2A novel m6A reader Prrc2a controls oligodendroglial specification and myelination显示文摘While N6-methyladenosine (m6A), the most abundant internal modification in eukaryotic mRNA, is linked to cell differentiation and tissue development, the biological significance of m6A modification in mammalian glial development remains unknown. Here, we identify a novel m6A reader, Prrc2a (Proline rich coiled-coil 2A), which controls oligodendrocyte specification and myelination. Nestin-Cre-mediated knockout of Prrc2a induces significant hypomyelination, decreased lifespan, as well as locomotive and cognitive defects in a mouse model. Further analyses reveal that Prrc2a is involved in oligodendrocyte progenitor cells (OPCs) proliferation and oligodendrocyte fate determination. Accordingly, oligodendroglial-lineage specific deletion of Prrc2a causes a similar phenotype of Nestin-Cre-mediated deletion. Combining transcriptome-wide RNA-seq, m6A-RIP-seq and Prrc2a RIP-seq analysis, we find that Olig2 is a critical downstream target gene of Prrc2a in oligodendrocyte development. Furthermore, Prrc2a stabilizes Olig2 mRNA through binding to a consensus GGACU motif in the Olig2 CDS (coding sequence) in an m6A-dependent manner. Interestingly, we also find that the m6A demethylase, Fto, erases the m6A modification of Olig2 mRNA and promotes its degradation. Together, our results indicate that Prrc2a plays an important role in oligodendrocyte specification through functioning as a novel m6A reader. 3Mild hypothermia combined with a scaffold of Ng Rsilenced neural stem cells/Schwann cells to treat spinal cord injury显示文摘Because the inhibition of Nogo proteins can promote neurite growth and nerve cell differentiation, a cell-scaffold complex seeded with Nogo receptor(Ng R)-silenced neural stem cells and Schwann cells may be able to improve the microenvironment for spinal cord injury repair. Previous studies have found that mild hypothermia helps to attenuate secondary damage in the spinal cord and exerts a neuroprotective effect. Here, we constructed a cell-scaffold complex consisting of a poly(D,L-lactide-co-glycolic acid)(PLGA) scaffold seeded with Ng R-silenced neural stem cells and Schwann cells, and determined the effects of mild hypothermia combined with the cell-scaffold complexes on the spinal cord hemi-transection injury in the T9 segment in rats. Compared with the PLGA group and the Ng R-silencing cells + PLGA group, hindlimb motor function and nerve electrophysiological function were clearly improved, pathological changes in the injured spinal cord were attenuated, and the number of surviving cells and nerve fibers were increased in the group treated with the Ng R-silenced cell scaffold + mild hypothermia at 34°C for 6 hours. Furthermore, fewer pathological changes to the injured spinal cord and more surviving cells and nerve fibers were found after mild hypothermia therapy than in injuries not treated with mild hypothermia. 5The efficiency and toxicity of dodecafluoro-2-methylpentan-3-one in suppressing lithium-ion battery fire显示文摘Currently,the effective and clean suppression of lithium-ion battery(LIB)fires remains a challenge.The present work investigates the use of various inhibitor doses(Xin)of dodecafluoro-2-methylpentan-3-one(C_(6) F_(12)O)in 300 Ah LIBs,and systematically examines the thermal and toxic hazards of the extinguished batteries via real scale combustion and gas analysis.The inhibitor is shown to be completely effective.The inhibition mechanism involves a combination of chemical inhibition and physical cooling.While the chemical inhibition effect tends to saturate with increasing Xin,the physical cooling remains effective at higher inhibitor doses.However,extinguishing the battery fire with a high Xin of C_(6)F_(12)O is found to incur serious toxicity problems.These results are expected to provide a guideline for the design of inhibitor doses for the suppression of LIB fires.Yujun Liu Kai Yang Mingjie Zhang Shi Li Fei Gao Qiangling Duan Jinhua Sun Qingsong Wang 2022Journal of Energy Chemistry2022,31,2:10
