| 5 | Integrated DNA-based/biochemical screening for early diagnosis of multiple endocrine neoplasia type 2A (MEN2A)显示文摘Multiple endocrine neoplasia type 2A (MEN2A), a subtype of MEN2, is characterized by medullary thyroid cancer, pheochromocytoma, and primary hyperparathyroidism. A Han Chinese pedigree with MEN2A was investigated following confirmation of the proband's diagnosis by pathological findings and DNA/biochemical screening. DNA samples from 4 other family members were collected and exon 5, 8, 10, 11, 13, 16 and 18 of the RET proto-oncogene were sequenced and then analyzed. A missense mutation of TGG (Trp) to TGC (Cys) at codon 634 (the classic MEN2A mutation) in exon 11 of the RET gene was detected in 3 family members, including the proband. Sequencing data were compared with the human gene mutation database. Elevated serum calcitonin level was detected initially; medullary thyroid carcinoma was revealed in the 3 cases and adrenal pheochromocy-toma was also found in the proband. Elective operations were successfully performed on the adrenal and thyroid glands because of pheochromocytoma and medullary thyroid carcinoma. Our case study confirms that integrated DNA-based/biochemical screening is crucial for early diagnosis of MEN2A and is helpful in the screening of their relatives. | 9 | Cation-vacancy induced Li+ intercalation pseudocapacitance at atomically thin heterointerface for high capacity and high power lithium-ion batteries显示文摘It is challenging to create cation vacancies in electrode materials for enhancing the performance of rechargeable lithium ion batteries (LIBs). Herein, we utilized a strong alkaline etching method to successfully create Co vacancies at the interface of atomically thin Co_(3−x)O_(4)/graphene@CNT heterostructure for high-energy/power lithium storage. The creation of Co-vacancies in the sample was confirmed by high-resolution scanning transmission electron microscope (HRSTEM), X-ray photoelectron spectroscopy (XPS) and electron energy loss near-edge structures (ELNES). The obtained Co_(3−x)O_(4)/graphene@CNT delivers an ultra-high capacity of 1688.2 mAh g^(−1) at 0.2 C, excellent rate capability of 83.7% capacity retention at 1 C, and an ultralong life up to 1500 cycles with a reversible capacity of 1066.3 mAh g^(−1). Reaction kinetic study suggests a significant contribution from pseudocapacitive storage induced by the Co-vacancies at the Co_(3−x)O_(4)/graphene@CNT interface. | 10 | Evaluation of a deep-learning model for multispectral remote sensing of land use and crop classification显示文摘High-resolution deep-learning-based remote-sensing imagery analysis has been widely used in land-use and crop-classification mapping. However, the influence of composite feature bands, including complex feature indices arising from different sensors on the backbone, patch size, and predictions in transferable deep models require further testing. The experiments were conducted in six sites in Henan province from2019 to 2021. This study sought to enable the transfer of classification models across regions and years for Sentinel-2 A(10-m resolution) and Gaofen PMS(2-m resolution) imagery. With feature selection and up-sampling of small samples, the performance of UNet++ architecture on five backbones and four patch sizes was examined. Joint loss, mean Intersection over Union(m Io U), and epoch time were analyzed, and the optimal backbone and patch size for both sensors were Timm-Reg Net Y-320 and 256 × 256, respectively. The overall accuracy and Fscores of the Sentinel-2 A predictions ranged from 96.86% to 97.72%and 71.29% to 80.75%, respectively, compared to 75.34%–97.72% and 54.89%–73.25% for the Gaofen predictions. The accuracies of each site indicated that patch size exerted a greater influence on model performance than the backbone. The feature-selection-based predictions with UNet++ architecture and upsampling of minor classes demonstrated the capabilities of deep-learning generalization for classifying complex ground objects, offering improved performance compared to the UNet, Deeplab V3+, Random Forest, and Object-Oriented Classification models. In addition to the overall accuracy, confusion matrices,precision, recall, and F1 scores should be evaluated for minor land-cover types. | 16 | A simple and sensitive assay for apurinic/apyrimidinic endonuclease 1activity based on host-guest interaction of β-cyclodextrin polymer and pyrene显示文摘Apurinic/apyrimidinic endonuclease 1(APE1) plays an important role in protecting the fidelity of genetic material and is also a promising biomarker of cancers. Herein, a simple and sensitive fluorescent method for APE1 activity detection was designed based on host-guest interaction between β-cyclodextrin polymer(β-CDP) and pyrene. In this method, pyrene-labelled DNA probes(AP-S1 S2) and β-CDP functioned as fluorescent signal producer and enhancer, respectively. When APE1 was absent, pyrene on the AP-S1 S2 could not enter the cavity of β-CDP because of steric hindrance, leading to a weak fluorescence intensity/anisotropy. When APE1 was present, it would recognize and cleave AP site in APS1 S2, thus producing a dissociated pyrene-labelled oligonucleotide segment. Pyrene labelled at the 50 end of oligonucleotide segment could be easily trapped into the cavity of β-CDP via host-guest interaction, leading to a significant enhancement of fluorescence intensity/anisotropy. This assay offered a dynamic range of 0.05 U/m L–5 U/m L and an estimated detection limit of 0.05 U/m L. | 17 | Stable perovskite solar cells with 23.12% efficiency and area over 1 cm^(2) by an all-in-one strategy显示文摘To realize the commercialization of perovskite solar cells(PSCs), it is required to overcome the remaining challenges in device enlargement and operational stability. Here, we report an all-in-one strategy by integrating the oxidation of hole-transport material(HTM) with the formation of the passivation layer, which simultaneously solved the stability issues caused by HTM oxidation and realized the uniform defects in passivation over a large area. The resulting devices achieved a certified PCE of23.12% on average with an aperture area of 1.04 cm^(2) and are reproducible with high operational stability because of the exclusion of air exposure, hygroscopic Li-TFSI, and the lithium-based wastes, maintaining ca. 90% of their initial PCEs after operation at the maximum power point under continuous 1 sun illumination for 1,600 h.
