1Plant-derived phosphocholine facilitates cellular uptake of anti-pulmonary fibrotic HJT-sRNA-m7显示文摘Pulmonary fibrosis, a progressive chronic disease with a high mortality rate, has limited treatment options. Currently, lung transplantation remains the only effective treatment. Here we report that a small RNA, HJT-sRNA-m7, from a Chinese herbal medicine Hong Jing Tian(HJT, RHODIOHAE CRENULATAE RADIX ET RHIZOMA, Rhodiola crenulata) can effectively reduce the expressions of fibrotic hallmark genes and proteins both in alveolar in vitro and in mouse lung tissues in vivo. We also discovered over one hundred oil-soluble chemicals from HJT decoctions, most of which are found in lipid extracts from other Chinese herbals decoctions, including Pu Gong Ying(PGY, TARAXACI HERBA, Taraxacum mongolicum), Chuan Xin Lian(CXL, changed to 'ANDROGRAPHIS HERBA, Andrographis paniculata'), and Jin Yin Hua(JYH, lonicera japonica or Honeysuckle). We identified the active component in these decoctions as two forms of phosphocholines, PC(18:0/18:2) and PC(16:0/18:2). These PCs potentially could form liposomes with small RNAs to enter human alveolar and gastric cells. 2Large-scale analysis of small RNAs derived from traditional Chinese herbs in human tissues显示文摘Plant-derived microRNAs have recently been reported to function in human blood and tissues. Controversy was immediately raised due to possible contamination and the lack of large sample sizes. Here, we report thousands of unique small RNAs derived from traditional Chinese medicine(TCM) herbs found in human blood cells and mouse lung tissues using a large-scale analysis.We extracted small RNAs from decoctions of 10 TCM plants(Ban Zhi Lian, Chai Hu, Chuan Xin Lian, Di Ding Zi Jin, Huang Qin, Jin Yin Hua, Lian Qiao, Pu Gong Ying, Xia Ku Cao, and Yu Xing Cao) and obtained millions of RNA sequences from each herb. We also obtained RNA-Seq data from the blood cells of humans who consumed herbal decoctions and from the lung tissues of mice administered RNAs from herbal decoctions via oral gavage. We identified thousands of unique small RNA sequences in human blood cells and mouse lung tissues. Some of these identified small RNAs from Chuan Xin Lian and Hong Jing Tian could be mapped to the genomes of the herbs, confirming their TCM plant origin. 4FKF1 F-box protein promotes flowering in part by negatively regulating DELLA protein stability under long-day photoperiod in Arabidopsis^FA显示文摘FLAVIN-BINDING KELCH REPEAT F-BOX 1(FKF1)encodes an F-box protein that regulates photoperiod flowering in Arabidopsis under long-day conditions(LDs).Gibberellin(GA)is also important for regulating flowering under LDs.However,how FKF1 and the GA pathway work in concert in regulating flowering is not fully understood.Here,we showed that the mutation of FKF1 could cause accumulation of DELLA proteins,which are crucial repressors in GA signaling pathway,thereby reducing plant sensitivity to GA in flowering.Both in vitro and in vivo biochemical analyses demonstrated that FKF1 directly interacted with DELLA proteins.Furthermore,we showed that FKF1 promoted ubiquitination and degradation of DELLA proteins.Analysis of genetic data revealed that FKF1 acted partially through DELLAs to regulate flowering under LDs.In addition,DELLAs exerted a negative feedback on FKF1 expression.Collectively,these findings demonstrate that FKF1 promotes flowering partially by negatively regulating DELLA protein stability under LDs,and suggesting a potential mechanism linking the FKF1 to the GA signaling DELLA proteins.Jindong Yan Xinmei Li Bingjie Zeng Ming Zhong Jiaxin Yang Piao Yang Xin Li Chongsheng He Jianzhong Lin Xuanming Liu Xiaoying Zhao 2020Journal of Integrative Plant Biology2020,62,11:8
