| 3 | Small molecule inhibitors reveal allosteric regulation of USP14 via steric blockade显示文摘The ubiquitin system is important for drug discovery,and the discovery of selective small-molecule inhibitors of deubiquitinating enzymes (DUBs) remains an active yet extremely challenging task. With a few exceptions,previously developed inhibitors have been found to bind the evolutionarily conserved catalytic centers of DUBs,resulting in poor selectivity. The small molecule IU1 was the first-ever specific inhibitor identified and exhibited surprisingly excellent selectivity for USP14 over other DUBs. However,the molecular mechanism for this selectivity was elusive. Herein,we report the high-resolution co-crystal structures of the catalytic domain of USP14 bound to IU1 and three IU1 derivatives. All the structures of these complexes indicate that IU1 and its analogs bind to a previously unknown steric binding site in USP14,thus blocking the access of the C-terminus of ubiquitin to the active site of USP14 and abrogating USP14 activity. Importantly,this steric site in USP14 is very unique,as suggested by structural alignments of USP14 with several known DUB X-ray structures. These results,in conjunction with biochemical characterization,Indicate a coherent steric blockade mechanism for U5P14 inhibition by compounds of the IU series. | 4 | Attribution of PM_(2.5) exposure in Beijing-Tianjin-Hebei region to emissions:implication to control strategies显示文摘The Beijing-Tianjin-Hebei(BTH) region is one of the most heavily polluted regions in China,with both high PM_(2.5) concentrations and a high population density.A quantitative source-receptor relationship can provide valuable insights that can inform effective emission control strategies.Both source apportionment(SA) and source sensitivity(SS) can provide such information from different perspectives.In this study,both methods are applied in northern China to identify the most significant emission categories and source regions for PM_(2.5) exposure in BTH in 2013.Despite their differences,both models show similar distribution patterns for population and simulated PM_(2.5) concentrations,resulting in overall high PM_(2.5) exposure values(approximately 110 μg/m~) and particularly high exposure values during the winter(approximately 200 μ/m~).Both methods show that local emissions play a dominant role(70%),with some contribution from surrounding provinces(e.g.,Shandong) via regional transport.The two methods also agree on the priority of local emission controls:both identify industrial,residential,and agricultural emissions as the top three categories that should be controlled locally.In addition,the effect of controlling agricultural ammonia emissions is approximately doubled when the co-benefits of reducing nitrate are considered.The synthesis of SA and SS for addressing specific categories of emissions provides a quantitative basis for the development of emission control strategies and policies for controlling PM_(2.5)in China. | Xin Li Qiang Zhang Yang Zhang Lin Zhang Yuxuan Wang Qianqian Zhang Meng Li Yixuan Zheng Guannan Geng Timothy J.Wallington Weijian Han Wei Shen Kebin He | 2017 | Science Bulletin2017,62,13: | 15 |
| 5 | Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. | 6 | The Low Energy X-ray telescope (LE) onboard the Insight-HXMT astronomy satellite显示文摘The Low Energy X-ray telescope(LE) is one of the three main instruments of the Insight-Hard X-ray Modulation Telescope(Insight-HXMT). It is equipped with Swept Charge Device(SCD) sensor arrays with a total geometrical area of 384 cm^2 and an energy band from 0.7 to 13 ke V. In order to evaluate the particle induced X-ray background and the cosmic X-ray background simultaneously, LE adopts collimators to define four types of Field Of Views(FOVs), i.e., 1.6°×6°, 4°×6°, 50°-60°×2°-6 oand the blocked ones which block the X-ray by an aluminum cover. LE is constituted of three detector boxes(LEDs) and an electric control box(LEB) and achieves a good energy resolution of 140 e V@5.9 ke V, an excellent time resolution of 0.98 ms, as well as an extremely low pileup(<1%@18000 cts/s). | 9 | Wide-steering-angle high-resolution optical phased array显示文摘Optical phased array(OPA)technology is considered a