1Vitamin B2 functionalized iron oxide nanozymes for mouth ulcer healing显示文摘Mouth ulcer is associated with inflammation and high risk of bacterial infection, which aggravates the patient's condition.Currently, there is no effective treatment for mouth ulcer. Herein, we report that vitamin-modified iron oxide nanoparticles improve the healing of mouth ulcer through anti-inflammation and antibacterial activities. We discovered that vitamin B2(VB2)modified iron oxide nanoparticles performed enhanced peroxidase-like, catalase-like, and superoxide dismutase(SOD)-like activities, acting as typical iron oxide nanozymes(IONzymes) with triad activities. In particular, VB2 modification significantly improved the SOD-like activity, thus providing a reactive oxygen species(ROS)-scavenging ability. Cellular antioxidant experiments showed that vitamin B2 modified IONzymes(VB2-IONzymes) protect human oral keratinocytes(HOK) and BALB/3 T3 cells from hydrogen peroxide(H2O2), and these cells have high biocompatibility to eukaryotic cells. In addition, VB2-IONzymes exerted an antibacterial activity against Streptococcus mutans, Staphylococcus aureus, and Escherichia coli. Importantly, VB2-IONzymes accelerated the recovery of mouth ulcer and reduced the local secretion of inflammatory factors in mouse ulcer model via ROS scavenging and antibacterial activity. 2CD4^(+)CD25^(+)but not CD4^(+)Foxp3^(+) T cells as a regulatory subset in primary biliary cirrhosis显示文摘Increasing evidence indicates a role for regulatory T cells(Tregs)in the immune response and in autoimmune diseases,but the role of Tregs and cytokines in autoimmune hepatic diseases remains largely unclear and controversial,especially in patients with primary biliary cirrhosis(PBC).This study was undertaken to investigate Tregs and different cytokines in the liver and peripheral blood of PBC patients.We found that these patients demonstrated a reduction of CD4^(+)CD25^(+) T cells but elevated CD4^(+)Foxp3^(+) T cells in peripheral blood mononuclear cells(PBMCs)and CD41 T cells.The percentage of CD4^(+)CD25^(+) T cells in PBMCs was negatively correlated with elevated plasma interferon(IFN)-c levels.A liver-specific analysis showed that the frequency of Foxp31 Tregs,transforming growth factor(TGF)-b1 and IFN-c were increased in PBC patients.Our findings suggest that an imbalance between CD4^(+)CD25^(+) Tregs and cytotoxic cytokines plays a crucial role in the pathogenesis of PBC while the role of Foxp3 needs further investigation.Dandan Wang Huayong Zhang Jun Liang Zhifeng Gu Qiang Zhou Xiangshan Fan Yayi Hou Lingyun Sun 2010Cellular & Molecular Immunology2010,7,6:9
3Effect of shrub-grass vegetation coverage and slope gradient on runoff and sediment yield under simulated rainfall显示文摘Evaluating the benefits of sediment and runoff reduction in different vegetation types is essential for studying the mechanisms of soil and water conservation on the Loess Plateau.The experiment was conducted in shrub-grass plots with nine levels of mixed vegetation coverage from 0%to 70%,three slopes(10,15,and 20)and two rainfall intensities(1.0 and 2.5 mm/min).The results showed that the vegetation coverage and slope gradient significantly affect runoff and sediment yield.Shrub-grass vegetation coverage had a significant effect on the runoff start-time,runoff flow velocity,runoff rate,and soil erosion rate on hillslopes.Mixed vegetation coverage could effectively delay the runoff starttime and decrease the runoff flow velocity.However,the effects of the slope gradient on runoff and sediment yield are opposite to those of vegetation coverage.Shrub-grass vegetation coverage could effectively increase runoff and sediment yield reduction benefits,while their benefits were affected by the rainfall intensity.At the 1.0 mm/min rainfall intensity,the reduction in the sediment production rate was greater than that under the 2.5 mm/min intensity.However,when the shrub-grass vegetation coverage exceeded 42%,the runoff reduction benefit was more obvious at higher rainfall intensities.The cumulative sediment yield increased with increasing cumulative runoff,and the rate of increase in the cumulative runoff was greater than that of the cumulative sediment yield with increasing of shrub-grass vegetation coverage.Moreover,there was a power function relationship between cumulative sediment yield and cumulative runoff yield(P<0.05).Our paper is expected to provide a good reference on the ecological environment and vegetation construction on the Loess Plateau.Dandan Han Jingcheng Deng Chaojun Gu Xingmin Mu Peng Gao Jianjian Gao 2021International Journal of Sediment Research2021,36,1:8