5Sintilimab versus docetaxel as second-line treatment in advanced or metastatic squamous non-small-cell lung cancer:an open-label,randomized controlled phase 3 trial(ORIENT-3)显示文摘Background:Treatment options for Chinese patients with locally advanced or metastatic squamous-cell non-small-cell lung cancer(sqNSCLC)after failure of first-line chemotherapy are limited.This study(ORIENT-3)aimed to evaluate the efficacy and safety of sintilimab versus docetaxel as second-line treatment in patients with locally advanced or metastatic sqNSCLC.Methods:ORIENT-3 was an open-label,multicenter,randomized controlled phase 3 trial that recruited patients with stage IIIB/IIIC/IV sqNSCLC after failure with first-line platinum-based chemotherapy.Patients were randomized in a 1:1 ratio to receive either 200 mg of sintilimab or 75 mg/m^(2) of docetaxel intravenously every 3 weeks,stratified by the Eastern Cooperative Oncology Group performance status.The primary endpoint was overall survival(OS)in the full analysis set(FAS).Secondary endpoints included progression-free survival(PFS),objective response rate(ORR),disease control rate(DCR),duration of response(DoR)and safety.Results:Between August 25,2017,and November 7,2018,290 patients were randomized.For FAS,10 patients fromthe docetaxel armwere excluded.Themedian OS was 11.79(n=145;95%confidence interval[CI],10.28-15.57)months with sintilimab versus 8.25(n=135;95%CI,6.47-9.82)months with docetaxel(hazard ratio[HR]:0.74;95%CI,0.56-0.96;P=0.025).Sintilimab treatment significantly prolonged PFS(median 4.30 vs.2.79 months;HR:0.52;95%CI,0.39-0.68;P<0.001)and showed higher ORR(25.50%vs.2.20%,P<0.001)and DCR(65.50%vs.37.80%,P<0.001)than the docetaxel arm.The median DoRwas 12.45(95%CI,4.86-25.33)months in the sintilimab arm and 4.14(95%CI,1.41-7.23)months in the docetaxel arm(P=0.045).Treatment-related adverse events of grade≥3were reported in 26(18.1%)patients in the sintilimab arm and 47(36.2%)patients in the docetaxel arm.Exploratory biomarker analysis showed potential predictive values of expression levels of two transcription factors,including OVOL2(HR:0.35;P<0.001)and CTCF(HR:3.50;P<0.001),for sintilimab treatment.Conclusions:Compared with docetaxel,sintilimab significantly improved the OS,PFS,and ORR of Chinese patients with previously treated locally advanced or metastatic sqNSCLC.Yuankai Shi Lin Wu Xinmin Yu Puyuan Xing Yan Wang Jianying Zhou Airong Wang Jianhua Shi Yi Hu Ziping Wang Guangyu An Yong Fang Sanyuan Sun Caicun Zhou Changli Wang Feng Ye Xingya Li Junye Wang Mengzhao Wang Yunpeng Liu Yanqiu Zhao Ying Yuan Jifeng Feng Zhendong Chen Jindong Shi Tao Sun Gang Wu Yongqian Shu Qisen Guo Yi Zhang Yong Song Shucai Zhang Yuan Chen Wei Li Hongrui Niu Wenwei Hu Lijun Wang Jianan Huang Yang Zhang Ying Cheng Zhengdong Wu Bo Peng Jiya Sun Christoph Mancao Yanqi Wang Luyao Sun 2022Cancer Communications2022,42,12:8
6Long-distance ranging with high precision using a soliton microcomb显示文摘Laser-based light detection and ranging(lidar)plays a significant role in both scientific and industrial areas.However,it is difficult for existing lidars to achieve high speed,high precision,and long distance simultaneously.Here,we demonstrate a high-performance lidar based on a chip-scaled soliton microcomb(SMC)that can realize all three specialties simultaneously.Aided by the excellent properties of ultrahigh repetition rate and the smooth envelope of the SMC,traditional optical frequency comb(OFC)-based dispersive interferometry is heavily improved and the measuring dead zone induced by the mismatch between the repetition rate of the OFC and resolution of the optical spectrum analyzer is totally eliminated.Combined with an auxiliary dual-frequency phase-modulated laser range finder,the none-dead-zone measurable range ambiguity is extended up to 1500 m.The proposed SMC lidar is experimentally implemented in both indoor and outdoor environment.In the