3A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 4Precise orbit determination of Beidou Satellites with precise positioning显示文摘Chinese Beidou satellite navigation system constellation currently consists of eight Beidou satellites and can provide preliminary service of navigation and positioning in the Asia-Pacific Region.Based on the self-developed software Position And Navigation Data Analysis(PANDA) and Beidou Experimental Tracking Stations (BETS),which are built by Wuhan University,the study of Beidou precise orbit determination,static precise point positioning (PPP),and high precision relative positioning,and differential positioning are carried out comprehensively.Results show that the radial precision of the Beidou satellite orbit determination is better than 10 centimeters.The RMS of static PPP can reach several centimeters to even millimeters for baseline relative positioning.The precision of kinematic pseudo-range differential positioning and RTK mode positioning are 2-4 m and 5-10 cm respectively,which are close to the level of GPS precise positioning.Research in this paper verifies that,with support of ground reference station network,Beidou satellite navigation system can provide precise positioning from several decimeters to meters in the wide area and several centimeters in the regional area.These promising results would be helpful for the implementation and applications of Beidou satellite navigation system.SHI Chuang ZHAO QiLe LI Min TANG WeiMing HU ZhiGang LOU YiDong ZHANG HongPing NIU XiaoJi LIU JingNan 2012Science China Earth Sciences2012,55,7:61

8China National Medical Products Administration approval summary:anlotinib for the treatment of advanced non-small cell lung cancer after two lines of chemotherapy显示文摘Background:On May 8,2018,the China National Medical Products Administration(NMPA)approved anlotinib,an orally administered anti-angiogenesis inhibitor,for the treatment of patients with advanced non-small cell lung can-cer(NSCLC)who have progressed after treatment with two or more lines of prior systemic chemotherapy.Main body of the abstract:China NMPA reviewed and inspected a regional double-blinded,placebo-controlled,Phase III trial comparing the overall survival(OS)of NSCLC patients between the anlotinib and placebo arms.A total of 437 patients were randomized(2:1)to receive either anlotinib(n=294)or placebo(n=143)once daily on a 2-week on and 1-week off schedule.Patients with epidermal growth factor receptor(EGFR)or activating anaplastic lymphoma kinase(ALK)genomic tumor aberrations should have disease progression on NMPA-approved therapy.Anlotinib is the first NMPA-approved drug for patients with advanced NSCLC who have progressed on at least two lines of prior systemic chemotherapies in China.The approval was based on a statistically and clinically significant improvement in median OS with anlotinib(9.46 months)compared with placebo[6.37 months;hazard ratio(HR])=0.70,95%confidence interval(CI)=0.55-0.89;two-sided log-rank P=0.002].The confirmed objective response rate(ORR)was 9.2%in the anlotinib arm and 0.7%in the placebo arm.The median duration of response(DoR)was 4.83 months,with a 95%CI of 3.31-6.97 months.The toxicity profile of anlotinib was consistent with that of known anti-angiogenesis inhibitors.Common adverse drug reactions(ADRs)in anlotinib-treated patients included hypertension(67.4%),hand-foot syndrome(43.9%),hemoptysis(14.0%),thyroid stimulating hormone(TSH)elevation(46.6%),and corrected QT interval(QTc)prolongation(26.2%).Short conclusion:Anlotinib demonstrated a clinically significant OS prolongation as a novel therapeutic option for advanced or metastatic NSCLC following at least two lines of chemotherapy.Ming Zhou Xiaoyuan Chen Hong Zhang Lin Xia Xin Tong Limin Zou Ruimin Hao Jianhong Pan Xiao Zhao Dongmei Chen Yuanyuan Song Yueli Qi Ling Tang Zhifang Liu Rong Gao Yuankai Shi Zhimin Yang 2019Cancer Communications2019,39,1:35
