2Usefulness of human epididymis protein 4 in predicting cytoreductive surgical outcomes for advanced ovarian tubal and peritoneal carcinoma显示文摘Objective: Human epididymis protein 4(HE4) is a promising biomarker of epithelial ovarian cancer(EOC). But its role in assessing the primary optimal debulking(OD) of EOC remains unknown. The purpose of this study is to elucidate the ability of preoperative HE4 in predicting the primary cytoreductive outcomes in advanced EOC, tubal or peritoneal carcinoma.Methods: We reviewed the records of 90 patients with advanced ovarian, tubal or peritoneal carcinoma who underwent primary cytoreduction at the Department of Obstetrics and Gynecology of Peking University People's Hospital between November 2005 and October 2010. Preoperative serum HE4 and CA125 levels were detected with EIA kit. A receiver operating characteristic(ROC) curve was used to determine the most useful HE4 cut-off value. Logistic regression analysis was performed to identify significant preoperative clinical characteristics to predict optimal primary cytoreduction.Results: OD was achieved in 47.7%(43/48) of patients. The median preoperative HE4 level for patients with OD vs. suboptimal debulking was 423 and 820 pmol/L, respectively(P<0.001). The areas under the ROC curve for HE4 and CA125 were 0.716 and 0.599, respectively(P=0.080). The most useful HE4 cut-off value was 473 pmol/L. Suboptimal cytoreduction was obtained in 66.7%(38/57) of cases with HE4 ≥473 pmol/L compared with only 27.3%(9/33) of cases with HE4 <473 pmol/L. At this threshold, the sensitivity, specificity, positive predictive value(PPV) and negative predictive value(NPV) for diagnosing suboptimal debulking were 81%, 56%, 67%, and 73%, respectively. Logistic regression analysis showed that the patients with HE4 ≥473 pmol/L were less likely to achieve OD(odds ratio =5.044, P=0.002).Conclusions: Preoperative serum HE4 may be helpful to predict whether optimal cytoreductive surgery could be obtained or whether extended cytoreduction would be needed by an interdisciplinary team.Zhijian Tang Xiaohong Chang Xue Ye Yi Li Hongyan Cheng Heng Cui 2015Chinese Journal of Cancer Research2015,27,3:10
3Clinical features of seizures after cerebral venous sinus thrombosis and its effect on outcome among Chinese Han population显示文摘background and purpose Clinical features of epileptic seizures after cerebral venous sinus thrombosis(CVST)among Chinese patients are not known,and it is still controversial whether seizures would affect the outcome of CVST.Methods In a Chinese hospital-based study of consecutive patients with CVST between 2003 and 2015,we described the clinical features of seizures and determined the predictors of seizure onset using multivariable logistic regression analysis.We also compared the in-hospital case-fatality and short-term functional outcome(modified Rankin Scale(mRS)at discharge)in patients with versus without seizures using ordinal regression analysis.results Among 151 patients with CVST,52(34.4%)presented seizures,of which 42(80.8%)were generalised seizures.Male gender(OR 6.32,95% CI 2.06 to 19.35,p=0.001),motor deficits(OR 4.89,95% CI 1.52 to 15.68,p=0.008),intracerebral haemorrhage(OR 3.93,95% CI 1.16 to 13.26,p=0.027),cerebral infarction(OR 3.78,95% CI 1.15 to 12.36,p=0.029)and superior sagittal sinus thrombosis(OR 3.38,95% CI 91.16 to 9.86,p=0.026)were independent predictors for seizures.The overall in-hospital case-fatality rate was 2.0%(3/151),and 21(13.9%)had mRS>2 at discharge.Compared with patients without seizures,patients with seizures were more likely to have a worse outcome(p=0.02)at discharge,independent of age,gender,clinical presentation,clot burden and presence of parenchymal lesions.conclusions In Chinese Han patients,compared with patients without seizures,patients with seizures after CVST had a worse outcome.Risk factors such as male gender,paresis,parenchymal lesion and superior sagittal sinus thrombosis were independently associated with seizure onset after CVST.Generalised seizure was the main form of seizures after CVST,which was obviously different to seizures after strokes of arterial origin.Hongyan Ding Yanan Xie Linxin Li Heling Chu Yuping Tang Qiang Dong Mei Cui 2017Stroke & Vascular Neurology2017,2,4:8