outdoor baseline field,real-time,high-speed(up to 35 k Hz)measurement of a long distance of^1179 m is achieved with a minimum Allan deviation of 5.6μm at an average time of 0.2 ms(27 nm at an average time of 1.8 s after high-pass filtering).The present SMC lidar approaches a compact,fast,high-precision,and none-dead zone long-distance ranging system,aimed at emerging applications of frontier basic scientific research and advances in industrial manufacturing.JINDONG WANG ZHIZHOU LU WEIQIANG WANG FUMIN ZHANG JIAWEI CHEN YANG WANG JIHUI ZHENG SAI T.CHU WEI ZHAO BRENT E.LITTLE XINGHUA QU WENFU ZHANG 2020Photonics Research2020,8,12:8
7Phot2-regulated relocation of NPH_3 mediates phototropic response to high-intensity blue light in Arabidopsis thaliana显示文摘Two redundant blue-light receptors, known as phototropins(phot1 and phot2), influence a variety of physiological responses, including phototropism, chloroplast positioning, and stomatal opening in Arabidopsis thaliana.Whereas phot1 functions in both low-and high-intensity blue light(HBL), phot2 functions primarily in HBL. Here, we aimed to elucidate phot2-specific functions by screening for HBLinsensitive mutants among mutagenized Arabidopsis phot1 mutants. One of the resulting phot2 signaling associated(p2 sa) double mutants, phot1 p2 sa2, exhibited phototropic defects that could be restored by constitutively expressing NON-PHOTOTROPIC HYPOCOTYL 3(NPH3), indicating that P2 SA2 was allelic to NPH3. It was observed that NPH3-GFP signal mainly localized to and clustered on the plasma membrane in darkness. This NPH3 clustering on the plasma High-Immembrane was not affected by mutations in genes encoding proteins that interact with NPH3, including PHOT1, PHOT2 and ROOT PHOTOTROPISM 2(RPT2). However, the HBL irradiationmediated release of NPH3 proteins into the cytoplasm was inhibited in phot1 mutants and enhanced in phot2 and rpt2-2 mutants. Furthermore, HBL-induced hypocotyl phototropism was enhanced in phot1 mutants and inhibited in the phot2 and rpt2-2 mutants. 9Xiyanping injection therapy for children with mild hand foot and mouth disease:a randomized controlled trial显示文摘OBJECTIVE: To evaluate the clinical effects of Xiyan-ping injection intervention in pediatric patients with mild hand foot and mouth disease(HFMD).METHODS: A total of 329 patients were stratified and block-randomized for symptomatic treatment of HFMD and assigned to one of the following groups: Western Medicine(group A, n = 103), Xiyanping injection(group B, n = 109), or Xiyanping injection and symptomatic treatment using Western Medicine(group C, n = 117). During the trial, fever,rash, ulcers of the mouth were observed among participants in each group before and after treatment, and conversion rates from mild to severe HFMD were measured.RESULTS: After 3-7 days' treatment, no significant differences in the conversion rates from mild to severe HFMD were observed among the three groups(P > 0.05). There was a significantly low number of patients with the onset time of antifebrile effect,vanished time of hand and foot rashes and cumulative time for the ulcers in the mouth vanished,among the three groups(P < 0.05, P < 0.01). The onset time of antifebrile effect of patients in groups B and C were markedly shorter compared with those in group A(P < 0.05, P < 0.01); there was no statically significant difference observed between groups B and C(P > 0.05). No significant differences were observed in the time for body temperature recovery among the three groups(P > 0.05). There were no significant differences in adverse effects among the three groups(P > 0.05).CONCLUSION: Xiyanping injection together with use of Western Medicine is most effective for symptomatic treatment of mild HFMD. 10Molecular cloning and expression of Pfu DNA polymerase gene and its application in long-distance PCR显示文摘A 2.3 kb DNA fragment containing Pfu DNA polA gene was amplified by PCR from total DNA of Pyrococcus furiosus and cloned into a pGEM-T vector. The recombinant clone pT-pfu was digested with Nco I and Xho I and the fragment was inserted into an expression vector pET3d-X. The Pfu polA gene was expressed in Escherichia coli BL21 (DE3). The gene product (Pfu) was purified with heat denaturation, polyethylenemine (PEI) precipitation and Bio-rex 70 ion-exchange chromatography. The recombinant Pfu was verified by protein N-terminal sequencing. 