2First experimental constraints on WIMP couplings in the effective field theory framework from CDEX显示文摘We present weakly interacting massive particles(WIMPs) search results performed using two approaches of effective field theory from the China Dark Matter Experiment(CDEX), based on the data from both CDEX-1B and CDEX-10 stages. In the nonrelativistic effective field theory approach, both time-integrated and annual modulation analyses were used to set new limits for the coupling of WIMP-nucleon effective operators at 90% confidence level(C.L.) and improve over the current bounds in the low mχregion. In the chiral effective field theory approach, data from CDEX-10 were used to set an upper limit on WIMP-pion coupling at 90% C.L. 3Brain imaging of mild cognitive impairment and Alzheimer's disease显示文摘The rapidly increasing prevalence of cognitive impairment and Alzheimer's disease has the potential to create a major worldwide healthcare crisis. Structural MRI studies in patients with Alzheimer's disease and mild cognitive impairment are currently attracting considerable interest. It is extremely important to study early structural and metabolic changes, such as those in the hippocampus, entorhinal cortex, and gray matter structures in the medial temporal lobe, to allow the early detection of mild cognitive impairment and Alzheimer's disease. The microstructural integrity of white matter can be studied with diffusion tensor imaging. Increased mean diffusivity and decreased fractional anisotropy are found in subjects with white matter damage. Functional imaging studies with positron emission tomography tracer compounds enable detection of amyloid plaques in the living brain in patients with Alzheimer's disease. 4Photocrosslinking silver nanoparticles–aloe vera–silk fibroin composite hydrogel for treatment of full-thickness cutaneous wounds显示文摘Damage to the skin causes physiological and functional issues.The most effective treatment approach is the use of wound dressings.Silk fibroin(SF)is a promising candidate biomaterial for regulating wound healing;however,its antibacterial properties and biological activity must be further improved.In this study,a photocrosslinking hydrogel was developed to treat full-thickness cutaneous wounds.The composite hydrogel(Ag-AV-SF hydrogel)was prepared by introducing the silver nanoparticles(AgNPs)and aloe vera(AV)as the modifiers.In vitro study exhibited great antibacterial ability,biocompatibility and cell-proliferation and-migration-promoting capacities.It also showed the pH-response releasing properties which release more AgNPs in a simulated chronic infection environment.The healing effect evaluation in vivo showed the healing-promoting ability of the Ag-AV-SF hydrogel was stronger than the single-modifiers groups,and the healing rate of it reached 97.02%on Day 21,higher than the commercial wound dressing,silver sulfadiazine(SS)cream on sale.Additionally,the histological and protein expression results showed that the Ag-AV-SF hydrogel has a greater effect on the pro-healing regenerative phenotype with M2 macrophages at the early stage,reconstructing the blood vessels networks and inhibiting the formation of scars.In summary,the Ag-AV-SF hydrogel developed in this study had good physical properties,overwhelming antibacterial properties,satisfactory biocompatibility and significantly promoting effect on cell proliferation,migration and wound healing.Overall,our results suggest that the Ag-AV-SF hydrogel we developed has great potential for improving the wound healing in clinical treatment.Yangkun Liu JinChuan Fan MingQi Lv Kepeng She Jiale Sun Qingqing Lu Changhao Han SongTao Ding Shuang Zhao GuiXue Wang YuChan Zhang GuangChao Zang 2021Regenerative Biomaterials2021,8,6:2
