| 1 | Numerical investigation of dual-porosity model with transient transfer function based on discrete-fracture model显示文摘Based on the characteristics of fractures in naturally fractured reservoir and a discrete-fracture model, a fracture network numerical well test model is developed.Bottom hole pressure response curves and the pressure field are obtained by solving the model equations with the finite-element method. By analyzing bottom hole pressure curves and the fluid flow in the pressure field, seven flow stages can be recognized on the curves. An upscaling method is developed to compare with the dual-porosity model(DPM). The comparisons results show that the DPM overestimates the inter-porosity coefficient λ and the storage factor ω. The analysis results show that fracture conductivity plays a leading role in the fluid flow. Matrix permeability influences the beginning time of flow from the matrix to fractures. Fractures density is another important parameter controlling the flow. | 2 | Heat shock protein 90 promotes RNA helicase DDX5 accumulation and exacerbates hepatocellular carcinoma by inhibiting autophagy显示文摘Objective:Hepatocellular carcinoma(HCC),the main type of liver cancer,has a high morbidity and mortality,and a poor prognosis.RNA helicase DDX5,which acts as a transcriptional co-regulator,is overexpressed in most malignant tumors and promotes cancer cell growth.Heat shock protein 90(HSP90)is an important molecular chaperone in the conformational maturation and stabilization of numerous proteins involved in cell growth or survival.Methods:DDX5 m RNA and protein expression in surgically resected HCC tissues from 24 Asian patients were detected by quantitative real-time PCR and Western blot,respectively.The interaction of DDX5-HSP90 was determined by molecular docking,immunoprecipitation,and laser scanning confocal microscopy.The autophagy signal was detected by Western blot.The cell functions and signaling pathways of DDX5 were determined in 2 HCC cell lines.Two different murine HCC xenograft models were used to determine the function of DDX5 and the therapeutic effect of an HSP90 inhibitor.Results:HSP90 interacted directly with DDX5 and inhibited DDX5 protein degradation in the AMPK/ULK1-regulated autophagy pathway.The subsequent accumulation of DDX5 protein induced the malignant phenotype of HCC by activating theβ-catenin signaling pathway.The silencing of DDX5 or treatment with HSP90 inhibitor both blocked in vivo tumor growth in a murine HCC xenograft model.High levels of HSP90 and DDX5 protein were associated with poor prognoses.Conclusions:HSP90 interacted with DDX5 protein and subsequently protected DDX5 protein from AMPK/ULK1-regulated autophagic degradation.DDX5 and HSP90 are therefore potential therapeutic targets for HCC. | Ting Zhang Xinrui Yang Wanping Xu Jing Wang Dawei Wu Zhixian Hong Shengxian Yuan Zhen Zeng Xiaodong Jia Shanshan Lu Rifaat Safadi Sen Han Zhihong Yang Leonard M.Neckers Suthat Liangpunsakul Weiping Zhou Yinying Lu | 2021 | Cancer Biology & Medicine2021,18,3: | 7 |

| 5 | The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. | 7 | Increased osteoblast adhesion on nanophase Ti6Al4V显示文摘The objective of the present study was to prepare a novel nanostructured surface of Ti6Al4V alloy by the severe plastic deformation (SPD) and the chemical treatment process and to evaluate the adhesion of osteoblast on the nanophase titanium alloy. In the in vitro study, the primary cultured osteoblasts of neonatal rat calvaria were cultured on the nanophase and the as-received smooth Ti6Al4V substrates. Then osteoblasts adhesion behaviors on different substrates were observed by the fluorescence mi-croscopy, scanning electron microscopy (SEM) and RT-PCR analysis. The results of our research showed increased osteoblast adhesion on the nanophase titanium alloy compared with the as-received case. On the nanophase substrate, the presence of extensive filopodia, strong cellular adhesion and early cellular confluency could be observed. In addition, the expression of the adhesion-related integrin β1 mRNA was also higher on the nanophase substrate. | 15 | AQR is a novel type 2 diabetes-associated gene that regulates signaling pathways critical for glucose metabolism显示文摘Type 2 diabetes mellitus(T2 DM) is a common metabolic disease influenced by both genetic and environmental factors. In this study, we performed an in-house genotyping and meta-analysis study using three independent GWAS datasets of T2 DM and found that rs3743121, located 1 kb downstream of AQR,was a novel susceptibility SNP associated with T2 DM. The risk allele C of rs3743121 was correlated with the increased expression of AQR in white blood cells, similar to that observed in T2 DM models. The knockdown of AQR in HepG2 facilitated the glucose uptake, decreased the expression level of PCK2,increased the phosphorylation of GSK-3β, and restored the insulin sensitivity. Furthermore, the suppression of AQR inhibited the mTOR pathway and the protein ubiquitination process. | 16 | Phytoplankton and chlorophyll a relationships with ENSO in Prydz Bay, East Antarctica显示文摘The historical data of phytoplankton and chlorophyll a(Chl a)(1990–2002)obtained during the Chinese National Antarctic Research Expedition(CHINARE)in the Prydz Bay have been integrated.The results showed that the temperature,salinity,nutrients,and oxygen of seawater changed when El Nino/La Nina occurred.The variation of biological communities reflected the response of ecosystem to environmental changes.During El Ni?o period,Chl a concentration and phytoplankton community structure changed significantly,and the relative proportion of diatoms increased while dinoflagellates decreased.During La Ni?a period,the proportion of diatoms decreased,but the golden-brown algae and blue-green algae increased significantly.The variation of phytoplankton population directly affected the biodiversity of the bay,which were also quite sensitive to the marine environment changes.Meanwhile,the satellite remote sensing data of 2002–2011(December–March)have been used to study the temporal connection change of Chl a and phytoplankton in the Prydz Bay.We found that there were significant differences in the monthly variation characteristics of satellite remote sensing Chl a and sea surface temperature(SST),which had some links with sea ice melting and El Ni?o/La Ni?a events.We found that the start time of bloom advanced,lagged or synchronized with the changes of the SST,and we also found the occurrence time of phytoplankton bloom corresponded with the sea ice melting inner bay.To some extent,this study will help us understand the relationships between ENSO events and the phytoplankton bloom in the Southern Ocean. | ZHANG HaiSheng HAN ZhengBing ZHAO Jun YU PeiSong HU ChuanYu SUN WeiPing Yang Dan ZHU GenHai LU Bing Hans-UIrich PETER Walter VETTER | 2014 | Science China Earth Sciences2014,57,12: | 1 |
