4Baicalin induces ferroptosis in bladder cancer cells by downregulating FTH1显示文摘Ferroptosis is a non-apoptotic regulated cell death caused by iron accumulation and subsequent lipid peroxidation.Currently,the therapeutic role of ferroptosis on cancer is gaining increasing interest.Baicalin an active component in Scutellaria baicalensis Georgi with anticancer potential various cancer types;however,the effects of baicalein on bladder cancer and the underlying molecular mechanisms remain largely unknown.In the study,we investigated the effect of baicalin on bladder cancer cells5637 and KU-19-19.As a result,we show baicalin exerted its anticancer activity by inducing apoptosis and cell death in bladder cancer cells.Subsequently,we for the first time demonstrate baicalin-induced ferroptotic cell death in vitro and in vivo,accompanied by reactive oxygen species(ROS) accumulation and intracellular chelate iron enrichment.The ferroptosis inhibitor deferoxamine but not necrostatin-1,chloroquine(CQ),N-acetyl-L-cysteine,L-glutathione reduced,or carbobenzoxy-valyl-alanyl-aspartyl-[O-methyl]-fluoromethylketone(Z-VAD-FMK) rescued baicalin-induced cell death,indicating ferroptosis contributed to baicalin-induced cell death.Mechanistically,we show that ferritin heavy chain1(FTH1) was a key determinant for baicalin-induced ferroptosis.Overexpression of FTH1 abrogated the anticancer effects of baicalin in both 5637 and KU19-19 cells.Taken together,our data for the first time suggest that the natural product baicalin exerts its anticancer activity by inducing FTH1-dependent ferroptosis,which will hopefully provide a prospective compound for bladder cancer treatment.Na Kong Xiaying Chen Jiao Feng Ting Duan Shuiping Liu Xueni Sun Peng Chen Ting Pan Lili Yan Ting Jin Yu Xiang Quan Gao Chengyong Wen Weirui Ma Wencheng Liu Mingming Zhang Zuyi Yang Wengang Wang Ruonan Zhang Bi Chen Tian Xie Xinbing Sui Wei Tao 2021Acta Pharmaceutica Sinica B2021,11,12:23
5Characterization of drug responses of mini patient-derived xenografts in mice for predicting cancer patient clinical therapeutic response显示文摘Background:Patient-derived organoids and xenografts(PDXs)have emerged as powerful models in functional diag-nostics with high predictive power for anticancer drug response.However,limitations such as engraftment failure and time-consuming for establishing and expanding PDX models followed by testing drug efficacy,and inability to subject to systemic drug administration for ex vivo organoid culture hinder realistic and fast decision-making in selecting the right therapeutics in the clinic.The present study aimed to develop an advanced PDX model,namely MiniPDX,for rapidly testing drug efficacy to strengthen its value in personalized cancer treatment.Methods:We developed a rapid in vivo drug sensitivity assay,OncoVee®MiniPDX,for screening clinically relevant regimens for cancer.In this model,patient-derived tumor cells were arrayed within hollow fiber capsules,implanted subcutaneously into mice and cultured for 7 days.The cellular activity morphology and pharmacokinetics were systematically evaluated.MiniPDX performance(sensitivity,specificity,positive and negative predictive values)was examined using PDX as the reference.Drug responses were examined by tumor cell growth inhibition rate and tumor growth inhibition rate in PDX models and MiniPDX assays respectively.The results from MiniPDX were also used to evaluate its predictive power for clinical outcomes.Results:Morphological and histopathological features of tumor cells within the MiniPDX capsules matched those both in PDX models and in original tumors.Drug responses in the PDX tumor graft assays correlated well with those in the corresponding MiniPDX assays using 26 PDX models generated from patients,including 14 gastric cancer,10 lung cancer and 2 pancreatic cancer.The positive predictive value of MiniPDX was 92%,and the negative predictive value was 81%with a sensitivity of 80%and a specificity of 93%.Through expanding to clinical tumor samples,Min-iPDX assay showed potential of wide clinical application.Conclusions:Fast in vivo MiniPDX assay based on capsule implantation was developed-to assess drug responses of both PDX tumor grafts and clinical cancer specimens.The high correlation between drug responses of paired MiniPDX and PDX tumor graft assay,as well as translational data suggest that MiniPDX assay is an advanced tool for personalized cancer treatment.Feifei Zhang Wenjie Wang Yuan Long Hui Liu Jijun Cheng Lin Guo Rongyu Li Chao Meng Shan Yu Qingchuan Zhao Shun Lu Lili Wang Haitao Wang Danyi Wen 2018Cancer Communications2018,38,1:20
