| 1 | Efficacy of Zhonglun'a-decoction-containing serum on fibroblastlike synoviocyte apoptosis in rats with collagen-induced arthritis via inhibiting Janus kinase/signal transducer and activator of transcription signaling pathway显示文摘OBJECTIVE: To investigate the efficacy of Zhonglun'a-decoction-containing serum(ZHONGL-CS)on the in vitro apoptosis of fibroblast-like synoviocytes(FLS) from rats with collagen-induced arthritis(CIA) by investigating the Janus kinase/signal transducer and activator of transcription(JAK/STAT) signaling pathway.METHODS: A CIA rat model was established using bovine type Ⅱ collagen. FLS were isolated, cultured and identified. A cell counting kit-8 was used to detect cell activity. The half maximal inhibitory concentration(IC50) was calculated. Experimental subjects were divided into control, CIA,ZHONGL-CS, JAK2 inhibitor AG490, and ZHONGLCS with AG490 groups. The in vitro cell cycle and apoptosis rate were detected in FLS by flow cytometry. Bcl-2-associated X protein(Bax), B-cell lymphoma 2(Bcl-2), caspase-3, cyclin D1, phosphorylated JAK2, STAT1, and STAT3 protein expressions in FLS were examined by Western blotting. JAK2, STAT1 and STAT3 mRNA levels were examined by quantitative real-time polymerase chain reaction.RESULTS: Compared with the CIA group, FLS proliferation was inhibited, the FLS G0/G1 cell cycle was arrested, and the rate of FLS apoptosis was increased in the ZHONGL-CS group. In the ZHONGLCS group, the protein levels of Bcl-2 and cyclin D1 were reduced compared with the CIA group and the levels of Bax and caspase-3 in FLS were increased. In the ZHONGL-CS group, the expressions of JAK2, STAT1, and STAT3 mRNA and the levels of phosphorylated JAK2, STAT1, and STAT3 proteins were reduced.CONCLUSION: ZHONGL-CS may induce FLS apoptosis in CIA rats. | 2 | Rheological responses of fumed silica suspensions under steady and oscillatory shear显示文摘Rheological experiments under steady and oscillatory shear were conducted for fumed silica suspen- sions in polyethylene glycol. Under steady shear the shear-thinning and thickening response were exhibited and the flow exponent N was determined. With the increase of concentration the flow exponent N showed a rapid increase, and it increased dramatically when the discontinuous shear-thickening took place. Oscillatory shear experiments were conducted at constant frequency and constant amplitude strain, respectively. The shear-thinning and the discontinuous shear-thickening behavior were observed under different constant frequencies from 10 to 80 rad/s. The correlation between complex modulus (G*) and sweep frequency (ω) was illuminated at γ =750%. It was found that the correlation between G* and ω could be fitted by equation: G*∝ωn. The indexes in shear-thinning region and shear-thickening were determined. The indexes were similar to some extent at shear-thinning region and increased dramati- cally to a much higher value when the shear-thickening occurred,especially at higher weight fractions. | 9 | Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment. In order to explore the pathogenic mechanism of Salmonella,phylogenetic analysis of major virulence factor genes in 33 Salmonella strains isolated from chicken was analyzed. According to the results,ivnA gene was successfully amplified from 33 Salmonella strains; ivnE gene was successfully amplified from 32 Salmonella strains. ivnA nucleotide sequences shared 72. 9%- 97. 6% homology among 12 sequenced Salmonella strains and shared 78. 9%- 97. 2% homology with those in GenB ank; ivnE nucleotide sequences shared over95. 3% homology among 23 sequenced Salmonella strains and shared 89. 6%- 98. 6% homology with those in GenB ank,which exhibited no genetic relationship to other organisms. | 17 | Isolation and Identification of Pathogenic Bacteria Causing Bovine Subclinical Mastitis in Eastern Hebei Province显示文摘In this study, the milk samples of 1 021 cows in eight dairy farms in Eastern Hebei Province were collected and detected with LMT reagent and somatic cell count for subclinical mastitis. Pathogenic bacteria in subclinical mastitis positive milk samples were isolated and identified.The results showed that 60.63%(619/1 021) of the sampled cows were diagnosed with subclinical mastitis, and mixed infections accounted for 88.21%(546/619) of the cases. In addition, 82 strains of 14 species were isolated from the subclinical mastitis positive milk samples, including 36 strains of Staphylococcus(43.90%), 33 strains of Streptococcus(40.24%), 8 strains of Enterobacteriaceae(9.76%) and 5 strains of Corynebacterium(6.10%), respectively.
