Cancer incidence and mortality in Shandong province,2012显示文摘Objective: Population-based cancer registration data in 2012 from all available cancer registries in Shandong province were collected by Shandong Center for Disease Control and Prevention(SDCDC). SDCDC estimated the numbers of new cancer cases and cancer deaths in Shandong province with compiled cancer incidence and mortality rates. Methods: In 2015, there were 21 cancer registries submitted data of cancer incidence and deaths occurred in 2012. All the data were checked and evaluated based on the National Central Cancer Registry(NCCR) criteria of data quality. Qualified data from 15 registries were used for cancer statistics analysis as provincial estimation. The pooled data were stratified by area(urban/rural), gender, age group(0, 1-4, 5-9, 10-14, …, 85+ years) and cancer type. New cancer cases and deaths were estimated using age-specific rates and corresponding provincial population in 2012. The Chinese census data in 2000 and Segi's population were applied for age-standardized rates. All the rates were expressed per 100,000 person-year.Results: Qualified 15 cancer registries(4 urban and 11 rural registries) covered 17,189,988 populations(7,486,039 in urban and 9,703,949 in rural areas). The percentage of cases morphologically verified(MV%) and death certificate-only cases(DCO%) were 66.12% and 2.93%, respectively, and the mortality to incidence rate ratio(M/I) was 0.60. A total of 253,060 new cancer cases and 157,750 cancer deaths were estimated in Shandong province in 2012. The incidence rate was 263.86/100,000(303.29/100,000 in males, 223.23/100,000 in females), the age-standardized incidence rates by Chinese standard population(ASIRC) and by world standard population(ASIRW) were 192.42/100,000 and 189.50/100,000 with the cumulative incidence rate(0-74 years old) of 22.07%. The cancer incidence, ASIRC and ASIRW in urban areas were 267.64/100,000, 195.27/100,000 and 192.02/100,000 compared to 262.32/100,000, 191.26/100,000 and 188.48/100,000 in rural areas, respectively. The cancer mortality was 164.47/100,000(207.42/100,000 in males, 120.23/100,000 in females), the age-standardized incidence rates by Chinese standard population(ASMRC) and by world standard population(ASMRW) were 117.54/100,000 and 116.90/100,000, and the cumulative mortality rate(0-74 years old) was 13.53%. The cancer mortality, ASMRC and ASMRW were 141.59/100,000, 101.17/100,000 and 100.33/100,000 in urban areas, and 173.79/100,000, 124.20/100,000 and 123.64/100,000 in rural areas, respectively. Cancers of the lung, stomach, liver, esophagus, colorectum, female breast, brain, leukemia, bladder and pancreas were the most common cancers, accounting for about 82.12% of all cancer new cases. Lung cancer, stomach cancer, liver cancer, esophageal cancer, colorectal cancer, female breast cancer, pancreatic cancer, brain tumor, leukemia and lymphoma were the leading causes of cancer death, accounting for about 89.01% of all cancer deaths. The cancer spectrum showed difference between urban and rural, males and females both in incidence and mortality rates. Conclusions: Cancer surveillance information in Shandong province is making great progress with the increasing number of cancer registries, population coverage and the improving data quality. Cancer registration plays a fundamental role in cancer control by providing basic information on population-based cancer incidence, mortality, survival and time trend. The disease burden of cancer is serious in Shandong province, and so cancer prevention and control in Shandong province should be enhanced including health education, health promotion, cancer screening and cancer care services. | Zhentao Fu Zilong Lu Yingmei Li jiyu Zhang Gaohui Zhang Xianxian Chen Jie Chu Jie Ren Haiyan Liu Xiaolei Guo | 2016 | Chinese Journal of Cancer Research2016,28,3: | 17
Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants, such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents, near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination, with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants, and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions. In contrast, the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite, indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants, mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents, since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations. Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together, these results suggest that the deployment of RBD-vaccines,through a third-dose boost, may be ideal for combating SARS-CoV-2 variants when necessary, especially for those carrying mutations that disrupt the NTD supersite. | Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie | 2021 | Cell Research2021,31,7: | 12
