1Effects of different dietary energy and protein levels and sex on growth performance,carcass characteristics and meat quality of F1 Angus x Chinese Xiangxi yellow cattle显示文摘Background:The experiment evaluated the effect of nutrition levels and sex on the growth performance,carcass characteristics and meat quality of F1 Angus × Chinese Xiangxi yellow cattle.Methods:During the background period of 184 d,23 steers and 24 heifers were fed the same ration,then put into a2×2×2 factorial arrangement under two levels of- dietary energy(TON:70/80%DM),protein(CP:11.9/14.3%DM)and sex(S:male/female) during the finishing phase of 146 d.The treatments were-(1) high energy/low protein(HELP),(2) high energy/high protein(HEHP),(3) low energy/low protein(LELP) and(4) low energy/high protein(LEHP).Each treatment used 6 steers and 6 heifers,except for HELP- 5 steers and 6 heifers.Results:Growth rate and final carcass weight were unaffected by dietary energy and protein levels or by sex.Compared with the LE diet group,the HE group had significantly lower dry matter intake(DMI,6.76 vs.7.48 kg DM/d),greater chest girth increments(46.1 vs.36.8 cm),higher carcass fat(19.9 vs.16.3%) and intramuscular fat content(29.9 vs.22.8%DM).The HE group also had improved yields of top and medium top grade commercial meat cuts(39.9 vs.36.5%).The dressing percentage was higher for the HP group than the LP group(53.4 vs.54.9%).Steers had a greater length increment(9.0 vs.8.3 cm),but lower carcass fat content(16.8 vs.19.4%) than heifers.The meat quality traits(shear force value,drip loss,cooking loss and water holding capacity) were not affected by treatments or sex,averaging 3.14 kg,2.5,31.5 and 52.9%,respectively.The nutritive profiles(both fatty and amino acid composition) were not influenced by the energy or protein levels or by sex.Conclusions:The dietary energy and protein levels and sex significantly influenced the carcass characteristics and chemical composition of meat but not thegrowth performance,meat quality traits and nutritive profiles.Lingyan Li Yuankui Zhu Xianyou Wang Yang He Binghai Cao 2014Journal of Animal Science and Biotechnology2014,5,4:21
2Toxicities and risk assessment of heavy metals in sediments of Taihu Lake, China, based on sediment quality guidelines显示文摘The occurrence, toxicities, and ecological risks of five heavy metals(Pb, Cu, Cd, Zn and Ni) in the sediment of Taihu Lake were investigated in this study. To evaluate the toxicities caused by the heavy metals, the toxicities induced by organic contaminants and ammonia in the sediments were screened out with activated carbon and zeolite. The toxicities of heavy metals in sediments were tested with benthic invertebrates(tubificid and chironomid).The correlations between toxicity of sediment and the sediment quality guidelines(SQGs)derived previously were evaluated. There were significant correlations(p < 0.0001) between the observed toxicities and the total risk quotients of the heavy metals based on SQGs,indicating that threshold effect level(TEL) and probable effect level(PEL) were reliable to predict the toxicities of heavy metals in the sediments of Taihu Lake. By contrast, the method based on acid volatile sulfides(AVS) and simultaneously extracted metals(SEM),such as ∑SEM/AVS and ∑SEM-AVS, did not show correlations with the toxicities. Moreover,the predictive ability of SQGs was confirmed by a total predicting accuracy of 77%. Ecological risk assessment based on TELs and PELs showed that the contaminations of Pb, Cu, Cd and Zn in the sediments of Taihu Lake were at relatively low or medium levels. 4Allele-aware chromosome-scale assembly of the allopolyploid genome of hexaploid Ma bamboo(Dendrocalamus latiflorus Munro)显示文摘Dendrocalamus latiflorus Munro is a woody clumping bamboo with rapid shoot growth.Both genetic transformation and clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein 9(Cas9)gene editing techniques are available for D.latiflorus,enabling reverse genetic approaches.Thus,D.latiflorus has the potential to be a model bamboo species.However,the genome sequence of D.latiflorus has remained unreported due to its polyploidy and large genome size.Here,we sequenced the D.latiflorus genome and assembled it into three allele-aware subgenomes(AABBCC),representingthe largest genome of a major bamboo species.We assembled 70 allelic chromosomes(2,737 Mb)for hexaploid D.latiflorus using both singlemolecule sequencing from the Pacific Biosciences(Pac Bio)Sequel platform and chromosome conformation capture sequencing(Hi-C).Repetitive sequences comprised 52.65%of the D.latiflorus genome.We annotated 135231 protein-coding genes in the genome based on transcriptomes from eight different tissues.Transcriptome sequencing using RNA-Seq and Pac Bio singlemolecule real-time long-read isoform sequencing revealed highly differential alternative splicing(AS)between non-abortive and abortive shoots,suggesting that AS regulates the abortion rate of bamboo shoots.This high-quality hexaploid genome and comprehensive strand-specific transcriptome datasets for this Poaceae family member will pave the way for bamboo research using D.latiflorus as a model species.Yushan Zheng Deming Yang Jundong Rong Liguang Chen Qiang Zhu Tianyou He Lingyan Chen Jing Ye Lili Fan Yubang Gao Hangxiao Zhang Lianfeng Gu 2022Journal of Integrative Plant Biology2022,64,3:6
