2Graft-accelerated virus-induced gene silencing facilitates functional genomics in rose flowers显示文摘Rose has emerged as a model ornamental plant for studies of flower development, senescence, and morphology, as well as the metabolism of floral fragrances and colors.Virus-induced gene silencing(VIGS) has long been used in functional genomics studies of rose by vacuum infiltration of cuttings or seedlings with an Agrobacterium suspension carrying TRV-derived vectors. However, VIGS in rose flowers remains a challenge because of its low efficiency and long time to establish silencing. Here we present a novel and rapid VIGS method that can be used to analyze gene function in rose,called ‘graft-accelerated VIGS', where axil ary sprouts are cut from the rose plant and vacuum infiltrated with Agrobacterium. The inoculated scions are then grafted back onto the plants to flower and silencing phenotypes can be observed within 5 weeks, post-infiltration. Using this new method, we successfully silenced expression of the RhDFR_1, RhA G, and RhNUDX_1 in rose flowers, and affected their color, petal number, as well as fragrance, respectively. This grafting method will facilitate high-throughput functional analysis of genes in rose flowers. 4Bolt support mechanism based on elastic theory显示文摘Mechanical model of anchorage surrounding rock considering tray effect was established based on elastic theory,in order to study the mechanism of bolt supporting.Elastic solutions of normal force at point in the interior of a semi-infnite solid were obtained by means of classical displacement function method in elasticity.The factors which influence stress of bolted surrounding rock,such as the length of bolt and tray effect,were analyzed.The absolute value of stress along bolt axes decreased rapidly with an increase in radical distance and the maximum appeared near ends of bolt.With increasing radical distance,the value of radical stress changed from positive to negative roughly and then increased to zero,with maximum at the middle of bolt.The evolution of hoop stress as radical distance increasing was similar with stress along bolt axes.With an increase in depth,the radical effect ranges of all normal stress components were reduced.These suggest that the effect from tray on stress along bolt axes of bolted surrounding rock could be neglected,except near surface of surrounding rock.Guo Xiaoqian Mao Xianbiao Ma Chao Huang Jinglong 2013International Journal of Mining Science and Technology2013,23,4:9
5Toripalimab plus chemotherapy as second-line treatment in previously EGFR-TKI treated patients with EGFR-mutant-advanced NSCLC:a multicenter phase-II trial显示文摘This multicenter phase-II trial aimed to investigate the efficacy,safety,and predictive biomarkers of toripalimab plus chemotherapy as second-line treatment in patients with EGFR-mutant-advanced NSCLC.Patients who failed from first-line EGFR-TKIs and did not harbor T790M mutation were enrolled.Toripalimab plus carboplatin and pemetrexed were administrated every three weeks for up to six cycles,followed by the maintenance of toripalimab and pemetrexed.The primary endpoint was objective-response rate(ORR).Integrated biomarker analysis of PD-L1 expression,tumor mutational burden(TMB),CD8+tumor-infiltrating lymphocyte(TIL)density,whole-exome,and transcriptome sequencing on tumor biopsies were also conducted.Forty patients were enrolled with an overall ORR of 50.0%and disease-control rate(DCR)of 87.5%.The median progression free survival(PFS)and overall survival were 7.0 and 23.5 months,respectively.The most common treatment-related adverse effects were leukopenia,neutropenia,anemia,ALT/AST elevation,and nausea.Biomarker analysis showed that none of PD-L1 expression,TMB level,and CD8+TIL density could serve as a predictive biomarker.Integrated analysis of whole-exome and transcriptome sequencing data revealed that patients with DSPP mutation had a decreased M2 macrophage infiltration and associated with longer PFS than those of wild type.Toripalimab plus chemotherapy showed a promising anti-tumor activity with acceptable safety profiles as the second-line setting in patients with EGFR-mutant NSCLC.DSPP mutation might serve as a potential biomarker for this combination.A phase-III trial to compare toripalimab versus placebo in combination with chemotherapy in this setting is ongoing(NCT03924050).Tao Jiang Pingyang Wang Jie Zhang Yanqiu Zhao Jianying Zhou Yun Fan Yongqian Shu Xiaoqing Liu Helong Zhang Jianxing He Guanghui Gao Xiaoqian Mu Zhang Bao Yanjun Xu Renhua Guo Hong Wang Lin Deng Ningqiang Ma Yalei Zhang Hui Feng Sheng Yao Jiarui Wu Luonan Chen Caicun Zhou Shengxiang Ren 2021Signal Transduction and Targeted Therapy2021,6,11:8
