| 4 | Manganese is critical for antitumor immune responses via cGAS-STING and improves the efficacy of clinical immunotherapy显示文摘CD8^+T cell-mediated cancer clearance is often suppressed by the interaction between inhibitory molecules like PD-1 and PD-L1,an interaction acts like brakes to prevent T cell overreaction under normal conditions but is exploited by tumor cells to escape the immune surveillance.Immune checkpoint inhibitors have revolutionized cancer therapeutics by removing such brakes.Unfortunately,only a minority of cancer patients respond to immunotherapies presumably due to inadequate immunity.Antitumor immunity depends on the activation of the cGAS-STING pathway,as STING-deficient mice fail to stimulate tumor-infiltrating dendritic cells(DCs)to activate CD8^+T cells.STING agonists also enhance natural killer(NK)cells to mediate the clearance of CD8^+T cell-resistant tumors.Therefore STING agonists have been intensively sought after.We previously discovered that manganese(Mn)is indispensable for the host defense against cytosolic dsDNA by activating cGAS-STING.Here we report that Mn is also essential in innate immune sensing of tumors and enhances adaptive immune responses against tumors.Mn-insufficient mice had significantly enhanced tumor growth and metastasis,with greatly reduced tumor-infiltrating CD8^+T cells.Mechanically,Mn^2+promoted DC and macrophage maturation and tumor-specific antigen presentation,augmented CD8^+T cell differentiation,activation and NK cell activation,and increased memory CD8^+T cells.Combining Mn^2+with immune checkpoint inhibition synergistically boosted antitumor efficacies and reduced the anti-PD-1 antibody dosage required in mice.Importantly,a completed phase 1 clinical trial with the combined regimen of Mn^2+and anti-PD-1 antibody showed promising efficacy,exhibiting type I IFN induction,manageable safety and revived responses to immunotherapy in most patients with advanced metastatic solid tumors.We propose that this combination strategy warrants further clinical translation. | Mengze Lv Meixia Chen Rui Zhang Wen Zhang Chenguang Wang Yan Zhang Xiaoming Wei Yukun Guan Jiejie Liu Kaichao Feng Miao Jing Xurui Wang Yun-Cai Liu Qian Mei Weidong Han Zhengfan Jiang | 2020 | Cell Research2020,30,11: | 27 |

| 6 | Report on Childhood Obesity in China (9): Sugar-sweetened Beverages Consumption and Obesity显示文摘Objective To explore the associations between sugar-sweetened beverage (SSB) consumption and obesity as well as obesity-related cardiometabolic disorders among children in China. Methods A total of 6974 (boys 3558, girls 3412) children aged 6-13 years participated in the study. Each participant's height, weight, waist circumference, fasting glucose, triglycerides, total cholesterol, high- density lipoprotein cholesterol, and low-density lipoprotein cholesterol were measured. The type of beverage consumption was determined using a self-administered questionnaire. Results SSBs were consumed regularly by 46.1% of the children. The prevalence [adjusted odds ratio (95% confidence internal (CI)] of obesity was 7.6% [as the reference group (ref.)], 10.1% [1.36(1.07, 1.74)], and 11.6% [1.46(1.21, 1.75)], among children who regularly drank milk, other beverages and SSBs, respectively. Regularly drinking SSBs elevated the likelihood of abdominal obesity [adjusted odds ratio (95% CI): 1.36 (1.17, 1.59)]. The prevalence [adjusted odds ratio (95% CI)] of obesity among children who regularly drank sports/caloric beverages, carbonated beverages, sweet tea, and plant protein beverages was 16.8% [2.00(1.31, 3.07)], 12.7% [1.52(1.23, 1.88)], 11.5% [1.52(1.18, 1.95)], and 10.4% [1.41(1.03, 1.94)], respectively, which was higher than that of regular milk drinkers [7.6 % (ref.)]. The prevalence [adjusted odds ratio (95% CI)] of abdominal obesity among children who regularly drank sweet tea, fruit/vegetable juices, and carbonated beverages was 17.7% [1.55(1.26, 1.90)], 16.2% [1.36(1.09, 1.70)], and 15.3% [1.24(1.03, 1.50)], respectively, which was much higher than that of regular milk drinkers [12.8% (ref.)]