1Collapse simulation of a super high-rise building subjected to extremely strong earthquakes显示文摘In recent years, super high-rise buildings (>500 m) are developing very quickly and become an important frontier of civil engineering. The collapse resistance of super high-rise buildings subjected to extremely strong earthquake is a critical problem that must be intensively studied. This paper builds up a nonlinear finite element model of the tallest building in China, Shang- hai Tower (632 m), and proposes the modeling method and failure criteria for different structural elements. The dynamic char- acters of this building are then analyzed, and the possible failure modes and collapse processes due to earthquakes are pre- dicted, as well as the corresponding collapse mechanism. 2Community structure of macrobenthos in coastal water off Rushan, southern Shandong Peninsula, and the relationships with environmental factors显示文摘To understand the present actuality of the marine ecosystem in the southern coastal water region of the Shandong Peninsula and the impact of the global change and the human activities to the marine ecosystem of the region, the macrobenthic community structure was researched based on data from 26 sampling stations carried out on four seasonal cruises from December 2006 to November 2007. The data was analyzed using PRIMER 6.0 and SPSS 15.0 software packages. The results showed that 236 macrobenthic species in total were collected from the research region by the field works. Most of the species belong to Polychaeta (76 species), Mollusca (75) and Crustacea (60). Of which, 33 species were common species by the four cruises. The dominant species were different among the four seasons, however, the polychaete species Nephtys oligobranchia and Sternaspis scutata were always dominant in the four seasons. The abundances and biomasses of the macrobenthos from the research region were variable in the four seasons. The results of CLUSTER and MDS analysis showed that the similarities of macrobenthic structures among the stations were low, most of the similarities were at about 40% of similarity values, only that of two stations were up to 60%. In accordance with the similarity values of the macrobenthic structures, the 26 stations were clustered as six groups at arbitrary similarity level of 30%. The ABC curve indicated that the marcofauna communities in the research region had not been disturbed distinctly. 3Identification and characterization of genes that control fat deposition in chickens显示文摘Background: Fat deposits in chickens contribute significantly to meat quality attributes such as juiciness, flavor,taste and other organoleptic properties. The quantity of fat deposited increases faster and earlier in the fastgrowing chickens than in slow-growing chickens. In this study, Affymetrix Genechip? Chicken Genome Arrays 32773transcripts were used to compare gene expression profiles in liver and hypothalamus tissues of fast-growing and slow-growing chicken at 8 wk of age. Real-time RT-PCR was used to validate the differential expression of genes selected from the microarray analysis. The mRNA expression of the genes was further examined in fat tissues. The association of single nucleotide polymorphisms of four lipid-related genes with fat traits was examined in a F2resource population.Results: Four hundred genes in the liver tissues and 220 genes hypothalamus tissues, respectively, were identified to be differentially expressed in fast-growing chickens and slow-growing chickens. Expression levels of genes for lipid metabolism(SULT1B1, ACSBG2, PNPLA3, LPL, AOAH) carbohydrate metabolism(MGAT4B, XYLB, GBE1, PGM1,HKDC1)cholesttrol biosynthesis(FDPS, LSS, HMGCR, NSDHL, DHCR24, IDI1, ME1) HSD17B7 and other reaction or processes(CYP1A4, CYP1A1, AKR1B1, CYP4V2, DDO) were higher in the fast-growing White Recessive Rock chickens than in the slow-growing Xinghua chickens. On the other hand, expression levels of genes associated with multicellular organism development, immune response, DNA integration, melanin biosynthetic process, muscle organ development and oxidation-reduction(FRZB, DMD, FUT8, CYP2C45, DHRSX, and CYP2C18) and with glycol-metabolism(GCNT2, ELOVL 6, and FASN), were higher in the XH chickens than in the fast-growing chickens. RT-PCR validated high expression levels of nine out of 12 genes in fat tissues. The G1257069A and T1247123C of the ACSBG2 gene were significantly associated with abdominal fat weight. The G4928024A of the FASN gene were significantly associated with fat bandwidth, and abdominal fat percentage. The C4930169T of the FASN gene was associated with abdominal fat weight while the A59539099G of the ELOVL 6 was significantly associated with subcutaneous fat. 5Structural basis for nucleosome-mediated inhibition of cGAS activity显示文摘Activation of cyclic GMP-AMP synthase(cGAS)through sensing cytosolic double stranded DNA(dsDNA)plays a pivotal role in innate immunity against exogenous infection as well as cellular regulation under stress.Aberrant activation of cGAS induced by self-DNA is related to autoimmune diseases.cGAS accumulates at