12018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. Based on the globally recent development and the evidence-based data in China, 2018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents was jointly prepared by the Pediatric Cardiology Society, Chinese Pediatric Society, Chinese Medical Association(CMA)/Committee on Pediatric Syncope, Pediatricians Branch, Chinese Medical Doctor Association(CMDA)/Committee on Pediatric Cardiology, Chinese College of Cardiovascular Physicians, Chinese Medical Doctor Association(CMDA)/Pediatric Cardiology Society, Beijing Pediatric Society, Beijing Medical Association(BMA). 2The state-of-the-art of the China Seismo-Electromagnetic Satellite mission显示文摘The China Seismo-Electromagnetic Satellite(CSES) mission was proposed in 2003 and approved in 2013 after ten years' scientific and engineering demonstrations. To meet the requirement of scientific objectives, the satellite is designed to be in a sunsynchronous orbit with an altitude of 507 km and descending node time of 14:00 LT. The CSES satellite carries 8 instruments,including search-coil magnetometer(SCM), electric field detector(EFD), high precision magnetometer(HPM), GNSS occultation receiver(GOR), plasma analyzer package(PAP), langmuir probe(LAP), high energetic particle package(HEPP) and detector(HEPD), and tri-band beacon(TBB), among which HEPD is provided by Italian Space Agency. The CSES satellite was launched successfully on February 2, 2018, and is planned to operate for 5 years. 3Over-expression of Metastasis-associated in Colon Cancer-1 (MACC1) Associates with Better Prognosis of Gastric Cancer Patients显示文摘Objective: The aim of this study was to detect metastasis-associated in colon cancer-1 (MACC1) expression in Chinese gastric cancer and analyze the relationship between MACC1 expression and postoperative survival. Methods: The expression of MACC1 and c-MET protein in a sample of 128 gastric cancer tissues was detected by immunohistochemistry. A retrospective cohort study on the prognosis was carried out and data were collected from medical records. Results: The positive rate of MACC1 protein expression in gastric cancer was 47.66%, higher than that in adjacent noncancerous mucosa (P<0.001). MACC1 protein expression was not related to the clinicopathological variables involved. Kaplan-Meier analysis revealed that the survival of MACC1 positive group tended to be better than that of MACC1 negative group, particularly in patients with stage III carcinoma (P=0.032). Cox regression analysis revealed that MACC1 protein over-expression in gastric cancer tended to be a protective factor with hazard ratio of 0.621 (P=0.057). Immunohistochemical analysis showed that the positive rate of c-MET protein expression was much higher in cases with positive MACC1 expression in gastric cancer (P=0.002), but P53 expression was not associated with MACC1 expression. 4Effects of Ce on catalytic combustion of methane over Pd-Pt/Al_2O_3 catalyst显示文摘Activity and stability of 1%Pd-0.2%Pt/Al2O3 and 1%Pd-0.2%Pt/0.6%Ce/Al2O3 catalysts prepared by impregnation method for catalytic combustion of methane in air were investigated. The catalysts before and after reaction were characterized by BET, CO chemisorption, XRD and XPS techniques. 5Olfactory ecto-mesenchymal stem cell-derived exosomes ameliorate murine Sjögren’s syndrome by modulating the function of myeloid-derived suppressor cells显示文摘Sjögren’s syndrome(SS)is a systemic autoimmune disease characterized by progressive inflammation and tissue damage in salivary glands and lacrimal glands.Our previous studies showed that myeloid-derived suppressor cells(MDSCs)exhibited impaired immunosuppressive function during disease progression in patients with SS and mice with experimental Sjögren’s syndrome(ESS),but it remains unclear whether restoring the function of MDSCs can effectively ameliorate the development of ESS.In this study,we found that murine olfactory ecto-mesenchymal stem cell-derived exosomes(OE-MSC-Exos)significantly enhanced the suppressive function of MDSCs by upregulating arginase expression and increasing ROS