promising solution for solid-state beam steering to supersede the traditional mechanical beam steering.As a key component of the LIDAR system for long-range detection,OPAs featuring a wide steering angle and high resolution without beam aliasing are highly desired.However,a wide steering range requires a waveguide pitch less than half of the wavelength,which is easily subjected to cross talk.Besides,high resolution requires a large aperture,and it is normally achieved by a high count number of waveguides,which complicates the control system.To solve the mentioned issues,we design two high-performance 128-channel OPAs fabricated on a multilayered SiN-on-SOI platform.Attributed to the nonuniform antenna pitch,only 128 waveguides are used to achieve a 4 mm wide aperture.Besides,by virtue of innovative dual-level silicon nitride(Si_(3)N_(4))waveguide grating antennas,the fishbone antenna OPA achieves a 100°×19.4°field of view(FOV)with divergence of 0.021°×0.029°,and the chain antenna OPA realizes a 140°×19.23°FOV with divergence of 0.021°×0.1°.To our best knowledge,140°is the widest lateral steering range in two-dimensional OPA,and 0.029°is the smallest longitudinal divergence.Finally,we embed the OPA into a frequency-modulated continuous-wave system to achieve 100 m distance measurement.The reflected signal from 100 m distance is well detected with 26 dBm input transmitter power,which proves that OPA serves as a promising candidate for transceiving optical signal in a LIDAR system. | YINGZHI LI BAISONG CHEN QUANXIN NA QIJIE XIE MIN TAO LANXUAN ZHANG ZIHAO ZHI YUXUAN LI XIAOBIN LIU XIANSHU LUO GUOQIANG LO FENGLI GAO XUEYAN LI JUNFENG SONG | 2021 | Photonics Research2021,9,12: | 7 |
| 10 | Predictive value of immune cell counts and neutrophil-to-lymphocyte ratio for 28-day mortality in patients with sepsis caused by intra-abdominal infection显示文摘Background:The current study aimed to evaluate the value of immune cell counts and neutrophilto-lymphocyte ratio(NLR)when attempting to predict 28-day mortality.Methods:We conducted an observational retrospective study that included consecutive septic patients.Severity scores on the first day and peripheral circulating immune cell counts(at day 1,day 3,day 5 and day 7 of admission)were collected during each patient’s emergency intensive care unit stay.We assessed the associations of peripheral circulating immune cell counts and NLR with the severity of illness.The relationships between 28-day mortality and peripheral circulating immune cell counts and NLR with were evaluated using Cox proportional cause-specific hazards models.Results:A total of 216 patients diagnosed with sepsis caused by IAI were enrolled.The lymphocyte counts(days 1,3,5 and 7)and monocyte counts(days 3,5 and 7)were significantly lower in nonsurvivors(n=72)than survivors(n=144).The NLR values at each time point were significantly higher in non-survivors.The day 1 lymphocyte counts,as well as the monocyte counts,were significantly lower in the highest-scoring group,when stratified by the Acute Physiology and Chronic Health Evaluation II and Sequential Organ Failure Assessment scores,than in the other groups(p<0.05).The day 1 NLR was significantly higher in the highest-scoring group than in the other groups(p<0.05).The day 5 and day 7 lymphocyte counts,day 3 and day 7 monocyte counts and day 7 NLR were significant predictors of 28-day mortality in the Cox proportional hazards models(day 5 lymphocyte count:hazard ratio,0.123(95%CI,0.055–0.279),p<0.001;day 7 lymphocyte count:hazard ratio,0.115(95%CI,0.052–0.254),p<0.001;day 3 monocyte count:hazard ratio,0.067(95%CI,0.005–0.861),p=0.038;day 7 monocyte count:hazard ratio,0.015(95%CI,0.001–0.158),p<0.001;day 7 NLR:hazard ratio,0.773(95%CI,0.659–0.905),p=0.001).Conclusions The results showed that circulating lymphocytes and monocytes were dramatically decreased within 7 days in non-survivors following sepsis from an IAI.Lymphocyte counts,monocyte counts and NLR appeared to be associated with the severity of illness,and they may serve as independent predictors of 28-day mortality in septic patients with IAIs. | Shuangqing Liu Yuxuan Li Fei She Xiaodong Zhao Yongming Yao | 2021 | Burns & Trauma2021,9,1: | 6 |