9Neuroprotective effect of bispectral index-guided fast-track anesthesia using sevoflurane combined with dexmedetomidine for intracranial aneurysm embolization显示文摘Dexmedetomidine has sedative, anxiolytic, analgesic, anti-sympathetic, and anti-shivering effects. Dexmedetomidine might be effective in combination with sevoflurane for anesthesia, but prospective randomized controlled clinical trials with which to verify this hypothesis are lacking. In total, 120 patients who underwent embolization of an intracranial aneurysm were recruited from Anhui Provincial Hospital and Renmin Hospital of Wuhan University of China and randomly allocated to two groups. After intraoperative administration of 2% to 3% sevoflurane inhalation, one group of patients received pump-controlled intravenous injection of 1.0 μg/kg dexmedetomidine for 15 minutes followed by maintenance with 0.3 μg/kg/h until the end of surgery; the other group of patients only underwent pump-controlled infusion of saline. Bispectral index monitoring revealed that dexmedetomidine-assisted anesthesia can shorten the recovery time of spontaneous breathing, time to eye opening, and time to laryngeal mask removal. Before anesthetic induction and immediately after laryngeal mask airway removal, the glucose and lactate levels were low, the S100β and neuron-specific enolase levels were low, the perioperative blood pressure and heart rate were stable, and postoperative delirium was minimal. 10New incompatible pair of TCM:Epimedii Folium combined with Psoraleae Fructus induces idiosyncratic hepatotoxicity under immunological stress conditions显示文摘Epimedii Folium(EF)combined with Psoraleae Fructus(PF)is a common modern preparation,but liver injury caused by Chinese patent medicine preparations containing EF and PF has been frequently reported in recent years.Zhuangguguanjiewan pills(ZGW),which contain EF and PF,could induce immune idiosyncratic liver injury according to clinical case reports and a nonhepatotoxic dose of lipopolysaccharide(LPS)model.This present study evaluated the liver injury induced by EF or PF alone or in combination and investigated the related mechanism by using the LPS model.Liver function indexes and pathological results showed that either EF or PF alone or in combination led to liver injury in normal rats;however,EF or PF alone could lead to liver injury in LPS-treated rats.Moreover,EF combined with PF could induce a greater degree of injury than that caused by EF or PF alone in LPS-treated rats.Furthermore,EF or PF alone or in combination enhanced the LPS-stimulated inflammatory cytokine production,implying that IL-1β,which is processed and released by activating the NLRP3 inflammasome,is a specific indicator of EF-induced immune idiosyncratic hepatotoxicity.Thus,EF may induce liver injury through enhancing the LPS-mediated proinflammatory cytokine production and activating the NLRP3 inflammasome.In addition,the metabolomics analysis results showed that PF affected more metabolites in glycerophospholipid and sphingolipid metabolic pathways compared with EF in LPS model,suggesting that PF increased the responsiveness of the liver to LPS or other inflammatory mediators via modulation of multiple metabolic pathways.Therefore,EF and PF combination indicates traditional Chinese medicine incompatibility,considering that it induces idiosyncratic hepatotoxicity under immunological stress conditions.Yuan Gao Zhilei Wang Jinfa Tang Xiaoyi Liu Wei Shi Nan Qin Xiaoyan Wang Yu Pang Ruisheng Li Yaming Zhang Jiabo Wang Ming Niu Zhaofang Bai Xiaohe Xiao 2020Frontiers of Medicine2020,14,1:32
11Terrestrial carbon sinks in China and around the world and their contribution to carbon neutrality显示文摘Enhancing the terrestrial ecosystem carbon sink(referred to as terrestrial C sink) is an important way to slow down the continuous increase in atmospheric carbon dioxide(CO_(2)) concentration and to achieve carbon neutrality target.To better understand the characteristics of terrestrial C sinks and their contribution to carbon neutrality,this review summarizes major progress in terrestrial C budget researches during the past decades,clarifies spatial patterns and drivers of terrestrial C sources and sinks in China and around the world,and examines the role of terrestrial C sinks in achieving carbon neutrality target.According to recent studies,the global terrestrial C sink has been increasing from a source of (-0.2±0.9) Pg C yr^(-1)(1 Pg=1015g)in the 1960s to a sink of (1.9±1.1) Pg C yr^(-1) in the 2010s.By synthesizing the published data,we estimate terrestrial C sink of 0.20–0.25 Pg C yr^(-1) in China during the past decades,and predict it to be 0.15–0.52 Pg C yr^(-1) by 2060.The