1Linking atmospheric pollution to cryospheric change in the Third Pole region: current progress and future prospects显示文摘The Tibetan Plateau and its surroundings are known as the Third Pole(TP). This region is noted for its high rates of glacier melt and the associated hydrological shifts that affect water supplies in Asia. Atmospheric pollutants contribute to climatic and cryospheric changes through their effects on solar radiation and the albedos of snow and ice surfaces; moreover, the behavior and fates within the cryosphere and environmental impacts of environmental pollutants are topics of increasing concern. In this review, we introduce a coordinated monitoring and research framework and network to link atmospheric pollution and cryospheric changes(APCC) within the TP region. We then provide an up-to-date summary of progress and achievements related to the APCC research framework, including aspects of atmospheric pollution’s composition and concentration, spatial and temporal variations, trans-boundary transport pathways and mechanisms, and effects on the warming of atmosphere and changing in Indian monsoon, as well as melting of glacier and snow cover. We highlight that exogenous air pollutants can enter into the TP’s environments and cause great impacts on regional climatic and environmental changes. At last, we propose future research priorities and map out an extended program at the global scale. 2The altered expression of inflammation-related microRNAs with microRNA-155 expression correlates with Th17 differentiation in patients with acute coronary syndrome显示文摘MicroRNAs(miRNAs)are a novel class of small,non-coding RNAs that play a significant role in both inflammatory and cardiovascular diseases.Immune cells,especially T helper(Th)cells,are critical in the development of atherosclerosis and the onset of acute coronary syndrome(ACS).To assess whether inflammation-related miRNAs(such as miR-155,146a,21,125a-5p,125b,31)are involved in the imbalance of Th cell subsets in patients with ACS,we measured the expression of related miRNAs in patients with acute myocardial infarction(AMI),unstable angina(UA),stable angina(SA)and chest pain syndrome(CPS);analyzed the relationship between miRNA expression and the frequency of Th cell subsets;and observed the co-expression of miR-155 and IL-17A in peripheral blood mononuclear cells(PBMCs)of patients with ACS.The results showed that the expression of miR-155 in the PBMCs of patients with ACS was decreased by approximately 60%,while the expression of both miR-21 and miR-146a was increased by approximately twofold.The expression patterns of miRNAs in plasma correlated with those in PBMCs,except for miR-21,which was increased by approximately sixfold in the AMI group and showed no significant difference between the UA group and the CPS group.We also found that the expression of miR-155 inversely correlated with the frequency of Th17 cells(r520.896,P,0.01)and that miR-155 was co-expressed with IL-17A in patients with ACS.In conclusion,our study revealed the expression patterns of inflammation-related miRNAs in patients with ACS and found that miR-155 may be associated with Th17 cell differentiation.Rui Yao Yulan Ma Youyou Du Mengyang Liao Huanhuan Li Wei Liang Jing Yuan Zhijun Ma Xian Yu Hong Xiao Yuhua Liao 2011Cellular & Molecular Immunology2011,8,6:32
3IL-17 sustains the plasma cell response via p38-mediated Bcl-xL RNA stability in lupus pathogenesis显示文摘Recent studies have demonstrated a central role for plasma cells in the development of autoimmune diseases,such as systemic lupus erythematosus(SLE).Currently,both the phenotypic features and functional regulation of autoreactive plasma cells during SLE pathogenesis remain largely unclear.In this study,we first found that a major subset of IL-17 receptor-expressing plasma cells potently produced anti-dsDNA IgG upon IL-17A(IL-17)stimulation in SLE patients and lupus mice.Using a humanized lupus mouse model,we showed that the transfer of Th17 cell-depleted PBMCs from lupus patients resulted in a significantly reduced plasma cell response and attenuated renal damage in recipient mice compared to the transfer of total SLE PBMCs.Moreover,long-term BrdU incorporation in lupus mice detected highly enriched long-lived BrdU+subsets among IL-17 receptor-expressing plasma cells.Lupus mice deficient in IL-17 or IL-17 receptor C(IL-17RC)exhibited a diminished plasma cell response and reduced autoantibody production with attenuated renal damage,while the adoptive transfer of Th17 cells triggered the plasma cell response and renal damage in IL-17-deficient lupus mice.In reconstituted chimeric mice,IL-17RC deficiency resulted in severely impaired plasma cell generation but showed no obvious effect on germinal center B cells.Further mechanistic studies revealed that IL-17 significantly promoted plasma cell survival via p38-mediated Bcl-xL transcript stabilization.Together,our findings identified a novel function of IL-17 in enhancing plasma cell survival for autoantibody production in lupus pathogenesis,which may provide new therapeutic strategies for the treatment of SLE.Kongyang Ma Wenhan Du Fan Xiao Man Han Enyu Huang Na Peng Yuan Tang Chong Deng Lixiong Liu Yulan Chen Jingjing Li Shiwen Yuan Qin Huang Xiaoping Hong Dajun Hu Xiaoyan Cai Quan Jiang Dongzhou Liu Liwei Lu 2021Cellular & Molecular Immunology2021,18,7:6