| 11 | Design and baseline data of a populationbased metabonomics study of eye diseases in eastern China: the Yueqing Ocular Diseases Investigation显示文摘Background:China is undergoing a massive transition toward an urban and industrial economy.These changes will restructure the demographics and economy which will eventually influence the future patterns of disease.The risk factors of vision-impairing eye diseases remain ambiguous and poorly understood.Metabolomics is an ideal tool to understand and shed light on the ocular disease mechanisms for earlier treatment.This article aims to describe the design,methodology and baseline data of the Yueqing Ocular Diseases Investigation(YODI),a developed county population-based study to determine the prevalence and primary causes of visual impairment;also with metabonomics analysis we aimed to identify,predict and suggest some preventive biomarkers that cause blindness.Methods:A population-based,cross-sectional study.Randomized clustering sampling was used to identify adults aged 50 years and older in Xiangyang Town,Yueqing county-level City.The interviews covered demographic,behavioral,ocular risk factors and mental health state.The ocular examination included visual acuity,autorefraction,intraocular pressure,anterior and posterior segment examinations,fundus photography,retinal tomography and angiography,and visual field testing.Anthropometric measurements included height and weight,waist and hip circumference,blood pressure,pulse rate,electrocardiogram,and abdominal ultrasound scan.A venous blood sample was collected for laboratory tests and metabonomics studies.Results:Of the 5319 individuals recruited for the YODI,4769(89.7%)subjects were enrolled for analyses.The median age was 62.0 years,and 45.6%were male.The educational level of illiteracy or semi-illiteracy,primary,middle and high school or above was 29.8%,45.5%,20.1%,and 3.3%,respectively.Majority of the participants were female,younger,and less educated when compared with nonparticipants.The average body mass index and waist-hip ratios were 24.4±3.4 kg/m^(2) and 0.9±0.1 respectively.Blood sample collection reached a sample size of 1909(479 from subjects with selfreported diabetes and 1430 from one-third of the 4290 subjects without self-reported diabetes).Conclusions:The YODI provides population-based data with a high response rate(89.7%)on the prevalence and primary causes of major vision-impairing eye diseases in developed county areas in eastern China.Metabonomics analysis from YODI will provide further association of metabolic characteristics with the visual impairment eye diseases.The risk prediction model could be created and has the potential to be generalized to developed eastern areas in China for prevention. | Yuxuan Deng Yuanbo Liang Sigeng Lin Liang Wen Jin Li Yue Zhou Meixiao Shen Jingwei Zheng Kemi Feng Yanting Sun Kwapong Willaim Robert Jia Qu Fan Lu | 2020 | Eye and Vision2020,7,1: | 4 |

| 14 | History,progress and future challenges of artificial blood vessels:a narrative review显示文摘Cardiovascular disease serves as the leading cause of death worldwide,with stenosis,occlusion,or severe dysfunction of blood vessels being its pathophysiological mechanism.Vascular replacement is the preferred surgical option for treating obstructed vascular structures.Due to the limited availability of healthy autologous vessels as well as the incidence of postoperative complications,there is an increasing demand for artificial blood vessels.From synthetic to natural,or a mixture of these components,numerous materials have been used to prepare artificial vascular grafts.Although synthetic grafts are more appropriate for use in medium to large-diameter vessels,they fail when replacing small-diameter vessels.Tissue-engineered vascular grafts are very likely to be an ideal alternative to autologous grafts in small-diameter vessels and are worthy of further investigation.However,a multitude of problems remain that must be resolved before they can be used in biomedical applications.Accordingly,this review attempts to describe these problems and provide a discussion of the generation of artificial blood vessels.In addition,we deliberate on current state-of-the-art technologies for creating artificial blood vessels,including advances in materials,fabrication techniques,various methods of surface modification,as well as preclinical and clinical applications.Furthermore,the evaluation of grafts both in vivo and in vitro,mechanical properties,challenges,and directions for further research are also discussed. | Ke Hu Yuxuan Li Zunxiang Ke Hongjun Yang Chanjun Lu Yiqing Li Yi Guo Weici Wang | 2022 | Biomaterials Translational2022,3,1: | 3 |