6Microstructure of premixed propane/air flame in the transition from laminar to turbulent combustion显示文摘In order to explore the flame structure and propagation behavior of premixed propane/air in the transi-tion from laminar to turbulent combustion, the high speed camera and Schlieren images methods were used to record the photograph of flame propagation process in a semi-vented pipe. Meanwhile, the super-thin thermocouple and ionization current probe methods were applied to detect the temperature distribution and reaction intensity of combustion reaction. The characteristics of propane/air flame propagation and microstructure were analyzed in detail by the experimental results coupled with chemical reaction thermodynamics. In the test, the particular tulip flame behavior and the formation process in the laminar-turbulent transition were disclosed clearly. 7Characteristics of flame spread over the surface of charring solid combustibles at high altitude显示文摘To explore the characteristics of flame spread over the surface of charring solid combustibles at high altitude, the whitewood with uniform texture was chosen to conduct a series of experiments in Lhasa and Hefei, with altitude of 3658 m and 50 m respectively. Several parameters, including the flame height, flame spread rate, flame temperature, surface temperature, were measured on samples with different width and inclinations. A quantitative analysis of flame spread characteristics over sample surface at high altitude was performed. Results showed that, in the environment of lower pressure and oxygen concentration at high altitude, the flame height and flame spread rate over sample surface decreased, but the flame temperature increased slightly. 8Flame propagation characteristics and flame structures of zirconium particle cloud in a small-scale chamber显示文摘Flame propagating through zirconium particle cloud in a small-scale vertical rectangle chamber was investigated experimentally.In the experiments,the zirconium quoted 99% purity was used and the diameter of particles was distributed 1-22 μm.The zirconium dust was dispersed into the chamber by air flow and ignited by an electrode spark.A high-speed video camera was used to record the images of the propagating flame.Micro-thermocouples,schlieren optical system and microscopic lens were used to obtain temperature profiles and flame structure,respectively.Based on the experimental results,flame propagation characteristics and flame structure of zirconium particle cloud were analyzed.The propagation velocity of the flame is quite slow in the initial 14 ms and then accelerates to maximum value.Subsequently,the propagation velocity of the flame almost keeps constant.The combustion zone width of zirconium particle cloud is 5-6 mm.Smaller particles burn mainly at the leading edge of combustion zone in the width of 1.4 mm followed by larger particles burning 1.4-6 mm behind the leading edge of the combustion zone.Gas phase flame is not seen in zirconium particle cloud and the combustion time of single zirconium particle is 1-5 ms,which depends on its original size.The preheated zone is 7-8 mm thickness ahead of the combustion zone and intensive chemical reaction takes place at 490 K.The maximum flame temperature increases at lower concentrations,reaches the maximum value,and then decreases slightly at higher concentrations.DING YiBin SUN JinHua HE XueChao WANG QiuHong YIN Yi XU Yao Chen XianFeng 2010Chinese Science Bulletin2010,55,34:8
9Design,methodology,and baseline of whole city‑million scale children and adolescents myopia survey(CAMS)in Wenzhou,China显示文摘Background:Myopia is the most common visual impairment in children and adolescents worldwide.This study described an economical and effective population-based screening pipeline and performed the project of a million scale children and adolescents myopia survey(CAMS),which will shed light on the further study of myopia from the level of epidemiology and precision medicine.Methods:We developed a novel population-based screening pattern,an intelligent screening process and internetbased information transmission and analysis system to carry out the survey consisting of school children in Wenzhou,China.The examination items include unaided distance visual acuity,presenting distance visual acuity,and noncycloplegic autorefraction.Myopia and high myopia were defined as spherical equivalent(SE)≤−1.00 diopters(D)and SE≤−6.00 D,respectively.Next,the reports of the vision checking were automatically sent to parents and the related departments.The CAMS project will be done two to four times annually with the support of the government.An online eyesight status information management system(OESIMS)was developed to construct comprehensive and efficient electronic vision health records(EVHRs)for myopia information inquiry,risk pre-warning,and further study.Results:The CAMS completed the first-round of screening within 30 days for 99.41%of Wenzhou students from districts and counties,in June 2019.A total of 1,060,925 participants were eligible for CAMS and 1,054,251(99.37%participation