2Generation of B Cell-Deficient Pigs by Highly Efficient CRISPR/Cas9-Mediated Gene Targeting显示文摘Generating B cell-deficient mutant is the first step to produce human antibody repertoires in large animal models. In this study, we applied the clustered regularly interspaced short palindromic repeat(CRISPR)/CRISPR-associated(Cas) system to target the JHregion of the pig Ig M heavy chain gene which is crucial for B cell development and differentiation. Transfection of Ig M-targeting Cas9 plasmid in primary porcine fetal fibroblasts(PFFs) enabled inducing gene knock out(KO) in up to 53.3% of colonies analyzed, a quarter of which harbored biallelic modification, which was much higher than that of the traditional homologous recombination(HR). With the aid of somatic cell nuclear transfer(SCNT) technology, three piglets with the biallelic Ig M heavy chain gene mutation were produced. The piglets showed no antibody-producing B cells which indicated that the biallelic mutation of the Ig M heavy chain gene effectively knocked out the function of the Ig M and resulted in a B cell-deficient phenotype. 4Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 6Occurrence of Aflatoxin B_(1),deoxynivalenol and zearalenone in feeds in China during 2018-2020显示文摘Background:The current study was conducted to investigate the individual and combined occurrence of aflatoxin B_(1)(AFB_(1)),deoxynivalenol(DON)and zearalenone(ZEN)in feeds from various Provinces of China during 2018 to 2020.A total of 3,507 feed samples,including 2,090 feed ingredients and 1,417 complete feed samples,were collected from different areas of China for mycotoxins analysis.Results:The individual contamination of AFB_(1),DON and ZEN were present in more than 81.9%,96.4% and 96.9% of feed samples,respectively,with average concentration ranges of AFB_(1) between 1.2-27.4μg/kg,DON between 458.0-1,925.4μg/kg and ZEN between 48.1-326.8μg/kg.Notably,0.9%,0.5% and 0.1% of feed ingredients,and 1.2-12.8%,0.9-2.9% and 0-8.9% of complete feeds for pigs,poultry and ruminants with AFB_(1),ZEN and DON that exceeded China’s safety standards,respectively.Moreover,more than 81.5%of feed ingredients and 95.7% of complete feeds were co-contaminated with various combinations of these mycotoxins.Conclusion:This study indicates that the feeds in China were universally contaminated with AFB_(1),DON and ZEN during the past 3 years.These findings highlight the significance of monitoring mycotoxin contaminant levels in the domestic animal feed,and the importance of carrying out feed administration and remediation strategies for mycotoxin control.Ling Zhao Lei Zhang Zijian Xu Xingda Liu Liyuan Chen Jiefan Dai Niel Alexander Karrow Lvhui Sun 2022Journal of Animal Science and Biotechnology2022,13,1:10

8Experimental and modeling study of NO emission under high CO_2 concentration显示文摘An experimental and numerical study of the NOx formation and reduction process in a designed coal combustion furnace under both traditional air atmosphere and O2/CO2 atmosphere was conducted, in an attempt to explore the chemistry mechanism of the experimentally observed NOx suppression under high CO2 concentration atmospheres. A simplified ‘chemically oriented’ approach, computational fluid dynamics (CFD)-chemical kinetics modeling method, was validated and used to model the experimental process. The high CO2 concentration’s chemical effect on NO reduction has been studied, and the differences in NOx reaction behaviors between O2/CO2 atmosphere and air atmosphere were analyzed by detailed chemical kinetic model. On the basis of investigations through elementary chemical reactions, it can be concluded that high CO2 concentration plays an important role on NOx conversion process during oxy-fuel combustion. Moreover, the dominant reaction steps and the most important reactions for NO conversion under different atmospheres were discussed. 9Sustained‑Release Nanocapsules Enable Long‑Lasting Stabilization of Li Anode for Practical Li‑Metal Batteries显示文摘A robust solid-electrolyte interphase(SEI)enabled by electrolyte additive is a promising approach to stabilize Li anode and improve Li cycling efficiency.However,the self-sacrificial nature of SEI forming additives limits their capability to stabilize Li anode for long-term cycling.Herein,we demonstrate nanocapsules made from metal–organic frameworks for sustained release of LiNO3 as surface passivation additive in commercial carbonate-based electrolyte.The nanocapsules can offer over 10 times more LiNO3 than the solubility of LiNO3.Continuous supply of LiNO3 by nanocapsules forms a nitride-rich SEI layer on Li anode and persistently remedies SEI during prolonged cycling.As a result,lifespan of thin Li anode in 50μm,which experiences drastic volume change and repeated SEI formation during cycling,has been notably improved.By pairing with an industry-level thick LiCoO2 cathode,practical Li-metal full cell demonstrates a remarkable capacity retention of 90%after 240 cycles,in contrast to fast capacity drop after 60 cycles in LiNO3 saturated electrolyte.Qianqian Liu Yifei Xu Jianghao Wang Bo Zhao Zijian Li Hao Bin Wu 2020Nano-Micro Letters2020,12,12:4

