| 2 | S100A8 promotes epithelial-mesenchymal transition and metastasis under TGF-β/USF2 axis in colorectal cancer显示文摘Background:The transforming growth factor-β(TGF-β)pathway plays a pivotal role in inducing epithelial-mesenchymal transition(EMT),which is a key step in cancer invasion and metastasis.However,the regulatory mechanism of TGF-βin inducing EMT in colorectal cancer(CRC)has not been fully elucidated.In previous studies,it was found that S100A8 may regulate EMT.This study aimed to clarify the role of S100A8 in TGF-β-induced EMT and explore the underlying mechanism in CRC.Methods:S100A8 and upstream transcription factor 2(USF2)expression was detected by immunohistochemistry in 412 CRC tissues.Kaplan-Meier survival analysis was performed.In vitro,Western blot,and migration and invasion assays were performed to investigate the effects of S100A8 and USF2 on TGF-β-induced EMT.Mouse metastasis models were used to determine in vivo metastasis ability.Luciferase reporter and chromatin immunoprecipitation assay were used to explore the role of USF2 on S100A8 transcription.Results:During TGF-β-induced EMT in CRC cells,S100A8 and the transcription factor USF2 were upregulated.S100A8 promoted cell migration and invasion and EMT.USF2 transcriptionally regulated S100A8 expression by directly binding to its promoter region.Furthermore,TGF-βenhanced the USF2/S100A8 signaling axis of CRC cells whereas extracellular S100A8 inhibited the USF2/S100A8 axis of CRC cells.S100A8 expression in tumor cells was associated with poor overall survival in CRC.USF2 expression was positively related to S100A8 expression in tumor cells but negatively related to S100A8-positive stromal cells.Conclusions:TGF-βwas found to promote EMT and metastasis through the USF2/S100A8 axis in CRC while extracellular S100A8 suppressed the USF2/S100A8 axis.USF2 was identified as an important switch on the intracellular and extracellular S100A8 feedback loop. | Si Li Jun Zhang Senmi Qian Xuesong Wu Liang Sun Tianyi Ling Yao Jin Wenxiao Li Lichao Sun Maode Lai Fangying Xu | 2021 | Cancer Communications2021,41,2: | 5 |
| 3 | Correlations Between Single Nucleotide Polymorphisms,Cognitive Dysfunction,and Postmortem Brain Pathology in Alzheimer's Disease Among Han Chinese显示文摘In this study, the distribution of five Alzheimer's disease(AD)-related single nucleotide polymorphisms(SNPs) in the Han population was examined in combination with the evaluation of clinical cognition and brain pathological analysis. The associations among SNPs,clinical daily cognitive states, and postmortem neuropathological changes were analyzed in 110 human brains from the Chinese Academy of Medical Sciences/Peking Union Medical College(CAMS/PUMC) Human Brain Bank.APOE ?4(OR = 4.482, P = 0.004), the RS2305421 GG genotype(adjusted OR = 4.397, P = 0.015), and the RS10498633 GT genotype(adjusted OR = 2.375,P = 0.028) were associated with a higher score on the ABC(Ab plaque score, Braak NFT stage, and CERAD neuritic plaque score) dementia scale. | 4 | Assessment of Changes in the Cesarean Scar and Uterus Between One and Two Years after Cesarean Section Using 3D T2w SPACE MRI显示文摘Objective Io evaluate changes in morphology of the cesarean scar and uterus between one and two years after cesarean section using high-resolution,three dimensional T2-weighted sampling perfection with application optimized contrast using diflerent flip angle evolutions Magnetic Resonance Imaging(3DT2w SPACE MRI).Methods This prospective study was performed to investigate morphological changes in the cesarean scars and uterus from one to two years after cesarean section using high-resolution,3D T2w SPACE MRI.The healthy volunteers having no childbearing history were recruited as the controls.All data were measured by two experienced radiologists.All data with normal distribution between the one-year and two-year groups were compared using a paired-sample t test or independent t test.Results Finally,46 women took a pelvic MR examination one year after cesarean section,and a subset of 15 completed the same examination again after two years of cesarean section.Both the uterine length and the anterior wall thickness after two years of cesarean section(5.75±0.46 and 1.45±0.35 cm)were significantly greater than those measured at one year(5.33±0.59 and 1.25±0.27 cm)(t=-2.363 and-2.175,P=0.033 and 0.048).No significant difference was shown in myometrial thickness two years after cesarean section(1.45±0.35 cm)with respect to the control group(1.58±0.21 cm,P=0.170).Nine women who underwent MRI twice were considered to have scar diverticula one year after cesarean section,and still had diverticula two years after cesarean section.The thickness,height,and width of the uterine scar showed no significant change from one to two years(all P>0.05).Conclusions 3D T2w SPACE MRI provides overall morphologic details and shows dynamic changes in the scar and the uterus between one and two years after cesarean section.Scar morphology after cesarean section reached relatively stable one year after cesarean section,and uterine morphology was closer to normal two years after cesarean section. | Yafei Qi Yonglan He Ning Ding Liangkun Ma Tianyi Qian Yuan Li Huadan Xue Zhengyu Jin | 2022 | Chinese Medical Sciences Journal2022,37,2: | 3 |

