1The Reference Genome of Tea Plant and Resequencing of 81 Diverse Accessions Provide Insights into Its Genome Evolution and Adaptation显示文摘Tea plant is an important economic crop,which is used to produce the world's oldest and most widely consumed tea beverages.Here,we present a high-quality reference genome assembly of the tea plant(Camellia sinensis var.sinensis)consisting of 15 pseudo-chromosomes.LTR retrotransposons(LTR-RTs)account for 70.38%of the genome,and we present evidence that LTR-RTS play critical roles in genome size expansion and the transcriptional diversification of tea plant genes through preferential insertion in promoter regions and introns.Genes,particularly those coding for terpene biosynthesis pro-teins,associated with tea aroma and stress resistance were significantly amplified through recent tandem duplications and exist as gene clusters in tea plant genome.Phylogenetic analysis of the sequences of 81 tea plant accessions with diverse origins revealed three well-differentiated tea plant populations,support-ing the proposition for the southwest origin of the Chinese cultivated tea plant and its later spread to western Asia through introduction.Domestication and modern breeding left significant signatures on hundreds of genes in the tea plant genome,particularly those associated with tea quality and stress resis-tance.The genomic sequences of the reported reference and resequenced tea plant accessions provide valuable resources for future functional genomics study and molecular breeding of improved cul-tivars of tea plants.Enhua Xia Wei Tong Yan Hou Yanlin An Linbo Chen Qiong Wu Yunlong Liu Jie Yu Fangdong Li Ruopei Li Penghui Li Huijuan Zhao Ruoheng Ge Jin Huang Ali Inayat Mallano Yanrui Zhang Shengrui Liu Weiwei Deng Chuankui Song Zhaoliang Zhang Jian Zhao Shu Wei Zhengzhu Zhang Tao Xia Chaoling Wei Xiaochun Wan 2020Molecular Plant2020,13,7:51

3Adjuvant chemotherapy with paclitaxel and cisplatin in lymph node-positive thoracic esophageal squamous cell carcinoma显示文摘Objective:No standard postoperative adjuvant chemotherapy has ever been established in node-positive esophageal squamous cell carcinoma(ESCC).This is a study to explore the effect of postoperative paclitaxel(PTX)and cisplatin(DDP)in lymph node-positive,completely resected thoracic ESCC patients.Methods:We conducted a prospective phase II trial.Patients had pathologically node-positive thoracic ESCC with negative margins.Outcomes of disease-free survival(DFS)and overall survival(OS)were compared with a matched historical control cohort.The postoperative chemotherapy regimen consisted of 4 to 6 cycles of PTX 150mg/m2 administered intravenously on d 1 followed by DDP 50 mg/m2 on d 2 every 14 d.Results:Forty-three patients were accrued from December 2007 to May 2012 at Cancer Hospital of Chinese Academy of Medical Sciences for adjuvant chemotherapy.The historical control group consisted of 80 patients who received complete resection but no adjuvant chemotherapy during the same period of time.Of the 43 patients with adjuvant chemotherapy,37(86.0%)patients completed 4 to 6 cycles of chemotherapy.The 3-year DFS rates were56.3%in the adjuvant group and 34.6%in the control group(P=0.006).The 3-year OS rates were 55.0%in the adjuvant group and 37.5%in the control group(P=0.013).Multivariate analysis revealed that postoperative chemotherapy was the significant predictor for improved OS(P=0.005).Conclusions:Biweekly adjuvant PTX and DDP might improve 3-year DFS and OS in lymph node-positive,curatively resected thoracic ESCC patients.These conclusions warrant further study in randomized phase III clinical trials.Liangze Zhang Weiwei Li Xiao Lyu Yan Song Yousheng Mao Shaoming Wang Jing Huang 2017Chinese Journal of Cancer Research2017,29,2:15
