5RNA-Seq Study Reveals AP2-Domain-Containing Signalling Regulators Involved in Initial Imbibition of Seed Germination in Rice显示文摘A number of internal signals are required for seed germination.However,the precise signalling responses in the initial imbibition of seed germination are not yet fully understood in rice.In this study,the RNA sequencing(RNA-Seq)approach was conducted in 8 h imbibed seeds to understand the signalling responses in the initial imbibition of rice seed germination.A total of 563 differentially expressed genes(DEGs)with at least 4-fold change were identified in 8 h imbibed seeds compared to dry seeds.MapMan analysis revealed that the majority of signalling response-related DEGs were hormone-and transcription factor-related genes,in which the largest number of DEGs belong to the AP2-domain-containing regulators,and their expressions were significantly induced in the initial imbibition of seed germination in rice.Moreover,at least five AP2-domain-containing transcription factor OsDREBs were identified in the initial imbibition of rice seed germination,and the expressions of 251 DEGs were putatively regulated by OsDREBs through the dehydration-responsive element(DRE)cis-element assay.It suggested that the OsDREBs might play important roles in the regulation of initial seed imbibition in rice.The identified genes provide a valuable resource to study the signalling regulation of seed germination in the future.HE Yongqi ZHAO Jia FENG Defeng HUANG Zhibo LIANG Jiaming ZHENG Yufei CHENG Jinping YING Jifeng WANG Zhoufei 2020Rice science2020,27,4:2
6Mechanical activation of spike fosters SARS-CoV-2 viral infection显示文摘The outbreak of SARS-CoV-2(SARS2)has caused a global COVID-19 pandemic.The spike protein of SARS2(SARS2-S)recognizes host receptors,including ACE2,to initiate viral entry in a complex biomechanical environment.Here,we reveal that tensile force,generated by bending of the host cell membrane,strengthens spike recognition of ACE2 and accelerates the detachment of spike’s S1 subunit from the S2 subunit to rapidly prime the viral fusion machinery.Mechanistically,such mechano-activation is fulfilled by force-induced opening and rotation of spike’s receptor-binding domain to prolong the bond lifetime of spike/ACE2 binding,up to 4 times longer than that of SARS-S binding with ACE2 under 10 pN force application,and subsequently by force-accelerated S1/S2 detachment which is up to~103 times faster than that in the no-force condition.Interestingly,the SARS2-S D614G mutant,a more infectious variant,shows 3-time stronger force-dependent ACE2 binding and 35-time faster force-induced S1/S2 detachment.We also reveal that an anti-S1/S2 non-RBD-blocking antibody that was derived from convalescent COVID-19 patients with potent neutralizing capability can reduce S1/S2 detachment by 3×106 times under force.Our study sheds light on the mechano-chemistry of spike activation and on developing a non-RBD-blocking but S1/S2-locking therapeutic strategy to prevent SARS2 invasion.Wei Hu Yong Zhang Panyu Fei Tongtong Zhang Danmei Yao Yufei Gao Jia Liu Hui Chen Qiao Lu Tenny Mudianto Xinrui Zhang Chuxuan Xiao Yang Ye Qiming Sun Jing Zhang Qi Xie Pei-Hui Wang Jun Wang Zhenhai Li Jizhong Lou Wei Chen 2021Cell Research2021,31,10:2

8Rock borehole shear tests in dam foundation of Xiangjiaba hydropower station显示文摘Xiangjiaba hydropower station is one of the cascade power stations on the Jinsha River, China. Due to the complicated geological conditions of its dam foundation, evaluating the rock mass quality and determining the mechanical parameters of rock masses are very important issues. To address these issues, several groups of rock borehole shear tests (RBSTs) were conducted on the black mudstone in the dam foundation of Xiangjiaba hydropower station in the second construction phase. Forty three groups of shear strengths of black mudstone samples were obtained from RBSTs, and the shear strength parameters (c and f ) were calculated using the least squares method. In addition, the limitations and merits of RBST employed in the Xiangjiaba hydropower station were discussed. Test results indicate that the shear strength parameters obtained from RBST have a good correlation with the results from sound wave test in borehole. 11Experimental study of suppressing the unbalanced response vibration of rotor system with a G-type integral squeeze film damper显示文摘Conventional squeeze film dampers have numerous challenges including lock up,bistable response and incoordinate precession.In order to resolve these nonlinear problems,a novel G-type integral squeeze film damper(GISFD)is proposed in this research.The experimental test rig is provided to investigate the rotor system with an unbalanced single disk.Numerical simulation results show that the structural design of GISFD is reasonable,which can ensure its safe and stable operation.The influence of different support stiffnesses on the first-order speed of the rotor system is analyzed.Experimental results show that GISFD can effectively suppress the unbalanced response vibration of the rotor.In a certain range,it is found that the suppression effect of GISFD increases with the increase in the kinematic viscosity of the damping fluid.When the silicone oil with kinematic viscosity coefficients v=30.0 cm^(2)/s is employed,the vibration reduction of GISFD is approximately 71.51%.Furthermore,the experimental results show that with the increase of the unbalance,there is a linear relationship between the unbalance and the corresponding amplitude of the unbalanced response.It is concluded that GISFD has excellent linear damping characteristics and reduces the sensitivity of the rotor system to the unbalanced mass.闫伟 He Lidong Zhang Yufei Zhang Yipeng Jia Xingyun 2021High Technology Letters2021,27,1:1

14Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND 16Heteroatom anchors Fe-Mn dual-atom catalysts with bi-functional oxygen catalytic activity for low-temperature rechargeable flexible Zn-air batteries显示文摘M-N-C(M=Fe,Co,Ni,etc.) catalyst owns high catalytic activity in the oxygen catalytic reaction which is the most likely to replace the Pt-based catalysts.But it is still a challenge to further increase the active site density.This article constructs the high-efficiency FeMn-N/S-C-1000 catalyst to realize ORR/OER bifunctional catalysis by hetero-atom,bimetal(Fe,Mn) doped simultaneously strategy.When evaluated it as bi-functional electro-catalysts,FeMn-N/S-C-1000 exhibits excellent catalytic activity(E_(1/2)=0.924 V,E_(j=10)=1.617 V) in alkaline media,outperforms conventional Pt/C,RuO_(2) and most non-precious-metal catalysts reported recently,Such outstanding performance is owing to N,S co-coordinated with metal to form multi-types of single atom,dual atom active sites to carry out bi-catalysis.Importantly,nitrite poison test provides the proof that the active sites of FeMn-N/S-C are more than that of single-atom catalysts to promote catalytic reactions directly.To better understand the local structure of Fe and Mn active sites,XAS and DFT were employed to reveal that FeMn-N_5/S-C site plays the key role during catalysis.Notably,the FeMn-N/S-C-1000 based low-temperature rechargeable flexible Zn-air also exhibits superior discharge performance and extraordinary durability at-40℃.This work will provide a new idea to design diatomic catalysts applied in low-temperature rechargeable batteries.Yuting He Hongtao Li Yi Wang Yufei Jia Yongning Liu Qiang Tan 2024Journal of Energy Chemistry2024,90,3:0
17An empirical approach to predict regional organic carbon in deep soils显示文摘Deep soil organic carbon(SOC)plays an important role in carbon cycling.Precisely predicting deep SOC at the regional scale is crucial for the accurate assessment of carbon sequestration potential in soils but has been challenging for a century.Herein,we developed a depth distribution function-based empirical approach to predict SOC in deep soils at the regional scale.We validated this approach with a dataset from four regions of the world and examined the application of this approach in China’s Loess Plateau.We found that among the reported depth distribution functions describing vertical patterns of SOC,the negative exponential function performed best in fitting SOC along the soil profile in various regions.Moreover,the parameters(i.e.,Ceand k)of the negative exponential function were linearly correlated to surface SOC(0–20 cm)and the changing rates of SOC within the topsoil(0–40 cm).Based on the above relationships,the empirical equations for predicting the negative exponential parameters are established.The validation results from site-specific and regional dataset showed that combining the negative exponential function and such empirical equations can precisely predict SOC concentration in soils down to 500 cm depth.Our study provides a simple,rapid and accurate method for predicting deep soil SOC at the regional scale,which could simplify the assessment of deep soil SOC in various regions.Jingjing WANG Xiaorong WEI Xiaoxu JIA Mingbin HUANG Zhipeng LIU Yufei YAO Ming’an SHAO 2023Science China Earth Sciences2023,66,3:0
18Regulation of different light conditions for efficient biomass production and protein accumulation of Spirulina platensis显示文摘Light plays an important role in the photosynthesis and metabolic process of microalgae.However,how different light conditions regulate the biomass production and protein accumulation of microalgae is mostly unknown.In this study,the influence of different light conditions,including light colors,densities,and light:dark cycles on the cell growth and biochemical composition of Spirulina platensis was symmetrically characterized.Under different colored lights,S.platensis all shows an increase trend within the increased light intensity ranges;however,each showing different optimal light intensities.At the same light intensity,different colored lights show different growth rate of S.platensis following the sequence of red>white>green>yellow>blue.The maximum growth rate and protein accumulation were determined as 21.88 and 5.10 mg/(L·d)when illuminated under red LED.The energy efficiency of different light sources was calculated and ranked as red>white>blue≈green>yellow.Transcriptomic analysis suggests that red light can promote cell growth and protein accumulation by upregulating genes related to photosynthesis,carbon fixation,and C-N metabolism pathways.This study provides a conducive and efficient way to promote biomass production and protein accumulation of S.platensis by regulating light conditions.Yufei ZHANG Xianjun LI Yuhui LI Shiqi LIU Yanrui CHEN Miao JIA Xin WANG Lu ZHANG Qiping GAO Liang ZHANG Daoyong YU Baosheng GE 2024Journal of Oceanology and Limnology2024,42,1:0
19Cloning and characterization of geranylgeranyl diphosphate synthetase from Pinus massoniana and its correlation with resin productivity显示文摘Geranylgeranyl pyrophosphate synthetase(GGPPS) has gained increasing attention as a key enzyme in terpene analysis.We designed specific primers based on plant GGPPS homologs and used reverse transcription polymerase chain reaction(RT-PCR) to obtain and identify Pin GGPPS,a GGPPS gene sequence from Pinus massoniana,using bioinformatics tools.Quantitative PCR analysis of Pin GGPPS expression levels in roots,pine needles,immature stems,and semilignified stems from 6-month-old P.massoniana showed that expression levels of Pin GGPPS were highest in pine needles,followed by immature stems and semilignified stems,and lowest in roots.When we examined the correlation between Pin GGPPS gene expression levels and resin productivity in 20 adult plants for 28 successive days,Pin GGPPS expression levels presented a substantially linear distribution when plotted against their corresponding resin yields.In summary,we characterized the gene Pin GGPPS for the first time in P.massoniana,and established a correlation between Pin GGPPS gene expression levels and resin productivity,suggesting the importance of theory and production practice for P.massoniana.Bowen Chen Yufei Xiao Junji Li Hailong Liu Hu Chen Jie Jia Nan Chao Ying Gai Xiangning Jiang 2018Journal of Forestry Research2018,29,2:0