518beta-glycyrrhetinic acid induces ROS-mediated apoptosis to ameliorate hepatic fibrosis by targeting PRDX1/2 in activated HSCs显示文摘Hepatic stellate cells(HSCs)are essential drivers of fibrogenesis.Inducing activated-HSC apoptosis is a promising strategy for treating hepatic fibrosis.18beta-glycyrrhetinic acid(18b-GA)is a natural compound that exists widely in herbal medicines,such as Glycyrrhiza uralensis Fisch,which is used for treating multiple liver diseases,especially in Asia.In the present study,we demonstrated that 18b-GA decreased hepatic fibrosis by inducing the apoptosis in activated HSCs.18b-GA inhibited the expression of a-smooth muscle actin and collagen type Ⅰ alpha-1.Using a chemoproteomic approach derived from activity-based protein profiling,together with cellular thermal shift assay and surface plasmon resonance,we found that 18b-GA covalently targeted peroxiredoxin 1(PRDX1)and peroxiredoxin 2(PRDX2)proteins via binding to active cysteine residues and thereby inhibited their enzymatic activities.18b-GA induced the elevation of reactive oxygen species(ROS),resulting in the apoptosis of activated HSCs.PRDX1 knockdown also led to ROS-mediated apoptosis in activated HSCs.Collectively,our findings revealed the target proteins and molecular mechanisms of 18b-GA in ameliorating hepatic fibrosis,highlighting the future development of 18b-GA as a novel therapeutic drug for hepatic fibrosis.Qian Zhang Piao Luo Liuhai Zheng Jiayun Chen Junzhe Zhang Huan Tang Dandan Liu Xueling He Qiaoli Shi Liwei Gu Jiahao Li Qiuyan Guo Chuanbin Yang Yin Kwan Wong Fei Xia Jigang Wang 2022Journal of Pharmaceutical Analysis2022,12,4:3

7Photocrosslinkable Col/PCL/Mg composite membrane providing spatiotemporal maintenance and positive osteogenetic effects during guided bone regeneration显示文摘Guided bone regeneration membranes have been effectively applied in oral implantology to repair bone defects.However,typical resorbable membranes composed of collagen(Col)have insufficient mechanical properties and high degradation rate,while non-resorbable membranes need secondary surgery.Herein,we designed a photocrosslinkable collagen/polycaprolactone methacryloyl/magnesium(Col/PCLMA/Mg)composite membrane that provided spatiotemporal support effect after photocrosslinking.Magnesium particles were added to the PCLMA solution and Col/PCLMA and Col/PCLMA/Mg membranes were developed;Col membranes and PCL membranes were used as controls.After photocrosslinking,an interpenetrating polymer network was observed by scanning electron microscopy(SEM)in Col/PCL and Col/PCL/Mg membranes.The elastic modulus,swelling behavior,cytotoxicity,cell attachment,and cell proliferation of the membranes were evaluated.Degradation behavior in vivo and in vitro was monitored according to mass change and by SEM.The membranes were implanted into calvarial bone defects of rats for 8 weeks.The Col/PCL and Col/PCL/Mg membranes displayed much higher elastic modulus(p<0.05),and a lower swelling rate(p<0.05),than Col membranes,and there were no differences in cell biocompatibility among groups(p>0.05).The Col/PCL and Col/PCL/Mg membranes had lower degradation rates than the Col membranes,both in vivo and in vitro(p<0.05).The Col/PCL/Mg groups showed enhanced osteogenic capability compared with the Col groups at week 8(p<0.05).The Col/PCL/Mg composite membrane represents a new strategy to display space maintenance and enhance osteogenic potential,which meets clinical needs.Feilong Wang Dandan Xia Siyi Wang Ranli Gu Fan Yang Xiao Zhao Xuenan Liu Yuan Zhu Hao Liu Yongxiang Xu Yunsong Liu Yongsheng Zhou 2022Bioactive Materials2022,7,7:1
8Efficacy on rabbits with arrhythmia: needling acupoint of Neiguan(PC6) at shallow or deep depth, and retaining needles for 10, 20, or 30 min显示文摘OBJECTIVE:To compare the effects of Neiguan(PC6)acupuncture at different depths and retention time on arrhythmia duration,myocardial tissue morphology,mRNA expression level of L-type calcium channelα1C subunit and Ca^(2+)-Mg^(2+)-At Pase activity in tachirrhythmia model of rabbits.METHODS:The tachyarrhythmia model was made by intravenous injection of barium chloride into the ears of rabbits.A total of 56 healthy adult male New Zealand big-eared white rabbits,apply the random number table method,divided into normal control group(group A),model group(group B),shallow needling Neiguan(PC6)10 min group(group C),shallow needling Neiguan(PC6)20 min group(group D),shallow needling Neiguan(PC6)30 min group(group E),deep needling Neiguan(PC6)10 min group(group F),deep