| 1 | Sensitivity of green and blue-green algae to methyl tert-butyl ether显示文摘The toxicity of methyl tert-butyl ether (MTBE) to Chlorella ellipsoidea and Aphanizomenon flos-aquae was tested and assessed for a 15-d incubation with concentrations of MTBE from high (2.00×104 mg/L) to low (2 mg/L). The results showed that the toxicity was low when the concentration of MTBE was in the range 1.00×104-2.00×104 mg/L (the greatest inhibition of growth-rate was 70%-71%, occurred during the day 1-5). Low concentrations (2-500 mg/L) stimulated algal growth up to the greatest effect of 85%-200% when the concentration of MTBE was 50-100 mg/L during day 3-5. The toxicity of MTBE (72-120 h EC50) was 6.65×103-9.58×103 mg/L for C. ellipsoidea and that is 1.14×104-2.00×104 mg/L for A. floc-aquae. | 2 | Effects of Epidural Spinal Cord Stimulation and Treadmill Training on Locomotion Function and Ultrastructure of Spinal Cord Anterior Horn after Moderate Spinal Cord Injury in Rats显示文摘Objective:To investigate the effects of epidural spinal cord stimulation(ESCS) and treadmill training on the locomotion function and ultrastructure of spinal cord anterior horn after moderate spinal cord injury in rats.Method:Nine adult female Sprague-Dawley rats were randomly distributed into three groups:①spinal cord injury group(SI,n=3).②spinal cord injury plus ESCS group(SE,n=3).③spinal cord injury plus treadmill training group(TT,n=3).All rats received a moderate spinal cord injury surgery.Four weeks after surgery,rats in SE group received an electrode implantation procedure,with the electrode field covering spinal cord segments L2-S1.Four weeks after electrode implantation,rats received subthreshold ESCS for 30 min/d.Rats in TT group received 4cm/s treadmill training for 30min/d.Rats in SI group received no intervention,as a control group.All procedures in these three groups lasted four weeks.The open field Basso,Beattie and Bresnahan(BBB) scale was used before and after intervention to evaluate rats' hindlimb motor function.Result:After four weeks intervention,rats in TT group improved their open field locomotion scores to 20.In contrast,no significant improvement was observed in groups SI and SE.The morphology of synapses and neurons were similar regardless of whether rats had undergone ESCS,treadmill training or not.Conclusion:ESCS alone was not sufficient to improve the walking ability of spinal cord injured rats.ESCS or treadmill training alone might not contribute to the changes of ultrastructure in anterior horn of spinal cord that underlie the recovery of walking ability.Further research is needed to understand the contributions of combination of ESCS and treadmill training to the rehabilitation of spinal cord injured rats. | WANG Yizhao HUANG Xiaolin XU Jiang XU Tao FANG Zhengyu XU Qi TU Xikai YANG Peipei | 2009 | 中国康复医学杂志2009,24,6: | 4 |
| 3 | Jugular arginine supplementation increases lactation performance and nitrogen utilization efficiency in lactating dairy cows显示文摘Background: Enhancing the post-ruminal supply of arginine(Arg), a semi-essential amino acid(AA), elicits positive effects on milk production. Our objective was to determine the effects of Arg infusion on milk production parameters and aspects of nitrogen(N) absorption and utilization in lactating dairy cows. Six lactating Chinese Holstein cows of similar body weight(508 ± 14 kg), body condition score(3.0 ± 0), parity(4.0 ± 0), milk yield(30.6 ±1.8 kg) and days in milk(20 ± 2 d) were randomly assigned to 3 treatments in a replicated 3 × 3 Latin square design with 21 d for each period(1 week for infusion and 2 weeks for washout). Treatments were 1) Control: saline;2) Arg group: saline + 9.42 g/L L-Arg;3) Alanine(Ala) group: saline + 19.31 g/L L-Ala(iso-nitrogenous to the Arg group). Milk production and composition, dry matter intake, apparent absorption of N, profiles of amino acids(AA) in blood,urea N in urine, milk, and blood, and gene expression of AA transporters were determined.Results: Compared with the Control or Ala group, the infusion of Arg led to greater expression of AA transporters(SLC7 A2 and SLC7 A8) and apparent uptake of free AA in the mammary gland, and was accompanied by greater milk yield, milk protein yield and milk efficiency(calculated by dividing milk yield over feed intake), together with lower concentration of urea N [regarded as an indicator of N utilization efficiency(NUE)] in blood and milk. | 6 | Astrocytic effect of low molecular weight heparin-superoxide dismutase conjugate in interleukin-6 overexpressing mice following local cerebral ischemia显示文摘BACKGROUND:Studies have shown that low molecular weight heparin-superoxide dismutase conjugate exhibits a remarkable neuroprotective effect. OBJECTIVE:To investigate the effect of low molecular weight heparin-superoxide dismutase conjugate on astrocytes in an interleukin-6(IL-6) overexpressing mice following local cerebral ischemia. DESIGN,TIME AND SETTING:Randomized,cytological,controlled,animal study was performed in the Department of Physiology and Neuroscience,Neurology and Biochemistry and Molecular Biology,Medical University of South Carolina from January 2005 to March 2005. MATERIALS:Nine IL-6 transgenic mice,irrespective of gender,were randomly divided into three groups:sham-operated,model,and treatment,with three mice in each group.With exception