6Simulated multi-scale watershed runoff and sediment production based on GeoWEPP model显示文摘The runoff and sediment yield data from the Qiaozidonggou, Qiaozixigou, and Lu'ergou watersheds, in the Loess Plateau of China are used to calibrate and validate the runoff and sediment yield simulated by GeoWEPP model of the WEPP Model at watershed scale. The indices of relative error, R, correlation coefficient, R 2 , and Nash-Suttcliffe efficiency coefficient En s are used to evaluate the model fit. The eco-hydrological responses in the Luoyugou and Lu'ergou watersheds are also forecast based on the WEPP Model. Meanwhile, the relation between vegetation pattern changes and sediment yield in the watershed is discussed, and the responses of runoff and sediment yield in the watersheds concerning forest growth stages are studied. The results show that the relative errors of simulated values of runoff and sediment yield are below 30%, the correlation coefficients are above 0.90, and the Nash-Suttcliffe efficiency coefficients are above 0.80. The simulation results present satisfactory performance, thus, the model could be used to simulate the runoff and sediment yield in these small watersheds. It is also observed that soil erosion tended to become severe as precipitation increased in the watershed, while soil erosion has a decreasing trend as forest cover increases and vegetation pattern is optimized. When the watershed is fully covered by forest, erosion and sediment yield are minimized. When the forest cover is about 30% and evenly distributed in the watershed, the erosion intensity is lower than if the forest cover is collectively distributed in the watershed. 7A Chromosome-Scale Genome Assembly of Paper Mulberry(Bmussonetia papyrifera)Provides New Insights into Its Forage and Papermaking Usage显示文摘Paper mulberry(Broussonetia papyrifera)is a well-known woody tree historically used for Cai Lun papermaking,one of the four great inventions of ancient China.More recently,Paper mulberry has also been used as forage to address the shortage of feedstuff because of its digestible crude fiber and high protein contents.In this study,we obtained a chromosome-scale genome assembly for Paper mulberry using integrated approaches,including Illumina and PacBio sequencing platform as well as Hi-C,optical,and genetic maps.The assembled Paper mulberry genome consists of 386.83 Mb,which is close to the estimated size,and 99.25%(383.93 Mb)of the assembly was assigned to 13 pseudochromosomes.Comparative genomic analysis revealed the expansion and contraction in the flavonoid and lignin biosynthetic gene families,respectively,accounting for the enhanced flavonoid and decreased lignin biosynthesis in Paper mulberry.Moreover,the increased ratio of syringyl-lignin to guaiacyl-lignin in Paper mulberry underscores its suitability for use in medicine,forage,papermaking,and barkcloth making.We also identified the rootassociated microbiota of Paper mulberry and found that Pseudomonas and Rhizobia were enriched in its roots and may provide the source of nitrogen for its stems and leaves via symbiotic nitrogen fixation.Collectively,these results suggest that Paper mulberry might have undergone adaptive evolution and recruited nitrogen-fixing microbes to promote growth by enhancing flavonoid production and altering lignin monomer composition.Our study provides significant insights into genetic basis of the usefulness of Paper mulberry in papermaking and barkcloth making,and as forage.These insights will facilitate further domestication and selection as well as industrial utilization of Paper mulberry worldwide.Xianjun Peng Hui Liu Peilin Chen Feng Tang Yanmin Hu Fenfen Wang Zhi Pi Meiling Zhao Naizhi Chen Hui Chen Xiaokang Zhang Xueqing Yan Min Liu Xiaojun Fu Guofeng Zhao Pu Yao Lili Wang He Dai Xuming Li Wei Xiong Wencai Xu Hongkun Zheng Haiyan Yu Shihua Shen 2019Molecular Plant2019,12,5:18

9Evolution of boundary layer ozone in Shijiazhuang,a suburban site on the North China Plain显示文摘The structure of the boundary layer affects the evolution of ozone(O3), and research into this structure will provide important insights for understanding photochemical pollution.In this study, we conducted a one-month observation(from June 15 to July 14, 2016) of the boundary layer meteorological factors as well as O3 and its precursors in Luancheng County,Shijiazhuang(37°53′N, 114°38′E). Our research showed that photochemical pollution in Shijiazhuang is serious, and the mean hourly maximum and mean 8-hr maximum O3 concentrations are 97.9 ± 26.1 and 84.4 ± 22.4 ppbV, respectively. Meteorological factors play a significant role in the formation of O3. High temperatures and southeasterly winds lead to elevated O3 values, and at moderate relative humidity(40%–50%) and medium boundary layer heights(1200–1500 m), O3 production sensitivity occurred in the transitional region between volatile organic compounds(VOC) and nitrogen oxides(NOx) limitations,and the O3 concentration was the highest. The vertical profiles of O3 were also measured by a tethered balloon. The results showed that a large amount of O3 was stored in the residual layer, and the concentration was positively correlated with the O3 concentration measured the previous day. 10Biofunctional magnesium coated Ti6Al4V scaffold enhances