. Conclusions Regular SSB consumption was positively related to obesity and abdominal obesity. | 7 | Occurrence and Fate of Antibiotics in the Aqueous Environment and Their Removal by Constructed Wetlands in China: A review显示文摘Overuse of antibiotics has become a serious ecological problem worldwide.There is growing concern that antibiotics are losing their effectiveness due to an increased antibiotic resistance in bacteria.During the last twenty years,consumption of antibiotics has increased rapidly in China,which has been cited as one of the world's worst abusers of antibiotics.This review summarizes the current state of antibiotic contamination in China's three major rivers(the Yangtze River,Yellow River,and Pearl River) and illustrates the occurrence and fate of antibiotics in conventional municipal wastewater treatment plants(WWTPs).The analytical data indicate that traditional WWTPs cannot completely remove these concerned pharmaceuticals,as seen in the large difference between the distribution coefficient(K_d) and the uneven removal efficiency of various types of antibiotics.Although constructed wetlands(CWs)offer a potential way to remove these antibiotics from water supplies,knowledge of their mechanisms is limited.There are four main factors affecting the performance of CWs used for the treatment of antibiotics in water supplies,the types and configurations of CWs,hydraulic load rates,substrates,and plants and microorganisms.Further researches focusing on these factors are needed to improve the removal efficiency of antibiotics in CWs. | GUAN Yidong WANG Bo GAO Yongxia LIU Wen ZHAO Xiaoli HUANG Xiaofeng YU Jianghuai | 2017 | Pedosphere2017,27,1: | 22 |
| 8 | Antioxidant properties of magnesium lithospermate B contribute to the cardioprotection against myocardial ischemia/reperfusion injury in vivo and in vitro显示文摘OBJECTIVE: To determine the cardioprotective effect of magnesium lithospermate B (MLB) on myocardial ischemia/reperfusion (MI/R) injury and to investigate the antioxidant potential in vivo and in vitro. METHODS: MI/R injury was induced by the occlusion of left anterior descending coronary artery for 30 min followed by reperfusion for 3 h in rats. After reperfusion, hearts were harvested to assess infarct size, histopathological damages, the levels of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), reduced glutathione (GSH) and malondialdehyde (MDA). Blood samples were col- lected to determine serum levels of creatine kinase-MB (CK-MB), cardiac troponin (cTnI) and lactate dehydrogenase (LDH). Furthermore, simulatedischemia/reperfusion (SI/R) injury in vitro was established by oxygen and glucose deprivation (OGD) for 2 h followed by 24-hour recovery period in cardiomyocytes. The activity of LDH in the cultured supernatant and the levels of intracellular reactive oxygen species (ROS), SOD and MDA in cardiomyocytes were also measured. Finally, cardiomyocytes apoptosis was determined with flow cytometry. RESULTS: MLB significantly limited infarct size, ameliorated histopathological damages and prevented leakage of CK-MB, cTnI and LDH. Additionally, SOD, CAT, GPx and GSH activities were notably increased by MLB, along with the MDA content decreased as compared with the model group in rats. In vitro study, MLB also decreased LDH activity in the cultured supernatant, increased SOD activity in cardiomyocytes, reduced intracellular ROS and MDA levels, and significantly suppressed cardiomyocytes apoptosis. CONCLUSION: MLB possessed remarkably cardioprotective effects on MI/R injury in vivo and in vitro. | 10 | Seasonal patterns and environmental control of ecosystem respiration in subtropical and temperate forests in China显示文摘Continuous measurement of carbon dioxide exchange using the eddy covariance (EC) technique was made at two ChinaFLUX forest sites including the young subtropical Pinus plantation (Qianyanzhou) and old temperate broad-leaved Korean pine mixed forest (Changbai Mountains) as part of the ChinaFLUX network. Seasonal patterns and environmental control of ecosystem respiration in the subtropical and temperate forests were evaluated by the often-used multiplicative model and Q10 model as a function of temperature and soil water content. The resuits suggested that ( i ) temperature was found to be a dominant factor in the ecosystem respiration, and most of the temporal variability of ecosystem respiration was explained by temperature. However, in the drought-stressed ecosystem, soil water content controlled the temporal variability of ecosystem respiration other than temperature effects, and soil water content became a dominat factor when severe drought affected the ecosystem respiration; (ii) the regression models analysis revealed that in the drier soil, ecosystem respiration was more sensitive to soil moisture than was expressed by the often-used multiplicative model. | 12 | Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. | 14 | Assessing the numbers of SNPs needed to establish molecular IDs and characterize the genetic diversityof soybean cultivars derived from Tokachi nagaha显示文摘The development of a core set of SNP molecular markers that could be widely used in soybean genetic research would greatly facilitate research into the genetic diversity of soybean.We conducted an analysis of Tokachi nagaha and 137 of