| 1 | Transcriptional analysis and histochemistry reveal that hypersensitive cell death and H2O2 have crucial roles in the resistance of tea plant (Camellia sinensis (L.) O. Kuntze) to anthracnose显示文摘Anthracnose causes severe losses of tea production in China.Although genes and biological processes involved in anthracnose resistance have been reported in other plants,the molecular response to anthracnose in tea plant is unknown.We used the susceptible tea cultivar Longjing 43 and the resistant cultivar Zhongcha 108 as materials and compared transcriptome changes in the leaves of both cultivars following Colletotrichum fructicola inoculation.In all,9015 and 8624 genes were differentially expressed between the resistant and susceptible cultivars and their controls(0 h),respectively.In both cultivars,the differentially expressed genes(DEGs)were enriched in 215 pathways,including responses to sugar metabolism,phytohormones,reactive oxygen species(ROS),biotic stimuli and signalling,transmembrane transporter activity,protease activity and signalling receptor activity,but DEG expression levels were higher in Zhongcha 108 than in Longjing 43.Moreover,functional enrichment analysis of the DEGs showed that hydrogen peroxide(H2O2)metabolism,cell death,secondary metabolism,and carbohydrate metabolism are involved in the defence of Zhongcha 108,and 88 key genes were identified.Protein–protein interaction(PPI)network demonstrated that putative mitogen-activated protein kinase(MAPK)cascades are activated by resistance(R)genes and mediate downstream defence responses.Histochemical analysis subsequently validated the strong hypersensitive response(HR)and H2O2 accumulation that occurred around the hyphal infection sites in Zhongcha 108.Overall,our results indicate that the HR and H2O2 are critical mechanisms in tea plant defence against anthracnose and may be activated by R genes via MAPK cascades. | Yuchun Wang Xinyuan Hao Qinhua Lu Lu Wang Wenjun Qian Nana Li Changqing Ding Xinchao Wang Yajun Yang | 2018 | Horticulture Research2018,5,1: | 11 |
| 2 | Dietary supplementation of Lonicera macranthoides leaf powder improves amino acid profiles in serum and longissimus thoracis muscle of growing-finishing pigs显示文摘A 60-days feeding trial was conducted to determine the effect of dietary supplementation of Lonicera macranthoides leaf(LML) on growth performance, meat quality, and free amino acid profiles in growingfinishing pigs. L. macranthoides leaves were harvested, air-dried and milled to powder. A total of 288 crossbred barrows(Large White x Landrace) with an average initial BW of 54.0 ± 3.4 kg were randomly allocated to 3 treatment groups and fed either a basal diet or a diet supplemented with 0.5% or 1.0% LML powder. Results showed that diet supplemented with LML powder did not affect average daily feed intake(ADFI) and feed: gain ratio(P> 0.05) in growing-finishing pigs. Moreover, diet supplemented with LML powder showed no significant effects on carcass traits(P > 0.05) including dressing percentage,backfat thickness and loin muscle area, and meat quality traits(P > 0.05) including meat color, intramuscular fat and cooking loss. However, diet supplemented with LML powder significantly improved free amino acid profiles in serum and longissimus thoracis muscle in growing-finishing pigs. Most importantly, dietary LML powder increased total free amino acids content(P < 0.001) and essential free amino acid content(P < 0.001) in longissimus thoracis muscle. | 3 | A MEMS Device for in-situ TEM Test of SCS Nanobeam显示文摘In-situ tensile testing in TEM (transmission electron microscopy) is a useful tool for studying mechanical properties of nano-structures because it can provide quanti-tative information on sample deformation at atomic scale. To facilitate in-situ TEM tensile tests of SCS (single crystal silicon) nanobeam, a MEMS tensile-testing chip was designed and fabricated. The chip was fabricated by means of bulk micro-machining and wafer bonding techniques. An SCS nanobeam, a comb drive ac-tuator, a force sensor