1Androgen receptor: structure, role in prostate cancer and drug discovery显示文摘Androgens and androgen receptors (AR) play a pivotal role in expression of the male phenotype. Several diseases, such as androgen insensitivity syndrome (AIS) and prostate cancer, are associated with alterations in AR functions. Indeed, androgen blockade by drugs that prevent the production of androgens and/or block the action of the AR inhibits prostate cancer growth. However, resistance to these drugs often occurs after 2-3 years as the patients develop castration-resistant prostate cancer (CRPC). In CRPC, a functional AR remains a key regulator. Early studies focused on the functional domains of the AR and its crucial role in the pathology. The elucidation of the structures of the AR DNA binding domain (DBD) and ligand binding domain (LBD) provides a new framework for understanding the functions of this receptor and leads to the development of rational drug design for the treatment of prostate cancer. 3Topographic differentiations of biological soil crusts and hydraulic properties in fixed sand dunes, Tengger Desert显示文摘Biological soil crusts(BSCs) play an important role in surface soil hydrology. Soils dominated with moss BSCs may have higher infiltration rates than those dominated with cyanobacteria or algal BSCs. However, it is unnown whether improved infiltration in moss BSCs is accompanied by an increase in soil hydraulic conductivity or water retention capacity. We investigated this question in the Tengger Desert, where a 43-year-old revegetation program has promoted the formation of two distinct types of BSCs along topographic positions, i.e. the moss-dominated BSCs on the interdune land and windward slopes of the fixed sand dunes, and the algal-dominated BSCs on the crest and leeward slopes. Soil water retention capacity and hydraulic conductivity were measured using an indoor evaporation method and a field infiltration method. And the results were fitted to the van Genuchten–Mualem model. Unsaturated hydraulic conductivities under greater water pressure(<–0.01 MPa) and water retention capacities in the entire pressure head range were higher for both crust types than for bare sand. However, saturated and unsaturated hydraulic conductivities in the near-saturation range(>–0.01 MPa) showed decreasing trends from bare sand to moss crusts and to algal crusts. 4S, C, O, H, and Pb isotopic studies for the Shuiyindong Carlin-type gold deposit, Southwest Guizhou, China: constraints for ore genesis显示文摘The Shuiyindong gold deposit is one of the most famous and largest Carlin-type gold deposits in China and is located in southwest Guizhou, in the eastern part of the Huijiabao anticline. The Shuiyindong's gold mineralization occurred in bioclastic limestone of the Permian Longtan Formation. Sulfur, carbon, hydrogen, oxygen, and lead isotopic compositions are reported in this paper. The properties and sources of ore-forming fluid have been discussed and a metallogenic model for the Shuiyindong gold deposit has been proposed. The d34 S values of stibnite, realgar, orpiment, pyrite from orebodies, and pyrite from quartz veins are similar to or slightly higher than the d34 S values of mantle sulfur. It is suggested that the sulfur of hydrothermal sulfides was likely of magmatic origin with minor heavy sulfur contributed from the country rocks. The measured d D values and calculated d18OH2 O values of inclusion fluid in quartz plotted within or below a magmatic hydrothermal fluid field far from the meteoric water line. This indicates that the ore-forming fluid for the main-stage gold mineralization could have been derived mainly from a magmatic source and mixed with a small amount of meteoric water. The carbon and oxygen isotopic compositions of calcites in the d18 O vs. d13 C diagram suggest that the CO2 in ore-forming fluid was derived from dissolution of bioclastic limestone and oxidation of sedimentary organic carbon in limestone. However, the d13 C values of ore-related calcites, which contain intergrown realgar and/or orpiment, are similar to those of mantle carbon. Although no igneous intrusive rock has been observed in the vicinity of the gold deposits, the possibility of mantle fluid integrated into the ore-forming fluid cannot be eliminated based on the d13 C values of ore-related calcites. The lead isotopes of sulfides are distributed near the growth curves of upper crust and orogenic belt in the plumbotectonic diagram. Their calculated Dc and Db values plotted within the magmatism field of crust-mantle subduction zone in the Dc- Db diagram. This suggests that the lead of sulfides has an intimate connection with magmatism. Our S, H, O, C, and Pb isotopic studies for the Shuiyindong Carlin-type gold deposit in Guizhou manifest a concordant possibility that the ore-forming fluid was mainly derived from magmatic fluid with minor contribution from the surrounding strata. 6Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Lung Cancer Patients显示文摘The prevalence of malnutrition is high among oncology patients in China. Although the Patient- Generated Subjective Global Assessment (PG-SGA) is widely recognized as an important nutritional assessment tool, it has not been validated for Chinese cancer patients. The purpose of this study was to conduct an analysis of nutrition screening and assessment data to validate the Chinese version of the PG-SGA in Chinese patients with lung cancer. This was an observational, cross-sectional study of 2,000 consecutive adult patients with lung cancer treated at several tertiary hospitals in China. Anthropometric and patient descriptive data were collected. The PG-SGA generated a score for the nutritional risk and a global rating for the nutritional status. The internal consistency, external consistency, and construct validity were evaluated, and a known-groups comparison and principal component analyses were conducted to evaluate the reliability and validity of the PG-SGA. The known-groups comparison demonstrated the ability of the PG-SGA to differentiate lung cancer patients of different nutritional and body mass index (BMI) groups. A confirmatory factor analysis supported the original four-factor structure of the PG-SGA. In terms of the internal and external consistency, the PG-SGA demonstrated intra-class correlation coefficients (ICC) up to 0.987. Alternative form validity was also demonstrated between Box 4 of the PG-SGA and the Karnofsky Performance Status score, with excellent external consistency (r = 0.972, P < 0.001). The construct validity was supported, and selected questionnaire dimensions were evident in the principal component analysis. Significant Spearman correlations (P < 0.001) were demonstrated. The Chinese version of the PG-SGA demonstrated significant reliability and sufficient exploratory properties to support its validity. It seems to be a valid tool that can be used to access the nutritional status of Chinese lung cancer patients. 8Development and utilization of a new chemically-induced soybean library with a high mutation density显示文摘Mutagenized populations have provided important materials for introducing variation and identifying gene function in plants. In this study, an ethyl methanesulfonate(EMS)-induced soybean(Glycine max) population,consisting of 21,600 independent M_2 lines, was developed.Over 1,000 M_(4(5))families, with diverse abnormal phenotypes for seed composition, seed shape, plant morphology and maturity that are stably expressed across different environments and generations were identified. Phenotypic analysis of the population led to the identification of a yellow pigmentation mutant, gyl, that displayed significantly decreased chlorophyll(Chl) content and abnormal chloroplast development. Sequence analysis showed that gyl is allelic to Minn Gold, where a different single nucleotide polymorphism variation in the Mg-chelatase subunit gene(ChlI1a) results in golden yellow leaves. A cleaved amplified polymorphic sequence marker was developed and may be applied to marker-assisted selection for the golden yellow phenotype in soybean breeding. 9Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Gastric Cancer Patients显示文摘Purpose Malnutrition is severe among gastric cancer patients in China. While the Patient-Generated Subjective Global Assessment (PG-SGA) is a widely used nutritional assessment tool, the validation for Chinese patients with gastric cancer has not been performed yet. The aim of this research is to validate the Chinese version of the PGSGA in Chinese gastric cancer patients by conducting analyses of nutrition screening and assessment data. Methods Two thousand consecutive adult patients with gastric cancer were included in this observational, cross-sectional study in several tertiary hospitals in China. A survey was made among those patients, which included several questionnaires and some anthropometric parameters. We calculated the parameters to validate the use of PG-SGA categorizing the nutritional status of gastric cancer patients. Then we performed statistical analysis to evaluate the internal consistency, external consistency, and construct validity. Results This study verified the ability of the PG-SGA to differentiate gastric cancer patients into different nutritional groups. The confirmatory factor analysis verified the original four-factor structure of the PG-SGA. The internal and external consistency was measured by a concordance analysis which showed an intraclass correlation coefficients (ICC) of 0.987. The correlations of the Box 4 score with the Karnofsky performance status score demonstrated alternative form validity with good external consistency (Pearson, r = 0.643, P < 0.001). All three anthropometric dimensions of the PG-SGA were supported by principal component analysis. Spearman correlations (P < 0.001) were significant in the majority of tests. 10Expression profile of microRNAs in gastrointestinal stromal tumors revealed by high throughput quantitative RT-PCR microarray显示文摘AIM: To investigate the microRNA(miRNA) expression profile in gastrointestinal stromal tumor(GIST) tissues that could serve as a novel diagnostic biomarker for GIST detection.METHODS: We performed a quantitative real-time quantitative reverse transcriptase polymerase chain reaction assay to analyze the expression of 1888 miRNAs in a sample set that included 54 GIST tissue samples.RESULTS: We found that dysregulation of several miRNAs may be related to the malignant potential of GISTs. Six of these miRNAs, hsa-let-7c, miR-218,miR-488#, miR-4683, miR-34c-5p and miR-4773, were selected as the final list of biomarkers to separate the malignant GISTs(M group) from the benign GISTs(B group). In addition, MiR-29b-2#, hsa-let-7c, miR-891 b, miR-218, miR-204, miR-204-3p, miR-628-5p,miR-744, miR-29c#, miR-625 and miR-196 a were used to distinguish between the borderline(BO group) and M groups. 