| 1 | Host metabolism dysregulation and cell tropism identification in human airway and alveolar organoids upon SARS-CoV-2 infection显示文摘The coronavirus disease 2019(COVID-19)pandemic is caused by infection with the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),which is spread primary via respiratory droplets and infects the lungs.Currently widely used cell lines and animals are unable to accurately mimic human physiological conditions because of the abnormal status of cell lines(transformed or cancer cells)and species differences between animals and humans.Organoids are stem cell-derived selforganized three-dimensional culture in vitro and model the physiological conditions of natural organs.Here we showed that SARS-CoV-2 infected and extensively replicated in human embryonic stem cells(hESCs)-derived lung organoids,including airway and alveolar organoids which covered the complete infection and spread route for SARS-CoV-2 within lungs.The infected ceils were ciliated,club,and alveolar type 2(AT2)cells,which were sequentially located from the proximal to the distal airway and terminal alveoli,respectively.Additionally,RNA-seq revealed early cell response to virus infection including an unexpected downregulation of the metabolic processes,especially lipid metabolism,in addition to the well-known upregulation of immune response.Further,Remdesivir and a human neutralizing antibody potently inhibited SARS-CoV-2 replication in lung organoids.Therefore,human lung organoids can serve as a pathophysiological model to investigate the underlying mechanism of SARS-CoV-2 infection and to discover and test therapeutic drugs for COVID-19. | Rongjuan Pei Jianqi Feng Yecheng Zhang Hao Sun Lian Li Xuejie Yang Jiangping He Shuqi Xiao Jin Xiong Ying Lin Kun Wen Hongwei Zhou Jiekai Chen Zhili Rong Xinwen Chen | 2021 | Protein & Cell2021,12,9: | 7 |
| 2 | Productive HBV infection of well-differentiated, hNTCP-expressing human hepatoma-derived(Huh7) cells显示文摘Feasible and effective cell models for hepatitis B virus(HBV) infection are required for investigating the complete lifecycle of this virus, including the early steps of viral entry. Resistance to dimethyl sulfoxide/polyethylene glycol(DMSO/PEG), h NTCP expression, and a differentiated state are the limiting factors for successful HBV infection models. In the present study, we used a hepatoma cell line(Hu7^(hDNTCPh)) to overcome these limiting factors so that it exhibits excellent susceptibility to HBV infection. To achieve this goal, different hepatoma cell lines were tested with 2.5% DMSO/4%PEG8000, and one resistant cell line(Huh7 D) was used to construct a stable h NTCP-expressing cell line(Hu7^(hDNTCPh)) using a recombinant lentivirus system. Then, the morphological characteristics and differentiation molecular markers of Hu7^(hDNTCPh) cells with or without DMSO treatment were characterized. Finally, the susceptibility of Hu7^(hDNTCPh) cells to HBV infection was assessed. Our results showed that Huh7 D cells were resistant to 2.5% DMSO/4% PEG8000, whereas the others were not. Hu7^(hDNTCPh) cells were established to express a high level of h NTCP compared to liver extracts, and Hu7^(hDNTCPh) cells rapidly transformed into a non-dividing, well-differentiated polarized phenotype under DMSO treatment. Hu7^(hDNTCPh) cells fully supported the entire lifecycle of HBV infection. | 6 | Changes in soil carbon sequestration and soil respiration following afforestation on paddy fields in north subtropical China显示文摘Aims Although many studies have reported net gains of soil organic carbon(SOC)after afforestation on croplands,this is uncertain for Chinese paddy rice croplands.Here,we aimed to evaluate the effects of affores-tation of paddy rice croplands on SOC sequestration and soil respiration(R).Such knowledge would improve our understanding of the efctive-ness of various land use options on greenhouse gas mitigation in China.Methods The investigation was conducted on the Chongming Island,north subtropical China.Field sites were reclaimed from coastal salt marshes in the 1960s,and soils were homogeneous with simple land use:histories.SOC stocks and R,levels were monitored over one year in a paddy rice cropland,an evergreen and a deciduous broad-leaved plantation established on previous paddy fields and a reference fal-low land site never cultivated.Laboratory incubation of soil under fast-changing temperatures was used to compare the temperature sensitivity(Q10)of SOC decomposition across land uses.Important Findings After 15-20 years of afforestation on paddy fields,SOC