11The blue light receptor CRY1 interacts with GID1 and DELLA proteins to repress GA signaling during photomorphogenesis in Arabidopsis显示文摘Light is a critical environmental cue that regulates a variety of diverse plant developmental processes.Cryptochrome 1(CRY1)is the major photoreceptor that mediates blue light-dependent photomorphogenic responses such as the inhibition of hypocotyl elongation.Gibberellin(GA)participates in the repression of photomorphogenesis and promotes hypocotyl elongation.However,the antagonistic interaction between blue light and GA is not well understood.Here,we report that blue light represses GA-induced degradation of the DELLA proteins(DELLAs),which are key negative regulators in the GA signaling pathway,via CRY1,thereby inhibiting the GA response during hypocotyl elongation.Both in vitro and in vivo biochemical analyses demonstrated that CRY1 physically interacts with GA receptors-GA-INSENSITIVE DWARF 1 proteins(GID1s)-and DELLAs in a blue light-dependent manner.Furthermore,we showed that CRY1 inhibits the association between GID1s and DELLAs.Genetically,CRY1 antagonizes the function of GID1s to repress the expression of cell elongation-related genes and thus hypocotyl elongation.Taken together,our findings demonstrate that CRY1 coordinates blue light and GA signali ng for plant photomorphogenesis by stabilizing DELLAs through the binding and in activation of GID1s,providing new in sights into the mechanism by which blue light antagonizes the function of GA in photomorphogenesis.Ming Zhong Bingjie Zeng Dongying Tang Jiaxin Yang Lina Qu Jindong Yan Xiaochuan Wang Xin Li Xuanming Liu Xiaoying Zhao 2021Molecular Plant2021,14,8:3
12Branched chain amino acids alter fatty acid profile in colostrum of sows fed a high fat diet显示文摘Background:Branched chain amino acids(BCAAs)are important substrates for milk protein synthesis in the mammary gland,and are tightly related to lipid metabolism.No study has been performed examining the role of BCAAs with high fat diets on milk fat synthesis.This study was designed to investigate the effect of dietary BCAAs on growth performance of piglets,progeny body weight,and milk fat composition in sows fed a high fat diet.Four diets(CON=control diet;HF=high fat diet with 8%soybean oil;HF-MB=HF plus 0.39%BCAAs;HF-HB=HF plus 0.78%BCAAs)were fed to sows from late gestation to weaning.Results:Compared to HF,BCAAs(HF-MB and HF-HB)increased the litter weight(P<0.05)and overall litter weight gain(P<0.05)at weaning and increased colostrum fat content by 27.3–35.8%(P<0.01).Fatty acid profiles between the two doses of BCAAs were similar.Compared with HF,HF-MB tended to decrease the percentage of C18:3n3(P=0.063)and increased the percentage of C18:1n9c(P=0.03).In addition,BCAAs in HF-MB increased the concentration of total fatty acid by 22.1%in colostrum(P=0.03)but decreased that in serum at parturition by 53.2%(P=0.027).The fatty acids in colostrum that increased with BCAAs were C15:0,C17:0,C20:3n6,C20:4n6,C20:5n3 and C22:6n3(P=0.00~0.04).Colostrum fatty acids of C20:0,C21:0,C22:0,C16:1,C20:1,C18:1n9c also tended to be increased(0.05Chang Ma Yajng Liu Shaoshuai Liu Crystal L.Lévesque Fengqi Zhao Jindong Yin Bing Dong 2020Journal of Animal Science and Biotechnology2020,11,2:3
