1Spirulina platensis aqueous extracts ameliorate colonic mucosal damage and modulate gut microbiota disorder in mice with ulcerative colitis by inhibiting inflammation and oxidative stress显示文摘Ulcerative colitis(UC)is a chronic and recurrent inflammatory bowel disease(IBD)that has become a major gastroenterologic problem during recent decades.Numerous complicating factors are involved in UC development such as oxidative stress,inflammation,and microbiota disorder.These factors exacerbate damage to the intestinal mucosal barrier.Spirulina platensis is a commercial alga with various biological activity that is widely used as a functional ingredient in food and beverage products.However,there have been few studies on the treatment of UC using S.platensis aqueous extracts(SP),and the underlying mechanism of action of SP against UC has not yet been elucidated.Herein,we aimed to investigate the modulatory effect of SP on microbiota disorders in UC mice and clarify the underlying mechanisms by which SP alleviates damage to the intestinal mucosal barrier.Dextran sulfate sodium(DSS)was used to establish a normal human colonic epithelial cell(NCM460)injury model and UC animal model.The mitochondrial membrane potential assay 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)and staining with Annexin V-fluorescein isothiocyanate(FITC)/propidium iodide(PI)and Hoechst 33258 were carried out to determine the effects of SP on the NCM460 cell injury model.Moreover,hematoxylin and eosin(H&E)staining,transmission electron microscopy(TEM),enzyme-linked immunosorbent assay(ELISA),quantitative real-time polymerase chain reaction(qPCR),western blot,and 16S ribosomal DNA(rDNA)sequencing were used to explore the effects and underlying mechanisms of action of SP on UC in C57BL/6 mice.In vitro studies showed that SP alleviated DSS-induced NCM460 cell injury.SP also significantly reduced the excessive generation of intracellular reactive oxygen species(ROS)and prevented mitochondrial membrane potential reduction after DSS challenge.In vivo studies indicated that SP administration could alleviate the severity of DSS-induced colonic mucosal damage compared with the control group.Inhibition of inflammation and oxidative stress was associated with increases in the activity of antioxidant enzymes and the expression of tight junction proteins(TJs)post-SP treatment.SP improved gut microbiota disorder mainly by increasing antioxidant enzyme activity and the expression of TJs in the colon.Our findings demonstrate that the protective effect of SP against UC is based on its inhibition of pro-inflammatory cytokine overproduction,inhibition of DSS-induced ROS production,and enhanced expression of antioxidant enzymes and TJs in the colonic mucosal barrier.Jian WANG Liqian SU Lun ZHANG Jiali ZENG Qingru CHEN Rui DENG Ziyan WANG Weidong KUANG Xiaobao JIN Shuiqing GUI Yinghua XU Xuemei LU 2022Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,6:3
219.34 cm2 large-area quaternary organic photovoltaic module with 12.36% certified efficiency显示文摘In this study, a quaternary blending strategy was applied in the fabrication of organic photovoltaic devices and large-area modules. As a result, the ultimate quaternary organic solar cells(OSCs) deliver 16.71% efficiency for small-area devices and 13.25% for large-area(19.34 cm2) modules(certified as 12.36%), which is one of the highest efficiencies for organic solar modules to date. 3Whole-genome Duplication Reshaped Adaptive Evolution in A Relict Plant Species,Cyclocarya paliurus显示文摘Cyclocarya paliurus is a relict plant species that survived the last glacial period and shows a population expansion recently.Its leaves have been traditionally used to treat obesity and diabetes with the well-known active ingredient cyclocaric acid B.Here,we presented three C.paliurus genomes from two diploids with different flower morphs and one haplotype-resolved tetraploid assembly.Comparative genomic analysis revealed two rounds of recent whole-genome duplication events and identified 691 genes with dosage effects that likely contribute to adaptive evolution through enhanced photosynthesis and increased accumulation of triterpenoids.Resequencing analysis of 45 C.paliurus individuals uncovered two bottlenecks,consistent with the known events of environmental changes,and many selectively swept genes involved in critical biological functions,including plant defense and secondary metabolite biosynthesis.We also proposed the biosynthesis pathway of cyclocaric acid B based on multi-omics data and identified key genes,in particular gibberellinrelated genes,associated with the heterodichogamy in C.paliurus species.Our study sheds light on evolutionary history of C.paliurus and provides genomic resources to study the medicinal herbs.Yinquan Qu Xulan Shang Ziyan Zeng Yanhao Yu Guoliang Bian Wenling Wang Li Liu Li Tian Shengcheng Zhang Qian Wang Dejin Xie Xuequn Chen Zhenyang Liao Yibin Wang Jian Qin Wanxia Yang Caowen Sun Xiangxiang Fu Xingtan Zhang Shengzuo Fang 2023Genomics, Proteomics & Bioinformatics2023,21,3:1