| 6 | Prediction of radiosensitivity in primary central nervous system germ cell tumors using dynamic contrast-enhanced magnetic resonance imaging显示文摘Objective: To evaluate the feasibility of dynamic contrast-enhanced magnetic resonance imaging(DCEMRI) for predicting tumor response to radiotherapy in patients with suspected primary central nervous system(CNS) germ cell tumors(GCTs).Methods: DCE-MRI parameters of 35 patients with suspected primary CNS GCTs were obtained prior to diagnostic radiation, using the Tofts and Kermode model. Radiosensitivity was determined in tumors diagnosed 2 weeks after radiation by observing changes in tumor size and markers as a response to MRI. Taking radiosensitivity as the gold standard, the cut-off value of DCE-MRI parameters was measured by receiver operating characteristic(ROC) curve. Diagnostic accuracy of DCE-MRI parameters for predicting radiosensitivity was evaluated by ROC curve.Results: A significant elevation in transfer constant(Ktrans) and extravascular extracellular space(Ve)(P=0.000), as well as a significant reduction in rate constant(Kep)(P=0.000) was observed in tumors. Ktrans, relative Ktrans, and relative Kep of the responsive group were significantly higher than non-responsive groups. No significant difference was found in Kep, Ve, and relative Ve between the two groups. Relative Ktrans showed the best diagnostic value in predicting radiosensitivity with a sensitivity of 100%, specificity of 91.7%, positive predictive value(PPV) of 95.8%, and negative predictive value(NPV) of 100%.Conclusions: Relative Ktrans appeared promising in predicting tumor response to radiation therapy(RT). | 17 | Delineating complex ferroelectric domain structures via second harmonic generation spectral imaging显示文摘Understanding the mechanisms and spatial correlations of crystallographic symmetry breaking in ferroelectric materials is essential to tuning their functional properties.While optical second harmonic generation(SHG)has long been utilized in ferroelectric studies,its capability for probing complex polar materials has yet to be fully realized.Here,we develop a SHG spectral imaging method implemented on a home-designed laser-scanning SHG microscope,and demonstrate its application for a model system of(K,Na)NbO3 single crystals.Supervised model fitting analysis produces comprehensive information about the polarization vector orientations and relative fractions of constituent domain variants as well as their thermal evolution across the polymorphic phase transitions.Multiple domains and phases are clearly delineated at different temperatures,suggesting the phase competitions in(K,Na)NbO3.Besides,we show that unsupervised matrix decomposition analysis can quickly and faithfully reveal domain configurations without a priori knowledge about specific material systems.The SHG spectral imaging method can be readily extended to other ferroelectric materials with potentials to be further enhanced. | Wei Li Yunpeng Ma Tianyi Feng Ziwan Du Yixuan Liu Sergei VKalinin Jing-Feng Li Qian Li | 2023 | Journal of Materiomics2023,9,2: | 0 |
| 18 | Study on myocardial infarction based on nationwide inpatient sample database:a bibliometric analysis from 2000 to 2022显示文摘Background:Studies on myocardial infarction(MI)based on large medical databases have become popular in recent years.The influence of the National Inpatient Sample(NIS),the largest collection of administrative healthcare data across the United States,on the field of MI has not been well investigated.This study aimed to quantify the contribution of NIS to MI research using bibliometric methods.Methods:We searched the Web of Science Core Collection database to identify publications on MI using NIS from 2000 to 2022.Bibliometric indicators,such as the number of publications,citations,and Hirsch index(H-index),were summarized by years,authors,organizations,and journals.VOSviewer and CiteSpace software were used to analyze the keywords and trends of the hot spots.Results:A total of 342 articles on MI based on NIS were included.A significant growth in outputs related to MI using the NIS from 2000 to 2020 was observed.The publications were mainly from the United States.The Mayo Clinic was the most prolific institution and had the most citations and the highest H-index.The American Journal of Cardiology ranked first among journals with the highest number of publications,citations,and H-index.Mortality and healthcare management are the main focuses of this field.Personalized risks and care are receiving increased attention.Conclusion:This study suggests that NIS significantly contributes to high-quality output in MI research.More efforts are needed to improve the impact of knowledge gained from the NIS on MI. | Tianyi Zhang Xue Yang Qian Zhuang Yuhua Liu Huajuan Bai Jianchao Liu Zhouheng Ye | 2024 | Emergency and Critical Care Medicine2024,4,1: | 0 |