chromosomes during mitosis or spontaneously in the nucleus.Binding of cGAS to the nucleosome competitively attenuates the dsDNA-mediated cGAS activation,but the molecular mechanism of the attenuation is still poorly understood.Here,we report two cryo-electron microscopy structures of cGAS-nucleosome complexes.The structures reveal that cGAS interacts with the nucleosome as a monomer,forming 1:1 and 2:2 complexes,respectively.cGAS contacts the nucleosomal acidic patch formed by the H2A-H2B heterodimer through the dsDNA-binding site B in both complexes,and could interact with the DNA from the other symmetrically placed nucleosome via the dsDNA-binding site C in the 2:2 complex.The bound nucleosome inhibits the activation of cGAS through blocking the interaction of cGAS with ligand dsDNA and disrupting cGAS dimerization.R236A or R255A mutation of cGAS impairs the binding between cGAS and the nucleosome,and largely relieves the nucleosome-mediated inhibition of cGAS activity.Our study provides structural insights into the inhibition of cGAS activity by the nucleosome,and advances the understanding of the mechanism by which hosts avoid the autoimmune attack caused by cGAS.Duanfang Cao Xiaonan Han Xiaoyi Fan Rui-Ming Xu Xinzheng Zhang 2020Cell Research2020,30,12:6
6Expression analysis of two novel cotton 14-3-3 genes in root development and in response to salt stress显示文摘14-3-3 proteins are phosphoserine-binding proteins that regulate the activities of a wide array of targets via direct protein-protein interactions,and may play an important role in response to biotic and abiotic stresses. In this study,two cDNAs (designated as Gh14-3-3b and Gh14-3-3c) encoding putative 14-3-3 proteins were isolated from cotton cDNA libraries. Gh14-3-3b gene encodes a pro-tein of 268 amino acids,while Gh14-3-3c gene encodes a protein of 261 amino acids. Real-time RT-PCR analysis revealed that both the Gh14-3-3b and Gh14-3-3c genes were preferentially expressed in roots. The transcript levels of both the genes were the highest in 3-day-old roots,and then dramatically decreased to relatively low levels in parallel with root development. 7New CFAR target detector for SAR images based on kernel density estimation and mean square error distance显示文摘A new constant false alarm rate (CFAR) target detector for synthetic aperture radar (SAR) images is developed. For each pixel under test, both the local probability density function (PDF) of the pixel and the clutter PDF in the reference window are estimated by the non-parametric density estimation. The target detector is defined as the mean square error (MSE) distance between the two PDFs. The CFAR detection in SAR images having multiplicative noise is achieved by adaptive kernel bandwidth proportional to the clutter level. In addition, for obtaining a threshold with respect to a given probability of false alarm (PFA), an unsupervised null distribution fitting method with outlier rejection is proposed. 9Non-anthracycline-containing docetaxel and cyclophosphamide regimen is associated with sustained worse outcome compared with docetaxel, anthracycline and cyclophosphamide in neoadjuvant treatment of triple negative and HER2-positive breast cancer patients:显示文摘Objective: A previous study demonstrated that non-anthracycline-containing docetaxel plus cyclophosphamide(TC) regimen was inferior to docetaxel, anthracycline and cyclophosphamide(TAC) in neoadjuvant treatment of triple-negative breast cancer(TNBC) and human epidermal growth factor receptor-2-(HER2)-positive breast cancer in a short-term follow-up. Herein, long-term follow-up survival outcomes have been investigated.Methods: TNBC or HER2-positive patients were randomized to receive 6 cycles of TC or TAC neoadjuvant treatment. The primary endpoint was pathological complete remission(p CR). Secondary endpoints included clinical response rate, event-free survival(EFS), and overall survival(OS).Results: A cohort of 96 patients consisted of 45 in TC and 51 in TAC arm. With a median follow-up period of53(range, 8–76) months, the patients achieving p CR post neoadjuvant chemotherapy exhibited superior EFS and OS than patients without p CR(P<0.05). TAC treatment resulted in consistently better EFS than TC treatment:the estimated 5-year EFS was 66.1% vs. 29.8%(P=0.002). Moreover, the estimated 5-year OS was also in favor of TAC: 88.4% vs. 51.6%(P<0.001). Multivariable analysis demonstrated that the treatment regimen was an independent prognostic factor, and patients treated with TAC had a superior EFS [hazard ratio(HR), 0.48; 95%confidence interval(95% CI), 0.26–0.90; P=0.021] and OS(HR, 0.20; 95% CI, 0.08–0.60; P=0.003).Conclusions: The updated long-term follow-up data demonstrated a sustained benefit in EFS and OS from anthracycline-containing TAC treatment, indicating that anthracycline is an essential and effective drug in this clinical trial.Xiaosong Chen Guolin Ye Chenfang Zhang Xinzheng Li Kunwei Shen 2016Chinese Journal of Cancer Research2016,28,6:4

