| 2 | Isolation,characterization,and phylogenic analysis of three new severe fever with thrombocytopenia syndrome bunyavirus strains derived from Hubei Province,China显示文摘Hubei Province is a major epidemic area of severe fever with thrombocytopenia syndrome bunyavirus(SFTSV) in China. However, to date, a few SFTSV strains have been isolated from Hubei Province, preventing effective studies of epidemic outbreaks. Here, we report three confirmed patients(2015–2016) with typical symptoms of severe fever with thrombocytopenia syndrome disease(SFTS) who were farmers resident in different regions in Hubei Province. Three new SFTSV strains were isolated from the serum samples of each patient. Characterization of viral growth properties showed that there were no significant differences in virus production. All strains were completely sequenced, and phylogenetic analysis showed that unlike the other strains from Hubei province, which belonged to the SFTSV C3 genotype, one of the three strains belonged to the SFTSV C2 genotype. | 3 | Study on Graphene Oxide Modified Inorganic Phase Change Materials and Their Packaging Behavior显示文摘We prepared the nano-inorganic phase-change 'alloy' materials through the modification of Na_2SO_4·10H_2O using Na_2HPO_4·12H_2O and GO nano-nucleating agent, and further investigated their thermophysical properties such as melting/solidification temperatures and enthalpies via differential scanning calorimetry. When the weight ratio of Na_2SO_4·10H_2O and Na_2HPO_4·12H_2O was 8:2 and the weight ratio of graphene oxide was 0.5% of phase change material, the phase change 'alloy' material showed excellent performances, specifically, the melting temperature and latent heat were found to be 22 ℃ and 190 J/g with a degree of subcooling decreased from 8.6℃ to 2.1℃. In order to extend the application of the phase change 'alloy' material to building energy saving field, it was adsorbed on expanded glass beads under vacuum and further covered with diatomite. | 4 | Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. | 5 | The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. | 6 | Degree of Brine Evaporation and Origin of the Mengyejing Potash Deposit:Evidence from Fluid Inclusions in Halite显示文摘The Mengyejing potash deposit is located in the southern port of the Simao Basin, Yunnan Province, and is hosted in mid-Cretaceous strata. The chemical compositions of fluid inclusions in halite crystals, collected from the level-610 adit in the deposit, were analysed by laser ablation inductively coupled plasma mass spectrometry(LA-ICP-MS). The results show that the brine is of the Na-K-Mg-Ca-Cl type and has K concentrations that are distinctly higher than those of Mg and Ca, unlike normal brines associated with Cretaceous halite. The high K concentrations indicate that the degree of evaporation of the ancient Mengyejing saline lake was very high, reaching the sylvite deposition stage but rarely reaching the carnallite deposition stage. The trajectory of the H and O isotopic compositions of the brines in the halite-hosted fluid inclusions corresponds to intense evaporation, indicating that the net evaporation exceeded the net inflow of brines. These brine compositions in halite-hosted fluid inclusions were likely formed by the dissolution of previously deposited K-bearing minerals by fresh continental and/or seawater, forming a type of modified seawater, with deep hydrothermal fluids potentially supplying additional potassium. | 7 | Cloning and expression of two sterol C-24 methyltransferase genes from upland cotton(Gossypium hirsuturm L.)显示文摘Brassinosteroids (BRs) are an important class of plant steroidal hormones that are essential in a wide variety of physiological proc-esses. Two kinds of intermediates,sitosterol and campesterol,play a crucial role in cell elongation,cellulose biosynthesis,and accumula-tion. To illuminate the effects of sitosterol and campesterol on the development of cotton (Gossypium hirsuturm L.) fibers through screening cotton fiber EST database and contigging the candidate ESTs,two key genes GhSMT2-1 and GhSMT2-2 controlling the sitos-terol biosynthesis were cloned from developing fibers of upland cotton cv. Xuzhou 142. The full length of GhSMT2-1 was 1,151 bp,in-cluding an 8 bp 5′-untranslated region (UTR),a 1,086 bp open reading frame (ORF),and a 57 bp 3′-UTR. GhSMT2-1 gene encoded a polypeptide of 361 amino acid residues with a predicted molecular mass of 40 kDa. The full length