7FOUR-PARAMETER AUTOMATIC TRANSMISSION TECHNOLOGY FOR CONSTRUCTION VEHICLE BASED ON ELMAN RECURSIVE NEURAL NETWORK显示文摘From the viewpoint of energy saving and improving transmission efficiency,the ZL50E wheel loader is taken as the study object. And the system model is analyzed based on the transmission system of the construction vehicle. A new four-parameter shift schedule is presented,which can keep the torque converter working in the high efficiency area. The control algorithm based on the Elman recursive neural network is applied,and four-parameter control system is developed which is based on industrial computer. The system is used to collect data accurately and control 4D180 power-shift gearbox of ZL50E wheel loader shift timely. An experiment is done on automatic transmission test-bed,and the result indicates that the control system could reliably and safely work and improve the efficiency of hydraulic torque converter. 9In vitro and in vivo degradation behavior of Mg-2Sr-Ca and Mg-2Sr-Zn alloys显示文摘Magnesium alloys with integration of degradability and good mechanical performance are desired for orthopedic implants.In this paper,Mg-2Sr-Ca and Mg-2Sr-Zn alloys were prepared and the degradation as well as the bone response were investigated.Compared with the binary Mg-2Sr alloys,the addition of Ca and Zn improved the in vitro and in vivo corrosion resistance.Mg-2Sr-Ca and Mg-2Sr-Zn alloys exhibited more uniform corrosion and maintained the configuration of the implants 4 weeks post-implantation.The in vivo corrosion rates were 0.85 mm/yr for Mg-2Sr-Zn and 1.10 mm/yr for Mg-2Sr-Ca in comparison with 1.37 mm/yr for Mg-2Sr.The in vitro cell tests indicated that Mg-2Sr-Ca and Mg-2Sr-Zn alloys exhibited higher MG63 cell viability than Mg-2Sr alloy.Furthermore,these two alloys can promote the mineralization and new bone formation without inducing any significant adverse effects and this sound osteogenic properties suggest its attractive clinical potential.Kai Chen Xinhui Xie Hongyan Tang Hui Sun Ling Qin Yufeng Zheng Xuenan Gu Yubo Fan 2020Bioactive Materials2020,5,2:6

11Profile of the culturable microbiome capable of producing acyl-homoserine lactone in the tobacco phyllosphere显示文摘Bacterial populations coexisting in the phyllosphere niche have important effects on plant health. Quorum sensing (QS) allows bacteria to communicate via diffusible signal molecules, but QS-dependent behaviors in phyllosphere bacterial populations are poorly understood. We investigate the dense and diverse N-acyl-homoserine lactone (AHL)-producing phyllosphere bacteria living on tobacco leaf surfaces via a culture-dependent method and 16S rRNA gene sequencing. Our results indicated that approximately 7.9%-11.7% of the culturable leaf-associated bacteria have the ability to produce AHL based on the assays using whole-cell biosensors. Sequencing of the 16S rRNA gene assigned the AHL-producing strains to two phylogenetic groups, with Gammaproteobacteria (93%) as the predominant group, followed by Alphaproteobacteria. All of the AHL-producing Alphaproteobacteria were affiliated with the genus Rhizobium, whereas the AHL-producing bacteria belonging to the Gammaproteobacteria mainly fell within the genera Pseudomonas, Acinetobacter, Citrobacter, Enterobacter, Pantoea and Serratia. The bioassays of supernatant extracts revealed that a portion of the strains have a remarkable AHL profile for AHL induction activity using the two different biosensors, and one compound in the active extract of a representative isolate, NTL223, corresponded to 3-oxo-hexanoyl-homoserine lactone. A large population size and diversity of bacteria capable of AHL-driven QS were found to cohabit on leaves, implying that cross-communication based AHL-type QS may be common in the phyllosphere. 12Combined detection and subclass characteristics analysis of CTCs and CTECs by SE-iFISH in ovarian cancer显示文摘Objective:Hematogenous metastasis is essential for the progression of ovarian cancer(OC),and circulating tumor cells(CTCs)are part of the metastatic cascade.However,the detection rate of CTC is low due to the use of less sensitive detection methods.Therefore,this study aimed to detect CTCs and circulating tumorigenic endothelial cells(CTECs)in patients with OC using subtraction enrichment and immunostaining and fluorescence in situ hybridization(SE-iFISH).Methods:We enrolled a total of 56 subjects,including 20 OC patients and 36 ovarian benign tumor patients.CTCs and CTECs were