5A gap-free and haplotype-resolved lemon genome provides insights into flavor synthesis and huanglongbing (HLB) tolerance显示文摘The lemon(Citrus limon;family Rutaceae)is one of the most important and popular fruits worldwide.Lemon also tolerates huan-glongbing(HLB)disease,which is a devastating citrus disease.Here we produced a gap-free and haplotype-resolved chromosome-scale genome assembly of the lemon by combining Pacific Biosciences circular consensus sequencing,Oxford Nanopore 50-kb ultra-long,and high-throughput chromatin conformation capture technologies.The assembly contained nine-pair chromosomes with a contig N50 of 35.6 Mb and zero gaps,while a total of 633.0 Mb genomic sequences were generated.The origination analysis identified 338.5Mb genomic sequences originating from citron(53.5%),147.4Mb frommandarin(23.3%),and 147.1Mb frompummelo(23.2%).The genome included 30528 protein-coding genes,and most of the assembled sequences were found to be repetitive sequences.Several significantly expanded gene families were associated with plant-pathogen interactions,plant hormone signal transduction,and the biosynthesis of major active components,such as terpenoids and f lavor compounds.Most HLB-tolerant genes were expanded in the lemon genome,such as 2-oxoglutarate(2OG)/Fe(II)-dependent oxygenase and constitutive disease resistance 1,cell wall-related genes,and lignin synthesis genes.Comparative transcriptomic analysis showed that phloem regeneration and lower levels of phloem plugging are the elements that contribute to HLB tolerance in lemon.Our results provide insight into lemon genome evolution,active component biosynthesis,and genes associated with HLB tolerance.Yixue Bao Ziyan Zeng Wei Yao Xiao Chen Mengwei Jiang Akbar Sehrish Bo Wu Charles A.Powell Baoshan Chen Jianlong Xu Xingtan Zhang Muqing Zhang 2023Horticulture Research2023,10,4:0
6Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND 7Integrated Rabies Surveillance-Hunan Province,China,2020显示文摘Introduction:The objective of this paper was to assess the epidemiology of rabies in Hunan Province,analyze the associated factors,understand the status of prevention and treatment after rabies exposure,evaluate the effectiveness of prevention and treatment,and provide a scientific basis for formulating effective prevention and control measures.Methods:The surveillance data of rabies in Hunan Province in 2020 were collected and analyzed by descriptive epidemiological method.Results:In 2020,a total of 59 cases of rabies were reported in Hunan Province,with an incidence rate of 0.09/100,000.Overall,42 cases(71.19%)were due to animal bites and 43 cases(72.88%)were of grade III.The proportion of hand and combined injury of hand was the highest(40.68%).A total of 603,261 cases of rabies exposure were reported from the rabies post-exposure prophylaxis(PEP)clinic in Hunan Province.Dogs were the main animal causing injuries,accounting for 74.21%.Only 83,418(13.84%)of the animals had a clear immune history,and a total of 11 dog attacks were reported in Hunan Province.The average immunity rate of dogs in the whole province was 30.98%.In 2020,554 dogs were sampled in the whole province;20 of them were positive for a positivity rate of 3.61%.Conclusions:Rabies in Hunan Province in 2020 had a relatively low prevalence.Failure to treat wounds,immunoglobulin injections,and vaccination after exposure were the main causes of rabies.Therefore,post-exposure management of rabies should be further strengthened to reduce the risk of rabies for high-risk populations.Hao Yang Shengbao Chen Zhihui Dai Longwei Luo Qianlai Sun Shanlu Zhao Ge Zeng Ziyan Liu Shixiong Hu 2022China CDC weekly2022,4,44:0

9Contact pattern,current immune barrier,and pathogen virulence determines the optimal strategy of further vaccination显示文摘Background:The current outbreak of novel coronavirus disease 2019 has caused a seriousdisease burden worldwide.Vaccines are an important factor to sustain the epidemic.Although with a relatively high-vaccination worldwide,the decay of vaccine efficacy andthe arising of new variants lead us to the challenge of maintaining a sufficient immunebarrier to protect the population.Method:A case-contact tracking data in Hunan,China,is used to estimate the contactpattern of cases for scenarios including school,workspace,etc,rather than ordinary susceptible population.Based on the estimated vaccine coverage and efficacy,a multi-groupvaccinated-exposed-presymptomatic-symptomatic-asymptomatic-removed model(VEFIAR)with 8 age groups,with each partitioned into 4 vaccination status groups isdeveloped.The optimal dose-wise vaccinating strategy is optimized based on the currentlyestimated immunity barrier of coverage and efficacy,using the greedy algorithm thatminimizes the cumulative cases,population size of hospitalization and fatality respectivelyin a certain future interval.Parameters of Delta and Omicron variants are used respectivelyin the optimization.Results:The estimated contact matrices of cases showed a concentration on middle ages,and has compatible magnitudes compared to estimations from contact surveys in otherstudies.The VEFIAR model is numerically stable.The optimal controled vaccination strategy requires immediate vaccination on the un-vaccinated high-contact population of age30e39 to reduce the cumulative cases,and is stable with different basic reproductionnumbers(R_(0)).As for minimizing hospitalization and fatality,the optimized strategy requires vaccination on the un-vaccinated of both aged 30e39 of high contact frequencyand the vulnerable older.Conclusion:The objective of reducing transmission requires vaccination in age groups ofthe highest contact frequency,with more priority for un-vaccinated than un-fully or fullyvaccinated.The objective of reducing total hospitalization and fatality requires not only toreduce transmission but also to protect the vulnerable older.The priority changes byvaccination progress.For any region,if the local contact pattern is available,then with thevaccination coverage,efficacy,and disease characteristics of relative risks in heterogeneouspopulations,the optimal dose-wise vaccinating process will be obtained and gives hintsfor decision-making.Xiaohao Guo Ziyan Liu Shiting Yang Zeyu Zhao Yichao Guo Guzainuer Abudurusuli Shanlu Zhao Ge Zeng Shixiong Hu Kaiwei Luo Tianmu Chen 2023Infectious Disease Modelling2023,8,1:0