beam and an electron beam window were integrated into the chip. With the on-chip comb-drive-actuator stretching the nanobeam and in-situ TEM observation, tensile test on a 90 nm-thick nanobeam was performed and the strain-stress relationship was obtained. | 4 | Circular RNAs in pulmonary hypertension:Emerging biological concepts and potential mechanism显示文摘Circular RNAs(circRNAs)are endogenous RNAs with a covalently closed single-stranded transcript.They are a novel class of genomic regulators that are linked to many important development and disease processes and are being pursued as clinical and therapeutic targets.Using the most powerful RNA sequencing and bioinformatics techniques,a large number of circRNAs have been identified and further functional studies have been performed.It is known that circRNAs act as potential biomarkers,sponges for microRNAs(miRNAs)and RNA-binding proteins(RBPs),and regulators of mRNA transcription.They also participate in the translation of peptides or proteins.Many types of circRNAs are dysregulated in plasma or lung tissues,and they may be involved in regulating the proliferation and apoptosis of pulmonary artery endothelial cells(PAECs)and pulmonary artery smooth muscle cells(PASMCs),leading to pulmonary vascular remodeling in pulmonary hypertension(PH).One possible mechanism is that circRNAs can regulate the function of PAECs and PASMCs by acting as miRNA sponge.However,other potential mechanisms of action of circRNAs are still being actively explored in PH.This paper presents a systematic review of the biogenesis,biological characterization,relevant underlying functions,and future perspectives for studies of circRNAs in the pathogenesis of PH. | Qian Wang Yuanyuan Sun Qinhua Zhao Wenhui Wu Lan Wang Yuqing Miao Ping Yuan | 2022 | Animal Models and Experimental Medicine2022,5,1: | 3 |
| 5 | Effects of catechins on litter size,reproductive performance and antioxidative status in gestating sows显示文摘This study was conducted to investigate the effects of catechins on reproductive performance, antioxidative capacity and immune function of gestating sows. A total of 60 cross-bred(Landrace × Large White) multiparious sows were blocked by body weight, parity and backfact and randomly allocated to 1 of 5 treatments: 0.100,200,300, or 400 mg/kg catechins. Dietary treatments were imposed from mating to d 40 of gestation of sows. At farrowing, litter total born, born alive, dead, and normal-(healthy piglets,>0.85 kg) and low-birth weight piglets(<0.85 kg) were recorded. Within 3.00 ± 0.50 days after farrowing litter size was standardized to 8,00 ± 1.50 piglets within treatment. The piglets were weighed at birth(d 1) and weaning(d 28). Sows serum samples were obtained from blood samples collected on d 40 of gestation for analyses of glutathione peroxidase(GSH-Px), superoxide dismutase(SOD), catalase(CAT),malondialdehyde(MDA), hydrogen peroxide(H2 O2), nitric oxide synthetase(NOS) and nitrogen monoxide(NO). Our results showed that supplementation of catechins at levels of 200 or 300 mg/kg led to improvements in litter born alive(P < 0.01) and piglet born healthy(P < 0.01) and a decrease in stillborn(P < 0.05) at farrowing when compared with the control. In comparison with the control, catechins at any supplemental levels all enhanced the serum SOD(P < 0.05) and CAT(P < 0.01) activities of sows at farrowing but no obvious differences in the serum GSH-Px and NOS activities were observed in this trial(P > 0.05). Sows received 200 mg catechin per kg diets showed a reduction(P < 0.05) of the serum MDA level at farrowing compared with all other treatments. Sows received all the levels of catechin showed a reduction(P < 0.05) of serum H2 O2 level compared with sows received the control diet on both d 40 of gestation and farrowing. | 6 | Role of miR-21-5p/FilGAP axis in estradiol alleviating the progression of monocrotaline-induced pulmonary hypertension显示文摘Background:Aberrant expression of microRNAs(miRNAs)has been associated with the pathogenesis of pulmonary hypertension(PH).It