of the sham-operated group,right middle cerebral artery occlusion was induced in the mice. Expression of glial fibrillary acidic protein,an astrocyte marker,was determined by immunohistochemistry.Low molecular weight heparin-superoxide dismutase conjugate was purchased from Biochemistry and Biotechnique Institute,Shandong University. METHODS:Two minutes prior to ischemia induction,0.5 mL/kg saline or 20 000 U/kg low molecular weight heparin-superoxide dismutase conjugate were administrated via the femoral artery in the model group and treatment group,respectively.The sham-operated group underwent the same protocols,with the exception of occlusion and treatment. MAIN OUTCOME MEASURES:The number of glial fibrillary acidic protein-positive cells was quantified under light microscopy(x200). RESULTS:In the sham-operated group,there were a large number of astrocytes in the IL-6 transgenic mice.However,the cell bodies were small,and the branches were few and thin.The number of astrocytes in the model group was remarkably less than the sham-operated group. Compared to the model and sham-operated groups,the number of astrocytes significantly increased,and the cell body became larger,following treatment with low molecular weight heparin-superoxide dismutase conjugate.Astrocytes exhibited hypertrophy and hyperplasia,and the processes became longer and thicker. BACKGROUND The effect and safety of ticlopidine irn the prevention of ischemic cerebral stroke in China has not been reported. METHODS 329 patients with TIA or mild ischemic cevebral stroke wasrandonmly assigned to ticlopidine group(165 case) or aspirin group (164 case) in this study.These patrents were randomly allocated to receive either 250mg trclopidine or 50mg aspirin daily and didnd take any other platelet antiaggregating drugs. Time of eacn follow up visit was one to two months. Follow up lasted for 6 to 18 months. RESULTS The event rate for stroke or death from any cause was 8.3% in ticlopidine group arid 14.9% in aspirin group. This repesented a risk reduction of 44.3%(95% cofidence interval, 0.29-0.94) for ticiopidine group as compared with aspirin group. The event raite for ischemic cerebral stroke or myocarction of ticlopidine group(7.0%)was lower than that cf aspirin group(14.8%)(P<0.05).A riskreduction of 52.7%(95% confidence interval,0.24-0.92) for ticlopidine group compared with aspirin group. The rate of adverse effects of ticlopidine group and aspirin group were 6.9% and 11.0% during the trial ,but this was not statistically significant(P<0.05).DISCUSSION and CONCLUSION Therapeutic efficacy for the prevention oi ischemic stroke of ticlopidine was better than that of aspirin, the rate of side effects in ticlopidine group and aspirin group are not statistically significant. OBJECTIVE: To investigate the intervention effects of LMWH-SOD conjugate on serum levels of nitric oxide (NO),glutathione peroxidase (GSH-Px),and myeloperoxidase (MPO) following cerebral ischemia/reperfusion injury. DESIGN,TIME AND SETTING: A randomized,controlled,and neurobiochemical experiment was performed at the Institute of Biochemical Pharmacy,School of Pharmaceutical Sciences,Shandong University between April and July 2004. MATERIALS: A total of 60 Mongolian gerbils of either gender were included in this study. Total cerebral ischemia/reperfusion injury was induced in 50 gerbils by occluding bilateral common carotid arteries. The remaining 10 gerbils received a sham-operation (sham-operated group). Kits of SOD,NO,and MPO were sourced from Nanjing Jiancheng Bioengineering Institute,China. LMWH,SOD,and LMWH-SOD conjugates were provided by Institute of Biochemistry and Biotechnique,Shandong University,China. METHODS: Fifty successful gerbil models of total cerebral ischemia/reperfusion injury were evenly randomized to five groups: physiological saline,LMWH-SOD,SOD,LMWH + SOD,and LMWH. At 2 minutes prior to ischemia,0.5 mL/65 g physiological saline,20 000 U/kg LMWH-SOD conjugate,20 000 U/kg SOD,a mixture of SOD (20 000 U/kg) and LMWH (LMWH dose calculated according to weight ratio,LMWH: SOD=23.6:51),and LMWH (dose as in the LMWH + SOD group) were administered through the femoral artery in each above-mentioned group,respectively. MAIN OUTCOME MEASURES: Serum levels of NO,MPO,and GSH-Px. RESULTS: Compared with 10 sham-operated gerbils,the cerebral ischemia/reperfusion injury gerbils exhibited decreased serum levels of GSH-Px and increased serum levels of NO and MPO (P<0.01). The serum level of GSH-Px was significantly upregulated in all groups,in particular in the LMWH-SOD group (P<0.01),compared with the physiological saline group (P<0.05-0.01). Following medical treatment,serum levels of NO and MPO were significantly downregulated in all groups,in particular in the LMWH-SOD group (P<0.01). Serum levels of GSH-Px,NO,and MPO in the LMWH-SOD group were close to those in the sham-operated group (P > 0.05). CONCLUSION: In cerebral ischemia/reperfusion injury,LMWH-SOD conjugate exhibits stronger neuroprotective effects on free radical scavenging,inflammation inhibition,and cytotoxicity inhibition than simple or combined application of LMWH and SOD by downregulating NO and MPO levels and upregulating the GSH-Px level. | Qingde Wang Guixiang Cui Hongxia Liu Yizhao Li Fengshan Wang | 2008 | Neural Regeneration Research2008,3,11: | 0 |
