3Pathophysiological mechanisms and therapeutic approaches in obstructive sleep apnea syndrome显示文摘Obstructive sleep apnea syndrome(OSAS)is a common breathing disorder in sleep in which the airways narrow or collapse during sleep,causing obstructive sleep apnea.The prevalence of OSAS continues to rise worldwide,particularly in middle-aged and elderly individuals.The mechanism of upper airway collapse is incompletely understood but is associated with several factors,including obesity,craniofacial changes,altered muscle function in the upper airway,pharyngeal neuropathy,and fluid shifts to the neck.The main characteristics of OSAS are recurrent pauses in respiration,which lead to intermittent hypoxia(IH)and hypercapnia,accompanied by blood oxygen desaturation and arousal during sleep,which sharply increases the risk of several diseases.This paper first briefly describes the epidemiology,incidence,and pathophysiological mechanisms of OSAS.Next,the alterations in relevant signaling pathways induced by IH are systematically reviewed and discussed.For example,IH can induce gut microbiota(GM)dysbiosis,impair the intestinal barrier,and alter intestinal metabolites.These mechanisms ultimately lead to secondary oxidative stress,systemic inflammation,and sympathetic activation.We then summarize the effects of IH on disease pathogenesis,including cardiocerebrovascular disorders,neurological disorders,metabolic diseases,cancer,reproductive disorders,and COVID-19.Finally,different therapeutic strategies for OSAS caused by different causes are proposed.Multidisciplinary approaches and shared decision-making are necessary for the successful treatment of OSAS in the future,but more randomized controlled trials are needed for further evaluation to define what treatments are best for specific OSAS patients.Renjun Lv Xueying Liu Yue Zhang Na Dong Xiao Wang Yao He Hongmei Yue Qingqing Yin 2023Signal Transduction and Targeted Therapy2023,8,6:8
4Graphene-based nanomaterials in biosystems显示文摘Graphene-based nanomaterials have emerged as a novel type of materials with exceptional physicochemical properties and numerous applications in various areas. In this review, we summarize recent advances in studying interactions between graphene and biosystems. We first provide a brief introduction on graphene and its derivatives, and then discuss on the toxicology and biocompatibility of graphene, including the extracellular interactions between graphene and biomacromolecules, cellular studies of graphene, and in vivo toxicological effects. 5A novel tomato SUMO E_3 ligase,SlSIZ_1,confers drought tolerance in transgenic tobacco显示文摘SUMOylation is an important post-translational modification process that regulates different cellular functions in eukaryotes.SIZ/PIAS-type SAP and Miz1(SIZ_1) proteins exhibit SUMO E_3 ligase activity,which modulates SUMOylation.However,SIZ_1 in tomato has been rarely investigated.In this study,a tomato SIZ_1 gene(Sl SIZ_1)was isolated and its molecular characteristics and role in tolerance to drought stress are described.Sl SIZ_1 was upregulated by cold,sodium chloride(Na Cl),polyethylene glycol(PEG),hydrogen peroxide(H_2O_2) and abscisic acid(ABA),and the corresponding proteins were localized in the nucleus.The expression of Sl SIZ_1 in Arabidopsis thaliana(Arabidopsis) siz1-2 mutants partially complemented the phenotypes of dwarf,cold sensitivity and ABA hypersensitivity.Sl SIZ_1 also exhibited the activity of SUMO E_3 ligase to promote the accumulation of SUMO conjugates.Under drought stress,the ectopic expression of Sl SIZ_1 in transgenic tobacco lines enhanced seed germination and reduced the accumulation of reactive oxygen species.SlSIZ_1 overexpression conferred the plants with improved growth,high free proline content,minimal malondialdehyde accumulation and increased accumulation of SUMO conjugates.Sl SIZ_1 is a functional homolog of Arabidopsis SIZ_1 with SUMO E_3 ligase activity.Therefore,overexpression of Sl SIZ_1 enhanced the tolerance of transgenic tobacco to drought stress.Song Zhang Kunyang Zhuang Shiju Wang Jinlian Lv Na'na Ma Qingwei Meng 2017Journal of Integrative Plant Biology2017,59,2:7
