1Meningeal lymphatic vessels regulate brain tumor drainage and immunity显示文摘Recent studies have shown that meningeal lymphatic vessels(MLVs),which are located both dorsally and basally beneath the skull,provide a route for draining macromolecules and trafficking immune cells from the central nervous system(CNS)into cervical lymph nodes(CLNs),and thus represent a potential therapeutic target for treating neurodegenerative and neuroinflammatory diseases.However,the roles of MLVs in brain tumor drainage and immunity remain unexplored.Here we show that dorsal MLVs undergo extensive remodeling in mice with intracranial gliomas or metastatic melanomas.RNA-seq analysis of MLV endothelial cells revealed changes in the gene sets involved in lymphatic remodeling,fluid drainage,as well as inflammatory and immunological responses.Disruption of dorsal MLVs alone impaired intratumor fluid drainage and the dissemination of brain tumor cells to deep CLNs(dCLNs).Notably,the dendritic cell(DC)trafficking from intracranial tumor tissues to dCLNs decreased in mice with defective dorsal MLVs,and increased in mice with enhanced dorsal meningeal lymphangiogenesis.Strikingly,disruption of dorsal MLVs alone,without affecting basal MLVs or nasal LVs,significantly reduced the efficacy of combined anti-PD-1/CTLA-4 checkpoint therapy in striatal tumor models.Furthermore,mice bearing tumors overexpressing VEGF-C displayed a better response to anti-PD-1/CTLA-4 combination therapy,and this was abolished by CCL21/CCR7 blockade,suggesting that VEGF-C potentiates checkpoint therapy via the CCL21/CCR7 pathway.Together,the results of our study not only demonstrate the functional aspects of MLVs as classic lymphatic vasculature,but also highlight that they are essential in generating an efficient immune response against brain tumors.Xueting Hu Qiuping Deng Lu Ma Qingqing Li Yidong Chen Yuhan Liao Fan Zhou Chen Zhang Linlin Shao Jun Feng Tubao He Weihai Ning Yan Kong Yingqing Huo Aibin He Bing Liu Jingjing Zhang Ralf Adams Yulong He Fuchou Tang Xiuwu Bian Jincai Luo 2020Cell Research2020,30,3:11
2EGTA reduces the inflorescence stem mechanical strength of herbaceous peony by modifying secondary wall biosynthesis显示文摘The mechanical strength of inflorescence stems is an important trait in cut flowers.Calcium ions(Ca^(2+))play a pivotal role in maintaining stem strength,but little is known about the underlying molecular mechanisms.In this study,we treated herbaceous peony(Paeonia lactiflora Pall.)with ethyl glycol tetraacetic acid(EGTA),an effective Ca^(2+)chelator,and used morphology indicators,spectroscopic analysis,histochemical staining,electron microscopy,and proteomic techniques to investigate the role of Ca^(2+)in inflorescence stem mechanical strength.The EGTA treatment reduced the mechanical strength of inflorescence stems,triggered the loss of Ca^(2+)from cell walls,and reduced lignin in thickened secondary walls in xylem cells as determined by spectroscopic analysis and histochemical staining.Electron microscopy showed that the EGTA treatment also resulted in significantly fewer xylem cell layers with thickened secondary walls as well as in reducing the thickness of these secondary walls.The proteomic analysis showed 1065 differentially expressed proteins(DEPs)at the full-flowering stage(S4).By overlapping the Kyoto encyclopedia of genes and genomes(KEGG)and gene ontology(GO)analysis results,we identified 43 DEPs involved in signal transduction,transport,energy metabolism,carbohydrate metabolism,and secondary metabolite biosynthesis.Using quantitative real-time polymerase chain reaction(qRT-PCR)analysis,we showed that EGTA treatment inhibited Ca^(2+)sensors and secondary wall biosynthesis-related genes.Our findings revealed that EGTA treatment reduced the inflorescence stem mechanical strength by reducing lignin deposition in xylem cells through altering the expression of genes involved in Ca^(2+)binding and secondary wall biosynthesis.Yuhan Tang Daqiu Zhao Jiasong Meng Jun Tao 2019Horticulture Research2019,6,1:2

