4China National Medical Products Administration approval summary:anlotinib for the treatment of advanced non-small cell lung cancer after two lines of chemotherapy显示文摘Background:On May 8,2018,the China National Medical Products Administration(NMPA)approved anlotinib,an orally administered anti-angiogenesis inhibitor,for the treatment of patients with advanced non-small cell lung can-cer(NSCLC)who have progressed after treatment with two or more lines of prior systemic chemotherapy.Main body of the abstract:China NMPA reviewed and inspected a regional double-blinded,placebo-controlled,Phase III trial comparing the overall survival(OS)of NSCLC patients between the anlotinib and placebo arms.A total of 437 patients were randomized(2:1)to receive either anlotinib(n=294)or placebo(n=143)once daily on a 2-week on and 1-week off schedule.Patients with epidermal growth factor receptor(EGFR)or activating anaplastic lymphoma kinase(ALK)genomic tumor aberrations should have disease progression on NMPA-approved therapy.Anlotinib is the first NMPA-approved drug for patients with advanced NSCLC who have progressed on at least two lines of prior systemic chemotherapies in China.The approval was based on a statistically and clinically significant improvement in median OS with anlotinib(9.46 months)compared with placebo[6.37 months;hazard ratio(HR])=0.70,95%confidence interval(CI)=0.55-0.89;two-sided log-rank P=0.002].The confirmed objective response rate(ORR)was 9.2%in the anlotinib arm and 0.7%in the placebo arm.The median duration of response(DoR)was 4.83 months,with a 95%CI of 3.31-6.97 months.The toxicity profile of anlotinib was consistent with that of known anti-angiogenesis inhibitors.Common adverse drug reactions(ADRs)in anlotinib-treated patients included hypertension(67.4%),hand-foot syndrome(43.9%),hemoptysis(14.0%),thyroid stimulating hormone(TSH)elevation(46.6%),and corrected QT interval(QTc)prolongation(26.2%).Short conclusion:Anlotinib demonstrated a clinically significant OS prolongation as a novel therapeutic option for advanced or metastatic NSCLC following at least two lines of chemotherapy.Ming Zhou Xiaoyuan Chen Hong Zhang Lin Xia Xin Tong Limin Zou Ruimin Hao Jianhong Pan Xiao Zhao Dongmei Chen Yuanyuan Song Yueli Qi Ling Tang Zhifang Liu Rong Gao Yuankai Shi Zhimin Yang 2019Cancer Communications2019,39,1:35
5Terrestrial carbon sinks in China and around the world and their contribution to carbon neutrality显示文摘Enhancing the terrestrial ecosystem carbon sink(referred to as terrestrial C sink) is an important way to slow down the continuous increase in atmospheric carbon dioxide(CO_(2)) concentration and to achieve carbon neutrality target.To better understand the characteristics of terrestrial C sinks and their contribution to carbon neutrality,this review summarizes major progress in terrestrial C budget researches during the past decades,clarifies spatial patterns and drivers of terrestrial C sources and sinks in China and around the world,and examines the role of terrestrial C sinks in achieving carbon neutrality target.According to recent studies,the global terrestrial C sink has been increasing from a source of (-0.2±0.9) Pg C yr^(-1)(1 Pg=1015g)in the 1960s to a sink of (1.9±1.1) Pg C yr^(-1) in the 2010s.By synthesizing the published data,we estimate terrestrial C sink of 0.20–0.25 Pg C yr^(-1) in China during the past decades,and predict it to be 0.15–0.52 Pg C yr^(-1) by 2060.The terrestrial C sinks are mainly located in the mid-and high latitudes of the Northern Hemisphere,while tropical regions act as a weak C sink or source.The C balance differs much among ecosystem types:forest is the major C sink;shrubland,wetland and farmland soil act as C sinks;and whether the grassland functions as C sink or source remains unclear.Desert might be a C sink,but the magnitude and the associated mechanisms are still controversial.Elevated atmospheric CO_(2) concentration,nitrogen deposition,climate change,and land cover change are the main drivers of terrestrial C sinks,while other factors such as fires and aerosols would also affect ecosystem C balance.The driving factors of terrestrial C sink differ among regions.Elevated CO_(2) concentration and climate change are major drivers of the C sinks in North America and Europe,while afforestation and ecological restoration are additionally important forcing factors of terrestrial C sinks in China.For future studies,we recommend the necessity for intensive and long-term ecosystem C monitoring over broad geographic scale to improve terrestrial biosphere models for accurately evaluating terrestrial C budget and its dynamics under various climate change and policy scenarios.Yuanhe Yang Yue Shi Wenjuan Sun Jinfeng Chang Jianxiao Zhu Leiyi Chen Xin Wang Yanpei Guo Hongtu Zhang Lingfei Yu Shuqing Zhao Kang Xu Jiangling Zhu Haihua Shen Yuanyuan Wang Yunfeng Peng Xia Zhao Xiangping Wang Huifeng Hu Shiping Chen Mei Huang Xuefa Wen Shaopeng Wang Biao Zhu Shuli Niu Zhiyao Tang Lingli Liu Jingyun Fang 2022Science China(Life Sciences)2022,65,5:32
