1Notch signaling in blood vessels: from morphogenesis to homeostasis显示文摘Notch signaling is an evolutionarily conserved intercellular signaling pathway that plays numerous crucial roles in vascular development and physiology.Compelling evidence indicates that Notch signaling is vital for vascular morphogenesis including arterial and venous differentiation and endothelial tip and stalk cell specification during sprouting angiogenesis and also vessel maturation featured by mural cell differentiation and recruitment.Notch signaling is also required for vascular homeostasis in adults by keeping quiescent phalanx cells from re-entering cell cycle and by modulating the behavior of endothelial progenitor cells.We will summarize recent advances of Notch pathway in vascular biology with special emphasis on the underlying molecular mechanisms.ZHANG Ping YAN XianChun CHEN Yan YANG ZiYan HAN Hua 2014Science China(Life Sciences)2014,57,8:15
2Endothelial cells-targeted soluble human Delta-like 4 suppresses both physiological and pathological ocular angiogenesis显示文摘Due to its essential roles in angiogenesis, Notch pathway has emerged as an attractive target for the treatment of pathologic angiogenesis. Although both activation and blockage of Notch signal can impede angiogenesis, activation of Notch signal may be more promising because it was shown that long-term Notch signal blockage resulted in vessel neoplasm. However, an in vivo deliverable Notch ligand with highly efficient Notch-activating capacity has not been developed. Among all the Notch ligands, Delta-like4(Dll4) is specifically involved in angiogenesis. In this study, we generated a novel soluble Notch ligand h D4 R, which consists of the Delta-Serrate-Lag-2 fragment of human Dll4 and an arginine-glycine-aspartate(RGD) motif targeting endothelial cells(ECs). We demonstrated that h D4 R could bind to ECs through its RGD motif and effectively triggered Notch signaling in ECs. Further, we confirmed that h D4 R could suppress angiogenesis in vitro as manifested by network formation assay and sprouting assay. More importantly, h D4 R efficiently repressed neonatal retinal angiogenesis and laser-induced choroidal neovascularization(CNV) as well in vivo. 3Suppress voltage decay of lithium-rich materials by coating layers with different crystalline states显示文摘Li-rich oxides are considered as the most commercial potential cathode materials due to the high theoretical specific discharge capacity. Here, ZrO_(2) in different crystalline states are applied as the coating layers to enhance the electrochemical performance of hollow Li[Li_(0.2)Mn_(0.54)Ni_(0.13)Co_(0.13)]O_(2) materials.Meanwhile, a series of characterizations(XRD, SEM, TEM, EDX etc.) are conducted to compare the effects of ZrO_(2) coating layer with different crystalline states on the host material. 5Current knowledge of Krüppel-like factor 5 and vascular remodeling: providing insights for therapeutic strategies显示文摘Vascular remodeling is a pathological basis of various disorders. Therefore, it is necessary to understand the occurrence, prevention, and treatment of vascular remodeling. Krüppel-like factor 5 (KLF5) has been identified as a significant factor in cardiovascular diseases during the last two decades. This review provides a mechanism network of function and regulation of KLF5 in vascular remodeling based on newly published data and gives a summary of its potential therapeutic applications. KLF5 modulates numerous biological processes, which play essential parts in the development of vascular remodeling, such as cell proliferation, phenotype switch, extracellular matrix deposition, inflammation, and angiogenesis by altering downstream genes and signaling pathways. 619.34 cm2 large-area quaternary organic photovoltaic module with 12.36% certified efficiency显示文摘In this study, a quaternary blending strategy was applied in the fabrication of organic photovoltaic devices and large-area modules. As a result, the ultimate quaternary organic solar cells(OSCs) deliver 16.71% efficiency for small-area devices and 13.25% for large-area(19.34 cm2) modules(certified as 12.36%), which is one of the highest efficiencies for organic solar modules to date. 7Endothelial Notch activation promotes neutrophil transmigration via downregulating endomucin to aggravate hepatic ischemia/reperfusion injury显示文摘Inflammatory leukocytes infiltration is orchestrated by mechanisms involving chemokines,selectins,addressins and other adhesion molecules derived from endothelial cells(ECs),but how they respond to inflammatory cues and coordinate leukocyte transmigration remain elusive.In this study,using hepatic ischemia/reperfusion injury(HIRI)as a model,we identified that endothelial Notch activation was rapidly