14Structures of the portal vertex reveal essential protein-protein interactions for Herpesvirus assembly and maturation显示文摘Dear Editor,Herpesviridae is a large family of double-stranded DNA(dsDNA)viruses that cause a variety of human diseases ranging from cold sores and chicken pox to congenital defects,blindness and cancer(Chayavichitsilp et al.,2009;Wang et al.,2018).In the past 70 years,substantial advances in our knowledge of the molecular biology of herpesviruses have led to insights into disease pathogenesis and management.However,the mechanism for capsid assembly that requires the ordered packing of about 4,000 protein subunits into the hexons,pentons and triplexes remains elusive.It is still a puzzle how initially identical subunits adopt both hexameric and pentameric conformations in the capsid and select the correct locations needed to form closed shells of the proper size.Biochemical and genetic studies have shown that the portal is involved in initiation of capsid assembly(Newcomb et al.,2005)and functions akin to a DNA-sensor coupling genome-packaging achieved by a genome-packaging machinery-“terminase complex”(Chen et al.,2020;Yunxiang Yang,2020)with icosahedral capsid maturation(Lokareddy et al.,2017).Structural investigations of the herpesvirus portal have proven challenging due to the small size of this dodecamer,which accounts for less than 1%of the total mass of the capsid protein layer and the technical difficulties involved in resolving non-icosahedral components of such large icosahedral viruses(diameter is∼1,250Å).Efforts of many investigators over two decades have made to reconstruct the cryo-electron microscopy(cryo-EM)structure of herpesvirus portal vertex and more recently near-atomic structures of two herpesvirus(herpes simplex virus type 1(HSV-1)and Kaposi’s sarcoma-associated herpesvirus(KSHV))portal vertices were reported(McElwee et al.,2018;Gong et al.,2019;Liu et al.,2019).Nan Wang Wenyuan Chen Ling Zhu Dongjie Zhu Rui Feng Jialing Wang Bin Zhu Xinzheng Zhang Xiaoqing Chen Xianjie Liu Runbin Yan Dongyao Ni Grace Guoying Zhou Hongrong Liu Zihe Rao Xiangxi Wang 2020Protein & Cell2020,11,5:2

16Low threshold random lasing in dye-doped and strongly disordered chiral liquid crystals显示文摘Random lasing was experimentally investigated in pyrromethene 597-doped strongly disordered chiral liquid crystals (CLCs) composed of the nematic liquid crystal SLC1717 and the chiral agent CB15. The concentration of the chiral agent tuned the bandgap, and disordered CLC microdomains were achieved by fast quenching of the mixture from the isotropic to the cholesteric phase. Random lasing and band edge lasing were observed synchronously, and their behavior changed with the spectral location of the bandgap. The emission band for band edge lasing shifted with the change of the bandgap, while the emission band for random lasing remained practically constant. 18Wavelength dependence of light propagation in a water suspension of anisotropic scatterers显示文摘We experimentally study the wavelength dependence of light propagation in a water suspension of lithium niobate microcrystalline particles.First,the ballistic transmission in the visible range of the suspension is measured.The nonlinear relationship is observed between the transport mean free path and the wavelength of the incident light.Secondly,we measure the coherent backscattering (CBS) of the sample at different wavelengths.The full width at half maximum of the CBS cone at 532 nm is about 1.24 times as large as that at 671 nm.The results indicate that the light with a long wavelength propagates further than the short wavelength light and the localization state of the short one is stronger.Finally,we investigate the light-controllable CBS experiments in the disordered materials of anisotropic scatterers,which show that the configuration of pump light with the longer wavelength and the probe light with the shorter wavelength performs better.SHI Fan ZHANG XinZheng LI Jun WANG PiDong XU Yan YU XuanYi XU JingJun 2011Science China(Physics,Mechanics & Astronomy)2011,54,11:2

20Development and Application of a High-Performance Triangular Shell Element and an Explicit Algorithm in OpenSees for Strongly Nonlinear Analysis显示文摘The open-source finite element software,OpenSees,is widely used in the earthquake engineering community.However,the shell elements and explicit algorithm in OpenSees still require further improvements.Therefore,in this work,a triangular shell element,NLDKGT,and an explicit algorithm are proposed and implemented in OpenSees.Specifically,based on the generalized conforming theory and the updated Lagrangian formulation,the proposed NLDKGT element is suitable for problems with complicated boundary conditions and strong nonlinearity.The accuracy and reliability of the NLDKGT element are validated through typical cases.Furthermore,by adopting the leapfrog integration method,an explicit algorithm in OpenSees and a modal damping model are developed.Finally,the stability and efficiency of the proposed shell element and explicit algorithm are validated through the nonlinear time-history analysis of a highrise building.Xinzheng Lu Yuan Tian Chujin Sun Shuhao Zhang 2019Computer Modeling in Engineering & Sciences2019,,9:1