13Liver resection versus liver transplantation for hepatocellular carcinoma within Milan criteria:a meta-analysis of 18,421 patients显示文摘Background:Outcomes after liver resection(LR)and liver transplantation(LT)for hepatocellular carcinoma(HCC)are heterogenous and may vary by region,over time periods and disease burden.We aimed to compare overall survival(OS)and disease-free survival(DFS)between LT versus LR for HCC within the Milan criteria.Methods:Two authors independently searched Medline and Embase databases for studies comparing survival after LT and LR for patients with HCC meeting the Milan criteria.Meta-analyses and meta-regression were conducted using random-effects models.Results:We screened 2,278 studies and included 35 studies with 18,421 patients.LR was associated with poorer OS[hazard ratio(HR)=1.44;95%confidence interval(CI):1.14-1.81;P<0.01]and DFS(HR=2.71;95%CI:2.23-3.28;P<0.01)compared to LT,with similar findings among intention-to-treat(ITT)studies.In uninodular disease,OS in LR was comparable to LT(P=0.13)but DFS remained poorer(HR=2.95;95%CI:2.30-3.79;P<0.01).By region,LR had poorer OS versus LT in North America and Europe(P≤0.01),but not Asia(P=0.25).LR had inferior survival versus LT in studies completed before 2010(P=0.01),but not after 2010(P=0.12).Cohorts that underwent enhanced surveillance had comparable OS after LT and LR(P=0.33),but cohorts undergoing usual surveillance had worse OS after LR(HR=1.95;95%CI:1.24-3.07;P<0.01).Conclusions:Mortality after LR for HCC is nearly 50%higher compared to LT.Survival between LR and LT were similar in uninodular disease.The risk of recurrence after LR is threefold that of LT.Jin Hean Koh Darren Jun Hao Tan Yuki Ong Wen Hui Lim Cheng Han Ng Phoebe Wen Lin Tay Jie Ning Yong Mark D.Muthiah Eunice X.Tan Ning Qi Pang Beom Kyung Kim Nicholas Syn Alfred Kow Brian K.P.Goh Daniel Q.Huang 2022Hepatobiliary Surgery and Nutrition2022,11,1:3
14Near-field surface deformation during the April 20,2013,Ms7.0 Lushan earthquake measured by 1-Hz GNSS显示文摘The April 20,2013,Ms7.0 Lushan earthquake was successfully recorded by closely spaced Continuous Global Positioning System (CGPS) stations owned by the Crustal Movement Observation Network of China (CMONC).The 1-Hz GNSS data from eight CGPS stations,which are located between 30 km and 200 km from the hypocenter,were processed within quasi-real-time.The near-field surface deformation indicated the following characteristics:the near-field movements were limited to several centimeters;the peak of the deformation wave was significantly larger than the static permanent offset;at the beginning of the event,the north wall of the fault moved to the southeast as the south wall moved to the southwest;station SCTQ,which was the closest station to the hypocenter at 30 km,had the largest static permanent displacement of 2 cm;the peaks of the deformation waves were 1.5 cm,5 cm and 3 cm,to the east,the south and vertically upward,respectively;and the peaks of velocity and acceleration,derived from the deformation,were 3.4 cm/s and 5.3 cm/s2,respectively.Liu Gang Zhao Bin Zhang Rui Huang Yong Wang Jun Nie Zhaosheng Qiao Xuejun Tan Kai 2013Geodesy and Geodynamics2013,4,2:3
15An unexpected similarity between antibiotic-resistant NDM-1 and beta-lactamaseⅡfrom Erythrobacter litoralis显示文摘NDM-1(New Delhi metallo-beta-lactamase)gene encodes a metallo-beta-lactamase(MBL)with high carbapenemase activity,which makes the host bacterial strain easily dispatch the last-resort antibiotics known as carbapenems and cause global concern.Here we present the bioinformatics data showing an unexpected similarity between NDM-1 and beta-lactamase II from Erythrobacter litoralis,a marine microbial isolate.We have further expressed these two mature proteins in E.coli cells,both of which present as a monomer with a molecular mass of 25 kDa.Antimicrobial susceptibility assay reveals that they share similar substrate specificities and are sensitive to aztreonam and tigecycline.The conformational change accompanied with the zinc binding visualized by nuclear magnetic resonance,Zn2+-bound NDM-1,adopts at least some stable tertiary structure in contrast to the metal-free protein.Our work implies a close evolutionary relationship between antibiotic resistance genes in environmental reservoir and in the clinic,challenging the antimicrobial resistance monitoring.Beiwen Zheng Shuguang Tan Jia Gao Huiming Han Jun Liu Guangwen Lu Di Liu Yong Yi Baoli Zhu George F.Gao 2011Protein & Cell2011,2,3:2
16Fabrication and Evaluation of Porous Keratin/chitosan(KCS) Scaffolds for Effectively Accelerating Wound Healing显示文摘Objective To develop a dressing with desired antibacterial activity, good water maintaining ability and mechanical properties for wound healing and skin regeneration. Methods The chitosan with different concentrations were added in keratin solution to form porous keratin/chitosan(KCS) scaffolds. The morphological characteristics, chemical composition, wettability, porosity, swelling ratio and degradation of the scaffolds were evaluated. The antibacterial activity was tested by using S. aureus and E. coli suspension for 2 h. And L929 fibroblast cells culture was used to evaluate the cytotoxicity of the KCS scaffolds. Results The adding of chitosan could increase the hydrophobicity, decrease porosity, swelling ratio and degradation rate of the KCS porous scaffolds. Mechanical properties of KCS scaffolds could be enhanced and well adjusted by chitosan. KCS scaffolds could obviously decrease bacteria number. The proliferation of fibroblast cells in porous KCS patch increased firstly and then decreased with the increase of chitosan concentration. It was appropriate to add 400 μg/m L chitosan to form porous KCS scaffold for achieving best cell attachment and proliferation compared with other samples.