and dynamically induced in liver sinusoidal endothelial cells(LSECs)in acute inflammation.In mice with EC-specific Notch activation(NICeCA),HIRI induced exacerbated liver damage.Consistently,endothelial Notch activation enhanced neutrophil infiltration and tumor necrosis factor(TNF)-αexpression in HIRI.Transcriptome analysis and further qRT-PCR as well as immunofluorescence indicated that endomucin(EMCN),a negative regulator of leukocyte adhesion,was downregulated in LSECs from NICeCA mice.EMCN was downregulated during HIRI in wild-type mice and in vitro cultured ECs insulted by hypoxia/re-oxygenation injury.Notch activation in ECs led to increased neutrophil adhesion and transendothelial migration,which was abrogated by EMCN overexpression in vitro.In mice deficient of RBPj,the integrative transcription factor of canonical Notch signaling,although overwhelming sinusoidal malformation aggravated HIRI,the expression of EMCN was upregulated;and pharmaceutical Notch blockade in vitro also upregulated EMCN and inhibited transendothelial migration of neutrophils.The Notch activation-exaggerated HIRI was compromised by blocking LFA-1,which mediated leukocyte adherence by associating with EMCN.Therefore,endothelial Notch signaling controls neutrophil transmigration via EMCN to modulate acute inflammation in HIRI.Peiran Zhang Kangyi Yue Xinli Liu Xianchun Yan Ziyan Yang Juanli Duan Congcong Xia Xinyuan Xu Mei Zhang Liang Liang Lin Wang Hua Han 2020Science China(Life Sciences)2020,63,3:1

10Preclinical characterization and comparison between CD3/CD19 bispecific and novel CD3/CD19/CD20 trispecific antibodies against B-cell acute lymphoblastic leukemia:targeted immunotherapy for acute lymphoblastic leukemia显示文摘The CD19-targeting bispecific T-cell engager blinatumomab has shown remarkable efficacy in patients with relapsed/refractory B-cell precursor acute lymphoblastic leukemia.However,several studies showed that blinatumomab has a short plasma half-life due to its low molecular weight,and thus its clinical use is limited.Furthermore,multiple trials have shown that approximately 30%of blinatumomab-relapsed cases are characterized by CD19 negative leukemic cells.Here,we design and characterize two novel antibodies,A-319 and A-2019.Blinatumomab and A-319 are CD3/CD19 bispecific antibodies with different molecular sizes and structures,and A-2019 is a novel CD3/CD19/CD20 trispecific antibody with an additional anti-CD20 function.Our in vitro,ex vivo,and in vivo experiments demonstrated that A-319 and A-2019 are potent antitumor agents and capable of recruiting CD3 positive T cells,enhancing T-cell function,mediating B-cell depletion,and eventually inhibiting tumor growth in Raji xenograft models.The two molecules are complementary in terms of efficacy and specificity profile.The activity of A-319 demonstrated superior to that of A-2019,whereas A-2019 has an additional capability to target CD20 in cells missing CD19,suggesting its potential function against CD19 weak or negative CD20 positive leukemic cells.Sisi Wang Lijun Peng Wenqian Xu Yuebo Zhou Ziyan Zhu Yushan Kong Stewart Leung Jin Wang Xiaoqiang Yan Jian-Qing Mi 2022Frontiers of Medicine2022,16,1:1

12Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND 13Fast modeling of gravity gradients from topographic surface data using GPU parallel algorithm显示文摘The gravity gradient is a secondary derivative of gravity potential,containing more high-frequency information of Earth’s gravity field.Gravity gradient observation data require deducting its prior and intrinsic parts to obtain more variational information.A model generated from a topographic surface database is more appropriate to represent gradiometric effects derived from near-surface mass,as other kinds of data can hardly reach the spatial resolution requirement.The rectangle prism method,namely an analytic integration of Newtonian potential integrals,is a reliable and commonly used approach to modeling gravity gradient,whereas its computing efficiency is extremely low.A modified rectangle prism method and a graphical processing unit(GPU)parallel algorithm were proposed to speed up the modeling process.The modified method avoided massive redundant computations by deforming formulas according to the symmetries of prisms’integral regions,and the proposed algorithm parallelized this method’s computing process.The parallel algorithm was compared with a conventional serial algorithm using 100 elevation data in two topographic areas(rough and moderate terrain).Modeling differences between the two algorithms were less than 0.1 E,which is attributed to precision differences between single-precision and double-precision float numbers.The parallel algorithm showed computational efficiency approximately 200 times higher than the serial algorithm in experiments,demonstrating its effective speeding up in the modeling process.Further analysis indicates that both the modified method and computational parallelism through GPU contributed to the proposed algorithm’s performances in experiments.Xuli Tan Qingbin Wang Jinkai Feng Yan Huang Ziyan Huang 2021Geodesy and Geodynamics2021,12,4:0