concentra-tion only slightly increased at the depth of 0--5 cm but decreased in deeper layers,which resulted in a net loss of SOC stock in the top 40cm.Seasonal increase of sOC was observed during the rice-growing period in croplands but not in afforested soils,suggest-ing a stronger SOC sequestration by paddy rice cropping.However,SOC sequestered under cropping was more labile,as indicated by its higher contents of dissolved organic carbon and microbial bio-mass.Also,paddy soils had higher annual R,than afforested soils;R,abruptly increased after paddy fields were drained and plowed and remained distinctively high throughout the dry farming period.Laboratory incubation revealed that paddy soils had a much higher Q10 of SOC decomposition than afforested soils.Given that tem-perature was the primary controller of R,in this region,it was con-cluded that despite the stronger SOC sequestration by paddy rice cropping,its SOC was less stable than in afforested systems and might be more easily released into the atmosphere under global warming. | Jun Cui Rongjuan Zhang Naishun Bu Huabin Zhang Boping Tang Zhaolei Li Lifeng Jiang Jiakuan Chen Changming Fang | 2013 | Journal of Plant Ecology2013,6,3: | 4 |

HDAC4 repressed the translocation of transcription factor IRF3 to the nucleus, thereby decreasing IRF3-mediated IFN-β expression. In particular, we also determined that HDAC4 can be phosphorylated and simultaneously block the phosphorylation of IRF3 at Ser386 and Ser396 by TBK1 and IKKε, respectively, by interacting with the kinase domain of TBK1 and IKKε. Furthermore, IFN-β may stimulate the expression of HDAC4. However, Huh7 cell line was the only highly permissive cell line for HCV infection and only a few clones were fully permissive. In this study, two different fully permissive clones of Huh7 cells, Huh7.5.1 and Huh7-Lunet-CD81 (Lunet-CD81) cells were compared for their responses upon HCV infection. The virus replication level was found slightly higher in Huh7.5.1 cells than that in Lunet-CD81 cells. Viability of Huh7.5.1 cells but not of Lunet-CD81 cells was reduced significantly after HCV infection. Further analysis showed that the cell cycle of infected Huh7.5.1 cells was arrested at G1 phase. The G1/S transition was blocked by HCV infection in Huh7.5.1 cells as shown by the cell cycle synchronization analysis. Genes related to cell cycle regulation was modified by HCV infection and gene interaction analysis in GeneSpring GX in Direct Interactions mode highlighted 31 genes. In conclusion, the responses of those two cell lines were different upon HCV infection. HCV infection blocked G1/S transition and cell cycle progress, thus reduced the cell viability in Huh7.5.1 cells but not in Lunet-CD81 cells. | 14 | Regulation of Hepatitis C Virus Replication and Gene Expression by the MAPK-ERK Pathway显示文摘The mitogen activated protein kinases-extracellular signal regulated kinases (MAPK-ERK) pathway is involved in regulation of multiple cellular processes including the cell cycle. In the present study using a Huh7 cell line Con1 with an HCV replicon, we have shown that the MAPK-ERK pathway plays a significant role in the modulation of HCV replication and protein expression and might influence IFN-α signalling. Epithelial growth factor (EGF) was able to stimulate ERK activation and decreased HCV RNA load while a MAPK-ERK pathway inhibitor U0126 led to an elevated HCV RNA load and higher NS5A protein amounts in Con1 cells. It could be further demonstrated that the inhibition of the MAPK-ERK pathway facilitated the translation directed by the HCV internal ribosome entry site. Consistently, a U0126 treatment enhanced activity of the HCV reporter replicon in transient transfection assays. Thus, the MAPK-ERK pathway plays an important role in the regulation of HCV gene expression and replication. In addition, cyclin-dependent kinases (CDKs) downstream of ERK may also be involved in the modulation of HCV replication since roscovitine, an inhibitor of CDKs had a similar effect to that of U0126. Modulation of the cell cycle progression by cell cycle inhibitor or RNAi resulted consistently in changes of HCV RNA levels. Further, the replication of HCV replicon in Con1 cells was inhibited by IFN-α. The inhibitory effect of IFN-α could be partly reversed by pre-incubation of Con-1 cells with inhibitors of the MAPK-ERK pathway and CDKs. | 19 | Polymerase mutations rtN238R,rtT240Y and rtN248H of hepatitis B virus decrease susceptibility to adefovir显示文摘Long term antiviral therapy with nucleos(t)ide analogs(NAs) may lead to