4Characteristics of Settings and Etiologic Agents of Foodborne Disease Outbreaks—China,2020显示文摘Introduction:Foodborne diseases are a growing public health problem and have caused a large burden of disease in China.This study analyzed epidemiological characteristics of foodborne diseases in China in 2020 to provide a scientific basis for prevention and control measures.Methods:Data were collected from 30 of 31 provincial-level administrative divisions(PLADs)in the mainland of China,excluding Xizang(Tibet)Autonomous Region,via the National Foodborne Disease Outbreaks Surveillance System・The number and proportion of outbreaks,illnesses,hospitalizations,deaths by setting,pathogen-food category pairs and etiology were calculated.Results:In 2020,7,073 foodborne disease outbreaks were reported,resulting in 37,454 illnesses and 143 deaths.Among the identified pathogens,microbial pathogens were the most common confirmed etiology,accounting for 41.7%of illnesses.Poisonous mushrooms caused the largest proportion of outbreaks(58.0%)and deaths(57.6%).For venues where foodborne disease outbreaks occur,household had the highest number of outbreaks(4,140)and deaths(128),and catering service locations caused the largest proportion of illnesses(59.9%).Outbreaks occurring between June and September accounted for 62.8%of total outbreaks.Conclusions:Foodborne disease outbreaks mainly occurred in households・Microbial pathogens remained the top cause of outbreak-associated illnesses.Poisonous mushrooms were ranked the top cause of deaths in private homes in China.The supervision and management of food safety and health education should be strengthened to reduce the burden of foodborne diseases・Publicity should be increased to reduce the incidence of mushroom poisonings in families,and supervision and management of food should be strengthened to reduce microbial contamination.Hongqiu Li Weiwei Li Yue Dai Yuyan Jiang Junhua Liang Santao Wang Maoqiang Zhuang Zheng Huang Lizi Xu Bo Xue Jikai Liu Haihong Han Sara M.Pires Ping Fu Yunchang Guo 2021China CDC weekly2021,3,42:14

6Coordinated Transcriptional Regulation by the UVB Photoreceptor and Multiple Transcription Factors for Plant UV-B Responses显示文摘Non-damaging ultraviolet B(UV-B)light promotes photomorphogenic development and stress acclimation through UV-B-specific signal transduction in Arabidopsis.UV-B irradiation induces monomerization and nuclear translocation of the UV-B photoreceptor UV RESISTANCE LOCUS 8(UVR8).However,it is not clear how the nuclear localization of UVR8 leads to changes in global gene expression.Here,we reveal that nuclear UVR8 governs UV-B-responsive transcriptional networks in concert with several previously known transcription factors,including ELONGATED HYPOCOTYL 5(HY5)and PHYTOCHROME INTERACTING FACTOR 4(PIF4).Based on the transcriptomic analysis,we identify MYB13 as a novel positive regulator in UV-B-induced cotyledon expansion and stress acclimation.MYB13 is UV-B inducible and is predominantly expressed in the cotyledons.Our results demonstrate that MYB13 protein functions as a transcription factor to regulate the expression of genes involved in auxin response and flavonoid biosynthesis through direct binding with their promoters.In addition,photoactivated UVR8 interacts with MYB13 in a UV-B-dependent manner and differentially modulates the affinity of MYB13 with its targets.Taken together,our results elucidate the cooperative function of the UV-B photoreceptor UVR8 with various transcription factors in the nucleus to orchestrate the expression of specific sets of downstream genes and,ultimately,mediate plant responses to UV-B light.Chongzhen Qian Zhiren Chen Qing Liu Weiwei Mao Yanling Chen Wei Tian Yan Liu Jiupan Han Xinhao Ouyang Xi Huang 2020Molecular Plant2020,13,5:13
7Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 8The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 10Direct generation of an ultrafast vortex beam in a CVD-graphene-based passively mode-locked Pr:LiYF4 visible laser显示文摘We report on the direct generation of passively mode-locked vortex lasers in the visible spectral region, for the first time to the best of our knowledge, using a Pr :Li YF4(Pr:YLF) crystal as the gain medium. A stable mode-locked TEM00 mode has been achieved with a maximum average output power of 75 m W using a graphene saturable absorber mirror. The mode-locked pulse width is measured to be as short as about 73.4 ps at a repetition rate of about 140 MHz, and the laser wavelength is at about 721 nm with spectral width of about 0.5 nm. By slightly misaligning the laser resonator, a first-order Laguerre-Gaussian mode(LG0,1) has also been obtained with output power reduced to about 22 m W. The achieved LG0,1 mode has been verified via a home made improved Fizeau interferometer. 