7Lignin provides mechanical support to herbaceous peony(Paeonia lactiflora Pall.)stems显示文摘Stem bending caused by mechanical failure is a major constraint for high-quality herbaceous peony(Paeonia lactiflora Pall.)cut flowers,but little is known about the underlying factors.In this study,two P.lactiflora cultivars,Xixia Yingxue(bending)and Hong Feng(upright),were used to investigate differences in stem bending.The results showed that the stem mechanical strength of Hong Feng was significantly higher than that of Xixia Yingxue,and the thickening of the secondary cell wall and the number of thickened secondary cell wall layers in Hong Feng were significantly higher than those in Xixia Yingxue.Moreover,compared with Xixia Yingxue,Hong Feng showed greater lignification of the cell wall and lignin deposition in the cell walls of the sclerenchyma,vascular bundle sheath and duct.All three types of lignin monomers were detected.The S-lignin,G-lignin,and total lignin contents and the activities of several lignin biosynthesis-related enzymes were higher in Hong Feng than in the other cultivar,and the S-lignin content was closely correlated with stem mechanical strength.In addition,113,974 full-length isoforms with an average read length of 2106 bp were obtained from the full-length transcriptome of P.lactiflora stems,and differential expression analysis was performed based on the comparative transcriptomes of these two cultivars.Ten lignin biosynthesis-related genes,including 26 members that were closely associated with lignin content,were identified,and multiple upregulated and downregulated transcription factors were found to positively or negatively regulate lignin biosynthesis.Consequently,lignin was shown to provide mechanical support to P.lactiflora stems,providing useful information for understanding the formation of P.lactiflora stem strength.Daqiu Zhao Yuting Luan Xing Xia Wenbo Shi Yuhan Tang Jun Tao 2020Horticulture Research2020,7,1:1
8Deterioration of Antioxidant Competence in Barley Lesion Mimic Mutant 194显示文摘A barley mutant,194,was observed to exhibit a leaf spot phenotype over the whole course of its growing period.In this study,the phenotype and antioxidant competence were studied in the lesion mimic mutant 194.Plant height was slightly higher in mutant 194 than in the wild type(WT).In addition,leaf spot per plant in mutant 194 was significantly higher than in WT.Antioxidant competence,as indicated by reactive oxygen species(ROS)accumulation,antioxidant enzyme activity,and the expression of antioxidant enzyme-encoding genes was also assessed in mutant 194.Compared to the WT,mutant 194 displayed a relatively higher accumulation of ROS,accompanied by lower activities of some antioxidant enzymes and downregulation of antioxidant enzyme-encoding genes.This demonstrated reduced antioxidant competence in mutant 194.The results suggested that this lower antioxidant competence of mutant 194 could lead to the accumulation of excessive ROS.This excess of ROS could induce programmed cell death and has the potential to promote disease resistance in mutant 194.Qunqun Hao Bo Lyu Yuhan Tang Deya Wang Yuanyuan Li Qingliang Li Yuhai Wang Wenqiang Wang 2019Phyton-International Journal of Experimental Botany2019,88,2:1
9Sensitivity-Enhanced Hot-Wire Anemometer by Using Cladding-Etched Fiber Bragg Grating显示文摘A sensitivity-enhanced hot-wire anemometer based on a cladding-etched optical fiber Bragg grating(FBG)coated with a layer of silver film and optically heated by using a 1480nm laser diode is demonstrated.The silver film absorbs the laser power to heat the FBG to a certain high temperature and the airflow cools down the FBG hot-wire with the cooling effect and hence the Bragg wavelength of the FBG is determined by the airflow velocity.Experimental measurement results show that the heating efficiency of the FBG hot wire is improved by 3.8times in magnitude by etching the fiber cladding from 125μm down to 73.4μm,and the achieved airflow velocity sensitivities,under a laser power of 200mW,are–3180pm/(m/s),–889pm/(m/s),–268pm/(m/s),and–8.7pm/(m/s)at different airflow velocities of 0.1m/s,0.5m/s,1.5m/s,and 17m/s,respectively.In comparison,the sensitivities are only–2193 pm/(m/s),–567 pm/(m/s),–161 pm/(m/s),and–4.9pm/(m/s)for the reference anemometer without cladding etching even at a much higher heating laser power of 530mW.These results prove that the method by using a cladding-etched FBG to improve sensitivity of FBG-based hot-wire anemometers works and the sensitivity is improved significantly.Yuhan TANG Xuke CHEN Jiarui ZHANG Dajuan LV Liangming XIONG Xinyong DONG 2023Photonic Sensors2023,13,3:0