Disease profile and plasma neutralizing activity of postvaccination Omicron BA.1 infection in Tianjin,China:a retrospective study显示文摘Dear Editor,SARS-CoV-2 Omicron variant BA.1 first emerged on the Chinese mainland on January 8,2022,in Tianjin and caused a large wave of infections.1 Four rounds of mass testing of its~14 million residents were launched on January 9,12,15,and 20,respectively.As of February 7,2022,a total of 430 individuals were tested positive for Omicron BA.1,with no new infections detected for the following 16 days.In the Chinese mainland, the most delivered vaccines are inactivated vaccines. As of January 8,2022,up to 93.2%of Tianjin’s residents had been vaccinated with at least one dose.2 We investigated the protection conferred by vaccination against Omicron BA.1 by examining the breakthrough infections among vaccinees as compared with unvaccinated cases. Our study population comprised all 430 cases during the outbreak of Omicron BA.1 in Tianjin. | Hong Zheng Yunlong Cao Xiaosu Chen Fengmei Wang Ye Hu Weiliang Song Yangyang Chai Qingqing Gu Yansong Shi Yingmei Feng Shuxun Liu Yan Xie Xiaoliang Sunney Xie Wentao Jiang Zhongyang Shen | 2022 | Cell Research2022,32,8: | 12
Human CMTM2/CKLFSF2 enhances the ligand-induced transactivation of the androgen receptor显示文摘CKLF (chemokine-like factor)-like MARVEL (MAL and related proteins for vesicle trafficking and mem-brane link domain) transmembrane domain containing (CMTM) is a novel gene family. One member of this family, CMTM2, also named chemokine-like factor superfamily 2 (CKLFSF2), is expressed highly in the testis and moderately in the prostate, marrow and peripheral blood cells. However, the function of human CMTM2 remains unknown. Here, we found that CMTM2 was upregulated in 5α-dihydrotestos- terone (DHT)-treated LNCaP cells. We investigated the relationship between CMTM2 and the androgen receptor. Our results showed that CMTM2 enhanced DHT-mediated androgen receptor (AR) transacti-vation and the expression of prostate specific antigen (PSA). We also observed that CMTM2 enhanced the AR protein level, which was reversed by silencing endogenous CMTM2 expression, which sug-gested that CMTM2 might play an important role in maintaining the AR protein level. We also found that CMTM2 suppressed Akt activation. A previous study showed that Akt could phosphorylate AR at Ser210 and Ser790 and lead to AR ubiquitylation and degradation as well as suppression of AR activity. Taken together, suppressing Akt activation and increasing the AR protein level might be one of the mechanisms for the CMTM2-mediated enhancement of AR transactivation. | LIU DaZhen YIN CaiHua ZHANG YingMei TIAN LinJie LI Ting LI Dan MA DaLong GUO YingLu WANG Ying | 2009 | Chinese Science Bulletin2009,54,6: | 7
Estrogen and insulin synergistically promote endometrial cancer progression via crosstalk between their receptor signaling pathways显示文摘Objective: Despite evidence that estrogens and insulin are involved in the development and progression of many cancers, their synergistic role in endometrial carcinoma(EC) has not been analyzed yet. Methods: Here, we investigated how estrogens act synergistically with insulin to promote EC progression. Cell growth in vitro and in vivo, effects of estradiol and insulin on apoptosis and cell cycle distribution, and expression and activation of estrogen receptor(ER), insulin receptor(InsR), and key proteins in the PI3K and MAPK pathways were examined after combined stimulation with estradiol and insulin. Results: Compared to EC cells treated with estradiol or insulin alone, those treated with both estradiol and insulin exhibited stronger stimulation. Estradiol significantly induced phosphorylation of InsR-β and IRS-1, whereas insulin significantly induced phosphorylation of ER-α. In addition, treatment with both insulin and estradiol together significantly increased the expression and phosphorylation of Akt, MAPK, and ERK. Notably, InsR-β inhibition had a limited effect on estradiol-dependent proliferation, cell cycle, and apoptosis, whereas ER-α inhibition had a limited insulin-dependent effect, in EC cell lines. Insulin and estradiol individually and synergistically promoted EC xenograft growth in mice. Conclusions: Estrogen and insulin play synergistic roles in EC carcinogenesis and progression by activating InsR-β and ER-α,promoting a crosstalk between them, and thereby resulting in the activation of downstream PI3K/Akt and MAPK/ERK signaling pathways. | Wenyan Tian Fei Teng Jinping Gao Chao Gao Guoyan Liu Yanfang Zhang Shizhu Yu Wei Zhang Yingmei Wang Fengxia Xue | 2019 | Cancer Biology & Medicine2019,16,1: | 6
