| 1 | De novo assembly of a Chinese soybean genome显示文摘Soybean was domesticated in China and has become one of the most important oilseed crops. Due to bottlenecks in their introduction and dissemination, soybeans from different geographic areas exhibit extensive genetic diversity. Asia is the largest soybean market; therefore, a high-quality soybean reference genome from this area is critical for soybean research and breeding.Here, we report the de novo assembly and sequence analysis of a Chinese soybean genome for 'Zhonghuang 13' by a combination of SMRT, Hi-C and optical mapping data. The assembled genome size is 1.025 Gb with a contig N50 of 3.46 Mb and a scaffold N50 of 51.87 Mb. Comparisons between this genome and the previously reported reference genome(cv. Williams82) uncovered more than 250,000 structure variations. A total of 52,051 protein coding genes and 36,429 transposable elements were annotated for this genome, and a gene co-expression network including 39,967 genes was also established. | 2 | Distribution characteristics of poly-and perfluoroalkyl substances in the Yangtze River Delta显示文摘In this work, a method was developed and optimized for the analysis of polyfluoroalkyl and/or perfluoroalkyl substances(PFASs) content in surface water and sediment samples with high instrumental response and good separation. Surface water and sediment samples were collected from the Yangtze River Delta(YRD) to analyze the distribution characteristics of perfluoroalkyl carboxylic acids(PFCAs), perfluoroalkane sulfonic acids(PFSAs), perfluoroalkyl phosphonic acids(PFPAs), perfluoroalkyl phosphinic acids(PFPiAs), and polyfluoroalkyl phosphoric acid diesters(di PAPs). The results showed that the total concentrations of PFCAs and PFSAs in YRD varied from 31 to 902 ng/L. PFCAs(≥ 11 carbons) and PFSAs(≥ 10 carbons atoms) were not detected in any surface water samples. The mean concentrations of all PFCAs and PFSAs in surface water from the sampling areas decreased in the following order:Yangtze river(191 ng/L) ≈ Taihu lake(189 ng/L) > Huangpu river(122 ng/L) ≈ Qiantang river(120 ng/L) > Jiaxing urban river(100 ng/L). Strong significant(p < 0.05) correlations between the concentrations of many of the compounds were found in the sampling areas, suggesting a common source for these compounds. Only perfluorooctanoic acid(PFOA) was observed in all sediment samples, at concentrations varying from 0.02 to 1.35 ng/g. | 5 | Serum macrophage migration inhibitory factor as a potential biomarker to evaluate therapeutic response in patients with allergic asthma:an exploratory study显示文摘Background:Previous studies have shown that macrophage migration inhibitory factor(MIF)is involved in the pathogenesis of asthma.This study aimed to investigate whether serum MIF reflects a therapeutic response in allergic asthma.Methods:We enrolled 30 asthmatic patients with mild-to-moderate exacerbations and 20 healthy controls,analyzing the parameter levels of serum MIF,serum total immunoglobulin E(tIgE),peripheral blood eosinophil percentage(EOS%),and fractional exhaled nitric oxide(FeNO).Lung function indices were used to identify disease severity and therapeutic response.Results:Our study showed that all measured parameters in patients were at higher levels than those of controls.After one week of treatment,most parameter levels decreased significantly except for serum tIgE.Furthermore,we found that serum MIF positively correlated with EOS%as well as FeNO,but negatively correlated with lung function indices.Receiver operator characteristic(ROC)curve analysis indicated that among the parameters,serum MIF exhibited a higher capacity to evaluate therapeutic response.The area under the curve(AUC)of MIF was 0.931,with a sensitivity of 0.967 and a specificity of 0.800.Conclusions:Our results suggested that serum MIF may serve as a potential biomarker for evaluating therapeutic response in allergic asthma with mild-to-moderate exacerbations. | Huiyuan ZHU Shaochun YAN Jingshuo WU Zhong ZHANG Xiaolin LI Zheng LIU Xing MA Lina ZHOU Lin ZHANG Mingming FENG Yiwei GENG Aixin ZHANG Sabina JANCIAUSKIENE Aiguo XU | 2021 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2021,22,6: | 3 |
| 6 | First experimental constraints on WIMP couplings in the effective field theory framework from CDEX显示文摘We present weakly interacting massive particles(WIMPs) search results performed using two approaches of effective field theory from the China Dark Matter Experiment(CDEX), based on the data from both CDEX-1B and CDEX-10 stages. In the nonrelativistic effective field theory approach, both time-integrated and annual modulation analyses were used to set new limits for the coupling of WIMP-nucleon effective operators at 90% confidence level(C.L.) and improve over the current bounds in the low mχregion. In the chiral effective field theory approach, data from CDEX-10 were used to set an upper limit on WIMP-pion coupling at 90% C.L. | 12 | Is tuberculosis patients management improved in the integrated TB control model in West China?A survey in Guizhou