4Clinical and pathological features of miR-10b and RHOC gene expression in hepatocellular carcinoma显示文摘Aberrant expression of microRNAs(miRNAs)was reported frequently in different human cancers.The major role of miRNA is targeting 30-UTR of coding gene and causing translational repression or mRNA degradation.miR-10b overexpression was reported to promote breast cancer metastasis by up-regulating RHOC expression.But its expression in hepatocellular carcinoma(HCC)remains unclear.Our study indicated that the expression of miR-10b was different in HCC and adjacent tissue samples,and reduced expression of miR-10b in HCC was related tovein invasion.High-level expression of RHOC was also related to vein invasion in HCC.But no correlation was found between miR-10b and RHOC expression.These results suggest that miR-10b and RHOC are independent predictors of HCC invasion and metastasis.Jingli Du Yulan Wang Hanjiang Fu Xiaofei Zheng Minggui Lin Mingzhou Guo Lixin Wei 2014Chinese Science Bulletin2014,59,19:4
5First results on ^(76)Ge neutrinoless double beta decay from CDEX-1 experiment显示文摘We report the first results on ^(76)Ge neutrinoless double beta decay from stage one of the China dark-matter experiment(CDEX).A p-type point-contact high-purity germanium detector with a mass of 994 g has been installed to detect neutrinoless double beta decay events, as well as to directly detect dark matter particles. An exposure of 304 kg d has been analyzed over a wide spectral band from 500 keV to 3 MeV. The average event rate obtained was about 0.012 counts per keV per kg per day over the 2.039 MeV energy range. The half-life of ^(76)Ge neutrinoless double beta decay derived based on this result is T^(0ν)1/2>6.4×10^(22) yr(90% C.L.). 7A light-addressable microfluidic device for label-free functional assays of bioengineered taste receptor cells via extracellular recording显示文摘The functional assay of chemical sensitive cells is of great importance for the preparation of sensitive elements towards the development of cell-based biosensors for chemical sensing. In this study, a novel light-addressable microfluidic device was developed by the integration of a light-addressable potentiometric sensor (LAPS) with a microfluidic chip for label-free functional assays of bioengineered taste receptor cells via extracellular recording. Extracellular potential changes of single bioengineered cells were recorded by LAPS. Microfluidic chip was capable of providing stable microenvironments for cell measurements w让h a well-defined concentration stimulus. Bioengineered taste receptor cells were utilized as a model of chemical sensitive cells and functional assayed by this light-addressable microfluidic device using bitter stimulation. The results indicate that this microfluidic device can efficiently monitor the membrane potential changes originated from bioengineered taste receptor cells in response to specific bitter stimulation. The bioengineered cells with responsive functions can be easily identified via the analysis on the firing rates of extracellular recording data. The expression of bitter receptors in the bioengineered taste receptor cells was further validated by the immunofluorescent staining results, which proved that the expression of specific bitter receptor was the main contribution to the responsive function of bioengineered cells. 