captured by subtraction enrichment(SE)and counted and classified according to immunofluorescence staining of tumor markers(TMs)carbohydrate antigen 125(CA125)and human epididymis protein 4(HE4)combined with fluorescence in situ hybridization(iFISH)of chromosome 8(Chr8)aneuploidy.The diagnostic value and subtype characteristics of CTCs and CTECs were investigated.Results:The detection rate of CTCs by SE-iFISH was high.Compared with CA125 and HE4,Chr8 aneuploidy was the major identification feature of CTC.CTC counts in OC were statistically higher than those in benign groups.CTC and CTEC with≥pentaploidy were detected in both groups,illustrating the poor diagnostic value of CTC or CTEC.Distributions of triploid and tetraploid CTC subtypes were significantly different,and combined detection of triploid and tetraploid CTCs showed the best diagnostic value.In contrast,the distribution of CTECs in the OC and benign groups had no statistically significant difference.Small CTCs accounted for over 1/3 of the total CTC count.We also found that small CTCs and CTECs primarily comprised triploid cells,while large CTCs and CTECs mainly comprised pentaploidy and beyond.Conclusions:The application of SE-iFISH offered a more comprehensive understanding of heterogeneous CTCs and CTECs in OC.Analysis of subclass characteristics of the CTCs and CTECs according to Chr8 aneuploidy and cell size may broaden their potential clinical utility and deepen mechanistic studies in OC.Hongyan Cheng Shang Wang Wenqing Luan Xue Ye Sha Dou Zhijian Tang Honglan Zhu Peter Ping Lin Yi Li Heng Cui Xiaohong Chang 2021Chinese Journal of Cancer Research2021,33,2:3
13Upwelling of Mantle-derived Material in Southeast China:Evidence from Noble Gas Isotopes显示文摘Shallow groundwater collected in Chaozhou,Huizhou,and Guangzhou allowed testing of concentrations and the isotope ratios of noble gases.Based on the calculated noble gas temperature(NGT)and the ratio of noble gas isotopes,the recharge temperature,recharge source,and residence time of groundwater can be calculated.In addition,the contribution of noble gas components from different sources to the sample components can be assessed.In the Huizhou area,according to the 1/Xe vs.Ne/Xe and NGT data,the shallow sandstone-confined water samples in the Shiba area and the unconfined water samples of the Huangshadong are in different temperature ranges,indicating that they have different recharge sources,both in time or space.The He components in the samples are calculated to obtain the content of radiogenic ^(4)He in the crust and to simulate the groundwater ages.The noble gas isotope ratios show the addition of mantle components into the basalt aquifers and sandstone aquifers in Chaozhou and Huizhou.Except for atmospheric and crustal sources,there is a certain proportion of mantle-derived components in the shallow underground cold water in Huizhou and Chaozhou.The noble gases in the Chaozhou groundwater have an obvious mantle signature,allowing speculation that there is a deep fluid carrying mantle characteristics.This upwelling of mantle-derived material might be caused by the India-Eurasia collision or that between the Philippine Sea Plate and the Eurasian Plate.WANG Shuai KUANG Jian HUANG Xuelian ZHANG Hongyan ZHANG Min QI Shihua HAN Yongjie XIAO Zhicai WANG Siqi TANG Ling 2022Acta Geologica Sinica(English Edition)2022,96,1:2
14Identification and characterization of the cellular subclones that contribute to the pathogenesis of mantle cell lymphoma显示文摘Mantle cell lymphoma(MCL)is a B-cell malignancy with poor clinical outcome and undefined pathogenesis.Development of clinically relevant cellular models for MCL research is an urgent need.Our preliminary observations lead the development of two novel hypotheses that we tested in this study:1.multicellular spheroid might be a unique growth mode of earlystage cells in MCL;2.MCL might be a polyclonal tumor.We made the following original observations that have not been reported:First,we have provided a new experiment method for enriching MCL early-stage cells and characterized the spheroid mode of growth as a unique feature of early-stage MCL cells in cell line as well as in clinical samples.Second,we have established a clinically relevant cellular model of MCL,the JeKo-1-spheroid cell line,that was highly enriched in early-stage sub-clones.JeKo-1-spheroid cells and the