needling Neiguan(PC6)20 min group(group G),deep needling Neiguan(PC6)30 min group(group H),7 animals in each group.Electrocardiograms were used to collect the duration of arrhythmia;hematoxylin-eosin staining method was performed on myocardial tissue,RT-PCR tested the expression ofα1C subunit mRNA,and the activity of Ca^(2+)-Mg^(2+)-ATPase were quantified by phosphorus determination method.RESULTS:The duration of arrhythmia in each acupuncture treatment group was shortened to varying degrees.Compare to the model group,the tissue damage from barium chloride inducing was improved in the acupuncture group.Compared to the model group,except for group E,most treatment groups had varying degrees of improvement with significantly down-regulated L-type calcium channelα1C subunit mRNA expressions level and increased Ca^(2+)-Mg^(2+)-ATPase activity.CONCLUSIONS:The effect of acupuncture at Neiguan(PC6)with different depths and retention time can reduce the duration of arrhythmia induced by barium chloride relatively,improve the induced pathological changes,down regulate L-type calcium channelα1C subunit mRNA expressions level and increase Ca^(2+)-Mg^(2+)-ATPase activity.Both the shallow and deep tissues of Neiguan(PC6)may be involved in transmitting acupuncture information.There is an optimal induction period for shallow needling at Neiguan(PC6)to reach the best therapeutic effect.GU Chunling CHENG Kai YANG Shouqin RAN Shaofeng TAI Hengyap GAO Xiaofeng PANG Dandan 2021Journal of Traditional Chinese Medicine2021,41,6:1

12Observation of CO2^++ dication in the dayside Martian upper atmosphere显示文摘Doubly charged positive ions(dications)are an important component of planetary ionospheres because of the large energy required for their formation.Observations of these ions are exceptionally difficult due to their low abundances;until now,only atomic dications have been detected.The Neutral Gas and Ion Mass Spectrometer(NGIMS)measurements made on board the recent Mars Atmosphere and Volatile Evolution mission provide the first opportunity for decisive detection of molecular dications,CO2^++in this case,in a planetary upper atmosphere.The NGIMS data reveal a dayside averaged CO2^++distribution declining steadily from 5.6 cm^−3 at 160 km to below 1 cm^−3 above 200 km.The dominant CO2^++production mechanisms are double photoionization of CO2 below 190 km and single photoionization of CO2^+at higher altitudes;CO2++destruction is dominated by natural dissociation,but reactions with atmospheric CO2 and O become important below 160 km.Simplified photochemical model calculations are carried out and reasonably reproduce the data at low altitudes within a factor of 2 but underestimate the data at high altitudes by a factor of 4.Finally,we report a much stronger solar control of the CO2^++density than of the CO2+density.Hao Gu Jun Cui DanDan Niu LongKang Dai JianPing Huang XiaoShu Wu YongQiang Hao Yong Wei 2020Earth and Planetary Physics2020,4,4:1

14Targeted gene panel provides advantages over whole-exome sequencing for diagnosing obesity and diabetes mellitus显示文摘A small fraction of patients diagnosed with obesity or diabetes mellitus has an underlying monogenic cause.Here,we constructed a targeted gene panel consisting of 83 genes reported to be causative for monogenic obesity or diabetes.We performed this panel in 481 patients to detect causative variants and compared these results with whole-exome sequencing(WES)data available for 146 of these patients.The coverage of targeted gene panel sequencing was significantly higher than that of WES.The diagnostic yield in patients sequenced by the panel was 32.9%with subsequent WES leading to three additional diagnoses with two novel genes.In total,178 variants in 83 genes were detected in 146 patients by targeted sequencing.Three of the 178 variants were missed by WES,although the WES-only approach had a similar diagnostic yield.For the 335 samples only receiving targeted sequencing,the diagnostic yield was 32.2%.In conclusion,taking into account the lower costs,shorter turnaround time,and higher quality of data,targeted sequencing is a more effective screening method for monogenic obesity and diabetes compared to WES.Therefore,this approach could be routinely established and used as a first-tier test in clinical practice for specific patients.Hairong Yu Haoyong Yu Rong Zhang Danfeng Peng Dandan Yan Yunjuan Gu Yuqian Bao Weiping Jia Hong Zhang Cheng Hu 2023Journal of Molecular Cell Biology2023,15,6:0