12Mortality outcomes of low-dose computed tomography screening for lung cancer in urban China:a decision analysis and implications for practice显示文摘Background: Mortality outcomes in trials of low-dose computed tomography(CT) screening for lung cancer are inconsistent. This study aimed to evaluate whether CT screening in urban areas of China could reduce lung cancer mortality and to investigate the factors that associate with the screening effect.Methods: A decision tree model with three scenarios(low-dose CT screening, chest X-ray screening, and no screening) was developed to compare screening results in a simulated Chinese urban cohort(100,000 smokers aged45-80 years). Data of participant characteristics were obtained from national registries and epidemiological surveys for estimating lung cancer prevalence. The selection of other tree variables such as sensitivities and specificities of low-dose CT and chest X-ray screening were based on literature research. Differences in lung cancer mortality(primary outcome), false diagnoses, and deaths due to false diagnosis were calculated. Sensitivity analyses were performed to identify the factors that associate with the screening results and to ascertain worst and optimal screening effects considering possible ranges of the variables.Results: Among the 100,000 subjects, there were 448,541, and 591 lung cancer deaths in the low-dose CT, chest X-ray, and no screening scenarios, respectively(17.2% reduction in low-dose CT screening over chest X-ray screening and 24.2% over no screening). The costs of the two screening scenarios were 9387 and 2497 false diagnoses and 7and 2 deaths due to false diagnosis among the 100,000 persons, respectively. The factors that most influenced death reduction with low-dose CT screening over no screening were lung cancer prevalence in the screened cohort, lowdose CT sensitivity, and proportion of early-stage cancers among low-dose CT detected lung cancers. 13Volumetric imaging of flame refractive index, density, and temperature using background-oriented Schlieren tomography显示文摘Volumetric imaging represents one of the major development trends of flow diagnostics due to both the advancement in hardware and the requirement for more information to further understand complicated turbulent and/or reactive flows. Backgroundoriented Schlieren tomography(BOST) has become increasingly popular due to its experimental simplicity. It has been demonstrated to be capable of simultaneously recovering the distributions of refractive index, density, and temperature of flows.However, its capability in thermometry has only been demonstrated under the axisymmetric assumption, which greatly limits its applicability. In this work, we dedicated to developing a cost-effective BOST system for the simultaneous retrieval of refractive index, density, and temperature distributions for the asymmetric flame. A few representative tomographic inversion algorithms were assessed as well. 16miR-137 inhibits melanoma cell proliferation through downregulation of GLO1显示文摘Late-stage melanoma is refractory to current therapies. MicroRNAs (miRNAs) can modulate many physiological and pathological processes of melanoma. Studies have demonstrated that miR-137 acts as a tumor suppressor by inhibiting the proliferation of melanoma cells through targeting multiple mRNAs. The glyoxalase system member glyoxalase 1 (GLO1) is the principal scavenging enzyme of methylglyoxal (MG), a toxic byproduct of glycolysis. Using 35S in vivo/vitro labelling analysis for dynamic proteomics (SiLAD), we found that miR-137 downregulated the expression of GLO1 in melanoma cells.Bioinformatics analysis predicted that GLO1 is a direct target of miR-137. This was validated by dual luciferase reporter assay.Quantitative RT-PCR (qRT-PCR) and western blot analysis indicated that miR-137 could decrease endogenous GLO1 expression.Furthermore, siRNA targeting of GLO1 mimicked inhibition of melanoma cell proliferation caused by miR-137 overexpression.Re-expression of GLO1 was able to restore miR-137-mediated suppression of melanoma cell proliferation. 