spheroid growing cells enriched from MCL patients exhibited comparably enhanced tumorigenic abilities and similar biological features.Third,Immunophenotypic analysis has revealed that MCL may be derived from precursor-B(pre-B),immature-B and mature-B cells,not only the mature-B cells as WHO classified in 2016.Fourth,MCL may be a polyclonal disease composed of CD19e/IgMe,CD19 e/IgMt,CD19t/IgMt three sub-clones,of which the CD19e/IgMt sub-clone might be the dominant sub-clone with the strongest tumorigenic ability.Fifth,CD19t/IgMe that differentiates MCL and normal B cells may represent a new marker for MCL early detection,minor residual disease monitoring after therapies and prognosis.Junling Tang Li Zhang Tiejun Zhou Zhiwei Sun Liangsheng Kong Li Jing Hongyun Xing Hongyan Wu Yongli Liu Shixia Zhou Jingyuan Li Mei Chen Fang Xu Jirui Tang Tao Ma Min Hu Dan Liu Jing Guo Xiaofeng Zhu Yan Chen Ting Ye Jianyu Wang Xiaoming Li H.Rosie Xing 2019Genes & Diseases2019,6,4:2
15Depression and stress levels increase risk of liver cancer through epigenetic downregulation of hypocretin显示文摘Recent studies suggest that Hypocretin (HCRT, Orexin) are involved in stress regulation of depression through the hypothalamic-pituitary-adrenal (HPA) axis. However, the molecular mechanism by which Hypocretin regulate neurobiological responses is unknown. Herein, the effects of chronic stress on the epigenetic modification of HCRT and its association with depression were explored with regard to a potential role in cancer progression. In the study, Sprague Dawley (SD) rats were used to establish an animal model of cancer with depression by administrating n-nitrosodiethylamine (DEN) and chronic unpredictable mild stress (CUMS). RNA-sequencing was used to detect differentially expressed genes in the hippocampus of rats and quantitative real-time polymerase chain reaction (qRT-PCR) was used to validate the results of RNA-sequencing. The status of HCRT promoter methylation was assessed by methylation specific polymerase chain reaction. Behavioral tests showed that rats exposed to CUMS had significant depressive-like behaviors. The number of liver tumors and tumor load in depressed rats exposed to CUMS was higher than in SD rats without CUMS. RNA-sequencing revealed that HCRT was one of the most siginificantly downregulated gene in the hippocampus of SD rats with CUMS compared to non-stressed group, which was validated by qRT-PCR. HCRT mRNA expression was downregulated and the promoter for HCRT was hyper-methylated in those with depression. These results identified a critical role for chronic psychological stressors in tumorigenesis and cancer progression, via epigenetic HCRT downregulation. 19A surface-eroding poly(1,3-trimethylene carbonate)coating for magnesium based cardiovascular stents with stable drug release and improved corrosion resistance显示文摘Magnesium alloys with integration of degradability and good mechanical performance are desired for vascular stent application.Drug-eluting coatings may optimize the corrosion profiles of magnesium substrate and reduce the incidence of restenosis simultaneously.In this paper,poly(trimethylene carbonate)(PTMC)with different molecular weight(50,000 g/mol named as PTMC5 and 350,000 g/mol named as PTMC35)was applied as drug-eluting coatings on magnesium alloys.A conventional antiproliferative drug,paclitaxel(PTX),was incorporated in the PTMC coating.The adhesive strength,corrosion behavior,drug release and biocompatibility were investigated.Compared with the PLGA control group,PTMC coating was uniform and gradually degraded from surface to inside,which could provide long-term protection for the magnesium substrate.PTMC35 coated samples exhibited much slower corrosion rate 0.05μA/cm^(2)in comparison with 0.11μA/cm^(2)and 0.13μA/cm^(2)for PLGA and PTMC5 coated counterparts.In addition,PTMC35 coating showed more stable and sustained drug release ability and effectively inhibited the proliferation of human umbilical vein vascular smooth muscle cells.Hemocompatibility test indicated that few platelets were adhered on PTMC5 and PTMC35 coatings.PTMC35 coating,exhibiting surface erosion behavior,stable drug release and good biocompatibility,could be a good candidate as a drug-eluting coating for magnesium-based stent.Hongyan Tang Shuangshuang Li Yuan Zhao Cunli Liu Xuenan Gu Yubo Fan 2022Bioactive Materials2022,7,1:1