osteogenesis and angiogenesis in vitro and in vivo for orthopedic application显示文摘The insufficient osteogenesis and osseointegration of porous titanium based scaffold limit its further application.Early angiogenesis is important for scaffold survival.It is necessary to develop a multifunctional surface on titanium scaffold with both osteogenic and angiogenic properties.In this study,a biofunctional magnesium coating is deposited on porous Ti6Al4V scaffold.For osseointegration and osteogenesis analysis,in vitro studies reveal that magnesium-coated Ti6Al4V co-culture with MC3T3-E1 cells can improve cell proliferation,adhesion,extracellular matrix(ECM)mineralization and ALP activity compared with bare Ti6Al4V cocultivation.Additionally,MC3T3-E1 cells cultured with magnesium-coated Ti6Al4V show significantly higher osteogenesisrelated genes expression.In vivo studies including fluorochrome labeling,micro-computerized tomography and histological examination of magnesium-coated Ti6Al4V scaffold reveal that new bone regeneration is significantly increased in rabbits after implantation.For angiogenesis studies,magnesium-coated Ti6Al4V improve HUVECs proliferation,adhesion,tube formation,wound-healing and Transwell abilities.HUVECs cultured with magnesium-coated Ti6Al4V display significantly higher angiogenesis-related genes(HIF-1αand VEGF)expression.Microangiography analysis reveal that magnesium-coated Ti6Al4V scaffold can significantly enhance the blood vessel formation.This study enlarges the application scope of magnesium and provides an optional choice to the conventional porous Ti6Al4V scaffold with enhanced osteogenesis and angiogenesis for further orthopedic applications.Peng Gao Bo Fan Xiaoming Yu Wenwen Liu Jie Wu Lei Shi Di Yang Lili Tan Peng Wan Yulin Hao Shujun Li Wentao Hou Ke Yang Xiaokang Li Zheng Guo 2020Bioactive Materials2020,5,3:15

12Optimizing aerobic biodegradation of dichloromethane using response surface methodology显示文摘Response surface methodology(RSM) was employed to evaluate the optimum aerobic biodegradation of dichloromethane(DCM) in pure culture.The parameters investigated include the initial DCM concentration,glucose as an inducer and hydrogen peroxide as terminal electron acceptor(TEA) .Maximum aerobic biodegradation effciency was predicted to occur when the initial DCM concentration was 380 mg/L,glucose 13.72 mg/L,and H2O2 115 mg/L.Under these conditions the aerobic biodegradation rate reached up to 93.18%,which was significantly higher than that obtained under original conditions.Without addition of glucose,degradation effciencies were 80%at DCM concentrations<326 mg/L.When concentrations of DCM were more than 480 mg/L,the addition of hydrogen peroxide did not help to significantly increase DCM degradation effciency.When DCM concentrations increased from 240 to 480 mg/L,the overall DCM degradation effciency decreased from 91%to 60%in the presence of H2O2 for 120 mg/L.WU Shijin,YU Xiang,HU Zhihang,ZHANG Lili,CHEN Jianmeng College of Biological and Environmental Engineering,Zhejiang University of Technology,Hangzhou 310032,China 2009Journal of Environmental Sciences2009,21,9:13

14GSA:Genome Sequence Archive显示文摘With the rapid development of sequencing technologies towards higher throughput and lower cost, sequence data are generated at an unprecedentedly explosive rate. To provide an efficient and easy-to-use platform for managing huge sequence data, here we present Genome Sequence Archive(GSA; http://bigd.big.ac.cn/gsa or http://gsa.big.ac.cn), a data repository for archiving raw sequence data. In compliance with data standards and structures of the International Nucleotide Sequence Database Collaboration(INSDC), GSA adopts four data objects(Bio Project, Bio Sample,Experiment, and Run) for data organization, accepts raw sequence reads produced by a variety of sequencing platforms, stores both sequence reads and metadata submitted from all over the world,and makes all these data publicly available to worldwide scientific communities. 15Effect of minor content of Gd on the mechanical and degradable properties of as-cast Mg-2Zn-xGd-0.5Zr alloys显示文摘RE-containing Mg alloys used as biodegradable medical implants exhibit good promising application due to their good mechanical properties and degradation resistance. In this work, effect of Gd on the microstructure, mechanical properties and biodegradation of as-cast Mg-2Zn-xGd-0.5Zr alloys was investigated. The results showed that there were mainly α-Mg, I-phase, W-phase and MgZn2 phase in Mg-Zn-Gd-Zr alloys. With increase of the Gd content, the strength of the alloys was enhanced due to the second phase strengthening and grain refinement. The degradation resistance of Mg-2Zn-0.5Zr alloy was increased by adding 0.5%–1% Gd due to the uniformly distributed second phases which acted as a barrier to prevent the pitting corrosion. However, increasing Gd content to 2% reduced the degradation resistance of the alloy due to the galvanic corrosion between the matrix and the second phases.The good degradation resistance and mechanical properties of as-cast Mg-2Zn-1Gd-0.5Zr alloy makes it outstanding for biomaterial application.Junxiu Chen Lili Tan Xiaoming Yu Ke Yang 2019Journal of Materials Science & Technology2019,35,4:13