the emergence of drug-resistance viral mutants in chronic hepatitis B virus(HBV) patient.The purpose of this study was to identify adefovir dipivoxil(ADV) resistance mutations of HBV polymerase and determine effective drugs to replace ADV.The reverse transcriptase(RT) coding region was PCR-amplified using HBV DNA extracted from patient blood samples and sequenced.Nineteen substitution mutations were detected.Among them,rtN238R,rtT240Y and rtN248H were often observed in patients receiving ADV administration.These three potential drug resistant sites were introduced into HBV replication-competent plasmids.The in vitro susceptibility of both wild-type(WT) and mutant-type(MT) HBV to NAs was analyzed by Southern blotting and quantitative real-time PCR.The rtN238R,rtT240Y and rtN248H substitutions had no obvious effect on HBV DNA replication or gene expression.The in vitro susceptibility analysis showed that rtN238R,rtT240Y and rtN248H substitutions were responsible for the reduced susceptibility to ADV,and demonstrated a 5.42-,2.89-and 5.72-fold increase in resistance towards ADV,respectively.However,HBV harbored these mutations retained normal susceptibility to LMV,LdT,ETV and TDF. | QIN Bo PEI RongJuan HE TingTing HUANG ZhaoHui PAN GuoShao TU ChunYu LU MengJi CHEN XinWen | 2013 | Chinese Science Bulletin2013,58,15: | 1 |
| 20 | A new way to analyze the traditional Chinese medicine syndrome:heat toxin syndrome in cerebral infarction显示文摘Objective:To establish a diagnostic system for heat toxin syndrome of acute cerebral infarction.Based on this toxin syndrome diagnostic system,the general principles of heat toxin development will be uncovered,and the critical turning point at which the heat toxin syndrome occurs will also be explored.Methods:In this study,a total of 271 hypertension patients with cerebral infarction within 72 h were recruited from the Affiliated Dongfang Hospital of the Beijing University of Chinese Medicine,the Affiliated Dongzhimen Hospital of Beijing University of Chinese Medicine,the Affiliated Renmin Hospital of Peking University,the Second Affiliated Hospital of Tianjin University of Traditional Chinese Medicine,the Affiliated Hospital of Shandong University of Traditional Chinese Medicine,the Affiliated Hospital of Changchun University of Traditional Chinese Medicine,the Affiliated Hospital of Hebei University of Traditional Chinese Medicine and China Meitan General Hospital from August,2008,to December,2009.The patients’Chinese medical information was recorded on days 1,3,5,7,and 14 during their hospitalizations.The medical records were recorded according to traditional Chinese medicine(TCM)theory and included the serum marker levels at the beginning and at the end of the trial.The time line was also analyzed.Results:The level of Hs-CRP,PAG,NSE,OX-LDL,and MMP-9 were abnormal and,were higher in CI patients compared to hypertension patients.In the study of the heat toxin diagnosis system,according to the entropy clustering results,30 combinations of the medical information can be sorted into the traditional syndromes,but 13 combinations cannot be sorted.To obtain more precise symptoms related to the heat toxins,a logistic regression equation was set up with the variables from the unsorted medical information;the dependent variables were fever and BP fluctuation.Weighted variables were obtained.MLP analysis demonstrated that the diagnosis model was stable and precise.The accuracy reached 83.82%.The ROC test showed that seven points of the diagnosis system was the best cutting point,with a sensitivity of 0.857 and a specificity of 0.955.Progressing stroke was related to heat toxin syndrome.When the turning point appeared,the combination of symptoms,such as coma,aphasia,gummy eyes,and halitosis,predicted the deterioration or recovery of CI.The heat toxin syndrome existed in every subtype of CI;however,the observed heat toxin levels were highest in PACI and lowest in LACI.Meanwhile,blood and sputum stasis syndromes transformed into heat toxicity were one source of heat toxin syndrome.Conclusion:Heat toxin syndrome,as well as qi/blood/sputum stasis,co-existed in the CI patients,and the transformation frequently appeared during the process.Three to five days after the onset of CI was the turning point,at which time several combinations of medical indicators make it possible to predict the development of CI. | Zhichen Zhang Shaozhen Ji Xuejie Yu Xianglan Jin Liping Zhang Rongjuan Guo Hong Zheng Mingqi Wang Yunling Zhang | 2014 | Journal of Traditional Chinese Medical Sciences2014,1,1: | 1 |