5Dynamic oxidation mechanism of carbon fiber reinforced SiC matrix composite in high-enthalpy and high-speed plasmas显示文摘This work employed an inductively coupled plasma wind tunnel to study the dynamic oxidation mechanisms of carbon fiber reinforced SiC matrix composite(C_(f)/SiC)in high-enthalpy and high-speed plasmas.The results highlighted a transition of passive/active oxidations of SiC at 800–1600℃and 1–5 kPa.Specially,the active oxidation led to the corrosion of the SiC coating and interruption of the SiO_(2) growth.The transition borders of active/passive oxidations were thus defined with respect to oxidation temperature and partial pressure of atomic O in the high-enthalpy and high-speed plasmas.In the transition and passive domains,the SiC dissipation was negligible.By multiple dynamic oxidations of C_(f)/SiC in the domains,the SiO_(2) thickness was not monotonously increased due to the competing mechanisms of passive oxidation of SiC and dissipation of SiO_(2).In addition,the mechanical properties of the SiC coating/matrix and the C_(f)/SiC were maintained after long-term dynamic oxidations,which suggested an excellent thermal stability of C_(f)/SiC serving in thermal protection systems(TPSs)of reusable hypersonic vehicles.Lingwei YANG Xueren XIAO Liping LIU Jie LUO Kai JIANG Xinxing HAN Changhao ZHAO Jun ZHANG Guolin WANG 2022Journal of Advanced Ceramics2022,11,2:1

7SUMOylation-triggered ALIX activation modulates extracellular vesicles circTLCD4-RWDD3 to promote lymphatic metastasis of non-small cell lung cancer显示文摘Lymph node(LN)metastasis is one of the predominant metastatic routes of non-small cell lung cancer(NSCLC)and is considered as a leading cause for the unsatisfactory prognosis of patients.Although lymphangiogenesis is well-recognized as a crucial process in mediating LN metastasis,the regulatory mechanism involving lymphangiogenesis and LN metastasis in NSCLC remains unclear.In this study,we employed high-throughput sequencing to identify a novel circular RNA(circRNA),circTLCD4-RWDD3,which was significantly upregulated in extracellular vesicles(EVs)from LN metastatic NSCLC and was positively associated with deteriorated OS and DFS of patients with NSCLC from multicenter clinical cohort.Downregulating the expression of EV-packaged circTLCD4-RWDD3 inhibited lymphangiogenesis and LN metastasis of NSCLC both in vitro and in vivo.Mechanically,circTLCD4-RWDD3 physically interacted with hnRNPA2B1 and mediated the SUMO2 modification at K108 residue of hnRNPA2B1 by upregulating UBC9.Subsequently,circTLCD4-RWDD3-induced SUMOylated hnRNPA2B1 was recognized by the SUMO interaction motif(SIM)of ALIX and activated ALIX to recruit ESCRT-III,thereby facilitating the sorting of circTLCD4-RWDD3 into NSCLC cell-derived EVs.Moreover,EV-packaged circTLCD4-RWDD3 was internalized by lymphatic endothelial cells to activate the transcription of PROX1,resulting in the lymphangiogenesis and LN metastasis of NSCLC.Importantly,blocking EV-mediated transmission of circTLCD4-RWDD3 via mutating SIM in ALIX or K108 residue of hnRNPA2B1 inhibited the lymphangiogenesis and LN metastasis of NSCLC in vivo.Our findings reveal a precise mechanism underlying SUMOylated hnRNPA2B1-induced EV packaging of circTLCD4-RWDD3 in facilitating LN metastasis of NSCLC,suggesting that EV-packaged circTLCD4-RWDD3 could be a potential therapeutic target against LN metastatic NSCLC.Xiayao Diao Chao Guo Hanhao Zheng Ke Zhao Yuming Luo Mingjie An Yan Lin Jiancheng Chen Yuanlong Li Yuting Li Xuehan Gao Jiaqi Zhang Mengxin Zhou Wenliang Bai Lei Liu Guige Wang Lanjun Zhang Xiaotian He Rusi Zhang Zhihua Li Changhao Chen Shanqing Li 2023Signal Transduction and Targeted Therapy2023,8,12:0

9An HGF-dependent positive feedback loop between bladder cancer cells and fibroblasts mediates lymphangiogenesis and lymphatic metastasis显示文摘Background Cancer-associated fibroblasts (CAFs) play a vital role in facilitating tumor progression through extensive reciprocal interplay with cancer cells. Tumor-derived extracellular vesicles (EVs) are the critical mediators involved in the crosstalk between cancer cells and stromal cells, contributing to the metastasis of cancers. Yet, the biological mechanisms of tumor-derived EVs in triggering CAFs phenotype to stimulate the lymph node (LN) metastasis of bladder cancer (BCa) are largely unknown. Here, we aimed to explore the effects and molecular mechanisms of tumor-derived EV-mediated CAFs phenotype in regulating BCa LN metastasis. Methods The high-throughput sequencing was utilized to identify the crucial long non-coding RNA (lncRNA) associated with CAF enrichment in BCa. The functional role of the transition of fibroblasts to CAFs induced by LINC00665-mediated EVs was investigated through the in vitro and in vivo assays. Chromatin isolation by RNA purification assays, fluorescence resonance energy transfer assays, cytokine profiling and patient-derived