of GhSMT2-2 was 1,166 bp,including an 18 bp 5′-UTR,a 1,086 bp ORF,and a 62 bp 3′-UTR. GhSMT2-2 gene encoded a polypeptide of 361 amino acid residues with a pre-dicted molecular mass of 40 kDa. The two deduced amino acid sequences had high homology with the SMT2 from Arabidopsis thaliana and Nicotiana tabacum. Furthermore,the typical conserved structures characterized by the sterol C-24 methyltransferase,such as region I (LDVGCGVGGPMRAI),region Ⅱ (IEATCHAP),and region Ⅲ (YEWGWGQSFHF),were present in both deduced proteins. Southern blotting analysis indicated that GhSMT2-1 or GhSMT2-2 was a single copy in upland cotton genome. Quantitative real-time RT-PCR analysis revealed that the highest expression levels of both genes were detected in 10 DPA (day post anthesis) fibers,while the lowest levels were observed in cotyledon and leaves. The expression level of GhSMT2-1 was 10 times higher than that of GhSMT2-2 in all the organs and tissues detected. These results indicate that the homologue of sterol C-24 methyltransferase gene was cloned from upland cotton and both GhSMT2 genes play a crucial role in fiber elongation. | 8 | Classification of the Gut Microbiota of Patients in Intensive Care Units During Development of Sepsis and Septic Shock显示文摘The gut microbiota of intensive care unit(ICU)patients displays extreme dysbiosis associated with increased susceptibility to organ failure,sepsis,and septic shock.However,such dysbiosis is difficult to characterize owing to the high dimensional complexity of the gut microbiota.We tested whether the concept of enterotype can be applied to the gut microbiota of ICU patients to describe the dysbiosis.We collected 131 fecal samples from 64 ICU patients diagnosed with sepsis or septic shock and performed 16S rRNA gene sequencing to dissect their gut microbiota compositions.During the development of sepsis or septic shock and during various medical treatments,the ICU patients always exhibited two dysbiotic microbiota patterns,or ICU-enterotypes,which could not be explained by host properties such as age,sex,and body mass index,or external stressors such as infection site and antibiotic use.ICU-enterotype I(ICU E1)comprised predominantly Bacteroides and an unclassified genus of Enterobacteriaceae,while ICU-enterotype II(ICU E2)comprised predominantly Enterococcus.Among more critically ill patients with Acute Physiology and Chronic Health Evaluation II(APACHE II)scores>18,septic shock was more likely to occur with ICU E1(P=0.041).Additionally,ICU E1 was correlated with high serum lactate levels(P=0.007).Therefore,different patterns of dysbiosis were correlated with different clinical outcomes,suggesting that ICU-enterotypes should be diagnosed as independent clinical indices.Thus,the microbial-based human index classifier we propose is precise and effective for timely monitoring of ICU-enterotypes of individual patients.This work is a first step toward precision medicine for septic patients based on their gut microbiota profiles. | Wanglin Liu Mingyue Cheng Jinman Li Peng Zhang Hang Fan Qinghe Hu Maozhen Han Longxiang Su Huaiwu He Yigang Tong Kang Ning Yun Long | 2020 | Genomics, Proteomics & Bioinformatics2020,18,6: | 7 |
| 9 | Phytosterol content and the campesterol:sitosterol ratio influence cotton fiber development: role of phytosterols in cell elongation显示文摘Phytosterols play an important role in plant growth and development,including cell division,cell elongation,embryogenesis,cellulose biosynthesis,and cell wall formation.Cotton fiber,which undergoes synchronous cell elongation and a large amount of cellulose synthesis,is an ideal model for the study of plant cell elongation and cell wall biogenesis.The role of phytosterols in fiber growth was investigated by treating the fibers with tridemorph,a sterol biosynthetic inhibitor.The inhibition of phytosterol biosynthesis resulted in an apparent suppression of fiber elongation in vitro or in planta.The determination of phytosterol quantity indicated that sitosterol and campesterol were the major phytosterols in cotton fibers;moreover,higher concentrations of these phytosterols were observed during the period of rapid elongation of fibers.Furthermore,the decrease and increase in campesterol:sitosterol ratio was associated with the increase and decease in speed of elongation,respectively,during the elongation stage.The increase in the ratio was associated with the transition from cell elongation to secondary cell wall synthesis.In addition,a number of phytosterol biosynthetic genes were down-regulated in the short fibers of ligon lintless-1mutant,compared to its near-isogenic wild-type TM-1.These results demonstrated that phytosterols play a crucial role in cotton fiber development,and particularly in fiber elongation. | Shasha Deng Ting Wei Kunling Tan Mingyu Hu Fang Li Yunlan Zhai Shue Ye Yuehua Xiao Lei Hou Yan Pei Ming Luo | 2016 | Science China(Life Sciences)2016,59,2: | 5 |