5Prognostic value of interim ^(18)F-FDG PET/CT in diffuse large B-cell lymphoma显示文摘Objective: Diffuse large B-cell lymphoma (DLBCL) is a heterogeneous disease. The prognostic factor currently used is not accurate enough to predict the outcomes of patients with DLBCL. The prognostic significance of interim PET/CT in DLBCL remains controversial. The aim of this study is to determine the predictive value of interim 18F-FDG PET/CT after first-line treatment in patients with DLBCL. Methods: Thirty-two patients with DLBCL underwent baseline, interim and post-treatment 18F-FDG PET/CT scans. Imaging results were analyzed for the survival of patients via software SPSS 13.0, retrospectively. Results: Thirty-one of the 32 patients were treated with R-CHOP regimen, and interim 18F-FDG PET/CT of 24 patients was performed after 2 cycles of treatment. After a median follow-up period of 16.7 months, the 2-year progression-free survival (PFS) rates were significantly different between the groups above and below SUV max cut-off value of 2.5 (P=0.039). No significant differences were found in the 2-year PFS rates if SUV max cut-off values were set as 2.0 and 3.0, respectively (P=0.360; P=0.113). 6Adsorptive removal of hydrophobic organic compounds by carbonaceous adsorbents:A comparative study of waste-polymer-based,coal-based activated carbon,and carbon nanotubes显示文摘Adsorption of the hydrophobic organic compounds(HOCs) trichloroethylene(TCE),1,3-dichlorobenzene(DCB),1,3-dinitrobenzene(DNB) and γ-hexachlorocyclohexane(HCH) on five different carbonaceous materials was compared.The adsorbents included three polymer-based activated carbons,one coal-based activated carbon(F400) and multiwalled carbon nanotubes(MWNT).The polymerbased activated carbons were prepared using KOH activation from waste polymers:polyvinyl chloride(PVC),polyethyleneterephthalate(PET) and tire rubber(TR).Compared with F400 and MWNT,activated carbons derived from PVC and PET exhibited fast adsorption kinetics and high adsorption capacity toward the HOCs,attributed to their extremely large hydrophobic surface area(2700 m 2 /g) and highly mesoporous structures.Adsorption of small-sized TCE was stronger on the tire-rubber-based carbon and F400 resulting from the pore-filling effect.In contrast,due to the molecular sieving effect,their adsorption on HCH was lower.MWNT exhibited the lowest adsorption capacity toward HOCs because of its low surface area and characteristic of aggregating in aqueous solution.Fei Lian Chun Chang Yang Du Lingyan Zhu Baoshan Xing Chang Liu 2012Journal of Environmental Sciences2012,24,9:4
7Effect of chronic corticosterone-induced depression on circadian rhythms and age-related phenotypes in mice显示文摘Disrupted circadian rhythms are a recognized effect of depression,and our previous article demonstrated an association between depression and premature aging,but the underlying mechanisms are not well understood.In the present study,we used a mouse model of chronic corticosterone (CORT)-treated depression to elucidate a mechanism by which depression may be associated with the circadian clock and mediate age-related phenotypes.Mice received a daily injection of 20 mg/kg CORT for 21 consecutive days,and the depression-like behaviors of mice were identified by the sucrose intake test,tail suspension test and open field test.Our findings indicated that CORT injection may be correlatedwith the circadian clock by impairing circadian rhythms or shifting the phase values of clock genes.We also showed that CORT-treated mice exhibited a significant gradual reduction in body weight gain with increased oxidative stress, including reduced activity of antioxidant-related enzymes,reduced glutathione:glutathione disulfide ratio and cytochrome (Cyt)-C level,and elevated reactive oxygen species content.Moreover, chronic CORT injection affected inflammatory responses,the production of mitochondrial ATP and telomere shortening,which may be associated with the Sirtuin 3 (SIRT3)signaling pathway. 8Prognostic value of pre-and post-transplantation 18F-fluorodeoxyglucose positron emission tomography results in non-Hodgkin lymphoma patients receiving autologous stem cell transplantation显示文摘Objective: High-dose chemotherapy(HDC) followed by autologous stem cell transplantation(ASCT) is the standard of care in the upfront or relapsed/refractory setting in some patients with non-Hodgkin lymphoma(NHL).However,a proportion of patients do not respond to ASCT.