15Overcoming the Limitation of Cs_(2)AgBiBr_(6) Double Perovskite Solar Cells Through Using Mesoporous TiO_(2) Electron Extraction Layer显示文摘Lead-free double perovskite Cs_(2)AgBiBr_(6) has gained increasing attention recently.However,the power conversion efficiency(PCE)of Cs_(2)AgBiBr_(6) perovskite solar cells(PSCs)is still low compared with their lead-based counterparts.Here,by using photoluminescence(PL),time-resolved photoluminescence(TRPL),and ultrafast transient absorption(TA)measurements,the unbalance between the electron and hole in diffusion and transfer,which limits the performance of the Cs_(2)AgBiBr_(6) PSCs,was further revealed.Considering this issue,a strategy of using the mesoporous TiO_(2) electron transport layer(ETL)to construct a bulk heterojunction in Cs_(2)AgBiBr_(6) PSCs was proposed.Consequently,the PCE had improved by over 24%comparing with that only used compact TiO_(2) ETL.Moreover,based on mesoporous TiO_(2),the unencapsulated Cs_(2)AgBiBr_(6) PSCs maintained 90%of their initial performance after approximately 1200 h of storage in a desiccator(humidity~30%).This work gives further understanding of Cs_(2)AgBiBr_(6) perovskite and demonstrates that a proper design of balancing the electron and hole diffusion can improve device performance.Dandan Zhao Chao Liang Bingzhe Wang Tanghao Liu Qi Wei Kaiyang Wang Hao Gu Sisi Wang Shiliang Mei Guichuan Xing 2022Energy & Environmental Materials2022,5,4:0
16Dissection of cellular and molecular mechanisms of aristolochic acid-induced hepatotoxicity via single-cell transcriptomics显示文摘Background:Aristolochic acids(AAs),a class of carcinogenic and mutagenic natural products from Aristolochia and Asarum plants,are well-known to be responsible for inducing nephrotoxicity and urothelial carcinoma.Recently,accumulating evidence suggests that exposure to AAs could also induce hepatotoxicity and even hepatocellular carcinoma,though the mechanisms are poorly defined.Methods:Here,we aimed to dissect the underlying cellular and molecular mechanisms of aristolochic acid I(AAI)-induced hepatotoxicity by using advanced single-cell RNA sequencing(scRNA-seq)and proteomics techniques.We established the first single-cell atlas of mouse livers in response to AAI.Results:In hepatocytes,our results indicated that AAI activated NF-κB and STAT3 signaling pathways,which may contribute to the inflammatory response and apoptosis.In liver sinusoidal endothelial cells(LSECs),AAI activated multiple oxidative stress and inflammatory associated signaling pathways and induced apoptosis.Importantly,AAI induced infiltration of cytotoxic T cells and activation of proinflammatory macrophage and neutrophil cells in the liver to produce inflammatory cytokines to aggravate inflammation.Conclusions:Collectively,our study provides novel knowledge of AAs-induced molecular characteristics of hepatotoxicity at a singlecell level and suggests future treatment options for AAs associated hepatotoxicity.Piao Luo Jiayun Chen Qian Zhang Fei Xia Chen Wang Yunmeng Bai Huan Tang Dandan Liu Liwei Gu Qingfeng Du Wei Xiao Chuanbin Yang Jigang Wang 2022Precision Clinical Medicine2022,5,4:0
17Effective depolymerization of alkali lignin using an attapulgite-Ce_(0.75)Zr_(0.25)O_(2)(ATP-CZO)-supported cobalt catalyst in ethanol/isopropanol media显示文摘Lignin is the world's greatest renewable aromatic biofeedstock,and it has promising applications in high value-added chemical products.Herein,N-Co/ATP-CZO was used as a catalyst for the depolymerization of alkali lignin in ethanol and isopropanol systems,and explored the effects of formic acid(FA)amount,reaction time,reaction temperature and other factors on the depolymerization of alkali lignin.Among them,formic acid serves as both catalytic and in situ-hydrogen donor.Ultimately,the highest yield of bio-oil(59.28%(mass)),including 30.05%(mass)of monomer,was obtained after a reaction of FA to alkali lignin mass ratio of 4 and 240°C for 8 h.Among the monomers,the yield of Guaiacol was the highest(5.94%(mass)),followed by 2-methoxy-4-methylphenol(5.74%(mass)).This study,the modification of attapulgite was carried out to reduce the acidity while enhancing the catalytic activity for depolymerization,and the selection of hydrogen donor was investigated.A feasible pathway for lignin depolymerization research was opened.Jiajia Chen Xinyu Lu Dandan Wang Pengcheng Xiu Xiaoli Gu 2023Chinese Journal of Chemical Engineering2023,57,5:0