its descendant soybean cultivars using 4044 SNP markers with the goal of determining the appropriate number of single-nucleotide polymorphisms(SNPs)needed to construct unambiguous molecular IDs and characterize genetic diversity based on a genetic distance matrix correlation method.When the number of SNPs was held constant,the number of accession pairs that could be distinguished increased as the polymorphism informative content(PIC)value of the SNPs increased.A core panel of 20 selected SNPs from 11 linkage groups with a mean PIC value of 0.3703 and a range of 0.3640–0.3749 was able to identify almost all of the accession pairs in our study[9445 pairs(99.92%)].The eight accession pairs that could not be identified with this core SNP set all originated from the same province and some of them had the same parental cultivars.The molecular IDs of the 138 accessions were constructed using the core 20 SNPs.It is known that both the number of SNPs and PIC values should be considered when SNPs are selected for use in the analysis of genetic diversity.In this study,when the PIC value was 0.3460,the correlation coefficient between the genetic distance matrices associated with a panel of 200 SNPs and the total population was>0.800,indicating satisfactory correlation.Our high-accuracy,high-resolution core SNP panel for germplasm fingerprinting and our findings about assessing genetic diversity will likely markedly improve the management and utilization efficiency of soybean germplasm resources. | Zhangxiong Liu Jun Li Xuhong Fan Nang Myint Phyu Sin Htwe Shuming Wang Wen Huang Jiyu Yang Lili Xing Lijun Chen Yinghui Li Rongxia Guan Ruzhen Chang Dechun Wang Lijuan Qiu | 2017 | The Crop Journal2017,5,4: | 10 |

| 19 | Xenograft tumors derived from malignant pleural effusion of the patients with non-small-cell lung cancer as models to explore drug resistance显示文摘Background:Non-small cell lung cancer(NSCLC)patients with epidermal growth factor receptor(EGFR)mutations or anaplastic lymphoma kinase(ALK)fusions show dramatic responses to specific tyrosine kinase inhibitors(TKIs);however,after 10-12 months,secondary mutations arise that confer resistance.We generated a murine xenograft model using patient-derived NSCLC cells isolated from the pleural fluid of two patients with NSCLC to investigate the mechanisms of resistance against the ALK-and EGFR-targeted TKIs crizotinib and osimertinib,respectively.Methods:Genotypes of patient biopsies and xenograft tumors were determined by whole exome sequencing(WES),and patients and xenograft-bearing mice received targeted treatment(crizotinib or osimertinib)accordingly.Xenograft mice were also treated for prolonged periods to identify whether the development of drug resistance and/or treatment responses were associated with tumor size.Finally,the pathology of patients biopsies and xenograft tumors were compared histologically.Results:The histological characteristics and chemotherapy responses of xenograft tumors were similar to the actual patients.WES showed that the genotypes of the xenograft and patient tumors were similar(an echinoderm microtu-bule-associated protein-like 4-ALK(EML4-ALK)gene fusion(patient/xenograft:CTC15035EML4-ALK)and EGFR L858R and T790M mutations(patient/xenograft:CTC15063EGFR L858R,T790M)).After continuous crizotinib or osimertinib treatment,WES data suggested that acquired ALK E1210K mutation conferred crizotinib resistance in the CTC15035EML4-ALK xenograft,while decreased frequencies of EGFR L858R and T790M mutations plus the appearance of v-RAF murine sarcoma viral oncogene homolog B(BRAF)G7V mutations and phosphatidylinositol-4-phosphate 3-kinase catalytic subunit type 2 alpha(PIK3C2A)A86fs frame shift mutations led to osimertinib resistance in the CTC15063EGFR L858R,T790M xenografts.Conclusions:We successfully developed a new method of generating drug resistance xenograft models from liquid biopsies using microfluidic technology,which might be a useful tool to investigate the mechanisms of drug resist-ance in NSCLC. | Yunhua Xu Feifei Zhang Xiaoqing Pan Guan Wang Lei Zhu Jie Zhang Danyi Wen Shun Lu | 2018 | Cancer Communications2018,38,1: | 7 |

A three-dimensional simulation model was established for the analysis of preparing the semisolid Al-3wt%Mg alloy. Through simulation and experiment, it is shown that the sloping angle of the plate greatly affects temperature and velocity distributions, and the temperature and velocity of the alloy at the exit of the sloping plate increase with the increase of the sloping angle. The alloy temperature decreases linearly from the pouring mouth to the exit. The alloy temperature at the exit increases obviously with the increase of pouring temperature.