12Overview to the Hard X-ray Modulation Telescope (Insight-HXMT) Satellite显示文摘As China’s first X-ray astronomical satellite, the Hard X-ray Modulation Telescope (HXMT), which was dubbed as Insight-HXMT after the launch on June 15, 2017, is a wide-band(1-250 ke V) slat-collimator-based X-ray astronomy satellite with the capability of all-sky monitoring in 0.2-3 Me V. 13Role of Heavy Metal Ions in the Formation of Oxyfluoride Glasses and Glass Ceramics显示文摘In order to investigate the influence of different heavy metal ions on the formation of the oxyfluoride glasses and glass ceramics, samples with different Pb F2/Cd F2 ratios have been prepared by the melting quenching and thermal treatment method. The different effects of Pb2 tand Cd2ton the glass network structure are investigated by FTIR and Raman spectra. During the formation of glass network structure, Pb2 tprefers to break the Sie Oe Si bond and subsequently bond to F for charge compensation, and Cd2 tprefers to break the SieO eA l bond and is surrounded by O2. Pb2 tand F gather together and form the fluoride nanocrystals,while Cd2 tremains in oxide matrix after thermal treatment. 14Temporal and spatial analysis of changes in snow cover in western Sichuan based on MODIS images显示文摘We developed a method for analyzing the change in snow cover using MODIS imagery.The method was applied to images of western Sichuan Province,China taken between 2002 and 2008.The model for extracting data on snow cover from MODIS images was created by spectral analysis.The multi-temporal snow layers were used to evaluate the temporal and spatial change in the area under snow cover between 2002 and 2008 using overlay and statistical analysis in ARCGIS.The majority(60.4%) of western Sichuan was rarely covered by snow and only 0.3% was covered by perennial snow in 2002.Snow cover was pri-marily distributed in Garzê and Aba.The area under snow cover was significantly and negatively correlated with the average monthly temperature and rainfall in 2002.The largest area under snow cover was measured in 2006 and the smallest was in 2007.Similarly,the area of snowmelt was the highest in 2006 and lowest in 2007.In general,the elevation of the snow line in-creased throughout the period 2002-2008;however,the elevation decreased in some years.Our results provide an important insight into the distribution of snow in this region,and may be useful for climate modeling and predicting the availability of water resources and the occurrence of floods and droughts.YANG CunJian ZHAO ZiJian NI Jing REN XiaoLan WANG Qin 2012Science China Earth Sciences2012,55,8:2

19Deletion of an electron donor-binding subunit of the NDH-1 complex, NdhS, results in a heat-sensitive growth phenotype in Synechocystis sp. PCC 6803显示文摘In cyanobacteria and higher plants, NdhS is suggested to be an electron donor-binding subunit of NADPH dehydrogenase(NDH-1) complexes and its absence impairs NDH-1-dependent cyclic electron transport around photosystem I(NDH-CET). Despite significant advances in the study of NdhS during recent years, its functional role in resisting heat stress is poorly understood.Here, our results revealed that the absence of NdhS resulted in a serious heat-sensitive growth phenotype in the unicellular cyanobacterium Synechocystis sp. strain PCC6803. Furthermore, the rapid and significant increase in NDH-CET caused by heat treatment was completely abolished, and the repair of photosystem II under heat stress conditions was greatly impaired when compared to that of other photosynthetic apparatus in the thylakoid membrane.