Province,China显示文摘Background:Tuberculosis(TB)patient management(TPM)is crucial to improve patient compliance to treatment.The coverage of TPM delivered by TB dispensaries or Centers for Disease Control and Prevention(CDC)was not high under the previous CDC model of TB control in China.In the integrated TB control model in China,TB patient management(TPM)was mainly delivered by lay health workers(LHWs)in primary health care(PHC)sectors.This study aims to investigate TPM delivery in resource-limited western China and to identify factors affecting TPM delivery by LHWs under the integrated TB control model.Methods:A stratified random sampling was used to select study sites.Pulmonary TB(PTB)patients≥15 years old from selected counties/districts in Guizhou Province were surveyed from August 2015 to May 2016.Structured questionnaires were used to collect data.Aχ^2 test and logistic regression were used to identify factors associated with self-administered treatment(non-TPM).Results:In total,638 PTB patients were included in the final analysis.Close to 30%of patients were ethnic minorities.More than 30%of patients were from counties with high TB burden,and 24.9%of patients had poor compliance to treatment.Only 37.1%of patients received TPM delivered by LHWs under the integrated TB control model throughout the treatment period.The main reasons for unwillingness to manage reported by patients included social stigma and no perceived need.Being ethnic minorities(OR=3.35)was a main factor associated with lower likelihood of receiving TPM,while living in areas with middle or high TB burden may increase the likelihood of receiving TPM(OR=0.17 and 0.25,respectively).Among current management approaches,more than 85%of patients chose phone reminder as their preferred TPM by LHWs.Conclusions:TPM under the integrated model in West China is still low and need further improvement,and the impeding factors of TPM need to be addressed.Strengthening patient-centered and community-based TPM and developing more feasible approaches of TPM delivery should be explored in future research in this region. | Jie Pu Wei Chen Wei-Xi Jiang Wei Xing Sheng-Xiang Liang Geng Wang Shi-Li Liu Hao Wu Ying Li Sheng-Lan Tang | 2019 | Infectious Diseases of Poverty2019,8,4: | 2 |

| 14 | Geant4 analysis and optimization of a double crystal phoswich detector for beta–gamma coincidence detection显示文摘In this study, a novel phoswich detector for beta–gamma coincidence detection is designed. Unlike the triple crystal phoswich detector designed by researchers at the University of Missouri, Columbia, this phoswich detector is of the semi-well type, so it has a higher detection efficiency. The detector consists of BC-400 and NaI:Tl with decay time constants of 2.4 and 230 ns, respectively.The BC-400 scintillator detects beta particles, and the Na I:Tl cell is used for gamma detection. Geant4 simulations of this phoswich detector find that a 2-mm-thick BC-400 scintillator can absorb nearly all of the beta particles whose energies are below 700 keV. Further, for a 2.00-cmthick NaI:Tl crystal, the gamma source peak efficiency for photons ranges from a maximum of nearly 90% at 30 keV to 10% at 1 MeV. | 15 | RNA-binding protein QKI regulates contact inhibition via Yes-associate protein in ccRCC显示文摘Contact inhibition adjusts organ size to the proper size and ensures the cultured cells growing to a monolayer.By regulating the downstream coordinator YAP,the evolutionarily conserved Hippo transduction pathway attunes cell growth and death in response to cell contact inhibition,polarity,self-renewal,and differentiation.Dysregulation of this pathway is involved in various diseases such as cancer.RNA-binding protein QKI regulates cell proliferation,metabolism,division,and immunity in various cancer models,but its role in cancer cell contact inhibition remains unclear.In this study,we aimed to clarify the relationship between QKI and YAP,and the role of their interaction in cell contact inhibition.We found a lower QKI expression level in sparse condition,whereas a higher expression level in confluent condition by western blot analysis and immunofluorescence assay.QKI knockdown elevated cell proliferation and invasion both in vitro and in vivo.Strikingly,the results of CCK-8 assay,colony formation assay,and transwell assay showed that the phenomenon was in accord with the expression level of pYAP and reverse with YAP.Higher levels of Wnt3a and β-catenin were also found in xenografts of QKI-knockdown clear cell renal cell carcinoma (ccRCC) CAKI-1 cells by western blot analysis and immumohistochemical staining.Finally,a positive correlation between QKI and pYAP was found in clinical specimens by immunohistochemistry.Thus,as a negative regulator of YAP,QKI attuned the cell contact inhibition,leading to inhibition of cancer cell proliferation and invasion through Wnt and GPCR pathway. | Zheng Zhu Di Wei Xi'an Li Fuli Wang Fei Yan Zibao Xing Zhao Yan Huanyu Lu Dongsheng Zhai Ziehen Ye Geng Zhang Ping Meng Yu Zheng Jiarui Yuan Zifan Lu Jianlin Yuan | 2019 | Acta Biochimica et Biophysica Sinica2019,51,1: | 2 |
