a heterologous icELISA method was developed for the quantitative detection of NT in cattle urine. The dynamic range was from 0.004 to 85.8 ng/mL, with a detection limit for the assay and IC 50 values of 0.002 and 0.55 ng/mL, respectively. Except for a high cross-reactivity (62%) to α-NT, negligible cross-reactivity to other compounds was observed. After optimization, 10% of methanol was used in the assay buffer, and a 20-fold dilution in cattle urine gave an inhibition curve almost the same as that in phosphate buffered saline. The correlation coefficient between the established icELISA and LC-MS/MS method was 0.9871. 15A Monte Carlo Neural Fictitious Self-Play approach to approximate Nash Equilibrium in imperfect-information dynamic games显示文摘Solving the optimization problem to approach a Nash Equilibrium point plays an important role in imperfect information games,e.g.,StarCraft and poker.Neural Fictitious Self-Play(NFSP)is an effective algorithm that learns approximate Nash Equilibrium of imperfect-information games from purely self-play without prior domain knowledge.However,it needs to train a neural network in an off-policy manner to approximate the action values.For games with large search spaces,the training may suffer from unnecessary exploration and sometimes fails to converge.In this paper,we propose a new Neural Fictitious Self-Play algorithm that combines Monte Carlo tree search with NFSP,called MC-NFSP,to improve the performance in real-time zero-sum imperfect-information games.With experiments and empirical analysis,we demonstrate that the proposed MC-NFSP algorithm can approximate Nash Equilibrium in games with large-scale search depth while the NFSP can not.Furthermore,we develop an Asynchronous Neural Fictitious Self-Play framework(ANFSP).It uses asynchronous and parallel architecture to collect game experience and improve both the training efficiency and policy quality.The experiments with th e games with hidden state information(Texas Hold^m),and the FPS(firstperson shooter)games demonstrate effectiveness of our algorithms.Li ZHANG Yuxuan CHEN Wei WANG Ziliang HAN Shijian Li Zhijie PAN Gang PAN 2021Frontiers of Computer Science2021,15,5:3
16Porous Carbon Architecture Assembled by Cross-Linked Carbon Leaves with Implanted Atomic Cobalt for High-Performance Li-S Batteries显示文摘The practical application of lithium-sulfur batteries is severely hampered by the poor conductivity,polysulfide shuttle effect and sluggish reaction kinetics of sulfur cathodes.Herein,a hierarchi-cally porous three-dimension(3D)carbon architecture assembled by cross-linked carbon leaves with implanted atomic Co-N4 has been deli-cately developed as an advanced sulfur host through a SiO_(2)-mediated zeolitic imidazolate framework-L(ZIF-L)strategy.The unique 3D architectures not only provide a highly conductive network for fast electron transfer and buffer the volume change upon lithiation-delithi-ation process but also endow rich interface with full exposure of Co-N4 active sites to boost the lithium polysulfides adsorption and conversion.Owing to the accelerated kinetics and suppressed shuttle effect,the as-prepared sulfur cathode exhibits a superior electrochemical perfor-mance with a high reversible specific capacity of 695 mAh g^(−1) at 5 C and a low capacity fading rate of 0.053%per cycle over 500 cycles at 1 C.This work may provide a promising solution for the design of an advanced sulfur-based cathode toward high-performance Li-S batteries.Ruirui Wang Renbing Wu Chaofan Ding Ziliang Chen Hongbin Xu Yongfeng Liu Jichao Zhang Yuan Ha Ben Fei Hongge Pan 2021Nano-Micro Letters2021,13,10:3

18The Fibrillin-1/VEGFR2/STAT2 signaling axis promotes chemoresistance via modulating glycolysis and angiogenesis in ovarian cancer organoids and cells显示文摘Background:Chemotherapy resistance is a primary reason of ovarian cancer therapy failure;hence it is important to investigate the underlying mechanisms of chemotherapy resistance and develop novel potential therapeutic targets.Methods:RNA sequencing of cisplatin-resistant and sensitive(chemoresis-tant and chemosensitive,respectively)ovarian cancer organoids was performed,followed by detection of the expression level of fibrillin-1(FBN1)in organoids and clinical specimens of ovarian cancer.Subsequently,glucose metabolism,angiogenesis,and chemosensitivity were analyzed in structural glycoprotein FBNl-knockout cisplatin-resistant ovarian cancer organoids and cell lines.To gain insights into the specific functions and mechanisms of action of FBN1 in ovarian cancer,immunoprecipitation,silver nitrate staining,mass spectrometry,immunofluorescence,Western blotting,and Forister resonance energy transfer-fluorescence lifetime imaging analyses were performed,followed by in vivo assays using vertebrate model systems of nude mice and zebrafish.Results:FBN1 expression was significantly enhanced in cisplatin-resistant ovar-ian cancer organoids and tissues,indicating that FBNI might be a key factor in chemoresistance of ovarian cancer.We also discovered that FBN1 sustained the energy stress and induced angiogenesis in vitro and in vivo,which promoted the