6Modeling CADASIL vascular pathologies with patient-derived induced pluripotent stem cells显示文摘Cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy(CADASIL)is a rare hereditary cerebrovascular disease caused by a NOTCH3 mutation.However,the underlying cellular and molecular mechanisms remain unidentified.Here,we generated non-integrative induced pluripotent stem cells(iPSCs)from fibroblasts of a CADASIL patient harboring a heterozygous NOTCH3 mutation(c.3226C>T,p.R1076C).Vascular smooth muscle cells(VSMCs)differentiated from CADASIL-specific iPSCs showed gene expression changes associated with disease phenotypes,including activation of the NOTCH and NF-kB signaling pathway,cytoskeleton disorganization,and excessive cell proliferation.In comparison,these abnormalities were not observed in vascular endothelial cells(VECs)derived from the patients iPSCs.Importantly,the abnormal upregulation of NF-kB target genes in CADASIL VSMCs was diminished by a NOTCH pathway inhibitor,providing a potential therapeutic strategy for CADASIL.Overall,using this iPSCbased disease model,our study identified clues for studying the pathogenic mechanisms of CADASIL and developing treatment strategies for this disease.Chen Ling Zunpeng Liu Moshi Song Weiqi Zhang Si Wang Xiaoqian Liu Shuai Ma Shuhui Sun Lina Fu Qun Chu Juan Carlos Izpisua Belmonte Zhaoxia Wang Jing Qu Yun Yuan Guang-Hui Liu 2019Protein & Cell2019,10,4:7
7Telomere-dependent and telomereindependent roles of RAP1 in regulating human stem cell homeostasis显示文摘RAP1 is a well-known telomere-binding protein, but its functions in human stem cells have remained unclea匚 Here we generated RAP1 -deficient human embryonic stem cells (hESCs) by using CRISPR/Cas9 technique and obtained RAP1-deficient human mesenchymal stem cells (hMSCs) and neural stem cells (hNSCs) via directed differentiation. In both hMSCs and hNSCs, RAP1 not only negatively regulated telomere length but also acted as a transcriptional regulator of RELN by tuning the methylation status of its gene promoter. 8GNP6,a novel allele of MOC1,regulates panicle and tiller development in rice显示文摘The yield of rice is mostly affected by three factors,namely,panicle number,grain number and grain weight.Variation in panicle and grain numbers is mainly caused by tiller and panicle branches generated from axillary meristems(AMs).MOC1 encodes a putative GRAS family nuclear protein that regulates AM formation.Although several alleles of MOC1 have been identified,its variation in germplasm resources remains unclear.In the present study we characterized a novel mocl allele named gnp6 which has a thymine insertion in the coding sequence of the SAW motif in the GRAS domain.This mutation causes arrested branch formation.The SAW motif is necessary for nuclear localization of GNP6/MOC1 where it functions as a transcription factor or co-regulator.Haplotype analysis showed that the coding region of GNP6/MOC1 was conserved without any non-synonymous mutations in 240 rice accessions.However,variation in the promoter region might affect the expression of it and its downstream genes.Joint haplotype analysis of GNP6/MOC1 and MOC3 showed that haplotype combinations H9,H10 and H11,namely MOC1-Hap1 in combination with MOC3-Hap3,MOC3-Hap4 or MOC3-Hap5 could be bred to promote branch formation.These findings will enrich the genetic resources available for rice breeders.Zhanying Zhang Xingming Sun Xiaoqian Ma Bingxia Xu Yong Zhao Zhiqi Ma Gangling Li Najeeb Ullah Khan Yinghua Pan Yuntao Liang Hongliang Zhang Jinjie Li Zichao Li 2021The Crop Journal2021,9,1:6
9Target classification using SIFT sequence scale invariants显示文摘On the basis of scale invariant feature transform(SIFT) descriptors,a novel kind of local invariants based on SIFT sequence scale(SIFT-SS) is proposed and applied to target classification.First of all,the merits of using an SIFT algorithm for target classification are discussed.Secondly,the scales of SIFT descriptors are sorted by descending as SIFT-SS,which is sent to a support vector machine(SVM) with radial based function(RBF) kernel in order to train SVM classifier,which will be used for achieving target classification.Experimental results indicate that the SIFT-SS algorithm is efficient for target classification and can obtain a higher recognition rate than affine moment invariants(AMI) and multi-scale auto-convolution(MSA) in some complex situations,such as the situation with the existence of noises and occlusions.Moreover,the computational time of SIFT-SS is shorter than MSA and longer than AMI.Xufeng Zhu Caiwen Ma Bo Liu Xiaoqian Cao 2012Journal of Systems Engineering and Electronics2012,23,5:5