18Absolute calibration of HY-2A and Jason-2 altimeters for sea surface height using GPS buoy in Qinglan, China显示文摘GPS buoy methodology is one of the main calibration methodologies for altimeter sea surface height calibration. This study introduces the results of the Qinglan calibration campaign for the HY-2A and Jason-2 altimeters. It took place in two time slices;one was from August to September 2014, and the other was in July 2015. One GPS buoy and two GPS reference stations were used in this campaign. The GPS data were processed using the real-time kinematic (RTK) technique. The fi nal error budget estimate when measuring the sea surface height (SSH) with a GPS buoy was better than 3.5 cm. Using the GPS buoy, the altimeter bias estimate was about -2.3 cm for the Jason-2 Geophysical Data Record (GDR) Version ‘D' and from -53.5 cm to -75.6 cm for the HY-2A Interim Geophysical Data Record (IGDR). The bias estimates for Jason-2 GDR-D are similar to the estimates from dedicated calibration sites such as the Harvest Platform, the Crete Site and the Bass Strait site. The bias estimates for HY-2A IGDR agree well with the results from the Crete calibration site. 19Auranofin mitigates systemic iron overload and induces ferroptosis via distinct mechanisms显示文摘Iron homeostasis is essential for health;moreover,hepcidin-deficiency results in iron overload in both hereditary hemochromatosis and iron-loading anemia.Here,we identified iron modulators by functionally screening hepcidin agonists using a library of 640 FDA-approved drugs in human hepatic Huh7 cells.We validated the results in C57BL/6J mice and a mouse model of hemochromatosis(Hfe^(−/−)mice).Our screen revealed that the anti-rheumatoid arthritis drug auranofin(AUR)potently upregulates hepcidin expression.Interestingly,we found that canonical signaling pathways that regulate iron,including the Bmp/Smad and IL-6/Jak2/Stat3 pathways,play indispensable roles in mediating AUR’s effects.In addition,AUR induces IL-6 via the NF-κB pathway.In C57BL/6J mice,acute treatment with 5 mg/kg AUR activated hepatic IL-6/hepcidin signaling and decreased serum iron and transferrin saturation.Whereas chronically treating male Hfe^(−/−)mice with 5 mg/kg AUR activated hepatic IL-6/hepcidin signaling,decreasing systemic iron overload,but less effective in females.Further analyses revealed that estrogen reduced the ability of AUR to induce IL-6/hepcidin signaling in Huh7 cells,providing a mechanistic explanation for ineffectiveness of AUR in female Hfe^(−/−)mice.Notably,high-dose AUR(25 mg/kg)induces ferroptosis and causes lipid peroxidation through inhibition of thioredoxin reductase(TXNRD)activity.We demonstrate the ferroptosis inhibitor ferrostatin significantly protects liver toxicity induced by highdose AUR without comprising its beneficial effect on iron metabolism.In conclusion,our findings provide compelling evidence that TXNRD is a key regulator of ferroptosis,and AUR is a novel activator of hepcidin and ferroptosis via distinct mechanisms,suggesting a promising approach for treating hemochromatosis and hepcidin-deficiency related disorders.Lei Yang Hao Wang Xiang Yang Qian Wu Peng An Xi Jin Weiwei Liu Xin Huang Yuzhu Li Shiyu Yan Shuying Shen Tingbo Liang Junxia Min Fudi Wang 2020Signal Transduction and Targeted Therapy2020,5,1:5
20Fenton process for degradation of selected chlorinated aliphatic hydrocarbons exemplified by trichloroethylene,1,1-dichloroethylene and chloroform显示文摘The degradation of selected chlorinated aliphatic hydrocarbons(CAHs)exemplified by trichloroethylene(TCE),1,1-dichloroethylene(DCE),and chloroform(CF)was investigated with Fenton oxidation process.The results indicate that the degradation rate was primarily affected by the chemical structures of organic contaminants.Hydroxyl radicals(·OH)preferred to attack the organic contaminants with an electron-rich structure such as chlorinated alkenes(i.e.,TCE and DCE).The dosing mode of Fenton’s reagent,particularly of Fe^(2+),significantly affected the degradation efficiency of studied organic compound.A new“time-squared”kinetic model,C=C_(oexp)(-k_(obst)^(2)),was developed to express the degradation kinetics of selected CAHs.This model was applicable to TCE and DCE,but inapplicable to CF due to their varied reaction rate constants towards·OH.Chloride release was monitored to examine the degree of dechlorination during the oxidation of selected CAHs.TCE was more easily dechlorinated than DCE and CF.Dichloroacetic acid(DCAA)was identified as the major reaction intermediate in the oxidation of TCE,which could be completely removed as the reaction proceeded.No reaction intermediates or byproducts were identified in the oxidation of DCE and CF.Based on the identified intermediate,the reaction mechanism of TCE with Fenton’s reagent was proposed.Zhimin QIANG Weiwei BEN Chin-Pao HUANG 2008Frontiers of Environmental Science & Engineering2008,2,4:5