14Tissue-specific transcriptomics reveals a central role of CcNST1 in regulating the fruit lignification pattern in Camellia chekiangoleosa,a woody oil-crop显示文摘Fruit lignification is of significant economic importance because it affects the quality of fruit and the production of seed oil.The specified lignification pattern in Camellia chekiangoleosa fruits plays critical roles in its seed oil yield,but little is known about how this lignification process is regulated.Here,we report on a comprehensive tissue-specific transcriptomics analysis conducted for C.chekiangoleosa fruit.By mining the differentially expressed genes,we found that lignin biosynthesis and transcriptional regulation pathways were significantly enriched in the lignified tissues.The homolog of NST-like transcription factor,CcNST1,was highly expressed in lignified seed coat and endocarp tissues;transgenic analyses of CcNST1 in Arabidopsis and hybrid poplar revealed the enhanced lignification levels of various tissues.Gene expression analysis of the transgenic lines uncovered potential downstream genes involved in the regulation of lignin biosynthesis.This work provides a valuable gene expression resource and identified the pivotal role of CcNST1 in regulating the lignin biosynthesis underlying fruit lignification.Chao Yan Ziyan Nie Zhikang Hu Hu Huang Xianjin Ma Sijia Li Jiyuan Li Xiaohua Yao Hengfu Yin 2022Forestry Research2022,2,1:0

16Three novel alleles of OsGS1 developed by base-editing-mediated artificial evolution confer glufosinate tolerance in rice显示文摘Only few glufosinate-tolerant genes,such as phosphinothricin acetyltransferase(PAT)and bialaphos resistance(bar)identified from Streptomyces,are currently available for developing genetically modified rice in agricultural application.Following the rapid development of genome editing technology,generation of novel glufosinate-tolerant gene resources through artificial evolution of endogenous genes is more promising and highly desirable in rice molecular breeding program.In this study,the endogenous Glutamine synthetase1(OsGS1)was artificially evolved by base-editing-mediated gene evolution(BEMGE)in rice cells to create novel alleles conferring glufosinate tolerance in rice germplasms.Two novel glufosinate-tolerant OsGS1 alleles(OsGS1-AVPS and OsGS1-+AF)and one reported tolerant allele(OsGS1-SGTA)were successfully identified from approximately 4200 independent hygromycin-tolerant calli.Germination assays and spray tests revealed that these three OsGS1 alleles confer glufosinate tolerance in rice.Furthermore,OsGS1-AVPS and OsGS1-SGTA were quickly deployed into the elite rice cultivar Nangeng 46 through precise base editing.Overall,our results demonstrate the feasibility of developing glufosinate-tolerant rice by editing an endogenous rice gene in molecular breeding programs.Bin Ren Yongjie Kuang Ziyan Xu Xuemei Wu Dawei Zhang Fang Yan Xiangju Li Xueping Zhou Guirong Wang Huanbin Zhou 2023The Crop Journal2023,11,2:0

19A multistimuli-responsive fluorescent hydrogel based on a fluorescence response to macromolecular segmental motion显示文摘Fluorescent hydrogels with fast and reversible responses have attracted extensive attention, and it remains a challenge to designmultistimuli-responsive fluorescent hydrogel through a facile and versatile method. Meanwhile, the segmental motion inhydrogels is of significance for the various functions of hydrogels such as chemical reactivity, self-healing, and mechanicalstrength, etc., however, it is difficult and complicated to in situ investigate the segmental motion under different conditions. In thiswork, a multistimuli-responsive fluorescent hydrogel was designed and fabricated by introducing a tetraphenylethylene (TPE)derivative as a nonaggregated crosslinker in the gel network. Since the intermolecular rotation of TPE at the crosslinking pointwas directly integrated with the dynamic conformational transition of the macromolecular network, the mobility of macromolecularsegments can be monitored by the fluorescence intensity of the hydrogel. The prepared hydrogel has promising fluorescenceresponses to temperature, pH, metal ions, and hydrogen bonding agents, and characterization of the fluorescence and the chainsegmental motion showed that the weaker the mobility of the network macromolecular chain is, the stronger the fluorescenceintensity is.