rate)were selected through data quality control,which comprised 1305 schools,and 580,609,251,050 and 170,967 elementary,middle,and high school students.The mean age of participants was 12.21±3.32 years(6-20 years),the female-to-male ratio was 0.82.The prevalence of myopia in elementary,middle,and high school students was 38.16%,77.52%,and 84.00%,respectively,and the high myopia incidence was 0.95%,6.90%,and 12.98%.Conclusions:The CAMS standardized myopia screening model involves automating large-scale information collection,data transmission,data analysis and early warning,thereby supporting myopia prevention and control.The entire survey reduced 90%of staff,cost,and time consumption compared with previous surveys.This will provide new insights for decision support for public health intervention.Liangde Xu Youyuan Zhuang Guosi Zhang Yunlong Ma Jian Yuan Changseng Tu MiaoMiao Li Wencan Wang Yaru Zhang Xiaoyan Lu Jing Li Xinting Liu Zhengbo Xue Meng Zhou Jie Sun Jinhua Bao Ming Li Fan Lu Hong Wang Jianzhong Su Jia Qu 2021Eye and Vision2021,8,1:7
10Self-organized criticality judgment and extreme statistics analysis of major urban fires显示文摘The spatial (economic loss) and temporal characteristics of urban fires were analyzed employing relevant statistical methods. A fractal structure in terms of the power-law relation between fire frequency and economic loss was found on a spatial scale, and an exponential relation between frequency and time interval was found on a temporal scale. Thus, urban fire does not meet the rigorous criteria of self-organized criticality. In addition, based on the spatial power-law distribution characteristics, a correlation model of the frequency and scale of loss due to urban fire was established using the extremum statistical method. 11Seasonal phenotypic flexibility in body mass,basal thermogenesis,and tissue oxidative capacity in the male Silky Starling(Sturnus sericeus)显示文摘Background: Acclimatization to winter conditions is an essential prerequisite for the survival of small birds in the northern temperate zone.Changes in photoperiod,ambient temperature and food availability trigger seasonal physiological and behavioral acclimatization in many passerines.Seasonal trends in metabolic parameters are well known in avian populations from temperate environments;however,the physiological and biochemical mechanisms underlying these trends are incompletely understood.In this study,we used an integrative approach to measure variation in the thermogenic properties of the male Silky Starling(Sturnus sericeus) at different levels or organization,from the whole organism to the biochemical.We measured body mass(Mb),basal metabolic rate(BMR),energy budget,the mass of selected internal organs,state 4 respiration and cytochrome c oxidase(COX) activity in the heart,liver and muscle.Methods: Oxygen consumption was measured using an open-circuit respirometry system.The energy intake of the birds were then determined using an oxygen bomb calorimeter.Mitochondrial state 4 respiration and COX activity in heart,liver and pectoral muscle were measured with a Clark electrode.Results: The results suggest that acclimatization to winter conditions caused significant change in each of the measured variables,specifically,increases in Mb,organ mass,BMR,energy intake and cellular enzyme activity.Furthermore,BMR was positively correlated with body mass,energy intake,the mass of selected internal organs,state 4 respiration in the heart,liver and muscle,and COX activity in the heart and muscle.Conclusions: These results suggest that the male Silky Starling's enhanced basal thermogenesis under winter conditions is achieved by making a suite of adjustments from the whole organism to the biochemical level,and provide further evidence to support the notion that small birds have high phenotypic plasticity with respect to seasonal changes.Min Li Yingqiao Sun Hongzhen Mao Jinhua Xu Weihong Zheng Jinsong Liu 2017Avian Research2017,8,4:6