| 10 | Measurements of ^(134)Cs and ^(137)Cs in urine and estimation of the internal dose of an adult exposed to the Chernobyl Accident显示文摘To estimate the internal dose of a Chinese visiting scholar internally contaminated because of the Cherno- byl Accident, the contents of 134Cs and 137Cs in urine were measured using a Ge(Li) γ-spectrometer. The internal doses were calculated based on data from the ICRP Publications. The effective doses from 134Cs and 137Cs were estimated to be 61 μSv and 98 μ Sv, respectively. The sum of 159 μSv was lower than the total effective dose (310 μSv), from the inhalation and ingestion of natural radionuclides. The dose of 131I was also reviewed referring to the UNSCEAR 2000 Report. The equivalent effective dose of 131I was estimated to be 2.9 mSv, 18 times more than the amount of 134Cs and 137Cs. | 11 | Synthesis and enhanced photocatalytic performance of Ag/AgCl/TiO^(2)nanocomposites prepared by ion exchange method显示文摘Ag/AgCl/TiO^(2)nanocomposite was synthesized through the combination of sol-gel process and hydrothermal(HT)method followed by ion exchange method in the paper.Transmission electron microscope,UVevis absorption property and X-ray diffraction were utilized to characterize the morphology,optical absorption property and phase structure of the as-prepared photocatalysts.The morphological and performance studies of Ag/AgCl/TiO^(2)revealed that a well-interconnected mixed phase heterojunction photocatalyst was achieved through as-prepared TiO^(2)and the subsequent ion exchange method.By the degradation of Rhodamine B under xenon lamp,Ag/AgCl/TiO^(2)exhibited a significant enhancement in photocatalytic activity compared with TiO^(2)and Ag/TiO^(2).The band gap of Ag/AgCl/TiO^(2)can reach to 2.53eV compared with 3.22eV of the prepared TiO^(2).Furthermore,the synthesis process and photoreaction mechanisms of TiO^(2),Ag/TiO^(2)and Ag/AgCl/TiO^(2)photocatalysts were proposed.It demonstrated that the enhancement of photocatalytic was mainly due to the reducing of electron-hole recombination in Ag/AgCl/TiO^(2). | Qionglian Yang Mingyu Hu Jun Guo Zhenhua Ge Jing Feng | 2018 | Journal of Materiomics2018,4,4: | 5 |
| 12 | Arterial relaxation is coupled to inhibition of mitochondrial fission in arterial smooth muscle cells:comparison of vasorelaxant effects of verapamil and phentolamine显示文摘Mitochondria are morphologically dynamic organelles which undergo fission and fusion processes. Our previous study found that arterial constriction was always accompanied by increased mitochondrial fission in smooth muscle cells, whereas inhibition of mitochondrial fission in smooth muscle cells was associated with arterial relaxation. Here, we used the typical vasorelaxants, verapamil and phentolamine, to further confirm the coupling between arterial constriction and mitochondrial fission in rat aorta. Results showed that phentolamine but not verapamil induced vasorelaxation in phenylephrine(PE)-induced rat thoracic aorta constriction. Verapamil, but not phentolamine, induced vasorelaxation in high K^+(KPSS)-induced rat thoracic aorta constriction. Pre-treatment with phentolamine prevented PEbut not KPSS-induced aorta constriction and pre-treatment with verapamil prevented both PE-and KPSSinduced aorta constriction. Transmission electron microscopy(TEM) results showed that verapamil but not phentolamine inhibited KPSS-induced excessive mitochondrial fission in aortic smooth muscle cells,and verapamil prevented both PE-and KPSS-induced excessive mitochondrial fission in aortic smoothmuscle cells. Verapamil inhibited KPSS-induced excessive mitochondrial fission in cultured vascular smooth muscle cells(A10). | 13 | Establishment of a Reverse Genetic System of Severe Fever with Thrombocytopenia Syndrome Virus Based on a C4 Strain显示文摘Severe fever with thrombocytopenia syndrome virus(SFTSV)is an emerging tick-borne bunyavirus that causes hemorrhagic fever-like disease(SFTS)in humans with a case fatality rate up to 30%.To date,the molecular biology