xenograft (PDX) model were performed to explore the underlying mechanism of LINC00665 in the LN metastasis of BCa. Results We found that CAFs are widely enriched in the tumor microenvironment of BCa, which correlated with BCa lymphangiogenesis and LN metastasis. We then identified a CAF-associated long non-coding RNA, LINC00665, which acted as a crucial mediator of CAF infiltration in BCa. Clinically, LINC00665 was associated with LN metastasis and poor prognosis in patients with BCa. Mechanistically, LINC00665 transcriptionally upregulated RAB27B expression and induced H3K4me3 modification on the promoter of RAB27B through the recruitment of hnRNPL. Moreover, RAB27B-induced EVs secretion endowed fibroblasts with the CAF phenotype, which reciprocally induced LINC00665 overexpression to form a RAB27B-HGF-c-Myc positive feedback loop, enhancing the lymphangiogenesis and LN metastasis of BCa. Importantly, we demonstrated that blocking EV-transmitted LINC00665 or HGF broke this loop and impaired BCa lymphangiogenesis in a PDX model. 11The effect of different Co phase structure(FCC/HCP)on the catalytic action towards the hydrogen storage performance of MgH_(2)显示文摘High hydrogen desorption temperature and sluggish reaction kinetics are the major limitations for the practical application of MgH_(2).In this study,Co particles with a face centered cubic(FCC)structure and a hexagonal close packed(HCP)structure were prepared facilely and proved to be good catalysts for magnesium hydride.Co particles with FCC structure presented better catalytic effect on MgH_(2)than that with HCP structure.Both 7%(mass)Co FCC and HCP particle modified MgH_(2)decreased the initial dehydrogenation temperature from 301.3℃ to approximately 195.0℃,but 7%(mass)Co with FCC structure modified MgH_(2)has a faster desorption rate,and around 6.5%(mass)H_(2)was desorbed in 10 min at325℃.Hydrogen uptake was detected at 70℃ under 3.25 MPa hydrogen pressure and 6.0%(mass)H_(2)was recharged in 40 min at 150℃.The hydrogen desorption and absorption activation energy for 7%(mass)FCC Co modified MgH_(2)was significantly decreased to(76.6±8.3)kJ·mol^(-1) and(68.3±6.0)kJ·mol^(-1),respectively.Thermodynamic property was also studied,the plateau pressures of MgH_(2)+7%(mass)FCC Co were determined to be 0.14,0.28,0.53 and 0.98 MPa for 300℃,325℃,350℃ and375℃.The decomposition enthalpy of hydrogen(ΔH)for MgH_(2)+7%(mass)FCC Co was(80.6±0.1)kJ·mol^(-1),5.8 kJ·mol^(-1)lower than that of as-prepared MgH_(2).Moreover,cycling performance for the first20 cycles revealed that the reaction kinetics and capacity of MgH_(2)-FCC Co composite remained almost unchanged.The result of density functional theory calculation demonstrated that cobalt could extract the Mg AH bond and reduced the decompose energy of magnesium hydride.Our paper can be presented as a reference for searching highly effective catalysts for hydrogen storage and other energy-related research fields.Liuting Zhang Haijie Yu Zhiyu Lu Changhao Zhao Jiaguang Zheng Tao Wei Fuying Wu Beibei Xiao 2022Chinese Journal of Chemical Engineering2022,35,3:0
12Correlation analysis of intestinal flora and pathological process of type 2 diabetes mellitus显示文摘Objective: To observe the relationship between the different stages of type 2 diabetes mellitus(T2DM)and the intestinal flora and verify its underlying mechanism.Methods: T2DM rats were generated by high-fat diet(HFD) combined with intraperitoneal streptozotocin(STZ) injection. The rats were divided into four groups: the control group(fed with normal feed for1 month), the HFD group(fed with HFD for 1 month), the T2DM group(HFD combined with STZ and blood glucose ≥11.1 m M), and the unformed T2DM model(Un-mod) group(HFD combined with STZ and blood glucose <11.1 m M). Feces were collected, and bacterial communities in the fecal samples were analyzed by 16S r RNA gene sequencing. The content of short-chain fatty acids(SCFAs) in feces was measured by gas chromatography. Western blot and quantitative real-time polymerase chain reaction were used to detect the expression of G protein-coupled receptor 41(GPR41) and GPR43.Results: At different stages of T2DM, the intestinal flora and SCFAs content of rats were significantly decreased(all P <.05). Our results indicated that g__Prevotella had a significant negative correlation, and