6A multi-resolution global land cover dataset through multisource data aggregation显示文摘Recent developments of 30 m global land characterization datasets(e.g., land cover, vegetation continues field) represent the finest spatial resolution inputs for global scale studies. Here, we present results from further improvement to land cover mapping and impact analysis of spatial resolution on area estimation for different land cover types. We proposed a set of methods to aggregate two existing 30 m resolution circa 2010 global land cover maps, namely FROM-GLC(Finer Resolution Observation and Monitoring-Global Land Cover) and FROM-GLC-seg(Segmentation), with two coarser resolution global maps on development, i.e., Nighttime Light Impervious Surface Area(NL-ISA) and MODIS urban extent(MODIS-urban), to produce an improved 30 m global land cover map—FROM-GLC-agg(Aggregation). It was post-processed using additional coarse resolution datasets(i.e., MCD12Q1, GlobCover2009, MOD44 W etc.) to reduce land cover type confusion. Around 98.9% pixels remain 30 m resolution after some post-processing to this dataset. Based on this map, majority aggregation and proportion aggregation approaches were employed to create a multi-resolution hierarchy(i.e., 250 m, 500 m, 1 km, 5 km, 10 km, 25 km, 50 km, 100 km) of land cover maps to meet requirements for different resolutions from different applications. Through accuracy assessment, we found that the best overall accuracies for the post-processed base map(at 30 m) and the three maps subsequently aggregated at 250 m, 500 m, 1 km resolutions are 69.50%, 76.65%, 74.65%, and 73.47%, respectively. Our analysis of area-estimation biases for different land cover types at different resolutions suggests that maps at coarser than 5 km resolution contain at least 5% area estimation error for most land cover types. 7Positive effects of tree species diversity on litterfall quantity and quality along a secondary successional chronosequence in a subtropical forest显示文摘Aims Litterfall,as an important link between aboveground and belowground processes,plays a key role in forest ecosystems.Here,we test for effects of tree species richness on litter production and litter quality in subtropical forest.The study further encompasses a factorial gradient of secondary succession that resulted from human exploitation.Given that a large percentage of subtropical forests are in secondary successional stages,understanding the role of biodiversity on forest re-growth after disturbance appears critical.Methods From January 2009 to December 2014,we monitored forest litterfall in 27 Comparative Study Plots that spanned a gradient of tree species richness(3-20 species)and secondary successional ages(~20 to 120 years)in Gutianshan Natural Nature Reserve,Zhejiang Province,China.The experiment is part of the biodiversity-ecosystem functioning research platform‘BEF-China’.Tree litterfall was collected in monthly intervals using litter traps.Samples were separated into leaf and non-leaf components.Leaf litter was further sorted into dominant and other species.Community level monthly leaf litter C and N contents were analysed through a full year.General linear mixed-effects models were applied to test for effects of tree species richness and successional age on litter quantity and leaf litter C/N.Important Findings Litterfall increased with species richness among and within successional age and this effect was consistent across years.Successionally older stands had higher litterfall and this effect was related to increased tree species richness.However,species richness did not change the intra-and inter-annual temporal stability of litterfall.Increasing tree species richness increased leaf litter quality(decreased C/N),while successional age had no effect.Our study indicates that more diverse forest stands produce more leaf litter and that this litter has higher N concentrations,which could promote forest growth through accelerated nutrient re-cycling.Yuanyuan Huang Yinlei Ma Ke Zhao Pascal A.Niklaus Bernhard Schmid Jin-Sheng He 2017Journal of Plant Ecology2017,10,1:18