^(18)F-fluorodeoxyglucose(FDG) positron emission tomography(PET)/computed tomography(CT) has been widely used for staging,response evaluation,and prognosis prediction.Here,we investigated the prognostic role of PET/CT in NHL patients before and after ASCT.Methods: A retrospective study was conducted at Peking University Cancer Hospital.All NHL patients who underwent ASCT between March 2010 and July 2016 were identified.Patients who had PET/CT scan before and after ASCT were included.Deauville criteria(5-point scale) were used to interpret PET scans.Univariate and multivariate survival analyses were performed using Cox regression.The predictive value of PET scanning was estimated by comparing the area under the receiver operating characteristic(ROC) curve.Results: In total,79 patients were enrolled in this study.In univariate analysis,pre-and post-ASCT PET result was identified as prognostic factors for 3-year progression-free survival(PFS) and overall survival(OS).Patients with negative pre-ASCT PET result demonstrated significantly better PFS(84.2% vs.54.2%) and OS(89.2% vs.63.6%) than patients with positive pre-ASCT PET result.PFS(91.6% vs.25.3%) and OS(96.5% vs.36.8%) were also significantly different between patients with negative and positive post-ASCT PET result.Multivariate analysis also showed a significant association between survival and post-ASCT PET result.ROC analysis revealed that the predictive value of post-ASCT PET result was superior to that of pre-ASCT PET result alone.Combined pre-and post-ASCT PET result is better for predicting outcomes in patients with NHL receiving transplantation.Deauville criteria score >3 was identified as the best cutoff value for post-ASCT PET.Conclusions: Post-ASCT PET result was more important than pre-ASCT PET result in predicting outcomes for NHL patients who underwent ASCT.The prognostic significance can be improved when combining preASCT PET result with post-ASCT PET result.Deauville criteria can be used for interpreting PET scans in this scenario.Zhitao Ying Lan Mi Xuejuan Wang Yuewei Zhang Zhi Yang Yuqin Song Xiaopei Wang WenZheng Ningjing Lin Meifeng Tu Yan Xie Lingyan Ping Chen Zhang Weiping Liu LijuanDeng Jun Zhu 2017Chinese Journal of Cancer Research2017,29,6:3
9Artificial intelligence-assisted automatic and index-based microbial single-cell sorting system for One-Cell-One-Tube显示文摘Identification,sorting,and sequencing of individual cells directly from in situ samples have great potential for in-depth analysis of the structure and function of microbiomes.In this work,based on an artificial intelligence(AI)-assisted object detection model for cell phenotype screening and a cross-interface contact method for single-cell exporting,we developed an automatic and index-based system called EasySort AUTO,where individual microbial cells are sorted and then packaged in a microdroplet and automatically exported in a precisely indexed,“One-Cell-One-Tube”manner.The target cell is automatically identified based on an AI-assisted object detection model and then mobilized via an optical tweezer for sorting.Then,a crossinterface contact microfluidic printing method that we developed enables the automated transfer of cells from the chip to the tube,which leads to coupling with subsequent single-cell culture or sequencing.The efficiency of the system for single-cell printing is>93%.The throughput of the system for single-cell printing is~120 cells/h.Moreover,>80%of single cells of both yeast and Escherichia coli are culturable,suggesting the superior preservation of cell viability during sorting.Finally,AI-assisted object detection supports automated sorting of target cells with high accuracy from mixed yeast samples,which was validated by downstream single-cell proliferation assays.The automation,index maintenance,and vitality preservation of EasySort AUTO suggest its excellent application potential for single-cell sorting.Zhidian Diao Lingyan Kan Yilong Zhao Jingyun Song Chen Wang Yang Liu Fengli Zhang Teng Xu Rongze Chen Xixian Wang Xiaoyan Jing Jian Xu Yuandong Li Bo Ma 2022mLife2022,1,4:2
10A fluorescent probe operating under weak acidic conditions for the visualization of HOCl in solid tumors in vivo显示文摘Levels of reactive oxygen species(ROS)in cancer cells or in the tumor microenvironment differ noticeably from those in normal cells and cellular microenvironments because ROS play important roles in all aspects of tumor physiology.However,due to the lack of adequate tools,it is difficult to study the relationship between ROS,especially certain types of ROS(e.g.,HOCl),and cancer.We report herein an HOCl-specific fluorescent probe,FDOCl-20,containing a thiocarbamide group as a receptor,for the visualization of HOCl in solid tumors in vivo.This probe displays high selectivity and sensitivity to HOCl,and is appropriate for use in acidic conditions,including the tumor microenvironment.Using FDOCl-20 as a tool,we can visualize HOCl in solid tumors in vivo.Importantly,the fluorescent intensity of FDOCl-20 is proportional to tumor volume.Thus,FDOCl-20 is a useful tool to investigate the relationship between HOCl and the physiological processes of tumors.Peng Wei Lingyan Liu Wei Yuan Jiajia Yang Ruohan Li Tao Yi 2020Science China Chemistry2020,63,8:2