6miR-137 inhibits melanoma cell proliferation through downregulation of GLO1显示文摘Late-stage melanoma is refractory to current therapies. MicroRNAs (miRNAs) can modulate many physiological and pathological processes of melanoma. Studies have demonstrated that miR-137 acts as a tumor suppressor by inhibiting the proliferation of melanoma cells through targeting multiple mRNAs. The glyoxalase system member glyoxalase 1 (GLO1) is the principal scavenging enzyme of methylglyoxal (MG), a toxic byproduct of glycolysis. Using 35S in vivo/vitro labelling analysis for dynamic proteomics (SiLAD), we found that miR-137 downregulated the expression of GLO1 in melanoma cells.Bioinformatics analysis predicted that GLO1 is a direct target of miR-137. This was validated by dual luciferase reporter assay.Quantitative RT-PCR (qRT-PCR) and western blot analysis indicated that miR-137 could decrease endogenous GLO1 expression.Furthermore, siRNA targeting of GLO1 mimicked inhibition of melanoma cell proliferation caused by miR-137 overexpression.Re-expression of GLO1 was able to restore miR-137-mediated suppression of melanoma cell proliferation. 8A critical epitope in CD147 facilitates memory CD4^(+) T-cell hyper-activation in rheumatoid arthritis显示文摘The abnormal activation of CD4^(+)CD45RO+memory T(Tm)cells plays an important role in the pathogenesis of rheumatoid arthritis(RA).Previous studies have shown that CD147 participates in T-cell activation.However,it remains unclear whether CD147 is involved in abnormal Tm-cell activation in RA patients.In this study,we demonstrated that CD147 was predominantly upregulated in Tm cells derived from RA patients.The anti-CD147 mAb 5A12 specifically inhibited Tm-cell activation and proliferation and further restrained osteoclastogenesis.Using a structural–functional approach,we depicted the interface between 5A12 and CD147.This allowed us to identify two critical residues,Lys63 and Asp65,as potential targets for RA treatment,as the double mutation K63A/D65A inhibited Tm-cell activation,mimicking the neutralization by 5A12.This study provides not only a theoretical basis for a“CD147-Tm/Osteoclast-RA chain”for the potential prevention and treatment of RA or other T-cell-mediated autoimmune diseases but also a new target for related drug design and development.Na Guo Sheng Ye Kui Zhang Xiaoling Yu Hongyong Cui Xiangmin Yang Peng lin Minghua Lv Jinlin Miao Yang Zhang Qing Han Rongguang Zhang Zhinan Chen Ping Zhu 2019Cellular & Molecular Immunology2019,16,6:6
9Synthesis and biological evaluation of novel triazole derivatives as antifungal agents显示文摘A series of 1-(benzylamino)-2-(2,4-difluorophenyl)-3-(1H-1,2,4-triazol-l-yl)propan-2-ols compounds were synthesized and evaluated for their antifungal activities in vitro.The results showed that compounds 6A and 6B exhibited good antifungal activity.Compound 6A8 showed the strongest antifungal activity,which was significantly higher than that of the lead compounds and positive-control drugs Fluconazole and Itraconazole.In particular,the antifungal activity of compound 6A8 against Candida albicans and Candida krusei(MIC80 both at 0.00097μg/mL) was 515 and 64 times that of Fluconazole,respectively.The structure-activity relationships of the synthesized compounds were discussed,and the docking model of the target compounds with fungal lanosterol 14α-demethylase (CYP51) was analyzed.Hui Tang Can-Hui Zheng Xiao-Hui Ren Jia Liu Na Liu Jia-Guo Lv Ju Zhu You-Jun Zhou 2013Chinese Chemical Letters2013,24,3:6
10The jasmonate-induced bHLH gene SlJIG functions in terpene biosynthesis and resistance to insects and fungus显示文摘Jasmonic acid(JA)is a key regulator of plant defense responses.Although the transcription factor MYC2,the master regulator of the JA signaling pathway,orchestrates a hierarchical transcriptional cascade that regulates the JA responses,only a few transcriptional regulators involved in this cascade have been described.Here,we identified the basic helix-loop-helix(bHLH)transcription factor gene in tomato(Solanum lycopersicum),METHYL JASMONATE(MeJA)-INDUCED GENE(SlJIG),the expression of which was strongly induced by MeJA treatment.Genetic and molecular biology experiments revealed that SlJIG is a direct target of MYC2.SlJIG knockout plants generated by gene editing had lower terpene contents than the wild type from the lower expression of TERPENE