10Highly sensitive ethanol gas sensor based on ultrathin nanosheets assembled Bi_2WO_6 with composite phase显示文摘Bismuth tungstate(Bi2 WO6) has many intriguing properties and has been the focus of studies in a variety of fields, especially photocatalysis. However, its application in gas-sensing has been seldom reported.Here, we successfully synthesized assembled hierarchical Bi2 WO6 which consists of ultrathin nanosheets with crystalline-amorphous composite phase by a one-step hydrothermal method. X-ray diffraction(XRD), X-ray photoemission spectroscopy(XPS), field-emission scanning electron microscopy(FESEM),and high-resolution transmission electron microscopy(HRTEM) techniques were employed to characterize its composition, morphology, and microstructure. By taking advantage of its unique microstructure,phase composition, and large surface area, we show that the resulting Bi2 WO6 is capable of detecting ethanol gas with quick response(7 s) and recovery dynamic(14 s), extremely high sensitivity(Ra/Rg= 60.8@50 ppm ethanol) and selectivity. Additionally, it has excellent reproducibility and long-term stability(more than 50 d). The Bi2 WO6 outperform the existing Bi2 WO6-based and most of the other state-of-the-art sensing platforms. 11A systems dynamic model of a coal-based city with multiple adaptive scenarios: A case study of Ordos, China显示文摘Cities based on coal resources have increasingly important social and economic roles in China. Their strategies for sustainable development, however, urgently need to be improved, which represents a huge challenge. Most observers believe that the continued progress of these cities relies on the optimization of scientific adaptive management in which social, economic, and ecological factors are incorporated. A systems perspective that combines policies, management priorities, and long-term policy impacts needs to be applied. To date, however, such an approach has not been adopted, which means it is difficult to implement adaptive management at the regional scale. In this study, we used various situations to develop a multiple adaptive scenario system dynamics model. We then simulated a range of policy scenarios, with Ordos in the Inner Mongolia Autonomous Region as a case study. Simulation results showed that the current strategy is not sustainable and predicted that the system would exceed the environmental capacity, with risks of resource exhaustion and urban decline in 2025–2035. Five critical policy variables, including the urban population carrying capacity, rates of water consumption and water recycling, and expansion of urban land cover, were identified during sensitivity analysis. We developed and compared six socio-economic scenarios. The urban area, represented by the urban population density, seemed to transition through five different stages, namely natural growth, rapid growth, stable oscillation, fading, and rebalancing. Our scenarios suggested that different policies had different roles through each stage. The water use efficiency management policy had a comprehensive far-reaching influence on the system behavior; land urbanization management functions dominated at the start, and population capacity management was a major control in the mid-term. 12Microstructure evolution and coercivity mechanism of hydrogenation-disproportionation-desorption-recombination(HDDR) treated Nd-Fe-B strip cast alloys显示文摘In this paper,we systematically investigated the microstructure evolution and coercivity mechanism of hydrogenation-disproportionation-desorption-recombination(HDDR)treated Nd-Fe-B strip cast alloys by transmission electron microscopy(TEM)and three-dimensional atom probe(3 DAP)analyses.The rodlike NdH_(2+x) phases with diameters of 10-20 nm are embedded intoα-Fe matrix,which hereditarily leads to textured grains in HDDR alloy.The migration of NdH_(2+x) from Nd-rich region toα-Fe matrix during hydrogen absorption process contributes to the uniform redistribution of Nd-rich phases after HDDR treatment The HDDR alloy with single domain grain sizes of 200-300 nm exhibits relatively low coercivity of 1.01 T that arises from pinning magnetic domain motion.The weak c-axis orientation of HDDR alloy results in a lower reverse magnetic field(coercivity)to reduce remanence to 0.Moreover,the direct contact of Nd_(2)Fe_(14)B grains and the high concentration of ferromagnetic elements(Fe content≈66.06 at%,Co content≈0.91 at%)in Nd-rich grain boundary layer lead to strong magnetostatic coupling effect among Nd_(2) Fe_(14)B grains.The nano-sized a-Fe inside Nd_(2) Fe_(14)B matrix makes the magnetization reversal easily and decreases the coercivity of HDDR alloy.Bin Ma Xiaoqian Bao Aizhi Sun Jiheng Li Xuexu Gao 2022Journal of Rare Earths2022,40,5:2