terrestrial C sinks are mainly located in the mid-and high latitudes of the Northern Hemisphere,while tropical regions act as a weak C sink or source.The C balance differs much among ecosystem types:forest is the major C sink;shrubland,wetland and farmland soil act as C sinks;and whether the grassland functions as C sink or source remains unclear.Desert might be a C sink,but the magnitude and the associated mechanisms are still controversial.Elevated atmospheric CO_(2) concentration,nitrogen deposition,climate change,and land cover change are the main drivers of terrestrial C sinks,while other factors such as fires and aerosols would also affect ecosystem C balance.The driving factors of terrestrial C sink differ among regions.Elevated CO_(2) concentration and climate change are major drivers of the C sinks in North America and Europe,while afforestation and ecological restoration are additionally important forcing factors of terrestrial C sinks in China.For future studies,we recommend the necessity for intensive and long-term ecosystem C monitoring over broad geographic scale to improve terrestrial biosphere models for accurately evaluating terrestrial C budget and its dynamics under various climate change and policy scenarios.Yuanhe Yang Yue Shi Wenjuan Sun Jinfeng Chang Jianxiao Zhu Leiyi Chen Xin Wang Yanpei Guo Hongtu Zhang Lingfei Yu Shuqing Zhao Kang Xu Jiangling Zhu Haihua Shen Yuanyuan Wang Yunfeng Peng Xia Zhao Xiangping Wang Huifeng Hu Shiping Chen Mei Huang Xuefa Wen Shaopeng Wang Biao Zhu Shuli Niu Zhiyao Tang Lingli Liu Jingyun Fang 2022Science China(Life Sciences)2022,65,5:32

13Efficacy and safety of Xiangsha Liujunzi granules for functional dyspepsia: A multi-center randomized double-blind placebo-controlled clinical study显示文摘AIM To assess the efficacy and safety of a Chinese herbal medicine(CHM), Xiangsha Liujunzi granules, in the treatment of patients with functional dyspepsia(FD). METHODS We performed a randomized, double-blind, placebocontrolled trial with patients from three centers. Two hundred and sixteen subjects diagnosed with FD according to ROME Ⅲ criteria and confirmed by upper gastrointestinal endoscopy and spleen-deficiency and Qi-stagnation syndrome were selected to receive Xiangsha Liujunzi granules or placebo for 4 wk in a 2:1 ratio by blocked randomization. The subjects also received follow-up after the 4-wk intervention. Herbal or placebo granules were dissolved in 300 m L of water. Participants in both groups were administered 130 m L(45 ℃) three times a day. Participants were evaluated prior to and following 4 wk of the intervention in terms of changes in the postprandial discomfort severity scale(PDSS) score, clinical global impression(CGI) scale score, hospital anxiety and depression scale(HADS) score, traditional Chinese medicine symptoms score(SS), scores of various domains of the 36-item short form health survey(SF-36), gastric emptying(GE) and any observed adverse effects. RESULTS Compared with the placebo group, patients in the CHM group showed significant improvements in the scores of PDSS, HADS, SS, SF-36 and CGI scale(P < 0.05 or P < 0.01). 15HIF-1α induces VE-cadherin expression and modulates vasculogenic mimicry in esophageal carcinoma cells显示文摘AIM:To investigate whether hypoxia inducible factor(HIF)-1αmodulates vasculogenic mimicry(VM)by upregulating VE-cadherin expression in esophageal squamous cell carcinoma(ESCC).METHODS:Esophageal squamous cancer cell lines Eca109 and TE13 were transfected with plasmids harboring small interfering RNAs targeting HIF-1αor VEcadherin.The proliferation and invasion of esophageal carcinoma cells were detected by MTT and Transwell migration assays.The formation of tubular networks of cells was analyzed by 3D culture in vitro.BALB/c nude mice were used to observe xenograft tumor formation.The relationship between the expression of HIF-1αand VE-cadherin,ephrin A2(Eph A2)and laminin5γ2(LN5γ2)was measured by Western blot and real-time polymerase chain reaction.RESULTS:Knockdown of HIF-1αinhibited cell proliferation(32.3%±6.1%for Eca109 cells and 38.6%±6.8%for TE13 cells,P<0.05).Both Eca109 and TE13cells formed typical tubular networks.The number of tubular networks markedly decreased when HIF-1αor VE-cadherin was knocked down.Expression of VEcadherin,Eph A2 and LN5γ2 was