17Microstructure,texture evolution and mechanical properties of cold rolled Ti-32.5Nb-6.8Zr-2.7Sn biomedical beta titanium alloy显示文摘Ti-32.5 Nb-6.8 Zr-2.7 Sn(TNZS,wt%) alloy was produced by using vacuum arc melting method,followed by solution treatment and cold rolling with the area reductions of 50% and 90%.The effects of cold rolling on the microstructure,texture evolution and mechanical properties of the experimental alloy were investigated by optical microscopy,X-ray diffraction,transmission electron microscopy and universal material testing machine.The results showed that the grains of the alloy were elongated along rolling direction and stress-induced α'' martensite was not detected in the deformed samples.The plastic deformation mechanisms of the alloy were related to {112} 111 type deformation twinning and dislocation slipping.Meanwhile,the transition from γ-fiber texture to α-fiber texture took place during cold rolling and a dominant {001} 110_(α-fiber) texture was obtained after 90% cold deformation.With the increase of cold deformation degree,the strength increased owing to the increase of microstrain,dislocation density and grain refinement,and the elastic modulus decreased owing to the increase of dislocation density as well as an enhanced intensity of {001} 110_(α-fiber)texture and a weakened intensity of {111} 112_(γ-fiber)texture.The 90% cold rolled alloy exhibited a great potential to become a new candidate for biomedical applications,since it possesses low elastic modulus(47.1 GPa),moderate strength(883 MPa) and high elastic admissible strain(1.87%),which are superior than those of Ti-6 Al-4 V alloy.Chunbo Lan Yu Wu Lili Guo Huijuan Chen Feng Chen 2018Journal of Materials Science & Technology2018,34,5:10

19Warming decreases photosynthates and yield of soybean [Glycine max(L.) Merrill] in the North China Plain显示文摘Understanding the responses of field crops such as soybean to climate warming is critical for economic development and adaptive management of food security. A field warming experiment was conducted using infrared heaters to investigate the responses of soybean phenology, photosynthetic characteristics, and yield to climate warming in the North China Plain. The results showed that 0.4 °C and 0.7 °C increases in soybean canopy air and soil temperature advanced anthesis stage by 3.8 days and shortened the length of entire growth stage by 4.5 days. Warming also decreased the leaf photosynthetic rate by 6.6% and 10.3% at the anthesis and seed filling stages, respectively, but increased the leaf vapor pressure deficit by 9.4%, 15.7%, and 14.1% at the anthesis, pod setting, and seed filling stages,respectively. However, leaf soluble sugar and starch were decreased by 25.6% and 20.5%,respectively, whereas stem soluble sugar was reduced by 12.2% at the anthesis stage under experimental warming. The transportation amount of leaf soluble sugar and contribution rate of transportation amount to seed weight were reduced by 58.2% and 7.7%, respectively,under warming. As a result, warming significantly decreased 100-seed weight and soybean yield by 20.8% and 45.0%, respectively. 20Porphyromonas gingivalis disrupts vascular endothelial homeostasis in a TLR-NF-κB axis dependent manner显示文摘Cardiovascular disease is still the leading cause of mortality worldwide.Vascular endothelial dysfunction is viewed as the initial step of most cardiovascular diseases.Many studies have indicated that periodontal pathogens,especially Porphyromonas gingivalis,are closely correlated with vascular endothelial homeostasis,but the function of P.gingivalis and the underlying mechanisms are still elusive.To illuminate the effects and elucidate the mechanisms of P.gingivalis on endothelial structural integrity,we developed P.gingivalis infection models in vivo and in vitro.Endothelial cell proliferation,differentiation and apoptosis were detected.Here,we showed that P.gingivalis can impair endothelial integrity by inhibiting cell proliferation and inducing endothelial mesenchymal transformation and apoptosis of endothelial cells,which reduce the cell levels and cause the endothelium to lose its ability to repair itself.A mechanistic analysis showed that TLR antagonist or NF-κB signalling inhibitor can largely rescue the damaged integrity of the endothelium caused by P.gingivalis,suggesting that TLR-NF-κB signalling plays a vital role in vascular endothelial homeostasis destroyed by P.gingivalis.These results suggest a potential intervention method for the prevention and treatment of cardiovascular disease.Mengru Xie Qingming Tang Shaoling Yu Jiwei Sun Feng Mei Jiajia Zhao Lili Chen 2020International Journal of Oral Science2020,12,3:9