and NO levels.Moreover,treatment with OE-MSC-Exos via intravenous injection markedly attenuated disease progression and restored MDSC function in ESS mice.Mechanistically,OE-MSC-Exo-secreted IL-6 activated the Jak2/Stat3 pathway in MDSCs.In addition,the abundant S100A4 in OE-MSC-Exos acted as a key factor in mediating the endogenous production of IL-6 by MDSCs via TLR4 signaling,indicating an autocrine pathway of MDSC functional modulation by IL-6.Taken together,our results demonstrated that OE-MSC-Exos possess therapeutic potential to attenuate ESS progression by enhancing the immunosuppressive function of MDSCs,possibly constituting a new strategy for the treatment of Sjögren’s syndrome and other autoimmune diseases.Ke Rui Yue Hong Qiugang Zhu Xiaofei Shi Fan Xiao Hailong Fu Qing Yin Yida Xing Xinfeng Wu Xiaodan Kong Huaxi Xu Jie Tian Shengjun Wang Liwei Lu 2021Cellular & Molecular Immunology2021,18,2:12

9QKI-5 regulates the alternative splicing of cytoskeletal gene ADD3 in lung cancer显示文摘Accumulating evidence indicates that the alternative splicing program undergoes extensive changes during cancer develop-ment and progression.The RNA-binding protein QKI-5 is frequently downregulated and exhibits anti-tumor activity in lung cancer.Howeve-r,little is known about the functional targets and regulatory mechanism of QKI-5.Here,we report that upregulation of exon 14 inclusion of cytoskeletal gene Adducin 3(ADD3)significantly correlates with a poor prognosis in lung cancer.QKI-5 inhibits cell proliferation and migration in part through suppressing the splicing of ADD3 exon 14.Through genome-wide mapping of QKI-5 binding sites in vivo at nucleotide resolution by iCLIP-seq analysis,we found that QKI-5 regulates alternative splicing of its target mRNAs in a binding position-dependent manner.By binding to multiple sites in an upstream intron region,QKI-5 represses the splicing of ADD3 exon 14.We also identified several QKI mutations in tumors,which cause dysregulation of the splicing of QKI targets ADD3 and NUMB.Taken together,our results reveal that QKI-mediated alternative splicing of ADD3 is a key lung cancer-associated splicing event,which underlies in part the tumor suppressor function of QKI.Jin-Zhu Wang Xing Fu Zhaoyuan Fang Hui Liu Feng-Yang Zong Hong Zhu Yan-Fei Yu Xiao-Ying Zhang Shen-Fei Wang Ying Huang jingyi Hui 2021Journal of Molecular Cell Biology2021,13,5:4
10First results on ^(76)Ge neutrinoless double beta decay from CDEX-1 experiment显示文摘We report the first results on ^(76)Ge neutrinoless double beta decay from stage one of the China dark-matter experiment(CDEX).A p-type point-contact high-purity germanium detector with a mass of 994 g has been installed to detect neutrinoless double beta decay events, as well as to directly detect dark matter particles. An exposure of 304 kg d has been analyzed over a wide spectral band from 500 keV to 3 MeV. The average event rate obtained was about 0.012 counts per keV per kg per day over the 2.039 MeV energy range. The half-life of ^(76)Ge neutrinoless double beta decay derived based on this result is T^(0ν)1/2>6.4×10^(22) yr(90% C.L.). 11Effects of IκBα and its mutants on NF-κB and p53 signaling pathways显示文摘AIM: To study the effects of IκBα and its mutants (IκBαM, IκBα243N, IκBαM244C) on NF-κB, p53 and their downstream target genes. The relationship of NF- κB, p53, and IκBα was further discussed.METHODS: pECFP-IκBα, pECFP-IκBαM (amino acides 1-317, Ser32, 36A), pECFP-IκBα243N (amino acides 1-243), pECFP-IκBα244C (amino acides 244-317), pEYFP-p65 and pp53-DsRed were constructed and transfected to ASTC-α-1 cells. Cells were transfected with pECFP-C1 as a control. 