18Research advances on internal processes affecting tropical cyclone intensity change from 2018–2022显示文摘This contribution summarizes the significant progress in a variety of topic areas related to internal tropical cyclone(TC)intensity change processes over 2018–2022 from the WMO Tenth International Workshop on Tropical Cyclones(IWTC-10).These topic areas include surface and boundary layer processes;TC internal structure and microphysical processes;and,radiation interactions with TCs.Recent studies better frame the uncertainty in the surface drag and enthalpy coefficients at high wind speeds.These parameters greatly impact TC intensity and it is therefore important that more direct measurements of these boundary layer parameters are made.Particularly significant scientific strides have been made in TC boundary layers.These advancements have been achieved through improved coupled models,large-eddy simulations,theoretical advance-ments,and detailed observations.It is now clear that the research field needs to better represent the eddy viscosity throughout the depth of the boundary layer.Furthermore,detailed study of coherent structures in TC boundary layers will likely be a propitious direction for the research community.Meanwhile,in-depth observational field campaigns and assiduous data analysis have made significant headway into verifying theory and modeling studies of intensification processes related to TC vortex alignment,efficient latent heating distributions,and overall 3D structure.Substantial efforts have also been made to better understand the intricate roles radiative processes play in TC evolution and intensity change.Finally,some promising progress has been made in the development of time-dependent theories of TC intensification and the predictability of internal TC intensity change.Overall,there have been well-earned gains in the understanding of intensity change processes intrinsic to the TC system,but the journey is not complete.This paper highlights some of the most relevant and important research areas that are still shedding new light into internal factors governing TC intensity change.Xiaomin Chen Christopher M.Rozoff Robert F.Rogers Kristen L.Corbosiero Dandan Tao Jian-Feng Gu Falko Judt Eric A.Hendricks Yuqing Wang Michael M.Bell Daniel P.Stern Kate D.Musgrave John A.Knaff John Kaplan 2023Tropical Cyclone Research and Review2023,12,1:0

20Neferine inhibits the progression of diabetic nephropathy by modulating the miR-17-5p/nuclear factor E2-related factor 2 axis显示文摘OBJECTIVE:To investigate the effect of Neferine(Nef)on diabetic nephropathy(DN)and to explore the mechanism of Nef in DN based on miRNA regulation theory.METHODS:A DN mouse model was constructed and treated with Nef.Serum creatinine(Crea),blood urea(UREA)and urinary albumin were measured in mice by kits,and renal histopathological changes and fibrosis were observed by hematoxylin-eosin staining and Masson staining.Renal tissue superoxide dismutase(SOD),malondialdehyde(MDA)and glutathione peroxidase(GSH-Px)activities were measured by enzyme-linked immunosorbent assay(ELISA).Western blotting was used to detect the expression of nuclear factor E2-related factor 2(Nrf2)/heme oxygenase 1(HO-1)signaling pathway-related proteins in kidney tissues.Quantitative reverse transcription-polymerase chain reaction(q RT-PCR)was used to detect the expression of miR-17-5p in kidney tissues.Subsequently,a DN in vitro model was constructed by high glucose culture of human mesangial cells(HMCs),cells were transfected with miR-17-5p mimic and/or treated with Nef,and we used q RTPCR to detect cellular miR-17 expression,flow cytometry to detect apoptosis,ELISAs to detect cellular SOD,MDA,and GSH-Px activities,Western blots to detect Nrf2/HO-1 signaling pathway-related protein expression,and dual luciferase reporter gene assays to verify the targeting relationship between Nrf2 and miR-17-5p.RESULTS:Administration of Nef significantly reduced the levels of blood glucose,Crea,and UREA and the expression of miR-17-5p,improved renal histopathology and fibrosis,significantly reduced MDA levels,elevated SOD and GSH-Px activities,and activated Nrf2 expression in kidney tissues from mice with DN.Nrf2 is a post-transcriptional target of miR-17-5p.In HMCs transfected with miR-17-5p mimics,the m RNA and protein levels of Nrf2 were significantly suppressed.Furthermore,miR-17-5p overexpression and Nef intervention resulted in a significant increase in high glucose-induced apoptosis and MDA levels in HMCs and a significant decrease in the protein expression of HO-1 and Nrf2.CONCLUSION:Collectively,these results indicate that Nef has an ameliorative effect on DN,and the mechanism may be through the miR-17-5p/Nrf2 pathway.HUANG Hongmei YANG Maojun LI Ting WANG Dandan LI Ying TANG Xiaochi YUAN Lu GU Shi XU Yong 2024Journal of Traditional Chinese Medicine2024,44,1:0