SYNTHASE(TPS)genes,rendering them more appealing to cotton bollworm(Helicoverpa armigera).Moreover,SlJIG knockouts exhibited weaker JA-mediated induction of TPSs,suggesting that SlJIG may participate in JA-induced terpene biosynthesis.Knocking out SlJIG also resulted in attenuated expression of JA-responsive defense genes,which may contribute to the observed lower resistance to cotton bollworm and to the fungus Botrytis cinerea.We conclude that SlJIG is a direct target of MYC2,forms a MYC2-SlJIG module,and functions in terpene biosynthesis and resistance against cotton bollworm and B.cinerea.Yunyun Cao Lun Liu Kangsheng Ma Wenjing Wang Hongmei Lv Ming Gao Xinman Wang Xichun Zhang Shuxin Ren Na Zhang Yang-Dong Guo 2022Journal of Integrative Plant Biology2022,64,5:4

13Interaction and independence on methane oxidation of landfill cover soil among three impact factors:water,oxygen and ammonium显示文摘To understand the influence patterns and interactions of three important environmental factors,i.e.soil water content,oxygen concentration,and ammonium addition,on methane oxidation,the soils from landfill cover layers were incubated under full factorial parameter settings.In addition to the methane oxidation rate,the quantities and community structures of methanotrophs were analyzed to determine the methane oxidation capacity of the soils.Canonical correspondence analysis was utilized to distinguish the important impact factors.Water content was found to be the most important factor influencing the methane oxidation rate and Type II methanotrophs,and the optimum value was 15%(w/w),which induced methane oxidation rates 10-and 6-times greater than those observed at 5%(w/w)and 20%(w/w),respectively.Ambient oxygen conditions were more suitable for methane oxidation than 3%oxygen.The addition of 100 mg-N·kg^(-1) drysoil of ammonium induced different effects on methane oxidation capacity when conducted at low or high water content.With regard to the methanotrophs,Type II was sensitive to the changes of water content,while Type I was influenced by oxygen content.Furthermore,the methanotrophic acidophile,Verrucomicrobia,was detected in soils with a pH of 4.9,which extended their known living environments.Pinjing HE Na YANG Wenjuan FANG Fan LV Liming SHAO 2011Frontiers of Environmental Science & Engineering2011,5,2:3
14A nanotheranostic agent based on Nd^(3+)-doped YVO_(4) with blood-brain-barrier permeability for NIR-Ⅱ fluorescence imaging/magnetic resonance imaging and boosted sonodynamic therapy of orthotopic glioma显示文摘The specific diagnosis and treatment of gliomas is a primary challenge in clinic due to their high invasiveness and blood-brain barrier(BBB)obstruction.It is highly desirable to find a multifunctional agent with good BBB penetration for precise theranostics.Herein,we design and construct a core-shell structured nanotheranostic agent(YVO_(4):Nd^(3+)-HMME@MnO_(2)-LF,marked as YHM)with YVO_(4)∶Nd^(3+) particles as the core and MnO_(2) nanosheets as the shell.Sonosensitizer hematoporphyrinmonomethyl ether(HMME)and lactoferrin(LF)were further loaded and modified on the surface,giving it a good ability to cross the BBB,near-infrared fluorescence imaging in the second window(NIR-Ⅱ)/magnetic resonance imaging(MRI)bimodality,and highly efficient sonodynamic therapy(SDT)of orthotopic gliomas.The YVO_(4):Nd^(3+)(25%)core exhibited good NIR-Ⅱ fluorescence properties,enabling YHM to act as promising probes for NIR-Ⅱ fluorescence imaging of vessels and orthotopic gliomas.MnO_(2) shell can not only provide O2 in the tumor microenvironments(TME)to significantly improve the healing efficacy of SDT,but also release Mn^(2+) ions to achieve T1-weight MRI in situ.Non-invasive SDT can effectively restrain tumor growth.This work not only demonstrates that multifunctional YHM is promising for diagnosis and treatment of orthotopic glioma,but also provides insights into exploring the theranostic agents based on rare earth-doped yttrium vanadate nanoparticles.Zhijia Lv Longhai Jin Yue Cao Hao Zhang Dongzhi Xue Na Yin Tianqi Zhang Yinghui Wang Jianhua Liu Xiaogang Liu Hongjie Zhang 2022Light(Science & Applications)2022,11,6:3