15Prognostic value of ^(18)F-fluorodeoxyglucose positron emission tomography using Deauville criteria in diffuse large B cell lymphoma treated with autologous hematopoietic stem cell transplantation显示文摘Objective: High-dose chemotherapy(HDC) followed by autologous hematopoietic stem cell transplantation(auto-HSCT) plays an important role in improving outcomes of diffuse large B cell lymphoma(DLBCL) patients.18 F-fluorodeoxyglucose(18 F-FDG) positron emission tomography(PET)/computed tomography(CT) has been widely accepted in response assessment and prediction of prognosis in DLBCL. Here, we report the value of 18 FFDG PET/CT pre-and post-HSCT in predicting outcomes of patients with DLBCL.Methods: DLBCL patients who had PET/CT scan before and after HSCT were included. PET results were interpreted based upon Deauville criteria. The prognostic value of 18 F-FDG PET/CT in auto-HSCT was evaluated.Results: Eighty-four patients were enrolled. In univariate analysis, pre-and post-HSCT PET findings were correlated with 3-year progression-free survival(PFS) [hazard ratio(HR)=4.391, P=0.001; HR=7.607, P<0.001] and overall survival(OS)(HR=4.792, P=0.008; HR=26.138, P<0.001). Patients receiving upfront auto-HSCT after firstline treatment had better outcomes than relapsed/refractory DLBCL patients(3-year PFS, P<0.001; 3-year OS,P<0.001). In the relapsed/refractory patients, pre-and post-HSCT PET findings were also associated with 3-year PFS(P=0.003 vs. P<0.001) and OS(P=0.027 vs. P<0.001). A significant correlation was observed between clinical response to chemotherapy before auto-HSCT and outcomes of patients in the entire cohort(3-year PFS, P<0.001;3-year OS, P<0.001) and in the subgroup of 21 patients with positive pre-HSCT PET(3-year PFS, P=0.084; 3-year OS, P=0.240). A significant association between survival and post-HSCT PET findings was observed in multivariate analysis(HR=5.168, P<0.001).Conclusions: PET results before and after HSCT are useful prognostic factors for DLBCL patients receiving HSCT. 17Phylogenetic Analysis among Maize Exserohilum turcicum Isolates from Yunnan Province by RAPD显示文摘[Objective] This study aimed to investigate the genetic variation of Yunnan Exserohilum turcicum isolates from molecular level,and provide theoretical basis for E. turcicum pathogenicity virulence differentiation and effective control of disease.[Method] A total of 56 E. turcicum isolates from some areas of Yunnan Province were analyzed by RAPD. Based on the genetic distance,a dendrogram was constructed.[Result] Ten genetic groups were formed in the dendrogram. The RAPD groups had no obvious correlation with geographic origins. 20Bidirectional differentiation of BMSCs induced by a biomimetic procallus based on a gelatin-reduced graphene oxide reinforced hydrogel for rapid bone regeneration显示文摘Developmental engineering strategy needs the biomimetic composites that can integrate the progenitor cells,biomaterial matrices and bioactive signals to mimic the natural bone healing process for faster healing and reconstruction of segmental bone defects.We prepared the gelatin-reduced graphene oxide(GOG)and constructed the composites that mimicked the procallus by combining the GOG with the photo-crosslinked gelatin hydrogel.The biological effects of the GOG-reinforced composites could induce the bi-differentiation of bone marrow stromal cells(BMSCs)for rapid bone repair.The proper ratio of GOG in the composites regulated the composites’mechanical properties to a suitable range for the adhesion and proliferation of BMSCs.Besides,the GOG-mediated bidirectional differentiation of BMSCs,including osteogenesis and angiogenesis,could be activated through Erk1/2 and AKT pathway.The methyl vanillate(MV)delivered by GOG also contributed to the bioactive signals of the biomimetic procallus through priming the osteogenesis of BMSCs.During the repair of the calvarial defect in vivo,the initial hypoxic condition due to GOG in the composites gradually transformed into a well-vasculature robust situation with the bi-differentiation of BMSCs,which mimicked the process of bone healing resulting in the rapid bone regeneration.As an inorganic constituent,GOG reinforced the organic photo-crosslinked gelatin hydrogel to form a double-phase biomimetic procallus,which provided the porous extracellular matrix microenvironment and bioactive signals for the bi-directional differentiation of BMSCs.These show a promised application of the bio-reduced graphene oxide in biomedicine with a developmental engineering strategy.Delong Jiao Ao Zheng Yang Liu Xiangkai Zhang Xiao Wang Jiannan Wu Wenjun She Kaige Lv Lingyan Cao Xinquan Jiang 2021Bioactive Materials2021,6,7:1