8SHP2 inhibition triggers anti-tumor immunity and synergizes with PD-1 blockade显示文摘Tyrosine phosphatase SHP2 is a promising drug target in cancer immunotherapy due to its bidirectional role in both tumor growth promotion and T-cell inactivation. Its allosteric inhibitor SHP099 is known to inhibit cancer cell growth both in vitro and in vivo. However, whether SHP099-mediated SHP2 inhibition retards tumor growth in vivo via anti-tumor immunity remains elusive. To address this, a CT-26 colon cancer xenograft model was established in mice since this cell line is insensitive to SHP099.Consequently, SHP099 minimally affected CT-26 tumor growth in immuno-deficient nude mice, but significantly decreased the tumor burden in CT-26 tumor-bearing mice with intact immune system.SHP099 augmented anti-tumor immunity, as shown by the elevated proportion of CD8tIFN-γtT cells and the upregulation of cytotoxic T-cell related genes including Granzyme B andPerforin, which decreased the tumor load. In addition, tumor growth in mice with SHP2-deficient T-cells was markedly slowed down because of enhanced anti-tumor responses. Finally, the combination of SHP099 and antiPD-1 antibody showed a higher therapeutic efficacy than either monotherapy in controlling tumor growthin two colon cancer xenograft models, indicating that these agents complement each other. 9Human umbilical cord-derived mesenchymal stem cell therapy in patients with COVID-19:a phase 1 clinical trial显示文摘No effective drug treatments are available for coronavirus disease 2019(COVID-19).Host-directed therapies targeting the underlying aberrant immune responses leading to pulmonary tissue damage,death,or long-term functional disability in survivors require clinical evaluation.We performed a parallel assigned controlled,non-randomized,phase 1 clinical trial to evaluate the safety of human umbilical cord-derived mesenchymal stem cells(UC-MSCs)infusions in the treatment of patients with moderate and severe COVID-19 pulmonary disease.The study enrolled 18 hospitalized patients with COVID-19(n=9 for each group).The treatment group received three cycles of intravenous infusion of UC-MSCs(3×107 cells per infusion)on days 0,3,and 6.Both groups received standard COVID-treatment regimens.Adverse events,duration of clinical symptoms,laboratory parameters,length of hospitalization,serial chest computed tomography(CT)images,the PaO2/FiO2 ratio,dynamics of cytokines,and IgG and IgM anti-SARS-CoV-2 antibodies were analyzed.No serious UC-MSCs infusion-associated adverse events were observed.Two patients receiving UC-MSCs developed transient facial flushing and fever,and one patient developed transient hypoxia at 12 h post UC-MSCs transfusion.Mechanical ventilation was required in one patient in the treatment group compared with four in the control group.All patients recovered and were discharged.Our data show that intravenous UC-MSCs infusion in patients with moderate and severe COVID-19 is safe and well tolerated.Phase 2/3 randomized,controlled,double-blinded trials with long-term follow-up are needed to evaluate the therapeutic use of UC-MSCs to reduce deaths and improve long-term treatment outcomes in patients with serious COVID-19.Fanping Meng Ruonan Xu Siyu Wang Zhe Xu Chao Zhang Yuanyuan Li Tao Yang Lei Shi Junliang Fu Tianjun Jiang Lei Huang Peng Zhao Xin Yuan Xing Fan Ji-Yuan Zhang Jinwen Song Dawei Zhang Yanmei Jiao Limin Liu Chunbao Zhou Markus Maeurer Alimuddin Zumla Ming Shi Fu-Sheng Wang 2020Signal Transduction and Targeted Therapy2020,5,1:16

11A multicenter,retrospective.epidemiologic survey of the clinical features and management of bone metastatic disease in China显示文摘Background:Bone metastases are common in patients with advanced cancer.Bisphosphonates(BPs) could prevent or delay the development of skeleton-related events(SREs).The present study aimed to identify the clinical features of and treatment strategies for Chinese patients with bone metastases.Methods:Consecutive cancer patients who had bone metastases and received BP treatment were enrolled.A questionnaire was developed to collect the patients' clinical data,as well as information on the diagnosis and management of bone metastases.Physicians' awareness of the guidelines and knowledge of the application of BP were also assessed.Results:A total of 3223 patients with lung cancer(36.5%),breast cancer(30.9%),prostate cancer(8.5%),and gastrointestinal cancer(5.7%) were included in this study.The sites of bone metastases were the thoracic spine(56.0%),lumbar spine(47.1%),ribs(32.6%),and pelvis(23.2%).The SRE frequency was the highest in patients with multiple myeloma(36.6%),followed by those with lung cancer(25.9%),breast cancer(20.2%),prostate cancer(18.2%),and gastrointestinal cancer(17.3%).Irradiation to the bone was the most frequent SRE(58%in lung cancer patients,45%in breast cancer patients,and 48%in prostate cancer patients).Our survey also showed that 45.5%of patients received BP within 3 months after their diagnosis of bone metastases,whereas the remaining 54.5%of patients did not receive BP treatment until at least 3 months after their diagnosis of bone metastases.The SRE frequency in the former group was significantly lower than that in the latter