11The chemical reprogramming of unipotent adult germ cells towards authentic pluripotency and de novo establishment of imprinting显示文摘Although somatic cells can be reprogrammed to pluripotent stem cells(PsCs)with pure chemicals,authentic pluripotency of chemically induced pluripotent stem celis(CipsCs)has never been achieved through tetraploid complementation assay.Spontaneous reprogramming of spermatogonial stem cells(ssCs)was another non-transgenic way to obtain PsCs,but this process lacks mechanistic explanation.Here,we reconstructed the trajectory of mouse SsC reprogramming and developed a five-chemical combination,boosting the reprogramming effciency by nearly 80-to 100-folds.More importantly,chemical induced germline-derived PsCs(5C-gPSCs),but not gpsCs and chemical induced pluripotent stem cells,had authentic pluripotency,as determined by tetraploid complementation.Mechanistically,ssCs traversed through an inverted pathway of in vivo germ ceil development,exhibiting the expression signatures and DNA methylation dynamics from spermatogonia to primordial germ cells and further to epiblasts.Besides,ssC-specific imprinting control regions switched from biallelic methylated states to monoallelic methylated states by imprinting demethylation and then re-methylation on one of the two alleles in 5c-gPsCs,which was apparently distinct with the imprinting reprogramming in vivo as DNA methylation simultaneously occurred on both alleles.Our work sheds ight on the unique regulatory network underpinning SsC reprogramming,providing insights to understand generic mechanisms for cell-fate decision and epigenetic-relateddisorders in regenerative medicine.Yuhan Chen Jiansen Lu Yanwen Xu Yaping Huang Dazhuang Wang Peiling Liang Shaofang Ren Xuesong Hu Yewen Qin Wei Ke Ralf Jauch Andrew Paul Hutchins Mei Wang Fuchou Tang Xiao-Yang Zhao 2023Protein & Cell2023,14,7:0
12A Statistical Algorithm for Retrieving Background Value of Absorbing Aerosol Index Based on TROPOMI Measurements显示文摘The ultraviolet aerosol index(UVAI) is essential for monitoring the absorbing aerosols during aerosol events. UVAI depends on the absorbing aerosol concentration, the viewing geometry, and the temporal drift of radiometric sensitivity. To efficiently detect absorbing aerosols with the highest precision and to improve the accuracy of long-term UVAI estimates,the background UVAI must be examined through the UVAI retrieval. This study presents a statistical method that calculates the background value of UVAI using TROPOspheric Monitoring Instrument(TROPOMI) observation data over the Pacific Ocean under clear-sky scenes. Radiative transfer calculations were performed to simulate the dependence of UVAI on aerosol type and viewing geometry. We firstly applied the background UVAI to reducing the effects of viewing geometry and the degradation of the TROPOMI irradiance measurements on the UVAI. The temporal variability of the background UVAI under the same viewing geometry and aerosol concentration was identified. Radiative transfer calculations were performed to study the changes in background UVAI using Aerosol Optical Depth from the Moderate Resolution Imaging Spectroradiometer(MODIS) and reflectance measurements from TROPOMI as input. The trends of the temporal variations in the background UVAI agreed with the simulations. Alterations in the background UVAI expressed the reflectance variations driven by the changes in satellite state. Decreasing trends in solar irradiance at 340 and 380 nm due to instrument degradation were identified. 13A domesticated Harbinger transposase forms a complex with HDA6 and promotes histone H3 deacetylation at genes but not TEs in Arabidopsis显示文摘In eukaryotes,histone acetylation is a major modification on histone N-terminal tails that is tightly connected to transcriptional activation.HDA6 is a histone deacetylase involved in the transcriptional regulation of genes and transposable elements(TEs)in Arabidopsis thaliana.HDA6 has been shown to participate in several complexes in plants,including a conserved SIN3 complex.Here,we uncover a novel protein complex containing HDA6,several Harbinger transposon-derived proteins(HHP1,SANT1,SANT2,SANT3,and SANT4),and MBD domain-containing proteins(MBD1,MBD2,and