involved in SFTSV infection remains obscure.There are seven major genotypes of SFTSV(C1-C4 and J1-J3)and previously a reverse genetic system was established on a C3 strain of SFTSV.Here,we reported successfully establishment of a reverse genetics system based on a SFTSV C4 strain.First,we obtained the 5’-and 3’-terminal untranslated region(UTR)sequences of the Large(L),Medium(M)and Small(S)segments of a laboratory-adapted SFTSV C4 strain through rapid amplification of cDNA ends analysis,and developed functional T7 polymerase-based L-,M-and S-segment minigenome assays.Then,fulllength cDNA clones were constructed and infectious SFTSV were recovered from co-transfected cells.Viral infectivity,growth kinetics,and viral protein expression profile of the rescued virus were compared with the laboratory-adapted virus.Focus formation assay showed that the size and morphology of the foci formed by the rescued SFTSV were indistinguishable with the laboratory-adapted virus.However,one-step growth curve and nucleoprotein expression analyses revealed the rescued virus replicated less efficiently than the laboratory-adapted virus.Sequence analysis indicated that the difference may be due to the mutations in the laboratory-adapted strain which are more prone to cell culture.The results help us to understand the molecular biology of SFTSV,and provide a useful tool for developing vaccines and antivirals against SFTS. | Mingyue Xu Bo Wang Fei Deng Hualin Wang Manli Wang Zhihong Hu Jia Liu | 2021 | Virologica Sinica2021,36,5: | 5 |
| 14 | Discovering unusual structures from exception using big data and machine learning techniques显示文摘Recently, machine learning(ML) has become a widely used technique in materials science study. Most work focuses on predicting the rule and overall trend by building a machine learning model. However,new insights are often learnt from exceptions against the overall trend. In this work, we demonstrate that how unusual structures are discovered from exceptions when machine learning is used to get the relationship between atomic and electronic structures based on big data from high-throughput calculation database. For example, after training an ML model for the relationship between atomic and electronic structures of crystals, we find AgO2 F, an unusual structure with both Ag3+and O22à, from structures whose band gap deviates much from the prediction made by our model. | 18 | The Variation of Stone Cell Content in 236 Germplasms of Sand Pear(Pyrus pyrifolia)and Identification of Related Candidate Genes显示文摘Stone cells have been described to substantially influence pear fruit quality,as lignin and cellulose are the main components of stone cells.However,there are limited studies on the relationship between the variation and molecular basis of stone cells,lignin and cellulose content among different pear varieties.Here,to reveal the variation of stone cell content within different cultivated species,we collected 236 germplasms of sand pear(Pyrus pyrifolia)at 50 days after flower blooming(DAFB),the key stage of stone cell formation.In our results,we measured the content of stone cells,lignin and cellulose and found that these contents ranged from2.82%to 29.00%,8.84%to 55.30%and 11.52%to 30.55%,respectively.Further analysis showed that the variation coefficient of stone cell,lignin and cellulose content was 39.10%,28.03%and 16.71%,respectively.Additionally,a significant correlation between stone cell,lignin and cellulose content were detected,and the correlation coefficient between the contents of stone cell and lignin(0.912)was higher than between the contents of stone cell and cellulose(0.796).Moreover,the average lignin content(29.73%)was higher than the average cellulose content(18.03%)in stone cells in pear fruits,indicating that lignin is the main component of stone cell in pears.Finally,on the basic of the transcriptome data,we identified 10 transcription factors belonging to bHLH,ERF,MYB,and NAC transcript families,which might be involved in lignin formation in stone cells.qRT-PCR experiments verified coincident trends between expression of candidate genes and stone cell content.This research laid foundation for future studies on genetic variation of stone cells in pear fruits and provided important gene resources for stone cell regulation. | Jiaying Zhang Jiaming Li Cheng Xue Runze Wang Mingyue Zhang Kaijie Qi Jing Fan Hongju Hu Shaoling Zhang Jun Wu | 2021 | Horticultural Plant Journal2021,7,2: | 3 |