16Effects of a combination of Japanese Raisin Tree Seed and Flower of Lobed Kudzuvine against acute alcohol-induced liver injury in mice显示文摘Objective:The Flower of Lobed Kudzuvine[Pueraria lobata(Willd.)Ohwi;Gehua in Chinese;GH]and Japanese Raisin Tree Seed(Hovenia dulcis Thunnb.;Zhijuzi in Chinese;ZJZ)are herbs that have been used in China for the treatment of alcohol intoxication and liver diseases.We aimed to evaluate the hepatoprotective potential of a combination of these in mice with acute alcohol-induced liver injury,and to elucidate the mechanisms involved.Methods:Male ICR mice were randomly allocated to six groups:a control group,an alcohol-administered group,and groups that were administered alcohol and one of silibinin,the GH,the ZJZ or a GH-ZJZ combination(at a ratio of 2:1).Animals were orally administered 56%alcohol(Er Guo-tou white spirit,0.12 mL/10 g/d)for 10 days and at the end of this period,hepatic biochemical indicators,antioxidant parameters,alcohol metabolic enzymes,and histopathologic changes were evaluated.Moreover,the expression of the signaling molecules KEAP1,NRF2,and AQP9 were measured by qRT-PCR and western blotting.Results:Compared with the model group,GH-ZJZ(2:1)had lower serum ALT(12.15±0.39,P紏.003),AST(104.07±1.03,P紏.001),and ALP(148.09±2.55,P紏.010)activities,and lower TC(1.97±0.05,P紏.001)and TG(1.54±0.07,P?.002)concentrations.GH-ZJZ(2:1)also significantly increased the hepatic activities of SOD and GSH(4.24±0.25 and 1.57±0.06,respectively;both P<.01),reduced the ROS and MDA concentrations(97.50±3.00 and 2.39±0.19,respectively;both P<.01),and upregulated Nrf2 expression(P<.01).GH-ZJZ(2:1)significantly reduced the expression of KEAP1 and AQP9 in the liver,compared with alcohol-administered mice(P<.01).Importantly,the GH-ZJZ combination caused a more marked improvement in acute liver injury than GH or ZJZ alone.Conclusion:We have demonstrated protective effects of GH-ZJZ(2:1)against acute alcohol-induced hepatic injury,and shown that these effects may be associated with improvements in lipid and alcohol metabolism,antioxidant capacity,and lipid peroxidation.Wan Xu Shaohong Chen Gansheng Zhong Haiyan Liu Linlin Xiu Xue Yu Feng Chen Na Li Yanmin Lv 2020Journal of Traditional Chinese Medical Sciences2020,7,1:2

20Amisulpride augmentation therapy improves cognitive performance and psychopathology in clozapine‑resistant treatment‑refractory schizophrenia:a 12‑week randomized,double‑blind,placebo‑controlled trial显示文摘Background:Although clozapine is an effective option for treatment-resistant schizophrenia(TRS),there are still 1/3 to 1/2 of TRS patients who do not respond to clozapine.The main purpose of this randomized,double-blind,placebocontrolled trial was to explore the amisulpride augmentation efficacy on the psychopathological symptoms and cognitive function of clozapine-resistant treatment-refractory schizophrenia(CTRS)patients.Methods:A total of 80 patients were recruited and randomly assigned to receive initial clozapine plus amisulpride(amisulpride group)or clozapine plus placebo(placebo group).Positive and Negative Syndrome Scale(PANSS),Scale for the Assessment of Negative Symptoms(SANS),Clinical Global Impression(CGI)scale scores,Repeatable Battery for the Assessment of Neuropsychological Status(RBANS),Treatment Emergent Symptom Scale(TESS),laboratory measurements,and electrocardiograms(ECG)were performed at baseline,week 6,and week 12.Results:Compared with the placebo group,amisulpride group had a lower PANSS total score,positive subscore,and general psychopathology subscore at week 6 and week 12(PBonferroni<0.01).Furthermore,compared with the placebo group,the amisulpride group showed an improved RBANS language score at week 12(PBonferroni<0.001).Amisulpride group had a higher treatment response rate(P=0.04),lower scores of CGI severity and CGI efficacy at week 6 and week 12 than placebo group(PBonferroni<0.05).There were no differences between the groups in body mass index(BMI),corrected QT(QTc)intervals,and laboratory measurements.This study demonstrates that amisulpride augmentation therapy can safely improve the psychiatric symptoms and cognitive performance of CTRS patients.Ming‑Huan Zhu Zhen‑Jing Liu Qiong‑Yue Hu Jia‑Yu Yang Ying Jin Na Zhu Ying Huang Dian‑Hong Shi Min‑Jia Liu Hong‑Yang Tan Lei Zhao Qin‑Yu Lv Zheng‑Hui Yi Feng‑Chun Wu Ze‑Zhi Li 2023Military Medical Research2023,10,4:1