is,however,not clear whether miRNAs are involved in estrogen rescue of PH.Methods:Fresh plasma samples were prepared from 12 idiopathic pulmonary arterial hypertension(IPAH)patients and 12 healthy controls undergoing right heart cath-eterization in Shanghai Pulmonary Hospital.From each sample,5μg of total RNA was tagged and hybridized on microRNA microarray chips.Monocrotaline-induced PH(MCT-PH)male rats were treated with 17β-estradiol(E_(2))or vehicle.Subgroups were cotreated with estrogen receptor(ER)antagonist or with antagonist of miRNA.Results:Many circulating miRNAs,including miR-21-5p and miR-574-5p,were mark-edly expressed in patients and of interest in predicting mean pulmonary arterial pres-sure elevation in patients.The expression of miR-21-5p in the lungs was significantly upregulated in MCT-PH rats compared with the controls.However,miR-574-5p showed no difference in the lungs of MCT-PH rats and controls.miR-21-5p was se-lected for further analysis in rats as E_(2) strongly regulated it.E_(2) decreased miR-21-5p expression in the lungs of MCT-PH rats by ERβ.E_(2) reversed miR-21-5p target gene FilGAP downregulation in the lungs of MCT-PH rats.The abnormal expression of RhoA,ROCK2,Rac1 and c-Jun in the lungs of MCT-PH rats was inhibited by E_(2) and miR-21-5p antagonist.Conclusions:miR-21-5p level was remarkably associated with PH severity in patients.Moreover,the miR-21-5p/FilGAP signaling pathway modulated the protective effect of E_(2) on MCT-PH through ERβ. | Xiaoyi Hu Qian Wang Hui Zhao Wenhui Wu Qinhua Zhao Rong Jiang Jinming Liu Lan Wang Ping Yuan | 2022 | Animal Models and Experimental Medicine2022,5,3: | 2 |

| 12 | Quantification of anaerobic ammonium-oxidizing bacteria in enrichment cultures by quantitative competitive PCR显示文摘The anaerobic ammonium-oxidizing (ANAMMOX) bacteria were enriched from a sequencing batch biofilm reactor (SBBR). A quantitative competitive polymerase chain reaction (QC-PCR) system was successfully developed to detect and quantify ANAMMOX bacteria in environmental samples. For QC-PCR system, PCR primer sets targeting 16S ribosomal RNA genes of ANAMMOX bacteria were designed and used. The quantification range of this system was 4 orders of magnitude, from 103 to 106 copies per PCR, corresponding to the detection limit of 300 target copies per mL. A 312-bp internal standard was constructed, which showed very similar amplification efficiency with the target amxC fragment (349 bp) over 4 orders of magnitude (103-106). The linear regressions were obtained with R2 of 0.9824 for 103 copies, 0.9882 for 104 copies, 0.9857 for 105 copies and 0.9899 for 106 copies, respectively. | 19 | Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment. | 20 | Selective photocatalytic oxidation of methane to C1 oxygenates by regulating sizes and facets over Au/ZnO显示文摘Photocatalytic oxidation of methane to value-added chemicals is a promising process under mild conditions,nevertheless confronting great challenges in efficiently activating C-H bonds and inhibiting over-oxidation.Herein,we propose a comprehensive strategy for the selective generation of reactive oxygen species(ROS)by regulating the sizes and facets of Au nanoparticles loaded on ZnO.For photocatalytic methane oxidation at ambient temperature,a high oxygenates yield of 36.4 mmol·g^(-1)·h^(-1) with a nearly 100%selectivity has been achieved over the optimized 1.0%Au/ZnO-9.6(1%Au with(111)facet and 9.6 nm size on ZnO)photocatalyst,exceeding most reported literatures.Mechanism investigations reveal that 1.0%Au/ZnO-9.6 with the medium size and Au(111)facet guarantees the favourable formation of superoxide radicals(·OOH)through mild oxygen reduction,ultimately leading to excellent photocatalytic methane oxidation performance.This work provides some guidance for the delicate design of photocatalysts for efficient photocatalytic methane oxidation and oxygen utilization. | Qiang Zhou Xinyu Wang Xiaojie Tan Qinhua Zhang Hao Yang Tao Xing Mingqing Wang Mingbo Wu Wenting Wu | 2024 | Nano Research2024,17,5: | 0 |