group(4.0%vs.42.3%,P < 0.05).In patients with more than 6 months of continuous BP treatment,the mean time to the first SRE was significantly longer than that in patients with less than 6 months of continuous BP treatment(7.2 vs.3.4 months,P < 0.05).In addition,12.2%of the physicians were not aware of the efficacy of BP in preventing and delaying SRE.Only half(52.3%) of the physicians agreed that the BP treatment should persist for at least 6 months unless it was intolerable.Conclusions:Our study suggested that prompt and persistent BP treatment was associated with a reduced risk of SREs.However,our survey also revealed that the proper application of BP was not as common as expected in China.Yunpeng Yang Yuxiang Ma Jin Sheng Yan Huang Yuanyuan Zhao Wenfeng Fang Shaodong Hong Ying Tian Cong Xue Li Zhang 2016Chinese Journal of Cancer2016,35,5:13
12Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 13The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 14Microvessel changes in the gerbil hippocampus after cerebral ischemia and reperfusion by Buyang Huanwu decoction pretreatment显示文摘Buyang Huanwu decoction(BYHWD) is a classic recipe for the prevention and treatment of ischemic cerebrovascular disease.Gerbils were pretreated with BYHWD,and then subjected to cerebral ischemia and reperfusion.Microvascular changes were determined with laser Doppler monitoring,tannic acid-ferric chloride mordant,and electron microscopy.Results showed that BYHWD pretreatment could enhance the function of hippocampal microvessels,prevent injury,and increase microvasular density and microvasular area density.Thus,these results suggest that BYHWD pretreatment could prevent microvascular occlusion,enhance the capacity of microvascular reperfusion,increase cerebral blood flow,and inhibit neuronal damage,and may be an effective therapy against brain ischemic injury.Xiaoguang Wu Yixue Li Haixia Liu Yuhua Yin Shumin Zhao Yuanyuan Guo 2011Neural Regeneration Research2011,6,9:11

16Comprehensive Evaluation of Ski Resort Development Conditions in Northern China显示文摘China′s bid for the 2022 Winter Olympic Games will promote rapid development of domestic ski tourism. More than 10 provinces in the northern China list ski tourism as a new source of growth in the tourism industry. Formulating a scientific evaluation system is an important foundation for ski resort development. By referring to the conditions of existing international ski resorts, a comprehensive index system for evaluating ski resort development was developed based on natural factors, human factors, athletic sports, and popular ski tourism. The system was composed of one target layer, two indices, seven sub-indices and twenty-five basic indices. Based on data collected during a field investigation in 2012 and on a statistical model, the development conditions of the following ski resorts were quantified: Yabuli Ski Resort in Heilongjiang Province and Beidahu Ski Resort in Jilin Province in Northeast China, Wanlong Ski Resort in Hebei Province and Nanshan Ski Resort in Beijing in the northern China, and Silk Road International Ski Resort in Uygur Autonomous Region of Xinjiang in the northwestern China. The resulting index values of development conditions at Beidahu, Yabuli, Wanlong, Nanshan, and Silk Road International Ski Resorts were 0.304, 0.278, 0.270, 0.214, and –0.025, respectively. Various natural and human factors exert positive and negative effects on the development potential of ski resorts. The development conditions of the five ski resorts were inferior to those of first-class international resorts. 17Monitoring dynamic changes of global land cover types:fluctuations of major lakes in China every 8 days during 2000–2010显示文摘Remote sensing images can be used to delineate variations in the area of lakes and to assess the influence of environmental changes and human activities.However,because lakes are dynamic,results obtained from individual images acquired on a single date are not representative and do not accurately reflect ongoing changes.In this study,we used 8-day moderate resolution imaging spectroradiometer(MODIS)composite data from 2000 to2010 to map water surface changes over 629 lakes in China.We combined automatic extraction of training data and support vector machine classification to derive the spatial distribution of these large water bodies.The producer’s and user’s accuracies for MODIS images were91.06%and 89.81%,respectively,when compared with interpretation results from 30 m resolution Landsat images taken on similar days.Area changes,variability,inundation intensity,and rainy seasons of the 629 lakes were analyzed based on this multi-temporal lake database.The total area of the 629 lakes increased over the study period,primarily as a result of the expansion of lake areas on