MBD4).We show that mutations of all four SANT genes in the sant-null mutant cause increased expression of the flowering repressors FLC,MAF4,and MAF5,resulting in a late flowering phenotype.Transcriptome deep sequencing reveals that while the SANT proteins and HDA6 regulate the expression of largely overlapping sets of genes,TE silencing is unaffected in sant-null mutants.Our global histone H3 acetylation profiling shows that SANT proteins and HDA6 modulate gene expression through deacetylation.Collectively,our findings suggest that Harbinger transposon-derived SANT domain-containing proteins are required for histone deacetylation and flowering time control in plants.Xishi Zhou Junna He Christos N.Velanis Yiwang Zhu Yuhan He Kai Tang Mingku Zhu Lisa Graser Erica de Leau Xingang Wang Lingrui Zhang W.Andy Tao Justin Goodrich Jian-Kang Zhu Cui-Jun Zhang 2021Journal of Integrative Plant Biology2021,63,8:0
14Age-Related Alterations in DNA Methylation and APETALA2(AP2)Levels in Herbaceous Peony(Paeonia lactiflora Pall.)显示文摘The ornamental and commercial values of herbaceous peony(Paeonia lactiflora Pall.)are directly related to its flower pattern.However,the molecular mechanisms underlying the type formation of P.lactiflora flowers have not been studied in great detail.Previous studies identified,using integrated multipleomics analysis,revealed that APETALA2(AP2)is an important candidate gene that modulates type formation of P.lactiflora flowers.To further reveal the expression mechanism of AP2 in P.lactiflora petals,we examined the profile of AP2 expression in the inner and outer petals of‘ZiFengyu’at various developmental stages using qRT-PCR and BSP+Miseq methylation analysis.Based on our data,the AP2 levels in the outer petals were obviously increased,relative to the inner petals.In addition,the S3 levels at the bloom stage were significantly higher than at the flower-bud stage S1,thereby promoting bloom stage S2,while declining stage S4.Using chromosome walking,the 2000 bp of the 5′-end upstream promoter region was achieved.This region harbored a CpG island(−665∼−872 bp),with multiple essential transcription factor binding sites(TFBS)such as NF-kappa B,GATA-1,Sp1,and C/EBP.Methylation sequencing revealed 7 methylated CpG sites in the CpG island region of the AP2 promoter,thereinto,the methylation ratio of the CpG-3 site in the inner petals was significantly higher than in the outer petals.Correlation analysis revealed a negative association between the level of methylation(CpG-3,CpG-6),and AP2 mRNA expression.CpG-3 was located on the Sp1 transcription factor binding site.Thus,we speculated that the CpG-3 methylation may inhibit transcription factor Sp1 binding to the gene promoter,thereby regulating AP2 expression.Herein,we examined the role of AP2 in the determination of flower patterns in P.lactiflora.Our conclusion will provide theoretical guidance for the molecular breeding of the flower pattern in P.lactiflora.Yanqing Wu Jiao Liu Yuhan Tang Daqiu Zhao Jun Tao 2022Phyton-International Journal of Experimental Botany2022,91,9:0
15Facile synthesis of ultrabright luminogens with specific lipid droplets targeting feature for in vivo two-photon fluorescence retina imaging显示文摘The application of fluorescent probes for in vivo retinal imaging is of great importance,which could provide direct and crucial imaging evidence for a better understanding of common eye diseases.Herein,a group of bright organic luminogens with typical electron-donating(D)and electron-accepting(A)structures(abbreviated as LDs-BDM,LDs-BTM,and LDs-BHM)was synthesized through a simple single-step reaction.They were found to be efficient solid-state emitters with high fluorescence quantum yields of above 70%(e.g.,83.7%for LDs-BTM).Their light-emission properties could be tuned by the modulation ofπ-conjugation effect with methoxy groups at different substituent positions.Their resulting fluorescent nanoparticles(NPs)were demonstrated as specific lipid droplets(LDs)targeting probes with high brightness,good biocompatibility,and satisfactory photostability.LDs-BTM NPs with a large two-photon absorption cross section(σ2=249 GM)were further utilized as ultrabright two-photon fluorescence(2PF)nanoprobes for in vivo retina imaging of live zebrafish by NIR excitation at an ultralow concentration(0.5μmol/L).Integrated histological structures at the tissue level and corresponding fine details at the cellular level of the embryonic retina of live zebrafish were clearly demonstrated.This is the first report of using ultrabright LDs-targeting nanoprobes to accurately measure fine details in the retina with 2PF microscopic technique.These good results are anticipated to open up a new avenue in the development of efficient 2PF emitters for non-invasive bioimaging of living animals.Huifang Su Tao Xie Yong U.Liu Yuhan Cui Wei Wen Ben Zhong Tang Wei Qin 2023Chinese Chemical Letters2023,34,6:0