dramatically inhibited,but the expression of matrix metalloproteinase 2 had no obvious change in HIF-1α-silenced cells.Knockdown of VE-cadherin significantly decreased expression of both Eph A2 and LN5γ2(P<0.05),while HIF-1αexpression was unchanged.The time for xenograft tumor formation was 6±1.2 d for Eca109 cells and Eca109cells transfected with HIF-1αNeo control short hairpin RNA(sh RNA)vector,and 8.4±2.1 d for Eca109 cells transfected with an sh RNA against HIF-1α.Knockdown of HIF-1αinhibited vasculogenic mimicry(VM)and tumorigenicity in vivo.CONCLUSION:HIF-1αmay modulate VM in ESCC by regulating VE-cadherin expression,which affects VM formation through Eph A2 and LN5γ2.Na-Na Tang Hong Zhu Hong-Jie Zhang Wei-Feng Zhang Hai-Lin Jin Lu Wang Pin Wang Gui-Jun He Bo Hao Rui-Hua Shi 2014World Journal of Gastroenterology2014,20,47:25

17Comparison of Electroacupuncture and Mild-Warm Moxibustion on Brain-Gut Function in Patients with Constipation-Predominant Irritable Bowel Syndrome: A Randomized Controlled Trial显示文摘Objective: To compare the effects of electroacupuncture(EA) and mild-warm moxibustion(Mox) therapies for constipation-predominant irritable bowel syndrome(C-IBS) patients. Methods: Sixty C-IBS patients were assigned to 2 groups by simple randomized method, i.e. EA group(30 cases) and Mox group(30 cases). Both EA and Mox treatments were performed on bilateral Tianshu(ST 25) and Shangjuxu(ST 37) for 30 min each time, 6 times per week, for 4 consecutive weeks. The gastrointestinal symptoms and psychological symptoms of the two groups were scored before and after treatment. The effects on the corresponding functional brain areas, namely the anterior cingulate cortex(ACC), insular cortex(IC) and prefrontal cortex(PFC) were observed by functional magnetic resonance imaging(f MRI) before and after treatment. Results: Compared with the Mox group, greater improvements in abdominal distension, defecation frequency, difficulty in defecation and stool features were observed in the EA group(all P<0.01), both Hamilton Anxiety Rating Scale and Hamilton Depression Rating Scale scores were significantly decreased in the EA group(all P<0.01). Finally, decreased activated voxel values were observed in the ACC, right IC and PFC brain regions of EA group with 150 m L colorectal distension stimulation(P<0.05 or P<0.01). Conclusions: Both EA and Mox could significantly improve some of the most intrusive symptoms of C-IBS patients, and EA was more effective than Mox. 18Wireless Communications with Programmable Metasurface:Transceiver Design and Experimental Results显示文摘Metasurfaces have drawn significant attentions due to their superior capability in tailoring electromagnetic waves with a wide frequency range, from microwave to visible light. Recently, programmable metasurfaces have demonstrated the ability of manipulating the amplitude or phase of electromagnetic waves in a programmable manner in real time, which renders them especially appealing in the applications of wireless communications. In this paper, we present the fundamental principle of applying programmable metasurface as transmitter for wireless communications. Then, we establish a prototype system of metasurface-based transmitter to conduct several experiments and measurements over the air, which practically demonstrate the feasibility of using programmable metasurfaces in future communication systems. By exploiting the dynamically controllable property of programmable metasurface, the design, implementation and experimental evaluation of the proposed metasurface-based wireless communication system are presented with the prototype, which realizes single carrier quadrature phase shift keying(QPSK) transmission over the air. In the developed prototype, the phase of the reflected electromagnetic wave of programma-ble metasurface is directly manipulated in real time according to the baseband control signal, which achieves 2.048 Mbps data transfer rate with video streaming transmission over the air. In addition, experimental result is provided to compare the performance of the proposed metasurface-based architecture against the conventional one. With the slight increase of the transmit power by 5 dB, the same bit error rate(BER) performance can be achieved as the conventional system in the absence of channel coding.