Distinct regulatory mechanism of immunoglobulin gene transcription in epithelial cancer cells显示文摘The restriction of immunoglobulin(Ig)expression to B lymphocytes is well established. However, several reports have confirmed that the Ig gene can be expressed in many non-B cancer cells and/or some normal cells. Our aim is to determine whether the Ig gene promoter can be activated in non-B cancer cells and to identify the regulatory mechanism for Ig gene expression. Our results show that the Ig promoter of VH4-59 was activated in several non-B cancer cell lines. Moreover, two novel positive regulatory elements,an enhancer-like element at 2800 to 2610 bp and a copromoter-like element at 2610 to 2300 bp,were identified in two epithelial cancer cell lines,HeLa S3 and HT-29.The octamer element(59-ATGCAAAT-39)located in the Ig promoter,a crucial element for B-cell-derived Ig gene transcription, was also very important for non-B-cell-derived Ig gene transcription.More importantly,we confirmed that octamer-related protein-1(Oct-1),but not Oct-2,was a crucial transcriptional factor for Ig gene transcription due to its ability to bind to the octamer element of the Ig promoter in epithelial cancer cells. These results suggested the presence of a distinct regulatory mechanism for Ig gene expression in non-B cancer cells. | Xiaohui Zhu Lina Wu Li Zhang Peng Hao Shuai Zhang Jing Huang Jie Zheng Yinan Liu Wenjun Li Yingmei Zhang Chunyan Zhou Youhui Zhang C Cameron Yin Xiaoyan Qiu | 2010 | Cellular & Molecular Immunology2010,7,4: | 5
Mesenchymal stem cell treatment improves outcome of COVID-19 patients via multiple immunomodulatory mechanisms显示文摘The infusion of coronavirus disease 2019(COVID-19)patients with mesenchymal stem cells(MSCs)potentially improves clinical symptoms, but the underlying mechanism remains unclear. We conducted a randomized,single-blind,placebo-controlled(29 patients/group)phase II clinical trial to validate previous findings and explore the potential mechanisms.Patients treated with umbilical cord-derived MSCs exhibited a shorter hospital stay(P=0.0198)and less time required for symptoms remission(P=0.0194)than those who received placebo.Based on chest images, both severe and critical patients treated with MSCs showed improvement by day 7(P=0.0099)and day 21(P=0.0084).MSC-treated patients had fewer adverse events.MSC infusion reduced the levels of C-reactive protein, proinflammatory cytokines, and neutrophil extracellular traps(NETs)and promoted the maintenance of SARS-CoV-2-specific antibodies. To explore how MSCs modulate the immune system,we employed single-cell RNA sequencing analysis on peripheral blood. Our analysis identified a novel subpopulation of VNN2+hematopoietic stem/progenitorlike(HSPC-like)cells expressing CSF3R and PTPRE that were mobilized following MSC infusion.Genes encoding chemotaxis factors—CX3CR1 and L-selectin—were upregulated in various immune cells.MSC treatment also regulated B cell subsets and increased the expression of costimulatory CD28 in T cells in vivo and in vitro. In addition,an in vivo mouse study confirmed that MSCs suppressed NET release and reduced venous thrombosis by upregulating kindlin-3 signaling.Together, our results underscore the role of MSCs in improving COVID-19 patient outcomes via maintenance of immune homeostasis. | Rongjia Zhu Tingdong Yan Yingmei Feng Yan Liu Hongcui Cao Gongxin Peng Yanlei Yang Zhen Xu Jingqi Liu Wei Hou Xiaoyue Wang Zhe Li Luchan Deng Shihua Wang Jing Li Qin Han Hongling Li Guangliang Shan Yinghao Cao Xingyan An Jianshe Yan Zhonghui Zhang Huafei Li Xuebin Qu Jiaqi Zhu Shumin Zhou Jiao Wang Fengchun Zhang Jinming Gao Ronghua Jin Dayong Xu Yan-Qing Ma Tao Huang Shuang Peng Zhi Zheng Ilia Stambler Eric Gilson Lee Wei Lim Alexey Moskalev Antonio Cano Sasanka Chakrabarti Brun Ulfhake Huanxing Su Haoying Xu Sihuan Xu Feng Wei Holly MBrown-Borg Kyung-Jin Min Georgina Ellison-Hughes Calogero Caruso Kunlin Jin Robert Chunhua Zhao | 2021 | Cell Research2021,31,12: | 5