8Cryosphere as a temporal sink and source of microplastics in the Arcticregion显示文摘Microplastics(MPs)pollution has become a serious environmental issue of growing global concern due to the increasing plastic production and usage.Under climate warming,the cryosphere,defined as the part of Earth’s layer characterized by the low temperatures and the presence of frozen water,has been experiencing significant changes.The Arctic cryosphere(e.g.,sea ice,snow cover,Greenland ice sheet,permafrost)can store and release pollutants into environments,making Arctic an important temporal sink and source of MPs.Here,we summarized the distributions of MPs in Arctic snow,sea ice,seawater,rivers,and sediments,to illustrate their potential sources,transport pathways,storage and release,and possible effects in this sentinel region.Items concentrations of MPs in snow and ice varied about 1-6 orders of magnitude in different regions,which were mostly attributed to the different sampling and measurement methods,and potential sources of MPs.MPs concentrations from Arctic seawater,river/lake water,and sediments also fluctuated largely,ranging from several items of per unit to>40,000 items m^(-3),100 items m^(-3),and 10,000 items kg^(-1) dw,respectively.Arctic land snow cover can be a temporal storage of MPs,with MPs deposition flux of about(4.9-14.26)×10^(8) items km^(-2) yr^(-1).MPs transported by rivers to Arctic ocean was estimated to be approximately 8-48 ton/yr,with discharge flux of MPs at about(1.65-9.35)×10^(8) items/s.Average storage of MPs in sea ice was estimated to be about 6.1×10^(18) items,with annual release of about 5.1×10^(18) items.Atmospheric transport of MPs from long-distance terrestrial sources contributed significantly to MPs deposition in Arctic land snow cover,sea ice and oceanic surface waters.Arctic Great Rivers can flow MPs into the Arctic Ocean.Sea ice can temporally store,transport and then release MPs in the surrounded environment.Ocean currents from the Atlantic brought high concentrations of MPs into the Arctic.However,there existed large uncertainties of estimation on the storage and release of MPs in Arctic cryosphere owing to the hypothesis of average MPs concentrations.Meanwhile,representatives of MPs data across the large Arctic region should be mutually verified with in situ observations and modeling.Therefore,we suggested that systematic monitoring MPs in the Arctic cryosphere,potential threats on Arctic ecosystems,and the carbon cycle under increasing Arctic warming,are urgently needed to be studied in future.Yulan Zhang Tanguang Gao Shichang Kang Deonie Allen Zhaoqing Wang Xi Luo Ling Yang Jinlei Chen Zhaofu Hu Pengfei Chen Wentao Du Steve Allen 2023Geoscience Frontiers2023,14,4:1
9SARS-CoV-2 immunity and functional recovery of COVID-19 patients 1-year after infection显示文摘The long-term immunity and functional recovery after SARS-CoV-2 infection have implications in preventive measures and patient quality of life.Here we analyzed a prospective cohort of 121 recovered COVID-19 patients from Xiangyang,China at 1-year after diagnosis.Among them,chemiluminescence immunoassay-based screening showed 99%(95%CI,98–100%)seroprevalence 10–12 months after infection,comparing to 0.8%(95%CI,0.7–0.9%)in the general population.Total anti-receptor-binding domain(RBD)antibodies remained stable since discharge,while anti-RBD IgG and neutralization levels decreased over time.A predictive model estimates 17%(95%CI,11–24%)and 87%(95%CI,80–92%)participants were still 50%protected against detectable and severe re-infection of WT SARS-CoV-2,respectively,while neutralization levels against B.1.1.7 and B.1.351 variants were significantly reduced.All non-severe patients showed normal chest CT and 21%reported COVID-19-related symptoms.In contrast,53%severe patients had abnormal chest CT,decreased pulmonary function or cardiac involvement and 79%were still symptomatic.Our findings suggest long-lasting immune protection after SARS-CoV-2 infection,while also highlight the risk of immune evasive variants and long-term consequences for COVID-19 survivors.Yan Zhan Yufang Zhu Shanshan Wang Shijun Jia Yunling Gao Yingying Lu Caili Zhou Ran Liang Dingwen Sun Xiaobo Wang Zhibing Hou Qiaoqiao Hu Peng Du Hao Yu Chang Liu Miao Cui Gangling Tong Zhihua Zheng Yunsheng Xu Linyu Zhu Jin Cheng Feng Wu Yulan Zheng Peijun Liu Peng Hong 2021Signal Transduction and Targeted Therapy2021,6,11:0

11Performances of a prototype point-contact germanium detector immersed in liquid nitrogen for light dark matter search显示文摘The CDEX-10 experiment searches for light weakly interacting massive particles, a form of dark matter, at the China Jinping Underground Laboratory, where approximately 10 kg of germanium detectors are arranged in an array and immersed in liquid nitrogen. Herein, we report on the experimental apparatus, detector characterization, and spectrum analysis of one prototype detector. Owing to the higher rise-time resolution of the CDEX-10 prototype detector as compared with CDEX-1 B, we identified the origin of an observed category of extremely fast events. For data analysis of the CDEX-10 prototype detector, we introduced and applied an improved bulk/surface event discrimination method. The results of the new method were compared to those of the CDEX-1 B spectrum.