| 15 | Infection and Colonization of Pathogenic Fungus Fusarium proliferatum in Rice Spikelet Rot Disease显示文摘Rice spikelet rot disease(RSRD), caused by Fusarium proliferatum, is an emerging disease. So far, the effects of diseased rice floral organs as well as the primary infection sites and stages of this pathogen are not determined. We investigated changes in the floral organs, along with the infection processes of the pathogen in plants inoculated with F. proliferatum and labelled with a green fluorescent protein during different growth stages of rice. The results showed that RSRD is not a systemic infectious disease, which has negative effects on the fertility of the infected rice. F. proliferatum caused brown colored anthers, crinkled pistils and ovaries, pollen grain deformities and anther indehiscence. The number of pollen grains on the stigmas decreased significantly in the infected spikelets, and the anther dehiscence and seed-setting rate successively declined by 69% and 73%, respectively, as a result of the infection. The initial infection stage occurred at the pollen cell maturity stage, and the primary invasion sites were determined to be the anthers of rice. | 16 | A Monte Carlo Neural Fictitious Self-Play approach to approximate Nash Equilibrium in imperfect-information dynamic games显示文摘Solving the optimization problem to approach a Nash Equilibrium point plays an important role in imperfect information games,e.g.,StarCraft and poker.Neural Fictitious Self-Play(NFSP)is an effective algorithm that learns approximate Nash Equilibrium of imperfect-information games from purely self-play without prior domain knowledge.However,it needs to train a neural network in an off-policy manner to approximate the action values.For games with large search spaces,the training may suffer from unnecessary exploration and sometimes fails to converge.In this paper,we propose a new Neural Fictitious Self-Play algorithm that combines Monte Carlo tree search with NFSP,called MC-NFSP,to improve the performance in real-time zero-sum imperfect-information games.With experiments and empirical analysis,we demonstrate that the proposed MC-NFSP algorithm can approximate Nash Equilibrium in games with large-scale search depth while the NFSP can not.Furthermore,we develop an Asynchronous Neural Fictitious Self-Play framework(ANFSP).It uses asynchronous and parallel architecture to collect game experience and improve both the training efficiency and policy quality.The experiments with th e games with hidden state information(Texas Hold^m),and the FPS(firstperson shooter)games demonstrate effectiveness of our algorithms. | Li ZHANG Yuxuan CHEN Wei WANG Ziliang HAN Shijian Li Zhijie PAN Gang PAN | 2021 | Frontiers of Computer Science2021,15,5: | 3 |

| 19 | Total flavonoids from Ganshanbian (Herba Hyperici Attenuati) effect the expression of CaL-α1C and K_(ATP)-Kir6.1 mRNA of the myocardial cell membrane in myocardial ischemia-reperfusion arrhythmia rats显示文摘OBJECTIVE: To observe the impact of total flavonoids from Ganshanbian(Herba Hyperici Attenuati)on the expression of vascular smooth muscle membrane L-type calcium channel alpha1 C subunit(CaL-α1C) and ATP-sensitive K+channel(KATP)-Kir6.1mRNA, and explore the mechanisms of the antiarrhythmic effect of Ganshanbian(Herba Hyperici Attenuati)total flavonoids.METHODS: The treatment group was fed total flavonoids from Ganshanbian(Herba Hyperici Attenuati)for 7 days by gavage with 100 mg·kg﹣1·d﹣1.The blank control group and model control group were given the same amount of normal saline for 7 d.Arrhythmias were induced by performing a myocardial ischemia-reperfusion and electrocardiogram was observed. Reverse transcription-polymerase chain reaction was used to detect the expression of CaL-α1CandKATP-Kir6.1 mRNA in the myocardial cell membrane of all groups of rats.