g__Ruminococcus_torques_group and g__lachnoclastic had a significant positive correlation with blood glucose. The content of SCFAs, in particular acetate and butyrate, in rat feces of different stages of T2DM were significantly reduced, as well as GPR41 and GPR43 expression. The results in the Un-mod group were similar to the T2DM group, and the expression of GPR41 and GPR43 proteins were significantly higher than those in the T2DM group(both P <.001).Conclusion: The intestinal flora-SCFAs-GPR41/GPR43 network may be important in the development of T2DM. 15A comprehensive review on polyurethane modified asphalt: Mechanism, characterization and prospect显示文摘Polyurethane(PU),with excellent physical and chemical properties and high designability,is one of the ideal materials for asphalt modification in the future.In this paper,based on the limitations of traditional asphalt modifiers,the preparation process,relative advantages and development prospects of PU as asphalt modifiers are described.Subsequently,the spatial structure,physical and chemical properties of PU synthetic raw materials were combined with the modification properties of PU to analyze the effect and influence of PU on asphalt modification.Specifically,polyurethane modified asphalt(PUMA)is divided into thermoplastic polyurethane modified asphalt(TP-PUMA)and thermosetting polyurethane modified asphalt(TS-PUMA).The gain effect of TPPUMA in high-temperature performance,low-temperature performance,aging resistance,fatigue resistance,weathering performance and bonding performance is obvious.In addition,it has good storage stability.With excellent road performance,TS-PUMA makes up for the shortcomings of epoxy asphalt in terms of lowtemperature performance and compatibility.Finally,due to the development trend of functional diversification of modified asphalt,the research basis and status of several new modified asphalts based on PU properties are described.Because the systematic study of PUMA is insufficient,this paper proposes corresponding research.To provide guidance and ideas for the research of PU modified asphalt.Peiliang Cong Changhao Liu Zhiyu Han Yuanfeng Zhao 2023Journal of Road Engineering2023,3,4:0
16Mulberry leaf flavonoids activate BAT and induce browning of WAT to improve type 2 diabetes via regulating the AMPK/SIRT1/PGC-1α signaling pathway显示文摘Mulberry (Morus alba L.) leaf is a well-established traditional Chinese botanical and culinary resource. It has found widespread application in the management of diabetes. The bioactive constituents of mulberry leaf, specifically mulberry leaf flavonoids (MLFs), exhibit pronounced potential in the amelioration of type 2 diabetes (T2D). This potential is attributed to their ability to safeguard pancreatic β cells, enhance insulin resistance, and inhibit α-glucosidase activity. Our antecedent research findings underscore the substantial therapeutic efficacy of MLFs in treating T2D. However, the precise mechanistic underpinnings of MLF’s anti-T2D effects remain the subject of inquiry. Activation of brown/beige adipocytes is a novel and promising strategy for T2D treatment. In the present study, our primary objective was to elucidate the impact of MLFs on adipose tissue browning in db/db mice and 3T3-L1 cells and elucidate its underlying mechanism. The results manifested that MLFs reduced body weight and food intake, alleviated hepatic steatosis, improved insulin sensitivity, and increased lipolysis and thermogenesis in db/db mice. Moreover, MLFs activated brown adipose tissue (BAT) and induced the browning of inguinal white adipose tissue (IWAT) and 3T3-L1 adipocytes by increasing the expressions of brown adipocyte marker genes and proteins such as uncoupling protein 1 (UCP1) and beige adipocyte marker genes such as transmembrane protein 26 (Tmem26), thereby promoting mitochondrial biogenesis. Mechanistically, MLFs facilitated the activation of BAT and the induction of WAT browning to ameliorate T2D primarily through the activation of AMP-activated protein kinase (AMPK)/sirtuin 1 (SIRT1)/peroxisome proliferator-activated receptor-gamma coactivator 1α (PGC-1α) signaling pathway. These findings highlight the unique capacity of MLF to counteract T2D by enhancing BAT activation and inducing browning of IWAT, thereby ameliorating glucose and lipid metabolism disorders.