the Tibetan Plateau.There were 12 lakes with a maximum area[1,000 km2,and six of these decreased in area from 2000to 2010.The shrinkages of Poyang Lake and Dongting Lake were-54.76 and-25.08 km2/a,respectively.The area of lakes on Tibetan Plateau,in northern Xinjiang,northeastern Inner Mongolia,and northeastern China varied little,while lakes on the Yangtze Plain,in southern Inner Mongolia,and central Xinjiang fluctuated considerably.Inundation intensity increased for lakes on the Tibetan Plateau,in northern Xinjiang,Heilongjiang,and Jilin,while inundation extent decreased in central Xinjiang,southern Tibet,southern Inner Mongolia,Sichuan,and on the Yangtze Plain.This study is an attempt to develop high-frequency specific land cover maps to improve applicability of general land cover maps.The lake products serve as an important supplement to hydrologic data.The lake database enables the generation of new land surface process models,which could improve the precision of simulations,based on more accurate observations of dynamic lake systems.Fangdi Sun Yuanyuan Zhao Peng Gong Ronghua Ma Yongjiu Dai 2014Chinese Science Bulletin2014,59,2:10
18Myeloid-specific targeting of Notch ameliorates murine renal fibrosis via reduced infiltration and activation of bone marrowderived macrophage显示文摘Macrophages play critical roles in renal fibrosis.However,macrophages exhibit ontogenic and functional heterogeneities,and which population of macrophages contributes to renal fibrosis and the underlying mechanisms remain unclear.In this study,we genetically targeted Notch signaling by disrupting the transcription factor recombination signal binding protein-JK(RBP-J),to reveal its role in regulation of macrophages during the unilateral ureteral obstruction(UUO)-induced murine renal fibrosis.Myeloid-specific disruption of RBP-J attenuated renal fibrosis with reduced extracellular matrix deposition and myofibroblast activation,as well as attenuated epithelial-mesenchymal transition,likely owing to the reduced expression of TGF-β.Mean while,RBP-J deletion significantly hampered macrophage infiltration and activation in fibrotic kidney,although their proliferation appeared unaltered.By using macrophage clearance experiment,we found that kidney resident macrophages made negligible contribution,but bone marrow(BM)-derived macrophages played a major role in renal fibrogenesis.Further mechanistic analyses showed that Notch blockade reduced monocyte emigration from BM by down-regulating CCR2 expression.Finally,we found that myeloid-specific Notch activation aggravated renal fibrosis,which was mediated by CCR2^+macrophages infiltration.In summary,our data have unveiled that myeloid-specific targeting of Notch could ameliorate renal fibrosis by regulating BM-derived macrophages recruitment and activation,providing a novel strategy for intervention of this disease.Yali Jiang Yuanyuan Wang Pengfei Ma Dongjie An Junlong Zhao Shiqian Liang Yuchen Ye Yingying Lu Peng Zhang Xiaowei Liu Hua Han Hongyan Qin 2019Protein & Cell2019,10,3:10
19DDIT4 promotes gastric cancer proliferation and tumorigenesis through the p53 and MAPK pathways显示文摘Background:Gastric cancer(GC)is one of the most common malignancies worldwide,particularly in China.DNA damage-inducible transcript 4(DDIT4)is a mammalian target of rapamycin inhibitor and is induced by various cellular stresses;however,its critical role in GC remains poorly understood.The present study aimed to investigate the poten-tial relationship and the underlying mechanism between DDIT4 and GC development.Methods:We used western blotting,real-time polymerase chain reaction,and immunohistochemical or immunoflu-orescence to determine DDIT4 expression in GC cells and tissues.High-content screening,cell counting kit-8 assays,colony formation,and in vivo tumorigenesis assays were performed to evaluate cell proliferation.Flow cytometry was used to investigate cell apoptosis and cell cycle distribution.Results:DDIT4 was upregulated in GC cells and tissue.Furthermore,downregulating DDIT4 in GC cells inhibited proliferation both in vitro and in vivo and increased 5-fluorouracil-induced apoptosis and cell cycle arrest.In contrast,ectopic expression of DDIT4 in normal gastric epithelial cells promoted proliferation and attenuated chemosensitivity.Further analysis indicated that the mitogen-activated protein kinase and p53 signaling pathways were involved in the suppression of proliferation,and increased chemosensitivity upon DDIT4 downregulation.Conclusion:DDIT4 promotes GC proliferation and tumorigenesis,providing new insights into the role of DDIT4 in the tumorigenesis of human GC.Feng Du Lina Sun Yi Chu Tingyu Li Chao Lei Xin Wang Mingzuo Jiang Yali Min Yuanyuan Lu Xiaodi Zhao Yongzhan Nie Daiming Fan 2018Cancer Communications2018,38,1:10