30 h after the transfection, location patterns of NF-κB, p53 and IκBα (IκBαM, IκBα243N, IκBα244C) were observed by a laser scanning microscope (LSM510/ConfoCor2, Zeiss). RNA extraction and reverse transcription were performed in cells transfected or co-transfected with different plasmids. Effects of IκBα and its mutants on the transprition level of NF-κB, NF-κB downstream target gene TNF-α, p53 and p53 downstream target gene Bax were observed by real time QT-PCR. In all experiments β-actin was reference. Results are expressed as the target/reference ratio of the sample divided by the target/reference ratio of the control. Different transfected cells were incubated with CCK-8 for 2 h in the incubator. Then the absorbance at 450 nm was measured by using a microplate reader. RESULTS: Cells that were transfected with p53- DsRed revealed a predominant nuclear localization. YFP-p65 mainly existed in the cytoplasm. Cells were transfected with CFP-IκBα, CFP-IκBαM, and CFP- IκBα243N respectively and revealed a predominant cytosolic localization. However, cells transfected of CFP- IκBα244C revealed a predominant nuclear localization. The mRNA levels of p65, TNF-α, p53 and Bax in CFP-IκBα transfected cells did not change significantly, while in YFP-p65/CFP-IκBα co-transfected cells, IκBα decreased the transcription of p65 downstream gene TNF-α (2.24 ± 0.503) compared with the YFP-p65/ CFP-C1 co-transfected cells (5.08 ± 0.891) (P < 0.05). Phosphorylation defective IκBα (IκBαM) decreased the transcription levels of all the four genes compared with the control (P < 0.05). The N terminus of IκBα (IκBα243N) increased the transcription of NF-κB (1.84 ± 0.176) and TNF-α (1.51 ± 0.203) a little bit. However, the C terminus of IκBα (IκBα244C) increased the transcription of NF-κB, TNF-α, p53 and Bax significantly (8.29 ± 1.662, 14.16 ± 2.121, 10.2 ± 0.621, 3.72 ± 0.346) (P < 0.05). The CCK-8 experiment also showed that IκBα244C and p53 synergistically mediate apoptosis. CONCLUSIONS: IκBα and its mutants (IκBαM, IκBα243N, IκBαM244C) have different effects on NF- κB and p53 signaling pathways, according to their different structures. IκBαM bounds with NF-κB and p53 in cytoplasm steadily, and inhibits both of the two signaling pathways. p53 and IκBα244C may be co-factor in inducing apoptosis. 12Effect of Chronic Noise Exposure on Expression of N-Methyl-D-Aspartic Acid Receptor 2B and Tau Phosphorylation in Hippocampus of Rats显示文摘Objective To study the effect of chronic noise exposure on expression of N-methyl-D-aspartic acid receptor 2B (NR2B) and tau phosphorylation in hippocampus of rats. Methods Twenty-four male SD rats were divided in control group and chronic noise exposure group. NR2B expression and tau phosphorylation in hippocampus of rats were detected after chronic noise exposure (100 dB SPL white noise, 4 h/d×30d) and their mechanisms underlying neuronal apoptosis in hippocampus of rats with TUNEL staining. Results The NR2B expression decreased significantly after chronic noise exposure which resulted in tau hyperphosphorylation and neural apoptosis in hippocampus of rats. Immunohistochemistry showed that the tau hyperphosphorylation was most prominent in dentate gyrus (DG) and CA1 region of rat hippocampus. This compound is also a potent toxin suppressing the bone marrow. Accordingly, benzene causes serious hematological adverse effects, such as pancytopenia and aplastic anemia. In the 1920s, the carcinogenicity of benzene was identified in humans, in which it was shown to cause acute myelocytic leukemia (AML)[2]. The reports highlighting the first cases of hematopoietic disease associated with benzene appeared in China in the late 1950s[3]. 15The first data release(DR1) of the LAMOST regular survey显示文摘The Large sky Area Multi-Object Fiber Spectroscopic Telescope(LAMOST) general survey is a spectroscopic survey that will eventually cover approximately half of the celestial sphere and collect 10 million spectra of stars, galaxies and QSOs. Objects in both the pilot survey and the first year regular survey are included in the LAMOST DR1. The pilot survey started in October 2011 and ended in June 2012, and the data have been released to the public as the LAMOST Pilot Data Release in August 2012. The regular survey started in September 2012, and completed its first year of operation in June 2013. The LAMOST DR1 includes a total of 1202 plates containing 2 955 336 spectra, of which 1 790 879 spectra have observed signalto-noise ratio(SNR) ≥ 10. All data with SNR ≥ 2 are formally released as LAMOST DR1 under the LAMOST data policy. This data release contains a total of 2 204 696 spectra, of which 1 944 329 are stellar spectra, 12 082 are galaxy spectra and 5017 are quasars. The DR1 not only includes spectra, but also three stellar catalogs with measured parameters: late A,FGK-type stars with high quality spectra(1 061 918 entries), A-type stars(100 073 entries), and M-type stars(121 522 entries). 