13Psoralen inhibits hepatitis B viral replication by down-regulating the host transcriptional machinery of viral promoters显示文摘The hepatitis B virus(HBV) is a global public health challenge due to its highly contagious nature. It is estimated that almost 300 million people live with chronic HBV infection annually. Although nucleoside analogs markedly reduce the risk of liver disease progression, the analogs do not fully eradicate the virus. As such, new treatment options and drugs are urgently needed. Psoralen is a nourishing monomer of Chinese herb and is known to inhibit virus replication and inactivate viruses. In this study, we evaluated the potential of psoralen as an anti-HBV agent.Quantitative PCR and Southern blot analysis revealed that psoralen inhibited HBV replication in Hep G2.2.15 cells in a concentration-dependent manner. Moreover, psoralen was also active against the 3TC/ETV-dual-resistant HBV mutant. Further investigations revealed that psoralen suppressed both HBV RNA transcription and core protein expression. The transcription factor FOXO1, a known target for PGC1α co-activation, binds to HBV precore/core promoter enhancer II region and activates HBV RNA transcription. Co-immunoprecipitation showed that psoralen suppressed the expression of FOXO1, thereby decreasing the binding of FOXO1 co-activator PGC1αto the HBV promoter. 14Extracellular Polysaccharide from Rhizopus nigricans Inhibits Hepatocellular Carcinoma via miR-494-3p/TRIM36 Axis and Cyclin E Ubiquitination显示文摘Background and Aims:This study was designed to uncov-er the mechanism for extracellular polysaccharide(EPS1-1)-mediated effects on hepatocellular carcinoma(HCC)devel-opment.Methods:HCC cells were treated with EPS1-1,miR-494-3p mimic,sh-TRIM36,and pcDNA3.1-TRIM36.The levels of miR-494-3p and TRIM36 were measured in nor-mal hepatocytes,THLE-2,and HepG2 and HuH7HCC cell lines,along with the protein expression of cyclin D/E and p21.The proliferation,cell cycle,and apoptosis of HCC cells were assayed.The interactions between miR-494-3p and TRIM36,and between TRIM36 and cyclin E were assessed.Finally,the expression and localization of TRIM36 and cyclin E were monitored,and tumor apoptosis was detected,in tumor xenograft model.Results:EPS1-1 suppressed HCC cell proliferation and cyclin D/E expression and promoted apoptosis and p21 expression.miR-494-3p was upregulated and TRIM36 was downregulated in HCC cells.Transfection with miR-494-3p mimic or sh-TRIM36 facilitated HCC cell proliferation and the expression of cyclin D/E protein but they inhibited apoptosis and p21 expression in the pres-ence of EPS1-1.Overexpression of TRIM36 further con-solidated EPS1-1-mediated inhibition of HCC proliferation,cyclin D/E,and the promotion of apoptosis and p21 expres-sion.Those effects were reversed by miR-494-3p overex-pression.TRIM36 was a target gene of miR-494-3p,and TRIM36 induced cyclin E ubiquitination.EPS1-1 suppressed cyclin E expression,promoted TRIM36 expression and tu-mor apoptosis,all of which were abrogated by increasing the expression of miR-494-3p in vivo.Conclusions:EPS1-1 protected against HCC by limiting its proliferation and sur-vival through the miR-494-3p/TRIM36 axis and by inducing cyclin E ubiquitination.Haixiong Yan XiaoQian Ma Ze Mi Zhenhu He Pengfei Rong 2022Journal of Clinical and Translational Hepatology2022,10,4:2

19Salmonella-mediated blood-brain barrier penetration,tumor homing and tumor microenvironment regulation for enhanced chemo/bacterial glioma therapy显示文摘Chemotherapy is an important adjuvant treatment of glioma,while the efficacy is far from satisfactory,due not only to the biological barriers of blood-brain barrier(BBB)and blood-tumor barrier(BTB)but also to the intrinsic resistance of glioma cells via multiple survival mechanisms such as upregulation of P-glycoprotein(P-gp).To address these limitations,we report a bacteria-based drug delivery strategy for BBB/BTB transportation,glioma targeting,and chemo-sensitization.Bacteria selectively colonized into hypoxic tumor region and modulated tumor microenvironment,including macrophages repolarization and neutrophils infiltration.Specifically,tumor migration of neutrophils was employed as hitchhiking delivery of doxorubicin(DOX)-loaded bacterial outer membrane vesicles(OMVs/DOX).By virtue of the surface pathogen-associated molecular patterns derived from native bacteria,OMVs/DOX could be selectively recognized by neutrophils,thus facilitating glioma targeted delivery of drug with significantly enhanced tumor accumulation by 18-fold as compared to the classical passive targeting effect.Moreover,the P-gp expression on tumor cells was silenced by bacteria typeⅢsecretion effector to sensitize the efficacy of DOX,resulting in complete tumor eradication with 100%survival of all treated mice.In addition,the colonized bacteria were finally cleared by anti-bacterial activity of DOX to minimize the potential infection risk,and cardiotoxicity of DOX was also avoided,achieving excellent compatibility.This work provides an efficient trans-BBB/BTB drug delivery strategy via cell hitchhiking for enhanced glioma therapy.Ze Mi Qing Yao Yan Qi Jinhai Zheng Jiahao Liu Zhenguo Liu Hongpei Tan Xiaoqian Ma Wenhu Zhou Pengfei Rong 2023Acta Pharmaceutica Sinica B2023,13,2:1