cisplatin-resistance of ovarian cancer.Knockout of FBN1 combined with treat-ment of the antiangiogenic drug apatinib improved the cisplatin-sensitivity of ovarian cancer cells.Mechanistically,FBN1 mediated the phosphorylation of vascular endothelial growth factor receptor 2(VEGFR2)at the Tyrl054 residue,which activated its downstream focal adhesion kinase(FAK)/protein kinase B(PKB or AKT)pathway,induced the phosphorylation of signal transducer and activator of transcription 2(STAT2)at the tyrosine residue 690(Tyr690),pro-moted the nuclear translocation of STAT2,and ultimately altered the expression of genes associated with STAT2-mediated angiogenesis and glycolysis.Conclusions:The FBN1/VEGFR2/STAT2 signaling axis may induce chemore-sistance of ovarian cancer cells by participating in the process of glycolysis and angiogenesis.The present study suggested a novel FBNl-targeted therapy and/or combination of FBN1 inhibition and antiangiogenic drug for treating ovarian cancer.Ziliang Wang Wei Chen Ling Zuo Midie Xu Yong Wu Jiami Huang Xu Zhang Yongheng Li Jing Wang Jing Chen Husheng Wang Huizhen Sun 2022Cancer Communications2022,42,3:3
19Mercury in soil,vegetable and human hair in a typical mining area in China:Implication for human exposure显示文摘Concentrations of total mercury(T-Hg) and methylmercury(MeHg) in soil, vegetables, and human hair were measured in a mercury mining area in central China. T-Hg and MeHg concentrations in soil ranged from 1.53 to 1054.97 mg/kg and 0.88 to 46.52 μg/kg, respectively.T-Hg concentrations was correlated with total organic carbon(TOC) content(R^2= 0.50, p<0.01)and pH values(R^2= 0.21, p<0.05). A significant linear relationship was observed between MeHg concentrations and the abundance of sulfate-reducing bacteria(SRB)(R^2= 0.39, p<0.05) in soil.Soil incubation experiments amended with specific microbial stimulants and inhibitors showed that Hg methylation was derived from SRB activity. T-Hg and MeHg concentrations in vegetables were 24.79 – 781.02 μg/kg and 0.01 – 0.18 μg/kg, respectively; levels in the edible parts were significantly higher than in the roots(T-Hg: p<0.05; MeHg: p<0.01). Hg species concentrations in rhizosphere soil were positively correlated to those in vegetables(p<0.01), indicating that soil was an important source of Hg in vegetables. Risk assessment indicated that the consumption of vegetables could result in higher probable daily intake(PDI) of T-Hg than the provisional tolerable daily intake(PTDI) for both adults and children. In contrast, the PDI of MeHg was lower than the reference dose. 20Evidence from pseudomorphous β-quartz phenocryst for decompression of rock-forming and ore-forming processes in Shapinggou porphyry Mo deposit显示文摘The Shapinggou porphyry molybdenum(Mo) deposit, located in Jinzhai County, Anhui Province, China, is the largest in the Qinling-Dabie Mo Metallogenic Belt. The intrusive rocks in the Shapinggou Mo ore district formed in the Yanshanian can be divided into two stages based on zircon U-Pb dating and geochemical features. This study focuses on the late stage intrusions(quartz syenite and granite porphyry), which are closely genetically related to molybdenum mineralization. Petrographic observations identified two quartz polymorphs in the quartz syenite and granite porphyry, which were derived from the same magmatic sources and similar evolutionary processes. The quartzes were identified as a xenomorphic β-quartz within quartz syenite, while the quartz phenocrysts within the granite porphyry were pseudomorphous b-quartz, characterized by a hexagonal bipyramid crystallography. The pseudomorphous b-quartz phenocrysts within the granite porphyry were altered from b-quartz through phase transformation. These crystals retained b-quartz pseudomorph. Combined with titanium-inzircon thermometry, quartz phase diagrams, and granitic Q-Ab-Or-H_2O phase diagrams, it is suggested that the quartz syenite and granite porphyry were formed under similar magmatic origins, including similar depths and magmatic crystallization temperatures. However, the β-quartz within quartz syenite indicated that the crystallization pressure was greater than 0.7 GPa, while the original b-quartz within the granite porphyry was formed under pressures between 0.4 and 0.7 GPa. The groundmass of the granite porphyry which formed after the phenocryst indicated a crystallizing pressure below 0.05 GPa. This indicates that the granite porphyry was formed under repetitive and rapid decompression. The decompression was significant as it caused the exsolution of the ore-forming fluids, and boiling and material precipitation during the magmatic-fluid process. The volumetric difference during the phase transformation from b-quartz to β-quartz caused extensive fracturing on the granite porphyry body and the wall rocks. As the main ore-transmitting and ore-depositing structures, these fractures benefit the hydrothermal alteration and stockwork-disseminated mineralization of the porphyry deposit. It is considered that the pseudomorphous β-quartz phenocrysts of the porphyritic body are metallogenic indicators within the porphyry deposits.