18奥曲肽持续皮下泵入治疗恶性肠梗阻的临床观察(英文)显示文摘Objective: The aim of the study was to observe the effectiveness of continuously subcutaneous-pumped octreotide infusion in palliative treatment of malignant bowel obstruction (MBO).Methods: Clinical data were retrospectively analyzed in 26 carcinoma patients complicated with MBO,in the Affiliated Hospital of Liaoning University of Traditional Chinese Medicine,China,from March 2007 to April 2009.All 26 carcinoma patients with MBO were divided into two groups: the controlling group (CG,routine therapy,15 patients) and the octreotide group (OG,11 patients).The octreotide group received routine therapy combined with octreotide (0.3 mg/d) by 24 hours continuously subcutaneous octreotide infusion.The changes of curative effectiveness related to symptoms,the times of recovering exsufflation and defecation,the average drain of gastrointestinal drainage tube,the duration of gastrointestinal drainage tube and the rate of extubation,were observed and compared between the two groups.Results: After treatment,remarkable changing rates of MBO related symptoms were 81.8% (9/11) in OG,and 46.7% (7/15) in CG (P < 0.05).The 78% of SG and 30% of CG recovered the exsufflation and defecation,and the mean times they needed were 3.2 d and 5.8 d,respectively (P < 0.05).The durations of gastrointestinal drainage tube of OG and CG were (5 ± 1.2) d and (10 ± 2.3) d,respectively,and the rates of extubation were 54.5% and 20%,respectively.The improvement rate in the octreotide group was better than that in the controlling group and the difference was significant (72.7% and 26.7%,P < 0.05).Conclusion: The administration of octreotide in combination with routine treatment can be very effective in the treatment of MBO.It can relieve the symptoms of MBO effectively and improve the quality of life of the end-stage patients.It has provided one kind of new treating thought and method for treatment of malignant bowel obstruction.Dongfeng Yin Yin Zhu Yuqing Xing Hong Gao Yuzhen Pan Lin Pan Guangyi Tang Xiangrong Xing Lijiang Zhou 2011The Chinese-German Journal of Clinical Oncology2011,10,1:2
19HYDRODYNAMIC ANALYSIS AND SIMULATION OF A SWIMMING BIONIC ROBOT TUNA显示文摘A dynamic model for undulatory locomotion was proposed to study the swimming mechanism of a developed bionic robot tuna. On the basis of inviscid hydrodynamics and rigid-body dynamics, the momentum and propulsive force required for propelling the swimming robot tuna’s flexible body was calculated. By solving the established dynamic equations and efficiency formula, the swimming velocity and propulsive efficiency of the bionic robot tuna were obtained. The relationship between the kinematic parameters of the robot tuna’s body curve and the hydrodynamic performances was established and discussed after hydrodynamic simulations. 20Relationship of HLA-A,-Cw Polymorphisms with HIV/AIDS in Chinese Yi Ethnic Group of Sichuan Province显示文摘The relationship of HLA-A, -Cw alleles on HIV infection and AIDS disease progression in the Chinese Yi ethnic group of Sichuan province were investigated. The genetic polymorphisms of HLA-A, -Cw alleles of 102 unrelated healthy Chinese Yi ethnic individuals, 68 HIV-1 infected and 21 HIV positive long-time survivors were typed by PCR-SSP assay. Statistic signifiance was determined by the χ2 test with the SPSS software. No significant differences were observed between the HLA-A, -Cw alleles of the 68 HIV-1 infected and 102 non-infected Chinese Yi control individuals. Whereas the prevalence of A*3601,Cw*14(01-03)and Cw*0304 was significantly higher in 21 long time survivors compared with 102 healthy controls with P values of 0.016, 0.016 and 0.000 by χ2 or the Fisher exact test respectively.