17Correction of measurement errors on sediment concentration sampled by stirring-sampling method from traditional runoff collection tanks显示文摘Stirring-sampling method is a widely adopted method to measure sediment concentrations in collection tanks of runoff plots,but with high systematic measurement errors.This research aimed to advance an approach for building correction equations to remove measurement errors in designed sediment concentration range.Experimental data of sediment measurement from the stirring-sampling method,with four representative soils,under the designed sediment concentrations(1,2,5,8,10,20,50,80,100,200,500,800,and 1000 kg/m^(3))were used to demonstrate the correction methodological process.Two correction methods(step-wise correction and universal correction)were suggested for the trial in this study based on the distribution of measurement errors.In the step-wise correction,the correction equations were made with a series of linear functions without intercept for the low concentration group(0-20 kg/m^(3)),a series of linear functions with intercept for the high(20-200 kg/m^(3))and extremely high(200-1000 kg/m^(3))concentration groups,consecutively.The correction equations were a series of power functions in the universal correction.For the step-wise correction,most of the relative errors of correction sediment concentrations were smaller than 15%and 10%under high and extremely high concentration groups,but the corrected accuracy was not good in the sediment concentration of 1,2,5 kg/m^(3)with the corrected relative errors of 0.20%-206.07%.For the universal correction,the corrected relative errors(0.19%-31.81%)of the four soils were low under the condition of extremely high sediment concentrations,but other corrected accuracies weren’t good with the corrected relative errors of 0.68%-1154.71%.The corrected accuracy of step-wise correction is higher than that of the universal correction,but the universal correction is more convenient.These results indicated that the correction equations could efficiently revise the measurement errors of the tested soils and that this method can be generalized to other soil types and was meaningful in monitoring soil erosion.Yuhan Huang Mingquan Zhao Hui Zhao Tingwu Lei Ju Tang Wei Wang 2022International Journal of Agricultural and Biological Engineering2022,15,6:0
18Autonomous vehicles for micro-mobility显示文摘Autonomous vehicles have been envisioned for more than 100 years.One of the first suggestions was a front cover of Scientific America back in 1916.Today,it is possible to get cars that drive autonomously for extended distances.We are also starting to see micro-mobility solutions,such as the Nuro vehicles for pizza delivery.Building autonomous cars that can operate in urban environments with a diverse set of road-users is far from trivial.Early 2018 the Contextual Robotics Institute at UC San Diego launched an effort to build a full stack autonomous vehicle for micro-mobility.The motivations were diverse:i)development of a system for operation in an environment with many pedestrians,ii)design of a system that does not rely on dense maps(or HD-maps as they are sometimes named),iii)design strategies to build truly robust systems,and iv)a framework to educate next-generation engineers.In this paper,we present the research effort of design,prototyping,and evaluation of such a vehicle.From the evaluation,several research directions are explored to account for shortcomings.Lessons and issues for future work are additionally drawn from this work.Henrik Christensen David Paz Hengyuan Zhang Dominique Meyer Hao Xiang Yunhai Han Yuhan Liu Andrew Liang Zheng Zhong Shiqi Tang 2021Autonomous Intelligent Systems2021,1,1:0