Enhanced strength and ductility in Ti46Al4Nb1Mo alloys via boron addition显示文摘To improve the strength and ductility of Ti Al alloys by second phase, Ti46 Al4 Nb1 Mo alloys doped with different B content(0.4%, 0.8%, 1.2%, 1.6% and 2.0%, atomic percent, hereafter in at.%, referred to as TNMx B) were prepared. Macro/microstructure evolution, mechanical properties and deformation mechanisms of the alloys were studied systematically. Results showed that the microstructure of TNM-0.4 B and TNM-0.8 B alloy remained columnar dendrites, and the secondary dendritic arms of columnar grains were more obvious. When the content of B is 1.2%, the columnar dendrites transformed to equiaxed grains, and theα2/γ lamellae colony size was further refined in TNM-1.6 B and TNM-2.0 B alloy. The morphologies and kinds of the borides were changed with increasing B content, XRD results showed that Ti B phase appeared with 1.6%B content, and both Ti B and Ti B2 phase formed in TNM-2.0 B alloy. There were straight and curved Ti B phases located around grain boundaries in TNM-0.4 B and TNM-0.8 B alloy, and when the content of B increased to 1.2%, the curved Ti B phases were reduced, while the tiny and straight Ti B phases increased. With further increasing B content to 1.6% and 2.0%, the tiny and straight Ti B phases were coarser. Compressive testing results showed that the mechanical properties of the TNM alloy were enhanced with increasing B content. The maximum strength and strain of TNM alloy were 2339 MPa and33.7% with 1.6% B addition. The compressive strength and strain were mainly enhanced via refinement of lamellar colony and formation of Ti B, and it is found that pile-up of dislocations and deformed twins promoted by Ti B are predominant in improving the mechanical properties of TNM alloys with higher strength and strain. | Yingmei Tan Ruirun Chen Hongze Fang Yangli Liu Hongzhi Cui Yanqing Su Jingjie Guo Hengzhi Fu | 2022 | Journal of Materials Science & Technology2022,,7: | 2
Clinical factors associated with composition of lung microbiota and important taxa predicting clinical prognosis in patients with severe community-acquired pneumonia显示文摘Few studies have described the key features and prognostic roles of lung microbiota in patients with severe community-acquired pneumonia(SCAP).We prospectively enrolled consecutive SCAP patients admitted to ICU.Bronchoscopy was performed at bedside within 48 h of ICU admission, and 16S rRNA gene sequencing was applied to the collected bronchoalveolar lavage fluid. The primary outcome was clinical improvements defined as a decrease of 2 categories and above on a 7-category ordinal scale within 14 days following bronchoscopy.Sixty-seven patients were included.Multivariable permutational multivariate analysis of variance found that positive bacteria lab test results had the strongest independent association with lung microbiota(R2=0.033;P=0.018),followed by acute kidney injury(AKI;R2=0.032;P=0.011)and plasma MIP-1βlevel(R2=0.027;P=0.044).Random forest identified that the families Prevotellaceae,Moraxellaceae, and Staphylococcaceae were the biomarkers related to the positive bacteria lab test results.Multivariable Cox regression showed that the increase inα-diversity and the abundance of the families Prevotellaceae and Actinomycetaceae were associated with clinical improvements. The positive bacteria lab test results,AKI,and plasma MIP-1βlevel were associated with patients’lung microbiota composition on ICU admission. The families Prevotellaceae and Actinomycetaceae on admission predicted clinical improvements. | Sisi Du Xiaojing Wu Binbin Li Yimin Wang Lianhan Shang Xu Huang Yudi Xia Donghao Yu Naicong Lu Zhibo Liu Chunlei Wang Xinmeng Liu Zhujia Xiong Xiaohui Zou Binghuai Lu Yingmei Liu Qingyuan Zhan Bin Cao | 2022 | Frontiers of Medicine2022,16,3: | 2

Farmers’Demand for Informal Risk Management Strategy and Weather Index Insurance:Evidence from China显示文摘In the absence of formal risk management strategies, agricultural production in China is highly vulnerable to climate change. In this study, field experiments were conducted with 344 households in Heilongjiang(Northeast China)and Jiangsu(East China)Provinces.Probit and logistic models and independent sample T-test were used to explore farmers’demand for weather index insurance,in contrast to informal risk management strategies, and the main factors that affect demand. The results show that the farmers prefer weather index insurance to informal risk management strategies, and farmers’characteristics have significant impacts on their adoption of risk management strategies. The variables non-agricultural labor ratio, farmers’risk perception, education, and agricultural insurance purchase experience significantly affect farmers’weather index insurance demand. The regression results show that the farmers’weather index insurance demand and the influencing factors in the two provinces are different.Farmers in Heilongjiang Province have a higher participation rate than those in Jiangsu Province. The government should conduct more weather index insurance pilot programs to help farmers understand the mechanism, and insurance companies should provide more types of weather index insurance to meet farmers’diversified needs. | Yingmei Tang Huifang Cai Rongmao Liu | 2021 | International Journal of Disaster Risk Science2021,12,2: | 1