15Effects of sample width on flame spread over horizontal charring solid surfaces on a plateau显示文摘Experiments were conducted on the Tibetan Plateau to investigate the effects of sample width (W) on flame spread over horizontal charring solid surfaces. The results show that the rate of spread is influenced by the combined effects of convection heat transfer and flame radiation. Moreover, two different regimes depending on sample width were found in flame spread behavior. The rate of spread decreased with width in the convective regime (W≤3 cm) but increased in the radiative regime (W>3 cm). 16Biodegradation of organochlorine pesticide endosulfan by bacterial strain Alcaligenes faecalis JBW4显示文摘The recently discovered endosulfan-degrading bacterial strain Alcaligenes faecalis JBW4 was isolated from activated sludge. This strain is able to use endosulfan as a carbon and energy source. The optimal conditions for the growth of strain JBW4 and for biodegradation by this strain were identified, and the metabolic products of endosulfan degradation were studied in detail. The maximum level of endosulfan biodegradation by strain JBW4 was obtained using broth at an initial pH of 7.0, an incubation temperature of 40°C and an endosulfan concentration of 100 mg/L. The concentration of endosulfan was determined by gas chromatography. Strain JBW4 was able to degrade 87.5% of α-endosulfan and 83.9% of β-endosulfan within 5 days. These degradation rates are much higher than the previously reported bacterial strains. Endosulfan diol and endosulfan lactone were the major metabolites detected by gas chromatography-mass spectrometry; endosulfan sulfate, which is a persistent and toxic metabolite, was not detected. These results suggested that A. faecalis JBW4 degrades endosulfan via a non-oxidative pathway. The biodegradation of endosulfan by A. faecalis is reported for the first time. 17Inhibiting effect of Al(OH)_3 and Mg(OH)_2 dust on the explosions of methane-air mixtures in closed vessel显示文摘The inhibiting effect of Al(OH)3 and Mg(OH)2 dust on explosion of methane-air mixtures was investigated by means of explosion parameter tests in a 20-liter closed vessel. The influences of varying methane concentration and dust concentration on explosion parameters were characterized based on the experimental data to determine the maximum explosion pressure, maximum rate of pressure rise, lower explosion limits and upper explosion limits. The inhibiting mechanisms of these kinds of dust were analyzed as well. The investigations indicate that Al(OH)3 and Mg(OH)2 dust can be used as inhibitors to prevent meth- ane explosion, however, their inhibiting effects are less than those of inert gas such as N2 and CO2 in that their dust can weaken the methane explosion but cannot totally eliminate it. The tests show that all of the explosion parameters with dust additives are strongly dependent on methane/air ratio and dust concentration, and Al(OH)3 dust has better performance than Mg(OH)2 dust in inhibiting methane explosion. The average percentage decreases of maximum explosion pressure and maximum rate of pressure rise with Al(OH)3 dust are 11.08% and 66.15%, respectively. Experiments also showed that there is a special phenomenon when methane explosion is inhibited by Al(OH)3 and Mg(OH)2 dust, in which is that during the process of explosion the maximum explosion pressure value first decreases then increases as dust concentration increases. The best dust concentra- tions to inhibit the explosion are 250 g/m3 with methane/air ratio at 9.5%, and 200 g/m3 with methane/air ratio at 7%. It is suggested that water vapor produced by the thermal decomposition of metal hydroxides makes the particles of descending dust combine, resulting in a decrease of the real dust concentration in the vessel. 18First experimental constraints on WIMP couplings in the effective field theory framework from CDEX显示文摘We present weakly interacting massive particles(WIMPs) search results performed using two approaches of effective field theory from the China Dark Matter Experiment(CDEX), based on the data from both CDEX-1B and CDEX-10 stages. In the nonrelativistic effective field theory approach, both time-integrated and annual modulation analyses were used to set new limits for the coupling of WIMP-nucleon effective operators at 90% confidence level(C.L.) and improve over the current bounds in the low mχregion. In the chiral effective field theory approach, data from CDEX-10 were used to set an upper limit on WIMP-pion coupling at 90% C.L. 20SERIALLY CONCATENATED CONTINUOUS PHASE MODULATION WITH REDUCED ITERATIVE DEMODULATION AND DETECTION显示文摘A reduced state Soft Input Soft Output (SISO) a posteriori probability algorithm for Seri-ally Concatenated Continuous Phase Modulation (SCCPM) is proposed in this paper. Based on the Reduced State Sequence Detection (RSSD),it has more general form compared with other reduced state SISO algorithms. The proposed algorithm can greatly reduce the state number,thus leads to the computation complexity reduction. It also minimizes the degradation in Euclidean distance with decision feedback in the reduced state trellis.
